1.Effect of flavonoids from Hippophae rhamniodes residues on blood lipoid metabolism and antioxidative activity in climacteric rats.

OBJECTIVETo study the effect of flavonoids from seed residues of Hippophae rhamnoides (FH) on the lipid metabolism and antioxidative activity in climacteric rats.
METHODMenopausal rats with aging were used in this experiment. The rats were fed with FH by gastrogavage for 13 weeks. The effect of drug on the lipid metabolism and the antioxidative activity were observed after the rats were killed.
RESULTSerum total cholesterol was decreased significantly in rats fed with FH, T-AOC and SOD in serum and liver were significantly higher than those in rats fed with water, and at the same time MDA was lower than that in rats fed with water.
CONCLUSIONFH can improve the climacteric rats' lipid metabolism, and enhance the antioxidation in climacteric rats.

2.Impact of targeted drug therapy on the prognosis of non small cell lung cancer patients with different mutations in the epidermal growth factor receptor gene

Objective To investigated the impact of patients with ad-vanced non small cell lung cancer (NSCLC) treated by the targeted drug tyrosine kinase inhibitor ( EGFR -TKI ) , and the prognosis of the pa-tients with different mutations in the epidermal growth factor receptor ( EGFR) gene.Methods A total of 69 cases of stageⅢb-ⅣNSCLC patients were selected, there were 21 cases of EGFR gene mutation (+) by DNA testing.Among them, there were 11 cases ( 52.4%) with out-side exon 19 mutations ( treatment group I) , 8 cases (38.1%) with out-side exon 21 mutations ( treatment groupⅡ) , 2 cases (9.5%) with out-side exon 20 mutations ( treatment groupⅢ) .Patients in treatment group were treated with standard regimens and oral TKI ( erlotinib hydrochlo-ride, 150 mg, once daily).The patients in control group were only trea-ted with standard treatment.All patients were followed up for 1 to 2 years, and the short -term efficacy and disease control rate were evaluated, and the progression-free survival rate and 1.5 year survival rate were investigated.Results After treatment, effective rate of treatment and control group ( 85.7%, 52.1%) and disease control rate ( 90.5%, 64.6%) had significant difference ( P<0.05 ); the difference of effective rate in three sub groups of treatment group also had statistically significance (72.7%,50.0%, 0%, P <0.05 ) .At follow -up of 1.5 years, the progression -free survival rate (28.6% and 8.3%) and total survival rate ( 47.6%, 16.7%) of treatment and control groups were significant difference, the significant differences were also observed in the three sub-groups in treatment group ( P<0.05).The treatment effect and the prognosis of patients with the exon 19 mutations in treatment group Ⅰ were better. Conclusion Good clinical effect of NSCLC patients with EGFR gene mutations was obtained by the treatment of TKI, but the prognosis of patients with different mutations of EGFR gene was different.
3.In vivo study of antifertility and anti-inflammatory effects of compound Kucen gel.

OBJECTIVETo study the antifertility and anti-inflammatory effects of compound Kucen gel in vivo.
METHODSAs antifertility experiment, we randomly divided 60 female SD rats into six groups of equal number: normal saline, blank gel, low-, medium- and high-dose compound Kucen gel (0.05, 0.10 and 0.15 g/g), and positive control (4% nonoxynol gel) to receive intravaginal administration of 200 microl of respective agent, followed by copulation with male rats in a 1:1 ratio. At 12 days after successful mating, the female rats were dissected for calculation of the embryos and the rate of contraception. As an anti-inflammatory trial, we established a mouse model of inflammation by applying xylene to the pinna, and equally randomized 60 Kunming mice to six groups as in the former experiment. We determined the degrees and average rates of swelling inhibition in the left ear.
RESULTSHigh-dose compound Kucen gel achieved a fertility-inhibition rate of 100% in the female rats, the number of embryos significantly lower than in the normal saline group (0.00 +/- 0.00 vs 11.00 +/- 2. 00, P < 0.05), but with no statistically insignificant difference from that of the positive control (0.00 +/- 0.00, P > 0.05). High-dose compound Kucen gel also markedly suppressed swelling in the left ear of the mice, with an inhibition rate of 52.3%, the average swelling degree significantly lower than in the normal saline group (10.17 +/- 2.56 vs 21.32 +/- 3.17, P < 0.01), but not remarkably different from that of the positive control (8.53 +/- 1.89, P > 0.05).
CONCLUSIONCompound Kucen gel, with its strong antifertility and anti-inflammatory effects, deserves further study and clinical application.

