1. Role of p38 MAP kinase in epigallocatechin-3-gallate-induced apoptosis of human gastric cancer MGC803 cells

Objective: To determine the role of p38 mitogen-activated protein kinase (MAP) kinase signal transduction pathways in epigallocatechin-3-gallate (EGCG)-induced apoptosis in human gastric cancer MGC803 cells. Methods: The viability of MGC803 cells was measured by MTT assay. Apoptosis of MGC803 cells was observed by AO/EB fluorescence microscopy and detected by flow cytometry with PI staining. Expression of p38MAPK and phosphorylated p38 (pp38) MAPK were determined by Western blot analysis. Results: EGCG induced apoptosis of MGC803 cells and apparently increased the activity of pp38MAPK in MGC803 cells. However, after interference with pp38MAPK inhibitor, the inhibitory effect of EGCG on MGC803 cells was significantly weakened. The apoptotic rate of the cells and the activity of pp38MAPK also decreased dramatically. Conclusions: EGCG can induce apoptosis of MGC803 cells. The effects could be markedly suppressed by pp38MAPK inhibitor, SB203580. EGCG induces apoptosis of MGC803 cells partly by activating p38 MAPK.
2.Expression and significance of Gadd45beta protein in human hepatocellular carcinoma

Objective To investigate the expression of Gadd45beta protein in human hepatocellular carcinoma(HCC) and its clinicopathologic significance.Methods Immunohistochemical method was used to detect the expressions of Gadd45beta in both hepatocellular carcinoma tissues and adjacent non-neoplastic liver tissues form 50 cases.Results The positive rates of Gadd45beta expression in the hepatocellular carcinoma tissues and adjacent non-neoplastic liver tissues were 26.0%(13/50) and 82.0%(41/50) respectively.There were significant difference for the positive rate between the two groups(P0.05).Conclusion Low expression of Gadd45beta protein in hepatocellular carcinoma tissues was exhibited,which may be related to hepatocarcinogenesis as well as biological behaviors and prognosis of HCC.

Objective To find out the material base of the induction and aggravation of psoriasis by psycological factors such as stress and depression etc. Methods Radioimmunoassay (RIA) was used to determine the effect of sera from 49 psoriatic patients and 34 healthy controls on the suppression of the lymphocyte transformation of normal mouse lymphocytes. Results The lymphocyte transformation rate of mouse lymphocytes treated with the sera of psoriatic patients was 41.55%, and that of controls was 68.09%. 4.Influence of UV on Gadd45?/? in human hepatoma carcinoma cell HepG2

Objective To investigate the change of Gadd45?/? in HepG2 cells after UV radiation.Methods HepG2 cells were cultured in vitro followed by radiation with UV (254nm,10 J/m2)and harvested at 1,2,4 h respectively.The induction of Gadd45?/? mRNA by UV was analyzed using RT-PCR,and the induction of Gadd45?/? protein was determined by Western blot assay at the given interval simultaneously.Results The peak values of Gadd45?/? mRNA appeared at 4 h and 1h after induction respectively,while the both of the peak value of Gadd45?/? protein were found at 1h after UV radiation.Conclusion Gadd45?/? could be rapidly induced by UV radiation in HepG2 cells.
5.Initial therapy indications of acute primary immune thrombocytopenia in children

