1.TRANSNEURONAL TRANSPORT OF HERPES SIMPLEX VIRUS TYPE 1 IN NEURONAL CONNECTIONS OF THE CEREBELLUM IN THE RAT

he herpes simplex virus type1 (HSV 1) was used for study on tracing neuronal connections of the cerebellum in the rat. The anterior or posterior lobe of the cerebellum in total 23 rats was injected by HSV 1 (HK 2 strain, from Institute of Virology, Chinese Academy of Preventive Medicine) with the titer of 10 -7 in a volume of 10 15?l for each case. Following a postoperative survival period of 1 5 or 10 days, the animals were perfused and their brains and spinal cords were sectioned and stained immunohistochemically by polycolonal antibodies raised in rabbit against HSV 1. Under the LM, the HSV 1 labelled neurons were shown to be a Golgi like appearance, with clearly labelled cell body and dendrites. It showed that the distributions of the labelled neurons in the CNS depended on the postoperative periods and injection sites. 1. After posterior lobe injection, the labelling was limited in the cerebellum, especially in Purkinje cells and the deep nuclei in 1 day of survival time. Two days later, the labelling could be seen in the vestibular, pontive nuclei and inferior olive nucleus. Anterogradely transneuronal labelling in the ventrolateral thalamic nuclus became apparant 3 days after injetions; 2. After anterior lobe injection, the red nucleus, cuneate nucleus, cuniform nucleus and interstitial nucleus of cajal were labelled after 3 days, in addition to the labelling as shown in those cases with injections in the posterior lobe. The results proved that HSV 1 (HK 2 strain) could be transported both retrogradely and anterogradely in CNS, while the transneuronal transport would mainly occur anterogradely. The distances of HSV 1 transport in neuronal pathways would depend upon the postoperative survival times. This indicates that HSV 1 (HK 2 strain) is a powerful tool for demonstrating the chains of synaptically connected neurons in CNS.
2.The flow cytometry of trabecular meshwork cells apoptosis in Monglia with primary angle closure glaucoma

Background Research showed that the morbidity rate of primary angle closure glaucoma (PACG) in Mongolian population is 3.02 times more than Han nationality population.To understand the cause and mechanism of PACG in Mongolia is of an important significance.Objective This study was to investigate the pathogenesis of Mongolian PACG.Methods Thirty-two eyes of 32 PACG patients in Mongolia and 40 eyes of 40 PACG patients of Han peoples were included in Inner Mongolia Autonomous Region People's Hospital according to the diagnosis criteria of glaucoma group of Chinese Medical Ophthalmology Association (version 1987),and 13 eyes of 13 normal Mongolia and 17 eyes of 17 normal Han peoples who suffered with ocular truma were recruited as controls.Intraocular pressure(IOP) was measured before surgery.The trabecular meshwork tissue was obtained from all the eyes during the operation.Annexinv-FITC/PI double staining was performed and the apoptosis rate of trabecula cells was tested with flow cytometry.Written informed consent was obtained initial of the study.Results The IOP value in Mongolia PACG group,Han PACG group,Mengolia normal group and Han normal group was (35.97±7.11)mmHg,(38.70± 6.82) mmHg,(14.69 ± 2.91) mmHg and (13.59 ± 2.91) mmHg,respectively,showing a significant difference among the 4 groups(F=106.144,P=0.000),and the IOP was significantly higher in the Mengolia PACG group and Han PACG group than the normal groups(P<0.05).The apoptosis rate of the cells was (7.14±0.67)%,(5.40±0.69) %,(5.86±0.91) % and(2.29±0.65) % in the Mongolia PACG group,Han PACG group,Mongolia normal group and Han normal group,respectively,with a significant difference among them (F =174.888,P =0.000),and apoptosis rate of the Mongolia PACG group was significantly higher than that of the Han PACG group and the Mongolia normal group (P<0.05).No significant difference was found between the Mongolia PACG group and the Han PACG group or between the Mongolia normal group and Han normal group (P>0.05).The cell apoptosis rate was increased with the elevation of IOP (b =0.990,F=10.209,P =0.009) with the regression equition Y =2.788 +0.092X.Conclusions The apoptosis rate of trabecula cells in Mongolian is higher than Han people.If these results are associated with the high incidence of Mengolia PACG is worth of study.
3.A comparative study on the quality of Radix Angelicae dahuricae Dispensing Granules produced by pharmaceutical factories

