1.Effect of different doses of cyclophosphamide on drug-resistance of implanted human lung cancer in nude mice

Objective To explore the effect of different doses of cyclophosphamide(CTX) on the drug-resistance of implanted human A549 lung cancer in nude mice,and investigate the anti-tumor effect and toxicity.Methods BALB/c nude mice bearing A549 lung carcinoma were randomized into low-dose metronomic(LDM) CTX group,maximum tolerate dose(MTD) CTX group and normal saline group.The changes of tumor volume,weight and toxicity were observed.Expression of glutathione S-transferase-?(GST-?) and lung resistance-related protein(LRP) in the tumor were detected by immunohistochemistry SP method.Results In LDM CTX group,the tumor grew fast at first,then slowed down gradually and grew slowly for a long time,while in MTD CTX group tumor grew slowly at first,then it accelerated.Side effects of chemotherapy:the weight of nude mice was lost persistently in MTD CTX group,while lost insignificantly in LDM CTX group.No obvious toxicity was observed in LDM CTX group.GST-? and LRP expression in LDM CTX group were not significantly different from those in the control,but significance was seen in MTD CTX group compared with those in the control(P
2.Experimental Research on Acute Toxicity and Long-term Toxicity of Mongolian Patent Drug Meng-Gen-Wu-Su-18 Pills

This study was aimed to observe the acute toxicity and long-term toxicity of Meng-Gen-Wu-Su-18 (MGWS-18) Pills, in order to provide references for safety application of this medicine in the clinical practice. MGWS-18 Pills suspension was intragastric administered to mice twice (0.2 mL/10 g) in 6 hours with maximal con-centration (0.4 g·mL-1). And the acute toxicity reaction was observed for 14 days. The dose of maximum, middle and minimum (3.67 g·kg-1, 1.84 g·kg-1, 0.92 g·kg-1) of MGWS-18 Pills were intragastric administered continuously to rats once a day for 180 days. The rats were observed 60 days after drug withdrawal. The results showed that the maximal tolerated dose (MTD) of MGWS-18 Pills was bigger than the dose of 16 g·kg-1 (which was equivalent to 436.36 times in clinical doses). There were significant differences on ALB, UREA, AST, TBIL, and CHOL between the control group and the maximum dose group of MGWS-18 Pills (P<0.05, or P<0.01) after 180 days of medica-tion. There were significant differences on ALB and UREA between the control group and the middle dose group (P< 0.05). There was no significant difference between the control group and the minimum dose group. Protein cast and degeneration necrosis at different levels of the epithelial cells of the proximal tubules were appeared in the maximum dose group after medication for 180 days. After 60 days of drug withdrawal, there were no significant dif-ferences on the general condition, body weight, hematological indexes, serum biochemical indexes, organ coefficient and etc. between the control group and each animal group. There was recovery tendency on the kidney damage of the maximum dose group. It was concluded that the basic safety intragastric administration dosage of MGWS-18 Pills in rats was 0.92 g·kg-1 (which was equivalent to 25 times in clinical doses).

4.Analysis of the reform trials in pilot public hospitals

Trials in public hospital reforms bear special significance for rational deployment of health resources and exploring the separation of hospital management from routine operations. The paper identified such setbacks in the present trials as easy technology measures, numerous models of separations, difficulty in setting pharmaceutical service fees, and inadequate governance. Authors also recommended such measures as searching for appropriate reforms for public hospitals locally, greater financial resources for completing the compensation mechanism of public hospitals, streamlining their management system for separation of management from operations, and improving governance. All these efforts are designed to enhance the public benefit nature of public hospitals.
5.Two Medical Cases of SHI Da-zhuo Using Integrated Traditional Chinese and Western Medicine for Left Ventricular Remodeling and Left Atrial Expansion after Acute Myocardial Infarction

This article summarized two medical cases of Professor SHI Da-zhuo treating acute myocardial infarction by Yiqi Huoxue Tuodu Shengji TCM therapy on the basis of conventional Western medicine therapy. Comparison of echocardiography before and after treatment showed that Yiqi Huoxue Tuodu Shengji TCM combined with conventional Western medicine therapy could inhibit the expansion of atrium and ventricle, improve the cardiac ejection fraction and improve the cardiac function after acute myocardial infarction, which could provide references for integrated traditional Chinese and Western medicine therapy for acute myocardial infarction.
6.Effect of Yiqi Huayu Recipe on Expression of Protein Kinase C and CyclinE in Rats with Gastric Precancerous Lesions

Objective To observe the effect of Yiqi Huayu Recipe on the expression of protein kinase C (PKC) and CyclinE in rats with gastric precancerous lesions (GPL). Methods The model of GPL was established mainly through N-methyl-N-nitro N-nitrosoguanidine (MNNG) together with other factors including alcohol stimulation and undue hunger and overeating. The survived rats after modeling were randomly divided into spontaneous recovery, and high-, moderate- and low-dose Yiqi Huayu group. After the corresponding treatment, the expression of PKC and CyclinE were detected by immunohistochemical method in each group. 7.Cigarette Smoke-Induced DNA Damage and Repair in Two Kinds of Human Lung Interstitial Cells

Objective To study the DNA damage and repair of normal lung interstitial cells and human lung adenocarcinoma cells exposed to cigarette smoke. Methods Cultured human embryo lung fibroblasts (HLF) and human lung adenocarcinoma A549 cells. Mainstream smoke was collected by using dimerhyl sulfoxide (DMSO) and phosphate buffer solution (PBS) as absorbents. MTT assay was used to test the cytotoxicity of the solutions of cigarette smoke, then selected the concentrations of the solutions with no obvious cytotoxicity to treat cells and detected DNA damage and repair by comet assay. Results As treated with original solutions or 1/2 dilution of DMSO cigarette smoke solutions only, the Viability of cells was below 80%, but it was beyond 80% when treated with PBS solutions. Polyethylene glycol (PEG) is extensively used to increasing the in vivo and in vitro stability of liposomes. However, PEGylated liposomes also produce some negative effects with further research, such as low cellular uptake, poor "endosomal escape" of pH sensitive liposome (PSL) and accelerated blood clearance (ABC) phenomenon, and this situation is referred as the "PEG dilemma". "PEG dilemma" posed severe challenges for the targeted delivery of PEGylated liposomes-loaded anticancer drugs, effective intracellular release of PEGylated PSL-encapsulated gene and protein drugs, and repeated administration of PEGylated liposomes. Therefore, it is urgent to solve the "PEG dilemma". This review focused on the definition, classification of "PEG dilemma", and discussed several possible approaches to overcome "PEG dilemma".
