6. Study on the tissue distribution of 3-n-butylphthalide in rats by HPLC
Chinese Pharmaceutical Journal 2012;47(19):1574-1577
OBJECTIVE: To study the tissue distribution of 3-n-butylphthalide (NBP) in rats. METHODS: NBP was analyzed on a Hypersil ODS2 column with the mobile phase consisting of 0.05 moL · L-1 sodium acetate-acetonitrile (50: 50, adjust pH to 4.5 with acetic acid). The flow rate was 1.0 mL · min-1 and the column temperature was 40°C. The wavelength of UV detector was set at 228 nm. A single dose of 360 mg · kg-1 NBP suspensions was administered orally to rats. The tissues of heart, brain, liver and kidney were taken at 1, 2, 4 and 8 h after administration. NBP concentrations in tissues were assayed by HPLC-UV method. RESULTS: The calibration curves for NBP in tissues were obtained over the concentration range of 0.0498 - 9.96 μg · mL-1 with good linearity and recoveries. The concentration of NBP at 1 h after administration was maximum in kidney followed by brain, heart and liver. CONCLUSION: The method is simple, sensitive, specific, accurate and stable. It can be used for the study on tissue distribution of NBP in rats. The results show that NBP is rapidly and widely distributed in rats and can cross the blood-brain barrier. 7.Association of BRCAl-related DNA repair genetic variations and high incidence of metastasis in triple-negative breast cancer
China Oncology 2009;19(7):552-556
Triple-negative breast cancer is defined by the absence of expression of oestrogen, progesterone, and Her-2 receptors. The clinicopathological characteristics include: higher histological grades, earlier relapse with worse prognosis, and a higher incidence of visceral metastases than bone metastases. Since it has no response to hormone treatment and targeted therapy for Her-2, chemotherapy became the main therapy. In addition, it may have a relationship with basal-like breast cancer and BRCAl-related breast cancer on the phenotype and molecular level. Inter-individual variations in DNA damage and repair have been associated with an increased risk of breast cancer. BRCA1 together with its associated genes such as BRCA2, ATM, RAD51 and CHEK2 play vital roles in different DNA repair pathways to preserve genome stability. Therefore variations of BRCA1 and its associated genes in multiple repair pathways may result in reduced DNA repair capacity and altered sensitivity to cytotoxic chemotherapeutic agents, which could promote incidence of distant metastases. Reduced DNA repair capacity and dysfunctional BRCA1 pathway in triple-negative breast cancer have been reported, however the detailed mechanisms are still unknown. Do the genetic variations of tumor cells or resistance of tumor cells to therapeutic drugs play the dominant role in high incidence of metastases in patients with triple-negative breast cancer? In this review, the association of dysfunctional BRCA1-related DNA repair pathways with high incidence of metastasis in triple-negative breast cancer would be introduced.

9.Mechanisms of secondary resistance to imatinib in gastrointestinal stromal tumors
Journal of International Oncology 2010;37(12):936-939
Imatinib is an effective tyrosine kinase inhibitor, used in the treatment of advanced or inoperable gastrointestinal stromal tumors. Despite its efficacy, resistance is often developed among a portion of patients. Resistance to imatinib can be divided into primary and secondary resistance, with the latter becoming the focus of many recent researches. Research on the mechanisms of secondary resistance may lead to the identification of novel therapeutic strategies.
10.Life quality improvement of spastic hemiplegia of stroke treated with fire-needle: a randomized controlled trial.
Chinese Acupuncture & Moxibustion 2015;35(11):1105-1109
OBJECTIVETo observe the life quality improvement of spastic hemiplegia of stroke treated with fire-needle.
METHODSSixty patients were randomly divided into a fire-needle group and a rehabilitation group, 30 cases in each one. In the two groups, the conventional treatment of neurology and rehabilitation strengthening limbs function were applied. In the fire-needle group, fire-needle was used at Jianyu (LI 15), Binao (LI 14), Quchi (LI 11), Yanglingquan (GB 34), Xuehai (SP 10) and Sanyinjiao (SP 6), etc. , once two days and continuously for 30 days (15 times). In the rehabilitation group, conventional rehabilitation exercises were given five times a week for 4 weeks (20 times). Before treatment, at the end of treating program and one month later after treatment, spasticity of upper and lower limbs was assessed by Ashworth scale and clinic spasticity index (CSI); limbs function was evaluated by Fugl-Meyer (FM) scale; life quality was estimated by patient-reported outcome (PRO). Clinical effects were compared between the two groups.
RESULTS(1) At the end of treating and one month later after treatment, the Ashworth scales were better than those before treatment in the two groups (P < 0.05, P < 0.01), and the scales of the fire-needle group were superior to those at the same time of the rehabilitation group (both P < 0.05). (2) At the end of treating and one month later after treatment, the CSIs were better than those be fore treatment in the fire-needle group (both P < 0.01), but not changed apparently in the rehabilitation group (both P > 0.05). And the indices of the fire-needle group were superior to those at the same time of the rehabilitation group (P < 0.05, P < 0.01). (3) The FM scales of upper and lower limbs at the end of treating and one month later after treatment were better than those before treatment in the two groups (all P 0.01), except the lower limbs at the end of treating in the rehabilitation group (P > 0.05). The FM scales of the fire-needle group were superior to those at the same time of the rehabilitation group (P < 0.05, P < 0.01). (4) At the end of treating and one month later after treatment, the PRO scales were better than those before treatment in the two groups (all P < 0.01), and the scales of the fire-needle group were superior to those at the same time of the rehabilitation group (P < 0.05, P < 0.01).
CONCLUSIONFire-needle can effectively relieve spastic hemiplegia of stroke, and improve limbs motor function and life quality. It is better than rehabilitation.