AIM:To compare the quality of Radix Angelicae dahuricae Dispensing Granules produced by different pharmaceutical factories.METHODS:The main coumarin compounds were identified by TLC and HPLC respectively;imperatorin was determined by HPLC.RESULTS:Imperatorin and other main coumarin compounds of Radix Angelicae dahuricae Dispensing Granules in the market could not be determined because of low content.CONCLUSION:Radix Angelicae dahuricae Dispensing Granules produced by different pharmaceutical factories in the market have distinct differences in quality,and were of poor quality in the mass.
4.Application of extracorporeal extended field radiotherapy in locally advanced cervical cancer

Objective To investigate the application value of extracorporeal extended field radiotherapy in locally advanced cervical cancer. Methods A total of one hundred and twenty patients with stage IIB?IVA cervical cancer in the First Affiliated Hospital of Hebei North University from June 2012 to June 2014 were randomly divided into two groups:the control group and the observation group,each with 60 cases. The routine concurrent chemoradiotherapy was adopted in the control group. The observation group was treated with extracorporeal extended field radiotherapy combined with concurrent chemotherapy. The patients have been followed up for three years. The total efficacy rate,toxicity reaction,local progression free survival time ( PFS) and survival rate of the two groups were compared. Results The total efficacy rate in the observation group was significantly higher than that of the control group ( 88. 3%( 53/60 ) vs. 73. 3%( 44/60 ) , χ2 = 4. 357, P=0. 037),while the incidences of toxicity reaction in the two groups were 18. 3%(11/60) and 16. 7%(10/60), the difference was not statistically significant (χ2=0. 058,P=0. 810) ,the percentages of I and II degree in the two groups were 3. 4%( 2/60 ) and 3. 4%( 2/60 ) , the difference between the two groups was not statistically significant (Z=0. 000,P=1. 000). The PFS value in the observation group was significantly longer than that of the control group ( 25. 6 months vs. 13. 8 months,χ2 = 25. 624, P= 0. 000 ) , and the survival rate in the observation group improved significantly ( 53. 3%( 32/60 ) vs. 33. 3%( 20/60 ) , the difference was statistically significant (χ2 = 4. 887, P= 0. 027 ) . Conclusion Extracorporeal extended field radiotherapy is safe and effective in the treatment of locally advanced cervical cancer.
5.Relationship between hypertriglyceridemic-waist phenotype and clustering of cardiovascular risk factors in adolescents

Objective The aim of the study was to explore the relationship between hypertriglyceridemic-waist(HTWC) phenotype and clustering of cardiovascular risk factors in adolescents. Methods A cross-sectional study was conducted in 3 136 Han adolescents aged 13-17 years in Qinhuangdao. The HTWC phenotype was defined as serum triglyceride concentrations ≥1. 47 mmol/ L and waist circumference ≥90 th percentile for age and sex. Results The prevalence of HTWC phenotype was 3. 3% . The prevalence of clustering of cardiovascular risk factors was 0. 4%in adolescents with normal waist circumference and serum triglyceride and 14. 7% in adolescents with HTWC phenotype. After adjustment for sex, age, body mass index, and family history of diabetes and hypertension, the prevalence of clustering of cardiovascular risk factors in adolescents with HTWC phenotype was 9. 286 fold(95% CI 3. 161-27. 282, P<0. 01) of that in adolescents with normal waist circumference and triglyceride. Conclusion There was a significant correlation between HTWC phenotype and clustering of cardiovascular risk factors, suggesting that this phenotype may be applied as a simple marker for identifying adolescents with clustering of cardiovascular risk factors.
6.Tibial bone transport over an intramedullary nail in combination with antibiotic-loaded calcium sulphate for treatment of segmental bone defect

