2.METTL3 regulates glucose transporter expression in placenta exposed to hyperglycemia through the mTOR signaling pathway

Background::Alterations in the placental expression of glucose transporters (GLUTs), the crucial maternal-fetal nutrient transporters, have been found in women with hyperglycemia in pregnancy (HIP). However, there is still uncertainty about the underlying effect of the high-glucose environment on placental GLUTs expression in HIP.Methods::We quantitatively evaluated the activity of mammalian target of rapamycin (mTOR) and expression of GLUTs (GLUT1, GLUT3, and GLUT4) in the placenta of women with normal pregnancies (CTRL, n = 12) and pregnant women complicated with poorly controlled type 2 diabetes mellitus (T2DM, n = 12) by immunohistochemistry. In addition, BeWo cells were treated with different glucose concentrations to verify the regulation of hyperglycemia. Then, changes in the expression of GLUTs following the activation or suppression of the mTOR pathway were also assessed using MHY1485/rapamycin (RAPA) treatment or small interfering RNA (siRNA)-mediated silencing approaches. Moreover, we further explored the alteration and potential upstream regulatory role of methyltransferase-like 3 (METTL3) when exposed to hyperglycemia. Results::mTOR, phosphorylated mTOR (p-mTOR), and GLUT1 protein levels were upregulated in the placenta of women with T2DM compared with those CTRL. In BeWo cells, mTOR activity increased with increasing glucose concentration, and the expression of GLUT1, GLUT3, and GLUT4 as well as GLUT1 cell membrane translocation were upregulated by hyperglycemia to varying degrees. Both the drug-mediated and genetic depletion of mTOR signaling in BeWo cells suppressed GLUTs expression, whereas MHY1485-induced mTOR activation upregulated GLUTs expression. Additionally, high glucose levels upregulated METTL3 expression and nuclear translocation, and decreasing METTL3 levels suppressed GLUTs expression and mTOR activity and vice versa. Furthermore, in METTL3 knockdown BeWo cells, the inhibitory effect on GLUTs expression was eliminated by activating the mTOR signaling pathway using MHY1485. Conclusion::High-glucose environment-induced upregulation of METTL3 in trophoblasts regulates the expression of GLUTs through mTOR signaling, contributing to disordered nutrient transport in women with HIP.
3.Association between rs579459 polymorphism of ABO gene and clinical severity of coronary heart disease in Chinese Han population

Objective To investigate the association of rs579459 polymorphism in ABO gene with clinical severity of coronary heart disease (CHD) in Chinese Han population. Methods Based on the results of study on the association with CHD conducted by our team ,the rs579459 polymorphism in ABO gene and clinical data of 515 Chinese Han patients with CHD and 908 healthy individuals were collected and analyzed with Plink 1.07 software. Results The rs579459 polymorphism in ABO gene using allele model showed a statistically significant difference between CHD and control group (OR :1.28 ,95% CI :1.11-1.65 ;P=0.027).When stratified by the severity of CHD ,the rs579459 polymorphism in ABO gene using allele model showed statistically significant difference between the severe CHD group and control group (OR :1.78 ,95% CI :1.25-2.34 ;P=1.85 × 10-4 ). Conclusions The rs579459 polymorphism of ABO gene is associated with susceptibility to CHD in Chinese Han population ,and also with the severity of CHD.
4.A study on changes in autonomic nervous system function in elderly coronary heart disease patients with hypertension

Objective:To investigate changes in autonomic nervous system function in elderly coronary heart disease patients with hypertension.Methods:Clinical data of 186 patients with coronary heart disease(CHD)from January 2017 to January 2021 in our hospital were retrospectively collected.Patients were divided into the CHD group(n=93)and the CHD with hypertension group(n=93)according to whether they had hypertension.All subjects underwent 24-hour dynamic electrocardiogram, and differences in heart rate variability(HRV)were analyzed and compared between the groups.Results:Values from time-domain parameters for HRV included the standard deviation of the normal-to-normal R-R intervals( SDNN)[(77.4±15.1)ms vs.(114.6±25.9)ms, t=11.97, P<0.05], the standard deviation of sequential five-minute R-R interval means( SDANN)[(66.8±14.5)ms vs.(97.4±25.0)ms, t=10.21, P<0.05], the SDNN index[(34.1±11.4)ms vs.(51.9±18.0)ms, t=8.07, P<0.05], the root mean square successive difference( rMSSD)[(26.1±13.9)ms vs.(36.1±27.2)ms, t=3.18, P<0.05], and the percentage of successive normal interbeat intervals greater than 50 milliseconds(PNN50)[(5.0±6.4)% vs.(8.3±11.0)%, t=2.53, P<0.05], and were lower in the CHD with hypertension group than in the CHD group.Values from frequency-domain parameters for HRV included low frequency(LF)[(168.9±202.8)ms vs.(443.6±663.6)ms, t=3.78, P<0.05], high frequency(HF)[(203.3±202.5)ms vs.(499.5±1222.7)ms, t=2.28, P<0.05], and the LF/HF ratio(1.0±0.7 vs.1.3±0.8, t=3.18, P<0.05), and were lower in the CHD with hypertension group than in the CHD group. Conclusions:Impairment of cardiac autonomic nervous system function is more serious in elderly CHD patients with hypertension than in elderly CHD patients.Heart rate variability is a valuable clinical indicator and may be used for treatment targets in evaluating dysfunction of the autonomic nervous system in CHD patients with hypertension.