4.Clinical trial of dexmedetomidine in the treatment of severe sepsis patients with acute lung and kidney injury

Objective To observe the influences of dexmedetomidine (DEX) on efficacy,inflammatory factors,liver and kidney functions in severe sepsis patients with acute lung and kidney injury.Methods A total of 60 severe sepsis patients with acute lung and kidney injury were randomly divided into treatment group and control group,30 cases in each group.Patients in two groups were maintained with regular treatment.Patients in treatment group were received DEX injection 1 μg · kg-1 · h-1 for early sedative treatment.While,patients in control group were received propofol injection for early sedative treatment.They were treated for 48 h.Time of mechanical ventilation,intensive care unit (ICU) stay time,and duration of continuous renal replacement therapy (CRRT) were recorded.Liver and kidney functions indexes and the levels of inflammatory factors in two groups before and after treatment were detected.Results ICU stay time and duration of CRRT in control group were (96.32 ± 13.51),(61.30 ±9.85)h,had significant difference with those in treatment group,which were (76.18 ± 11.32),(52.93 ± 9.21) h (P < 0.01).After operation,the levels of procalcitonin,C reactive protein,interleukin-6 and tumor necrosis factor in treatment group were(2.05 ±0.39),(56.36 ±9.94),(78.72 ± 11.33),(17.56 ±4.07)ng · L-1,had significant differences with those in eontrol group,which were (2.43 ±0.24),(79.91 ± 12.55),(92.07 ± 12.55),(24.41 ±4.25) ng · L-1 (all P < 0.01).After operation,the levels of alanine aminotransferase,aspartate transaminase,alkaline phosphatase,total bilirubin,blood urea nitrogen and serum creatinine in treatment group were (21.37 ± 4.73) U·L-1,(29.61 ±6.97)g · L-1,(67.27 ±7.26)U · L-1,(11.36 ±2.91)μmol · L-1,(7.03 ±2.03) mmol· L-1,(87.98 ±10.56)μmol · L-1,had significant difference with those in control group,which were (30.46±6.44)U· L-1,(38.60 ±7.53)g · L-1,(71.75 ±8.41)U · L-1,(14.72 ±2.29)μmol · L-1,(10.07 ± 3.12) mmol · L-1,(123.73 ± 11.74) μmol · L-1 (all P < 0.05).There was no significant adverse drug reaction in two groups.Conclusion Compared with propofol,dexmedetomidine has better clinical effect for severe sepsis patients with acute lung injury by improving the inflammatory factors and protecting organ.

With social development and technology progress, there is an increasing use of daily electronic products and radioactive medical devices, which increases the risk of exposure to radiation in medical practices, daily life activities, and work. Notably, the effects of radiation exposure on pregnant women and their offspring have attracted increasing attention. Comprehensive and accurate assessment and intervention of the effects of radiation exposure on pregnant women and their offspring are of great significance to reduce birth defects and achieve eugenics. This article reviews the effects of radiation exposure during pregnancy on offspring health based on animal experiments and clinical studies, and describes the possible mechanisms, so as to provide the basis for further studies.

Objective To investigate the impact of n?3 polyunsaturated fatty acid (n?3 PUFA) on function and expression of store?operated calcium channels (SOCC) in coronary artery smooth muscle cells (SMC) derived from diabetic rat. Methods A total of 180 healthy male Sprague?Dawley (SD) rats were randomly divided into normal group (N, n=45), placebo?treated diabetic group (D, n=45), lose dose n?3 PUFA treated diabetic group (DL, n=45) and high dose n?3 PUFAs treated diabetic group (DH, n=45). Streptozotocin?induced diabetic rat animal model was established by two consecutive intraperitoneal injections. After modeling, rats in group DL and DH were treated with 10 mg·kg-1·d-1 and 50 mg·kg-1·d-1 n?3 PUFAs respectively per gavage for eight weeks. After eight weeks, rat coronary artery SMC was isolated by enzyme digestion. Changes of cytosolic calcium concentration in coronary artery SMC were examined by calcium fluorescence imaging technique, coronary artery tension was detected by myograph system, and protein expressions of SOCC on coronary artery SMC were measured by Western blot. Results SOCC induced ΔF340/F380 of group N, D, DL and DH were 0.425±0.023, 0.838±0.037, 0.342±0.052 and 0.364± 0.045 respectively, which was significantly lower in group N, DL, DH than in group D (P<0.05). SOCC induced changes of tensions were 0.94±0.09, 1.95±0.18, 1.35±0.24 and 1.01±0.18 in the group N, D, DL and DH, respectively, which was significantly lower in group N and DH than in group D (P<0.05). Protein expressions of STIM1, Orai1 and TRPC1 were significantly higher in diabetic rat coronary SMC than in group N (P<0.05). STIM1 protein expressions were significantly lower in group DL and DH than in group D, and Orai1 and TRPC1 protein expressions were similar among group. Conclusions Coronary artery tension, cytosolic calcium concentration and protein expressions of SOCC are higher in diabetic rat coronary artery SMC when compared with normal rats. n?3 PUFA intervention could downregulate the protein expression of SOCC, reduce cytosolic calcium concentration and coronary artery tension, and is protective to the diabetic injury in coronary artery.
