Objective To study the effect of allicin on the growth and radiosensitivity of human pancreatic carcinoma BXPC3 cells. Methods BXPC3 cells were exposed to X-rays in the presence or absence of allicin.Cell proliferation was measured by MTT assay.Cell cycle distribution and apoptosis were detected by flow cytometry assay.Cell radiosensitivity and the influence of allicin on it was evaluated by colony formation assay. The expressions of Bax and Bcl-2 proteins were assayed by RT-PCR and Western blot. Results IC50 values of allicin on cell growth were 76.24,58.34 and 43.58 μmol/L under 12,24 and 48 h treatment, respectively.Treatment of cells with allicin obviously inhibited cell growth after irradiation and hence increased radiosensitivity (t =2.74,P < 0.05).This treament also enhanced radiation-induced cell cycle arrest at G2/M phase (t =11.41,P <0.05),apoptosis induction (t =12.36,P < 0.05),and Bax expression (t =4.83,P < 0.05),but it decreased Bcl-2 expression (t =3.69,P < 0.05).Conclusions Allicin could inhibit cell growth, induce cell cycle arrest and apoptosis via Bax/Bcl-2 pathway and hence increases radiosensitivity of BXPC3 cells.

Objective To explore the clinical effect of single use or combination of dydrogesterone and progestin in treatment of threatened abortion caused by uteal phase defect. Methods Totally 186 patients with threatened abortion caused by uteal phase defect accepted in The First Affiliated Hospital of Henan from April 2015 to April 2016 were selected and randomly divided into groups A,B,and C with 62 cases in each group.Patients in group A were given dydrogesterone, those in group B were given progestin, and those in group C were given dydrogesterone combined with progestin. Then the clinical effect, expression of hormones, treatment outcome, and adverse reaction were observed and compared. Results The total effective rates of groups A and B were 72.58% and 66.13%,respectively,which were obviously lower than 90.32% of group C with statistically significance (P <0.05).The expression levels of P,E2,and hCG of three groups after treatment were higher than those before, those in group C were the highest among them (P < 0.05).The successful treatment rates of groups A,B,and C were 83.87%,82.26%,and 95.16%,respectively,which had no great difference. Conclusion Combination use of dydrogesterone and progestin has better effective rate in treatment of threatened abortion caused by uteal phase defect compared to single use of these two drugs, which has good safety and worth of clinical application.

Objective To analyze the transcription level of new tumor suppressor gene SLC5A8 mRNA in colorectal cancer tissues. Methods Collected specimens of 23 cases with colorectal cancer and cut out carcinoma tissues and Pericarcinomatous tissue respectively, then used real time fluorescent quantitative PCR (RT-qPCR)to detect the transcription level of gene SLC5A8 mRNA,and the results of carcinoma tissues and pericarcinomatous tissues were analyzed by t test. Results The transcription level of new tumor suppressor gene SLC5A8 mRNA in colorectal cancer tissues is significantly lower than pericarcinomatous tissue (P =0.002).Conclusion The expression of gene SLC5A8 in colorectal cancer tissues is declining or missing, suggesting it has a certain relationship with the incidence of colorectal cancer.

A highly sensitive electrochemical deoxyribonucleic acid(DNA) biosensor based on multi-walled carbon nanotubes(MWNT)/Ag-TiO_2 composite film was developed. The solution containing Ag-TiO_2-MWNT composite was casted on the carbon paste electrode surface to form a robust film, which combine the advantages of the good biocompatibility of Ag-TiO_2 naocomposite and the fine conductivity, as well as the large active surface area of carbon nanotubes. The composite could greatly improve the immobilization capacity of the probe DNA. The morphologies and electrochemistry of the nanocomposite film were investigated by scanning electron microscopy and electrochemical techniques including electrochemical impedance spectroscopy and cyclic voltammetry, respectively. DNA hybridization events were monitored by a label-free method of electrochemical impedance spectroscopy. This label-free electrochemical impedance DNA biosensor showed high sensitivity and selectivity for phosphinothricin acetyltransferase gene sequence assay. The multicomponents films also displayed a high stability during repeated regeneration and hybridization process.

Objective To study the clinical efficacy of amiodarone combined with metoprolol in the treatment of arrhythmia.Methods100 cases of patients with coronary heart disease arrhythmia treated in our hospital from August 2015 to September 2016 were selected as the research object in this study. They were randomly divided into the control group and the experimental group, each group had a total of 50 cases.Patients in the control group were treated with conventional arrhythmia treatment, and the experimental group was treated with amiodarone combined with metoprolol on the basis of conventional treatment.Comparative analysis of the experimental group and the control group of patients with clinical therapeutic effect index.ResultsAfter the corresponding treatment, the experimental group of 50 patients, the therapeutic effect of 18 cases were invalid, 12 cases were effective, 20 cases were markedly effective, the total effective number of cases to 32 cases, the effective rate was 64.0%.In the control group of 50 patients, 29 patients were invalid, and the effective rate was effective in all of the 8 patients. The effective rate of the treatment was effective in all of the 13 patients, with a total effective rate of 42.0%.Therefore, the effective rate of the experimental group was significantly higher than that of the control group, with statistical difference (P<0.05).The diastolic blood pressure, systolic blood pressure and heart rate in the experimental group were significantly better than those in the control group (P<0.05).There was no significant difference between the experimental group and the control group in the incidence of adverse reactions, which was not statistically significant.ConclusionAmiodarone Combined with metoprolol for treatment of arrhythmia is better, more widely used, can improve the treatment efficiency to a great extent, the treatment of high safety, with further clinical promotion and application.

Objective:To express Tumstatin_(183-230)-TRAIL fusion protein and to observe its biological functions. Methods: SOE-ing PCR was employed to amplify the recombinant sequence of Tumstatin_(183-230)and TNF-related apoptosis-inducing ligand (TRAIL_(114-281)).An expression vector pMAL-Tu-T was constructed by inserting Tu-T sequence into pMAL-c_2;the vector was used to transfect E.coli BL21(DE3)and expression of MBP-Tu-T fusion protein was induced by IPTG.Amylose Resin columns were employed to purify the fusion protein. The biological functions of MBP-Tu-T protein was examined by inhibitory test of endothelial cell proliferation, standard tumor cell cytotoxic assay,in vitro tube formation inhibition, and electron microscopic observation(apoptosis). Results:The expression rate of MBP-Tu-T fusion protein in E.coli was about 20%. Purified recombinant protein obviously inhibited endothelial cell proliferation(IC_(50)12.5?g/ml),induced apoptosis of pancreatic cancer cells, and inhibited tube formation. Conclusion:Constructed MBP-Tu-T fusion protein is bifunctional, which lays a solid foundation for further investigation of antitumor effect of Tumstatin_(183-230)-TRAIL in vivo.
