Acta Anatomica Sinica 1955;0(03):-
Epitheloid hepatomic cell lines FSK 7901, FSK 7902 were established in cell culture from male rats with an average body weight 103.03?26.73 g sacrified 131?24.94 days after feeding with 107.1?25.65 mg. of diethylnitrosamine(DEN).The chromosome number of the majority of the cells from FSK 7901, FSK 7902 falls between a range of 63~84 with a mode of 81.The cells are very closely arranged in FSK 7901, but loosely in FSK 7902.The distribution of the smallest basophilic hepatomic cells of FSK 7901 are regularly around or within the new growth area, but in FSK 7902 are irregularly scattered. The acentrically displaced bean-shaped nuclei of moderate or large hepatomic cells with large juxtanuclear acidophilic area are more numerous and distinct in FSK 7901 than in FSK 7902.The growth of FSK 7902 is more rapid than FSK 7901 as judged from the main cell cycle time from population doubling time during lorgarthmic growth of culture and the average of mitotic index.

Acta Anatomica Sinica 1953;0(01):-
Liver carcinogenesis induced by diethylnitrosamine (DEN) was studied in 19 male albino rats under electron microscope. The effects of DEN on hepatocytes included following basic abnomalities:1. The characteristic features of the nucleus are irregulat in shape, increase of interchromatin granules, and decrease of nucleolus-associated chromatin. The nucleoli are enlarged, irregular in shape and showing remarkable deformity of nucleolonema.2. The characteristic features of the cytoplasm are as follows. The mitochondria increase or decrease in number and size, they are irregular in shape with some variation in cristal pattern. Endoplasmic reticulum (ER) increases in the width of the lumina, detachment of some ribosomes from the membraneous component of rough endoplasmic reticulum (RER) and the appearance of concentric bodies of ER (RER concentric body or SER concentric body).3. The characteristic features of the cell membrane are the increase in the number of microvilli facing the Disses' space, the enlargement of bile canaliculi with increase or decrease in number of microvilli. Plasmalemma connecting between the adjacent hepatoma cells may be straight, wavelike or interdigitated.

We prepared bovine epidermal keratin (EK) antibody, which can be used as a specific marker of cholangiocarcinoma(CC) cells to be distinguished from hepatocellular carcinoma (HCC) cells. The intrahepatically implanted solid tumors of FSK 7901 and FSK 7902 have the histological structures of CC and HCC, respectively. Using immunofluorescence and PAP techniques, we demonstrated that, in the solid tumors, FSK 7901 cells are definitely EK-positive, while most of the cells of FSK 7902 are EK-negative. Therefore, we considered FSK 7901 and FSK 7902 as CC and HCC, respectively. In the solid tumors of FSK 7902, few cells which are EK-positive may be those in which there are Mallory bodies or the structures are forming.

This paper presents the results of an ultrustructural study of the hepatoma cell lines FSK 7901, FSK 7902 of long term cultures about 2 years more. The size of cell varies. Distribution of the cell is single or in epitheloid monolayer. Pseudopodial processes or microvilli cover the perimeter of certain hepatoma cells. In the monolayer, the microvilli exist where the neighboring plasmalemmae are in loose contact. When their contact is close, the neighboring plasmalemmae may be straight, wavelike or interdigitated. The nucleus and enlarged nucleoli are irregu- lar in shape. The deformity of nucleolonema is remarkable.Rough endoplasmic reticulum (RER) and mitochondria are much less in number in the cytoplasm, but the free ribosomes is very abundant. The shape of mitochondria and their cristae are irregular. The Golgi complex is often or less arranged around 10 in a group, situated near the nucleus. Typical coucentric body from RER or SER has not been observed. Annulate lamellae are easily found. A peculiar structure, primordial rough-surfaced endoplasmic reticulum (PRER) is seen.Juxta-nuclear acidophilic area consists of a group of Goigi complex or Golgi complex and mitochondria.
5. Exploring active compounds of Jinhua Qinggan Granules for prevention of COVID-19 based on network pharmacology and molecular docking
Chinese Traditional and Herbal Drugs 2020;51(7):1685-1693
Objective: To explore the effective chemical constituents of Jinhua Qinggan Granules for treatment of coronavirus disease 2019 (COVID-19). Methods: The compounds and action targets of eleven herbal medicines in Jinhua Qinggan Granules were collected via TCMSP. The genes corresponding to the targets were queried by the UniProt database, then the “herbal medicine-compound-target” network was established by Cytoscape software. The gene ontology (GO) function enrichment analysis and KEGG pathway enrichment analysis were performed by DAVID to predict their mechanism. Molecular docking was used to analyze the binding force of the core effective compounds in the “herbal medicine-compound-target” network with SARS-CoV-2 3CL hydrolase and angiotensin converting enzyme II (ACE2). Results: The “herbal medicine-compound-target” network contained 154 compounds and 276 targets, and the key targets involved PTGS2, HSP90AB1, HSP90AA1, PTGS1, NCOA2, etc. GO function enrichment analysis revealed 278 items, including ATP binding, transcription factor activation and regulation of apoptosis process, etc. KEGG pathway enrichment screened 127 signaling pathways, including TNF, PI3K/Akt and HIF-1 signaling pathways related to lung injury protection. The results of molecular docking showed that formononetin, stigmasterol, beta-sitosterol, anhydroicaritin and other key compounds have a certain degree of affinity with SARS-CoV-2 3CL hydrolase and ACE2. Conclusion: The effective compounds in Jinhua Qinggan Granules regulate multiple signaling pathways via binding ACE2 and acting on targets such as PTGS2, HSP90AB1, HSP90AA1, PTGS1, NCOA2 for the prevention of COVID-19.

