Objective To explore the effects of endoplasmic reticulum (ER) stress in the hyperoxia-induced lung injury in premature rats. Methods Forty-eight premature Wistar rats were randomized into two groups 12 hours after birth:hyperoxia group (n=24) inhaled 95%oxygen and control group (n=24) inhaled air. Eight rats were sacriifced in each group on day 1, 3, 7 after the treatment and the left lungs were embedded. The pathological changes in the HE stained sections of lung tissues were observed. The expressions of ER related protein ERp57 and c/EBP homologous protein CHOP were detected by immuno histo-chemistry and the apoptosis of lung cells was detected by TUNEL analysis. Results The typical pathological characteristics of acute lung injury were observed in hyperoxia group. The expressions of ERp57 and CHOP were increased with the exposure time in hyperoxia group, and were signiifcantly higher than in control group (P<0.05). The apoptosis rate of lung cells in hyperoxia group was signiifcantly higher than in control group (P<0.01). There was signiifcant positive correlation between cell apoptosis index and expressions of Erp57 and c/EBP homogeneous protein. Conclusions ER stress initiated apoptosis participates and plays an important role in the process of hyperoxia-induced lung injury in premature rats.

Journal of Xi'an Jiaotong University(Medical Sciences) 1981;0(03):-
Objective To investigate the risk factors of complication rate after colorectal cancer operation. Methods This study recruited a total of 254 non-emergency colorectal cancer patients admitted to our hospital between December 2005 and December 2006,and then evaluated the effects of life style, preoperative factors and intraoperative factors on postoperative complications. Results Single factor analysis showed that the postoperative complication rate was not significantly affected by gender,age,obesity,palliative/curative resection, anesthesia style as well as preoperative drinking or smoking history.Preoperative complications(P=0.001),tumor stage and operation time(P=0.025) affected the postoperative complicatin rate.Multi-factor logistic regression analysis showed that preoperative complications were the only risk factor of postoperative complications [P=0.001,OR=5.871,(95% CI 2.958-11.651)].Conclusion Old age as such does not represent a contraindication for surgical treatment.Preoperative complications, operation time and tumor stage significantly affect the postoperative complication rate.Preoperative complications are the strongest risk factor of all. Therefore, reasonable perioperative managements and shortening operation time are the key to reducing postoperative complications.

Objective: To investigate the influence of TGF-β and IFN-γ on the proliferation, migration and invasion of melanoma cells. Methods: Melanoma cells were cultured in vitro. When tumor cells were confluent about 80% degree, cytokines were added into cell culture media. The concentration of TGF-β and IFN-β was 5ng/mL and 10ng/mL, respectively. Melanoma cells were divided into free-cytokine group, TGF-β group,IFN-γ group, TGF-β and IFN-γ group. Tumor cells in each group were then incubated for 8h, 16h, 24h, 32h,40h and 48h, respectively. After incubation, fixing and staining with SRB, the optical densities and percentage viability were then determined by absorption at 540 nm (A 540). The scarification of tumor cells in each group on the surface was created by a 2001μL pipette tube. The motility of tumor cells in each group was assessed by measuring the distance between scarifications. The speed of the scuffing closure was monitored after 12h.The invasive ability of melanoma cells was observed by transwell cultivation. The tumor cells that invaded through the Matrigel and adhered to the bottom of the outside membrane were determined by absorption at 595 nm (A595). Gelatin zymography assay was used to examine the levels of matrix metalloproteinases-2 (MMP-2) and matrix metalloproteinases-9 (MMP-9) activity when the tumor cells were treated with cytokines after 24h. MMP-2 and MMP-9 activity was demonstrated by gradation in the sodium dodecyl sulfate polyacryl-amide gel electrophoresis (SDS-PAGE) gelatin. MMP-2 and MMP-9 activity was determined by Image analy-sis Software. Results: TGF-β promoted the proliferation, migration and invasion of melanoma cells (P<0.05).However, IFN-γ inhibited the proliferation, migration and invasion of melanoma cells (P<0.05). The effect weakened or disappeared when both of them were used (P>0.05). Conclusion: In vitro, TGF-β may affect the inhibitory effect of IFN-γ on the proliferation, migration and invasion of melanoma cells. This study provided a better understanding of the relationship between tumor and inflammatory factors and established a good ba-sis for future research.
9. General considerations on bioanalytical methods for comparative evaluation of biosimilars immunogenicity
Chinese Journal of Clinical Pharmacology and Therapeutics 2021;26(7):721-728
Based on the principle of progressive comprehensive comparability, must be similar to reference drugs in terms of physicochemical properties, bioactivity, pharmacokinetics, efficacy, and safety (including immunogenicity). The objective of the immunogenicity assessment of biosimilars is to assess potential differences in the incidence and severity of human immune responses between biosimilars and reference drugs. No clinically significant differences in immune responses between the two products is a key factor in demonstrating biosimilarity. The influencing factors include subjects, sampling scheme, product factors and bioanalysis methods. An important issue in bioanalysis is the selection of two-assay (based on biosimilars and reference agents respectively) or one-assay (based on biosimilars respectively) as the best method for comparative immunogenicity assessment. In order to support the development of biosimilars, this paper focuses on the use of One-assay to evaluate immunogenicity based on biosimilars. The development and validation of an ADA assay for biosimilar drugs should include all validation of the original drug. In addition, biosimilars need to be compared with reference to confirmation thresholds, antigenic equivalence, and drug tolerance to assess the ability of biosimilars and reference drugs to bind in a similar manner to positive controls. ADA data analysis should be combined with PK/PD parameters and clinical events.
