Glucagon-like peptide-1 (GLP-1) is an ineretin hormone released from intestinal L-cells into circulation in response to dietary nutrient intake.Based on the recently finished studies,we reviewed the impact of GLP-1 on gastric emptying,satiety,and food intake.Meanwhile,the ehnical advantage and gastrointestinal adverse effects of long-acting GLP-1 analogue liraglutide were evaluated.

4.Experimental study of intratumor injection of ~(90)Y-SAg for the treatment of liver cancer in mice

Conclusion: Intratumor injection of ~(90)Y-SAg is a safe,feasible and effective method to treat liver cancer in mice.
5.INDUCING ARSENIC-RESISTANCE CELLS WITH HUMAN BONE MARROW-DERIVED MESENCHYMAL STEM CELLS

Objective To obtain stable arsenic-resistance cells,the fetal bone marrow mesenchymal stem cells(BMSCs)were exposed to low-level arsenite for 18 weeks. Methods Cells from 4 months fetal bone marrow were cultured in ?-MEM medium to obtain BMSCs.After 24h cytotoxicity test of the fetal BMSCs,we chosed 1?mol/L NaAsO_2 to be the dose under which the cell death-rate was 5%-10%.The fetal BMSCs were exposed to low-level arsenite.MTT was used to detect the survival rate and IC_(50) of arsenic-exposed cells and the control cells,which can reflect the change of arsenic tolerance.A hydride generationatomic fluorescence spectrometry method was used to detect arsenic in the arsenic-resistance cells and the control cells.In order to study the mechanism of arsenic-resistance,we also examined the intracellular GSH and GST content in the arsenic-resistance cells and the control cells. Results The fetal BMSCs were continuously exposed to 1?mol/L NaAsO_2.Parallel cells were cultured in medium without arsenic provided passage-matched control.After the fetal BMSCs were continuously exposed to low level NaAsO_2 for 18 weeks,cells exhibited dramatic resistance to acute arsenite toxicity.Compared to control cells,arsenic-resistance cells showed reduction in arsenic accumulation in cells.The GSH levels and GST activity of arsenic-resistance cells were higher than that of control cells.Conclusion Acquisition of stable arsenic-resistance in BMSCs was available by using cell culture and adding low-level arsenic.Our research established a solid basis for further study about the mechanisms of arsenic-resistance in human being.
6.Establishment of two-dimensional polyacrylamide gel electrophoresis for serum proteomics

Objective: To establish a two-dimensional polyacrylamide gel electrophoresis(2-DE) technique for serum proteomics.Methods: Various conditions of serum proteomic 2-DE were adjusted and optimized.Results: A steady 2-DE technique was established.Conclusions: Our established 2-DE technique of serum proteins can be effectively applied in serum proteomics of diseases.
7.Screening and identification of an anti-bacterial and cytotoxic marine halophilic strain XD20

Objective:To isolate and screen the active marine microorganisms from East China Sea and to identify the phenotypes of the isolated active strains.Methods: The samples were widely collected from East China Sea and the active strains were screened out through an anti-bacterial model and a cytotoxic model.The isolated strains were subjected to phenotypical study,salt-aggregation test and rDNA ITS sequencing.Neighbor-joining phylogenetic trees were drawn by comparing the results of rDNA ITS sequencing with sequences of described species in the BLAST server of the National Center for Biotechnology Information(NCBI).One of the strains,subsequently named XD20,was identified to species level.Results: XD20,a halophilic strain with a highest tolerable salt concentration of 10.0% and suitable concentration range of 2.5% to 7.5%,was collected and isolated on the site of C01(122?26′30″,30?43′48″) 30 meters under water.XD20 showed an inhibitory effect on the growth of Candida albicans and a cytotoxic activity toward SMMC7721 and HeLa cells.XD20 produced arthroconidia by mycelium break and the arthroconidia had similar size and morphology to those of Wallemia sebi.rDNA ITS sequencing showed that the sequence of XD20 had a higher similarity to those of Wallemia sebi and the differences were located in ITS1 and ITS2 areas,both belong to variable region.Conclusion: XD20,an active halophilic strain fungus,has been obtained from the sea for the first time,and has been identified to be Wallemia sebi(Fr.) Arx.
8.In vitro purification and culture of multipotent adult progenitor cells from human bone marrow by magnetic activated cell sorting

Objective:To establish a method for isolation,purification,and culture of human multipotent adult progenitor cells(MAPCs) in vitro,so as to lay a foundation for the application of hMAPCs in tissue-engineering research and clinical medicine.Methods: The mononuclear cells were obtained from bone marrow of healthy volunteers by gradient centrifugation and were subjected to adherence culture.The adherent cells were then subjected to magnetic activated cell sorting(MACS)(depletion selection with CD45 and GlyA microbeads).Then the purity of selected cells was identified by flow cytometer.The growth of the purified cells was observed and the expression of CD29,CD44,CD34,and HLA-DR was analyzed by flow cytometers.Results: (1)Averagely(5-10)?10~(4)/ml hMAPCs could be separated from 1?10~(6)/ml bone marrow mononuclear cells by MACS.The cell viability was similar before(96.7?1.7%) and after(96.0?2.4%) isolation.(2)The isolated MAPCs grew well in the self-designed culture medium and could be passaged for 20 generation.(3)The purity of the CD45~-and GlyA~-cells separated from bone marrow adherent cells was more than 98% as determined by flow cytometer.(4) In hMAPCs,the positive rate was 99.2% for CD29,98.3% for CD44,1.2% for CD34,and 5.3% for HLA-DR.Conclusion:(1)The bone marrow-derived hMAPCs can be purified by MACS through depletion selection of CD45 and GlyA microbeads.(2)The hMAPCs can be expanded in vitro in self-designed medium,maintaining a non-differentiation state for a long time.
9.Observation of Curative Effect on Treating The Old Osteoporosis with The Chinese Medicine Which Warm Kidney and Benefit Spleen Added with 1a(OH)D3

[Objective] To study the clinical effect of the decoction of warming kidney and notifying spleen added with 1a(OH)D3 on senile osteoporosis.[Method] Administer 180 cases with the decoction and 1a(OH)D3 3 tablets;test the BMD change of lumbar vertebra and left thighbone before and after treatment,record the low back pain as well.[Result] After 3 courses,BMD was increased by 2.02% for low back,it was by 1.82% for thighbone;there’s difference between before and after treatment;82 cases were markedly relieved on low back pain(45.56%),76(42.22%)much relieved;there’s no marked difference on Ca,P and AKP.[Conclusion]The decoction and 1a(OH)D3 can improve bone concentration,increase bone intensity,relieve pain and symptoms.
10.Immunoregulatory effect of 1,25-dihydroxyvitamin D_3 on human peripheral blood lymphocytes

The effects of 1,25-dihydroxyvitamin D_3 on PHA-and alloantigen-induced human peripher-al blood lymphocyte proliferation,expressions of IL-2R and TfR were investigated by ~3H-TdRincorporation and indirect immunofluorescence methods respectively,The results demonstratedthat 1,25— (OH)_2D_3 had an inhibitory effect on PHA-induced proliferation (IC_(50)=19.1nM),which could partially reversed by exogenous recombinant IL-2,1,25 (OH)_3D_3 at high doses(10-100nM) it had an inhibitory effect.In both PHA-stimulated and alloactivated lymphocytes1,25(OH)_2D_3 could inhibit the expression of TfR and had no effect on that of IL-2R.This studysuggests that 1,25 (OH)_2D_3 in an important immunoregulatory hormone,the properties of whichneed further clarification.