The aim of this study was to clone the goat RPL29 gene and analyze its effect on lipogenesis in intramuscular adipocytes. Using Jianzhou big-eared goats as the object, the goat RPL29 gene was cloned by reverse transcription-polymerase chain reaction (RT-PCR), the gene structure and expressed protein sequence were analyzed by bioinformatics, and the mRNA expression levels of RPL29 in various tissues and different differentiation stages of intramuscular adipocytes of goats were detected by quantitative real-time PCR (qRT-PCR). The RPL29 overexpression vector pEGFP-N1-RPL29 constructed by gene recombination was used to transfect into goat intramuscular preadipocytes and induce differentiation. Subsequently, the effect of overexpression of RPL29 on fat droplet accumulation was revealed morphologically by oil red O and Bodipy staining, and changes in the expression levels of genes related to lipid metabolism were detected by qRT-PCR. The results showed that the length of the goat RPL29 was 507 bp, including a coding sequence (CDS) region of 471 bp which encodes 156 amino acid residues. It is a positively charged and stable hydrophilic protein mainly distributed in the nucleus of cells. Tissue expression profiling showed that the expression level of this gene was much higher in subcutaneous adipose tissue and inter-abdominal adipose tissue of goats than in other tissues (P < 0.05). The temporal expression profile showed that the gene was expressed at the highest level at 84 h of differentiation in goat intramuscular adipocytes, which was highly significantly higher than that in the undifferentiated period (P < 0.01). Overexpression of RPL29 promoted lipid accumulation in intramuscular adipocytes, and the optical density values of oil red O staining were significantly increased (P < 0.05). In addition, overexpression of RPL29 was followed by a highly significant increase in ATGL and ACC gene expression (P < 0.01) and a significant increase in FASN gene expression (P < 0.05). In conclusion, the goat RPL29 may promote intra-muscular adipocyte deposition in goats by up-regulating FASN, ACC and ATGL.

ObjectiveHenoch-Schönlein purpura(HSP) is one of the dominant diseases in Mongolian medicine. Qishun Baolier(QSBLE), as the main prescription for the treatment of HSP, has significant clinical effect, but its mechanism is not yet clear. Baed on this, this study is intended to screen the differentially expressed proteins before and after treatment, and preliminarily explore the molecular mechanism of QSBLE in the treatment of HSP. MethodTaking oneself as the control, 30 HSP patients aged 6-45 years were collected, and QSBLE was taken orally at 12:00 and 24:00, respectively. The dose was adjusted according to age and the course of treatment was one week. The distribution of proteinuria, hematuria and skin purpura of all patients were determined before and after treatment. The serum samples of 10 patients with clinically significant remission after QSBLE treatment were randomly selected for proteomics. Isobaric tags for relative and absolute quantification(iTRAQ) combined with liquid chromatography tandem mass spectrometry(LC-MS/MS) was used to analyze the proteins in serum of HSP patients before and after treatment, and differential proteins were analyzed bioinformatically and the protein-protein interaction(PPI) networks were constructed. ResultA total of 378 proteins were identified from serum, including 18 differentially expressed proteins, of which 15 proteins were up-regulated and 3 proteins were down regulated. Bioinformatics showed that the differential proteins were mainly involved in biological processes such as immune response, immunoglobulin production, phagocytosis, adaptive immune response before and after treatment. Biological processes, pathways and proteins were used to construct the PPI network, the proteins represented by immunoglobulin heavy constant γ1(IGHG1), immunoglobulin λ-chain 7-43(IGLV7-43), gelsolin(GSN) and 60 kDa heat shock protein(HSPD1) were involved in biological processes and related pathways such as adaptive immune response, immunoglobulin production, leukocyte-mediated immunity, regulation of stress response, regulation of immune system processes, regulation of trauma response, and these proteins were at the center of the PPI network. ConclusionQSBLE may play a role in the treatment of HSP by regulating the expression of IGHG1, IGLV7-43, GSN, HSPD1 and other key proteins to affect immune-related biological processes.