Objective To explore the initial therapy indications of acute immune thrombocytopenia (ITP) in children based on the classification treatment.Methods Three hundred and eighty newly diagnosed ITP cases were enrolled in this study from Jan.1st 2012 to Apr.30th 2013 in Children's Hospital Affiliated to Shanghai Jiaotong University.In total 380 patients,there were 214 male cases (56.31%) and 166 female cases (43.68%).The cases were divided into observation group and therapy group according to the initial platelet count which was ≤ 30 × 109/L or the bleeding over moderate volume or with active bleeding.Platelet values were observed in the observation group weekly,adrenal cortical hormone and immunoglobulin treatment were adopted in the therapy group,cases were followed up to Aug.30th 2013,9 months on the average.Results Three hundred and five cases showed overall response (80.26%) and 75 cases showed no response(19.74%).One hundred and seventy-eight cases were divided into observation group (46.84%),in which 133 cases (74.72%) showed complete response or response.Two hundred and two cases were included in therapy group (53.16%),in which 167 cases (82.67%) were with complete response or response.There was no statistical difference between the 2 groups in curative effect (Z =-0.54,P =0.59).Forty-five cases in observation group were no response and accepted therapy,35 of them (77.78%) had response.There were equal efficiency in the initial therapy group and the subsequent therapy group (x2 =3.60,P =0.06).There was no difference between the age of onset,sex and season in 2 groups.Cases aged from 1 month to 1 year seemed to have a high incidence because of vaccination,and in the cases aged from 3 to 14 years the onset was related to infection.The children over 3 years old had higher risk factors in self-healing and the curative effect.There was no severe bleeding or adverse effect or dead cases in this study.Conclusions It is feasible to take platelet count ≤30 × 109/L as the threshold for initial therapy indications.Almost half of the cases could avoid overtreatment and pretherapy observation will not reduce the initial cure effect; no severe internal bleeding was observed in all the cases.
6.Risk factors of sexual dysfunction in aged men in Beijing : a multicenter community-based cross-sectional survey

Objective To investigate the prevalence of sexual dysfunction in aged men and associated risk factors in Beijing. Methods A cross-sectional study was performed in communities of Beijing involved 1656 men aged over 50 years.The International Index of Erectile Function-5 (IIEF-5),Brief Male Sexual Function Inventory for Urology ( O'Leary 1995 ) and the International Prostate Symptom Score (IPSS)questionnaires was recorded.The body mass index (BMI),prostate size was measured.The survey was conducted to make sure if the patients had diabetes,high blood pressure,hyperlipidemia,cerebrovascular history,and smoking and drinking situation.Pearson's X2 test and unconditional logistic regression were used to investigate the factors associated with sexual dysfunction. Results 1644 subjects were enrolled.The incidence of ED,reduction of sexual desire and defective ejaculation was 90.45%,60.04% and 38.81% respectively,and significantly different according to age ( P < O.05 ). Age was positively correlated with ED (3 =0.12,P<0.05),reduction of sexual desire (β =0.10,P<0.05) and defective ejaculation (β =0.10,P < 0.05 ) ; ED was significantly associated with BMI (β =0.07,P < 0.05 ).Hypertension and prostate size were risk factors for reduction of sexual desire and defective ejaculation; There was correlation between drinking and defective ejaculation ( β =- O.31,P < 0.05 ). Conelusions Compared with high prevalence of ED and lower sexual desire,the incidence of defective ejaculation were lower; this may reflect the sexual activities of aged male were more active compared with the less success of really erection.The prevalence of ED,reduction of sexual desire or defective ejaculation increased with age.BMI was the risk factor for ED.Enlarged prostate and hypertension was associated with reduction of sexual desire,and drinking was the risk factor for defective ejaculation.
7.Clinicopathologic study based on 2 056 cases of pulmonary resection of lung adenocarcinoma

Objective The international multidisciplinary lung adenocarcinoma classification was published by IASLC/ ATS/ERS in 2011.This study aimed to explore the clinicopathologic characteristics of lung adenocarcinoma based on IASLC/ATS/ERS classification and validate its clinical diagnostic and therapeutic value.Methods 2 056 cases of surgical resection from Shanghai Chest Hospital were classified according to the new classification and clinical information were retrospectively reviewed.The clinicopathologic characteristics based on new classification were analyzed statistically.Results Our data indicated that women were in high risk of lung adenocarcinoma; The average age of onset was 59-year-old; the female patients were younger than the male patients (58.7 years vs 60.2 years,P < 0.01) ; Average tumor diameter was 2.6 centimeter; right lung was more popular than left and superior lobe than the inferior one.Acinar predominant subtype and papillary predominant subtype were frequently observed.Micropapillary predominant subtype and solid predominant subtype were identified to be more aggressive than other histopathologic subtypes.Most patients were classified as stage Ⅰ(71.7%),which were predominantly stage Ⅰa (53.1%).Conclusion The new classification is superior to reflect the clinicopathologic characteristics of lung adenocarcinoma and satisfy clinical needs,especially contributing to change and update the surgical strategy of early stage lung adenocarcinoma.