Objective:To analyze the clinical outcomes of tibial bone transport over an intramedullary nail in combination with antibiotic-loaded calcium sulphate for treatment of segmental bone defect.Methods:A retrospective analysis was conducted by enrolling 11 surgically treated patients with tibial segmental bone defect after the debridement for tibial infection or osteomyelitis who were admitted in Lower Limb Surgery Ward of Traumatic Orthopedic Department, Xi′an Honghui Hospital Affiliated to Xi′an Jiaotong University from Jan. 2018 to Jan. 2020. The collected clinical materials and data included gender, age, injury mechanism, smoke or alcohol, comorbidities, intraoperative bleeding loss, bone defect length, resorption time of calcium sulphate, bone transport time, external fixation time, external fixation index, complications and Paley bone and functional criteria. SPSS 24.0 software was used to analyze the data.Results:Eleven patients were followed up for 8 to 31 months (average 23.2 months). All patients achieved bone healing and the infection was eradicated with no sign of recurrence. The mean length of defect was (8.1±1.6) cm, mean resorption time of calcium sulphate was (6.6±2.6) months, bone transport time was (11.4±2.8) weeks, external fixation time was (4.7±1.2) months, external fixation index was (0.58±0.07) month/cm and full weight bearing time was (6.1±1.4) months. The complication rate was 36.4% including deep vein thrombosis, delayed union of the docking site, pin tract infection and sterile draining of the wound. Paley bone evaluation results were excellent and good in 10 patients (90.9%) and functional results were excellent and good in 11 patients (100%).Conclusion:Tibial bone transport over an intramedullary nail in combination with antibiotic-loaded calcium sulphate is a safe, reliable and successful method for segmental bone defect and eradication of infection which reduces external fixation time and complication rate, allows patients perform weight bearing and return to daily life earlier.
7.Analysis on risk factor of chronic disease-related behaviors among adult residents in Liubei District, Liuzhou City

Objective To explore the risk factor of chronic disease-related behaviors such as smoking, drinking, exercising and eating habit among residents in Liubei District, Liuzhou City. Methods 2 487 adult residents in Liubei District were sampled by multistage sampling method and surveyed on their way of life and behavior risk factors. The content of questionnaire involved familial population information, past medical history, way of life and eating habit. EpiData 3.1 was applied to install database, SPSS 17.0 was used for data analysis, multivariate Logistic regression was used to analyze the risk factor of chronic disease-related behaviors. Results The rates of hypertension, diabetes, stroke, chronic obstructive pulmonary disease (COPD) were 13.63%,4.22%,3.62%,0.36% and 6.84%, respectively. The rate of smoking, drinking, doing physical exercise and eat breakfast regularly were 22.92%, 5.43%,52.87% and 93.07%. Multivariate Logistic regression analysis showed that older and smoking were risk factors of chronic disease, while physical exercise was protective factor of chronic disease. Conclusions Intervening measure taken in chronic disease demonstration zone were effective in the past 5 years, and health education on behavioral intervention should be carried on continuous to reduce incidence of chronic disease.
8.Impacts of passive smoking on learning and memory ability of mouse offsprings and intervention by antioxidants.

