1.Histological and immunohistochemical studies on the epididymal duct in the dromedary camel (Camelus dromedarius).

This study was conducted to underscore the spatial distribution of some biologically active proteins within the epididymal duct in the dromedary camel. Paraffin-embedded sections from different regions of epididymis were stained by conventional histological techniques and by immunohistochemistry. A battery of primary antibodies against six proteins (S100, alpha smooth muscle actin [alpha-SMA], connexin-43 [Cx43], galactosyltransferase [GalTase], angiotensin converting enzyme [ACE], and vascular endothelial growth factor [VEGF]) were used. The epididymal epithelium consisted of five cell populations: principal, basal, apical, dark, and halo cells. The histochemical findings indicated the absence of binding sites for VEGF and Cx43. The principal cells (PCs) showed variable immunoreactivity (IR) for ACE, S100, and GalTase throughout the whole length of the duct. The apical surfaces of most PCs (at the caput) and some PCs (at the corpus) exhibited intense ACE-IR, whereas those at the cauda displayed alternating negative and strong immunostaining. Similarly, moderate S100-IR was found in cytoplasm and nuclei of all PCs at the caput, few PCs at the corpus, and several PCs alternating with negative PCs at the cauda. In contrast, only some PCs showed weak to strong GalTase-IR in different regions. Apart from negative to weak positive S100-IR, basal cells failed to show IR for all other proteins. Apical cells displayed strong IR for ACE, S100, and GalTase with some regional differences. The peritubular and vascular smooth muscle cells revealed strong alpha-SMA-IR in all regions. In conclusion, the spatial distribution of different proteins in camel epididymis showed similarities and differences to other mammalian species. The region-specific topographic distribution of different proteins and cell types might indicate that the caput and cauda are metabolically more active than that of the corpus.

2.The nasoalveolar molding technique versus DynaCleft nasal elevator application in infants with unilateral cleft lip and palate

Background:
The introduction of presurgical nasoalveolar molding represented a significant departure from traditional molding methods. Developed by Grayson and colleagues in 1993, this technique combines an intraoral molding device with a nasal molding stent. This study aimed to compare the Grayson nasoalveolar molding appliance versus DynaCleft appliance as two methods of presurgical nasoalveolar molding.
Methods:
A single-blinded, randomized, parallel-arm clinical trial was conducted. Sixteen infants with complete unilateral cleft lip and palate were enrolled and divided into two groups of eight. Group 1 was treated with a modified Grayson nasoalveolar molding appliance that included a nasal stent, while group 2 was treated with DynaCleft elastic adhesive tape and an external nasal elevator. Standardized digital photographs of each infant were taken at baseline and post-treatment using a professional camera. Nine extraoral anthropometric measurements were obtained from each image using image measurement software.
Results:
The modified Grayson nasoalveolar appliance demonstrated a more significant improvement compared to DynaCleft in terms of alar length projection (on both sides), columella angle, and nasal tip projection. Symmetry ratios also showed enhancement, with significant improvements observed in nasal width, nasal basal width, and alar length projection (p< 0.05).
Conclusion
Both the modified Grayson nasoalveolar appliance and DynaCleft appear to be effective presurgical infant orthopedics treatment options, demonstrating improvements in nasolabial aesthetics. The modified Grayson appliance, equipped with a nasal stent, improved nasal symmetry more effectively than DynaCleft, resulting in a straighter columella and a more medially positioned nasal tip.
3.Fibrin Sealant and Lipoabdominoplasty in Obese Grade 1 and 2 Patients.

BACKGROUND: Ever since lipoabdominoplasty was first developed to achieve better aesthetic outcomes and less morbidity, the rate of seroma formation, especially in obese patients, has disturbed plastic surgeons. The aim of this study was to evaluate the effect of fibrin sealant in the prevention of seroma formation after lipoabdominoplasty in obese patients. METHODS: Sixty patients with a body mass index (BMI) between 30 and 39.9 were assigned randomly to 1 of 2 groups (30 patients each). Group A underwent lipoabdominoplasty with fibrin glue, while group B underwent traditional lipoabdominoplasty; both had closed suction drainage applied to the abdomen. The patients' demographics and postoperative complications were recorded. Seroma was detected using abdominal ultrasound examinations at two postoperative periods: between postoperative days 10 and 12 and, between postoperative days 18 and 21. RESULTS: The age range was 31 to 55 years (38.5+/-9.5 years) in group A and 25 to 58 years (37.8+/-9.1 years) in group B, while the mean BMI was 31.4 to 39.9 kg/m2 (32.6 kg/m2) in group A and 32.7 to 37.4 kg/m2 (31.5 kg/m2) in group B. In group A, the patients had a complication rate of 10% in group A versus 43% in group B (P<0.05). The incidence of seroma formation was 3% in the fibrin glue group but 37% in the lipoabdominoplasty-alone group (P<0.05). CONCLUSIONS: Lipoabdominoplasty with the use of autologous fibrin sealant is a very effective method that significantly reduces the rate of postoperative seroma.