Acute paraplegia is a rare presentation for a spinal cord ependymoma. Subarachnoid hemorrhage
(SAH) due to spinal ependymoma is also very rare. We report a 32-year-old woman who presented
with acute paraplegia and typical clinical signs of SAH with normal cerebral angiography, and further
diagnostic work-up revealed an spinal cord ependymoma as the source of the hemorrhage. There
is evidence that some spinal cord ependymomas have intratumoral hemorrhage, but most of these
bleedings occur without symptoms. We discuss the clinical and neuroradiological findings of this rare
case and review the literature related to this unusual presentation.431
9.Recent advances in the treatment of autoimmune encephalitis

Autoimmune encephalitis is a kind of inflammatory disease of central nervous system caused by abnormal immune response of body immune system to neuronal antigen,and is generally considered to be reversible encephalitis caused by noninfectious factors.Its characteristic manifestations include acute and subacute onset of cognitive dysfunction,epilepsy and mental disorder.With the discovery of related antibodies,summaries of clinical syndrome and application of new functional imaging instruments,the diagnosis of autoimmune encephalitis is increasingly standardized.The priority treatment of autoimmune encephalitis is immunomodulatory therapy,including glucocorticoid,immunoglobulin,plasma exchange and immunosuppressant.The other treatments could be the related tumor resection,electroshock therapy,etc.The symptoms in most patients can get substantial relief with active treatment.The present paper would focus on the research progress in treatment of autoimmune encephalitis.

To study the toxicokinetics of bakuchiol, hepatic and renal toxicity in rats after single oral administration of Psoraleae Fructus and combined with Glycyrrhizae Radix et Rhizoma, in order to provide scientific evidences for clinical safe medication use. A total of 35 SD rats were randomly divided into seven groups: vehicle (distilled water) control group, Glycyrrhizae Radix et Rhizoma group, positive control (aristolochic acid A) group, Psoraleae Fructus (40 g x kg(-1)) group( both male and female rats), Psoraleae Fructus and Glycyrrhizae Radix et Rhizoma (40 +20) g x kg(-1) group (both male and female rats). HPLC-UV method was used to determine the concentration of bakuchiol in rat plasma at different time points after single oral administration. Plasma alanine transaminase (ALT), aspartate aminotransferase (AST), blood urea nitrogen (BUN), plasma creatinine (Cr), N-acetyl-β-D-glucosaminidase (NAG) and kidney injury molecule 1 (Kim-1) were measured after administration for 24 h. The main toxicokinetics parameters of bakuchiol in rats exert significantly gender difference. When Psoraleae Fructus combination with Glycyrrhizae Radix et Rhizoma, the total area under the plasma concentration-time curve( AUC), C(max), and plasma clearance (CL) of bakuchiol were increased, respectively; CL, half-life (t½) were decreased, and T(max) were prolonged. The biochemical indicators (including ALT, AST, BUN, Cr and KIM-1 level) in different dose of Psoraleae Fructus groups, were found no statistically significant difference when compared with vehicle control group. The level of NAG in both Psoraleae Fructus and compatibility with Glycyrrhizae Radix et Rhizoma groups were significant increased (P < 0.05). There are obvious effects on toxicokinetics of bakuchiol in rats when Psoraleae Fructus combined with Glycyrrhizae Radix et Rhizoma. Renal toxicity induced by Psoraleae Fructus at high dose was observed after single oral administration and no liver damage in rats was found.