OBJECTIVETo determine the impact of passive smoking and the protective effect of antioxidants such as vitamin E and quercetin on learning and memory ability of mouse offsprings.
METHODSA passive smoking model of pregnant mice was established. Learning and memory ability was evaluated by the water maze test and long term potentiation (LTP). Nitric oxide (NO), content, nitric oxide synthase (NOS), acetylcholinesteras (Ache) activity in brain, vitamin E concentration, and reactive oxygen species (ROS) in serum were determined. The latency period (the time during which the mice swim from the starting position to the ending position) and errors (the number of mice entering the blind end) in control and antioxidant intervention groups were compared with those in the smoke exposure group after 6 days.
RESULTSThe latency period as well as errors in the air, control diet, tobacco smoke (TS), and vitamin E diet groups were decreased significantly as compared with the TS and control diet groups (P<0.05). LTP was restrained in the TS and control diet groups. LTP in all the antioxidant diet groups was significantly increased compared with the TS and control diet groups. In addition, NOS and acetylcholinesteras (Ache) activitiy was significantly higher in the TS and control diet groups than in the air and control diet group. NO content was not significantly different among the different groups, and significantly lower in the TS and vitamin E diet groups than in the TS group, control diet group, quercetin diet group, and mixture diet group (P<0.05). Vitamin E concentration and ROS activity in serum were correlated with the outcome of water maze and LTP.
CONCLUSIONPassive smoking reduces LTP formation by disturbing the hippocampus function of mice, by decreasing NOS and Ache activity and increasing NO content. Antioxidants (especially vitamin E) partially improve the learning and memory ability of offsprings whose mothers are exposed to tobacco smoke during pregnancy.

9.Serum magnesium levels and the prognosis of type 2 diabetes patients with community-acquired pneumonia

Objective:To explore the effects of type 2 diabetes mellitus (T2DM) on the levels of serum magnesium in patients with community-acquired pneumonia (CAP) and the effects of abnormal serum magnesium on the prognosis in T2DM patients with CAP.Methods:The study was a retrospective single-center study conducted in adult patients hospitalized with CAP at the First Hospital of Qinhuangdao between January 2015 and December 2018. These patients were divided into two groups according to the diagnosis of T2DM (control group and T2DM group). The reference range for serum magnesium is from 0.75 to 1.25 mmol/L. The primary endpoint was in-hospital mortality.Results:The prevalences of hypomagnesemia and hypermagnesemia were numerically higher in T2DM group than in control group (hypomagnesemia: 14.6% vs 12.0%, hypermagnesemia: 1.4% vs 0.7%), but there was no statistical difference ( P>0.05). In patients with T2DM, the in-hospital mortalities in patients with normal serum magnesium and hypomagnesemia were 5.9% and 12.7%, respectively; In multivariate logistic regression analysis, the in-hospital mortality of patients with hypomagnesemia was 5.629(95% CI 2.012-15.750, P=0.001)times as much as that of patients with normal serum magnesium. Conclusions:Abnormal serum magnesium was common in T2DM inpatients with CAP and hypomagnesemia was the most common. Hypomagnesemia was closely related to in-hospital mortality in T2DM patients with CAP. The assessment of serum magnesium levels on admission may be helpful in predicting the prognosis of T2DM patients with CAP.
10.Rosiglitazone promotes phosphorylation of AMPK protein and improves insulin resistance in db/db mice

Objective To observe the effect of rosiglitazone on the protein expression of AMPK and GLUT4 in peripheral tissue (liver, skeletal muscle and fat) of type 2 diabetic db/db mice and to prove that rosiglitazone can regulate the glucose metabolism in db/db mice partly through the AMPK pathway. Methods db/db mice were randomly divided into model group and rosiglitazone group according to their blood glucose.The db/m mice were normal control group.After 4 weeks of administration, fasting blood glucose was detected in each group.Western blot was used to detect the contents of AMPK, p-AMPK and GLUT4 in liver, skeletal muscle and adipose tissue. Results (1) Rosiglitazone significantly reduced the fasting blood glucose of db/db mice; (2)Rosiglitazone increased the level of AMPK phosphorylation in the liver, skeletal muscle and adipose tissue of db/db mice, and increased the content of GLUT4 protein in skeletal muscle and adipose tissue. Conclusion Rosiglitazone can increase the phosphorylation of AMPK and the expression of GLUT4 protein in the liver, muscle and fat tissue of db/db mice, and promote the uptake and utilization of glucose in peripheral tissue, suggesting that it can regulate glucose metabolism in db/db mice partly through the AMPK pathway.