4.Expression of Histo-blood Group Antigens in Tumor and Adjacent Normal Breast Tissues as Prognostic Markers of Breast Carcinoma

PURPOSE:
Aberrant glycosylation of the histo-blood group antigens (including the angina bullosa haemorrhagica [ABH]) is often observed during malignant transformation in most types of carcinomas. Data concerning their ethnic distributions are diverse which explains why their biological characteristics have to be studied in different populations. Our aim was to analyze the expression of the histo-blood group (specifically the ABH) antigens in breast carcinoma.
METHODS:
The expression of the histo-blood group (specifically the ABH) antigens was studied in 109 patients with breast carcinoma using immunohistochemistry. Statistical analysis was performed using χ² and Fisher analyses.
RESULTS:
The loss of expression of histo-blood group (ABH) antigens in breast carcinoma was observed in 81.13% of patients with blood group O, 37.93% with blood group A, and 96.30% with blood group B. One key finding of this study was that the loss of expression of the ABH antigen was also observed in normal tissues adjacent to the tumor. The loss of expression was associated with higher tumor grade (p < 0.05). Expression of H antigen was observed in 50% of cases with loss of expression of B antigen and was associated with human epidermal growth factor receptor 2 (HER2) overexpression (p < 0.05). The loss of H antigen in patients with blood group O was associated with estrogen receptor expression (p < 0.001). Incompatible A antigen in tumor was expressed in 20.75% of patients with blood group O.
CONCLUSION
Loss of the ABH antigens correlated with the Scarff-Bloom-Richardson histologic grading. H antigen was associated with HER2 overexpression in breast cancer. However, further studies are needed to determine the role of incompatible A antigen in mammary carcinogenesis.
5.Prevalence of methicillin-resistant Staphylococcus aureus on cellphones of healthcare workers in Najran University hospital, Najran City (Saudi Arabia)

Aims:
Mobile phones are used extensively by healthcare workers (HCWs) who are unaware of the amount of
contamination these phones carry and act as reservoirs for organisms causing hospital-acquired infections. This investigation was aimed to find the prevalence of methicillin-resistant Staphylococcus aureus (MRSA) contamination and antimicrobial resistance patterns of isolates from HCW’s cellphones.
Methodology and results:
This study was a hospital-based cross-sectional study. A total of 120 samples were isolated from HCWs’ cellphones and subjected to culture and sensitivity as per the standard guidelines. Five (18.1%) out of 120
collected samples were from 11 lab technicians’ phones and were positive for S. aureus and 6.25% were obtained from 48 nurses’ cellphone swabs.
Conclusion, significance and impact of study
The findings of this study reveal that HCWs’ cellphones could be apossible source of infection since a high prevalence of MRSA was found on lab technicians and nurse cellphone sample swabs. On the other hand, all S. aureus isolates were resistant to methicillin and ceftazidime. Also, no significant relationship between the prevalence of MRSA was detected.
6.The safety, efficacy and pharmaceutical quality of male enhancement nutraceuticals bought online: Truth versus claim.

OBJECTIVE:
Nutraceutical products are widely used for their claimed therapeutic benefits. However, falsified or adulterated nutraceuticals present a major health threat to consumers. This study investigates the pharmaceutical quality, safety and anti-inflammatory effects of six male enhancement nutraceuticals that claim to be 100% natural.
METHODS:
Three batches of six male enhancement products were tested to detect the presence and levels of adulterants via high-performance liquid chromatography (HPLC). The pharmaceutical quality of the selected nutraceuticals was tested with near infrared spectroscopy (NIR) and SeDeM. The cytotoxic effects of these products on HepG2 cells were determined through cell proliferation (XTT) and lactate dehydrogenase (LDH) cytotoxicity assays. Lastly, the in vitro inflammatory effects of these products were investigated using murine J774 macrophages through cytokine release analysis.
RESULTS:
HPLC analysis detected the presence of sildenafil citrate, a vasodilator, and the active ingredient in Viagra and Revatio, in all batches of the products we analyzed. Amount of sildenafil citrate ranged from 0.45 mg to 51.85 mg among different batches. NIR assessment showed inter- and intra-batch heterogeneity in product composition. Results of the XTT and LDH assays showed significant cytotoxic effects of the analyzed products. XTT analysis revealed that the viability of HepG2 treated with tested products varied from 27.57% to 41.43%. Interestingly, the male enhancement products also showed anti-inflammatory effects.
CONCLUSION
Despite their labeling as 100% natural, all products tested in this study contained levels of sildenafil citrate, which was not reported on the packaging. There was a lack of pharmaceutical uniformity among products of the same batch and across different batches. Additionally, the products we tested had cytotoxic effects. These study findings highlight the adulteration, poor quality and hazard of these nutraceuticals. Therefore, strict regulation of these products and standardization of the definition of nutraceuticals are urgently needed. Further, these falsely advertised products should be withdrawn from the market due to potential adverse effects on the health of their consumers.