This study aimed to investigate the effect of Tibetan medicine Ershiwuwei Songshi Pills(ESP) on the intestinal flora of non-alcoholic steatohepatitis(NASH) mice. Forty-eight male C57 BL/6 mice were randomly divided into the control group, model(methionine-choline-deficient, MCD) group, high-(0.8 g·kg~(-1)), medium-(0.4 g·kg~(-1)), and low-dose(0.2 g·kg~(-1)) ESP groups, and pioglitazone(PGZ, 10 mg·kg~(-1)) group, with eight mice in each group. Mice in the control group were fed with normal diet, while those in the remaining five groups with MCD diet for five weeks for inducing NASH. During modeling, they were gavaged with the corresponding drugs. The changes in body mass, daily water intake, and daily food intake were recorded. At the end of the experiment, the liver tissues were collected and stained with hematoxylin-eosin(HE) for observing the pathological changes, followed by oil red O staining for observing fat accumulation in the liver. The levels of serum aspartate aminotransferase(AST) and alanine aminotransferase(ALT) and triglyceride(TG) in liver tissue were measured. The changes in intestinal flora of mice were determined using 16 S rRNA high-throughput sequencing technology. The results showed that compared with the model group, the high-, medium-and low-dose ESP groups and the PGZ group exhibited significantly lowered AST and ALT in serum and TG in liver tissues and alleviated hepatocellular steatosis and fat accumulation in the liver. As demonstrated by 16 S rRNA sequencing, the abundance index and diversity of intestinal flora decreased in the model group, while those increased in the ESP groups. Besides, the Firmicutes to Bacteroidetes ratio decreased at the phylum level. In the alteration of the composition of intestinal flora, ESP reduced the abundance of Erysipelotrichia and Faecalibaculum but increased the abundance of Desulfovibrionaceae, Rikenellaceae, Lachnospiraceae, and Ruminococcaceae. This study has revealed that ESP has a protective effect against NASH induced by MCD diet, which may be related to its regulation of the changes in intestinal flora, alteration of the composition of intestinal flora, and inhibition of the intestinal dysbiosis.

The present study investigated the mechanism of the Tibetan medicine Ershiwuwei Songshi Pills(ESP) against the liver injury induced by acetaminophen(APAP) in mice based on the kelch-like ECH-associated protein 1(Keap1)/nuclear transcription factor E2 related factor 2(Nrf2) and Toll-like receptor 4(TLR4)/nuclear factor-kappa B(NF-κB) p65 signaling pathways. Kunming mice were randomly divided into a blank control group, a model group, an N-acetyl-L-cysteine(NAC) group, and high-(400 mg·kg~(-1)), medium-(200 mg·kg~(-1)), and low-dose(100 mg·kg~(-1)) ESP groups. After 14 days of continuous administration, except for those in the control group, the mice were intraperitoneally injected with 200 mg·kg~(-1) APAP. After 12 h, the serum and liver tissues of mice were collected. Hematoxylin-eosin(HE) staining was performed on pathological sections of the liver, and the levels of aspartate aminotransferase(AST) and alanine aminotransferase(ALT) in the serum and the levels of glutathione(GSH), malondialdehyde(MDA), superoxide dismutase(SOD), catalase(CAT), myeloperoxidase(MPO), and total antioxidant capacity(T-AOC) in liver tissue homogenate were detected to observe and analyze the protective effect of ESP on APAP-induced liver injury in mice. The serum levels of tumor necrosis factor-alpha(TNF-α), interleukin-1 beta(IL-1β), and interleukin-6(IL-6) were determined by enzyme-linked immunosorbent assay(ELISA). The protein expression of Nrf2, Keap1, TLR4, and NF-κB p65 in the liver was determined by Western blot. Quantitative real-time was used to determine the mRNA expression of glutamate-cysteine ligase catalytic subunit(GCLC), glutamate-cysteine ligase regulatory subunit(GCLM), heme oxygenase-1(HO-1), and NAD(P)H dehydrogenase quinone 1(NQO-1) in the liver to explore the mechanism of ESP in improving APAP-induced liver damage in mice. As revealed by results, compared with the model group, the ESP groups showed improved liver pathological damage, decreased ALT and AST levels in the serum and MDA and MPO content in the liver, increased GSH, SOD, CAT, and T-AOC in the liver, reduced TNF-α and IL-6 levels in the serum, down-regulated expression of Keap1 in the liver cytoplasm and NF-κB p65 in the liver nucleus, up-regulated expression of Nrf2 in the liver nucleus, insignificant change in TLR4 expression, and elevated relative mRNA expression levels of antioxidant genes GCLC, GCLM, HO-1, and NQO-1. ESP can reduce the oxidative damage and inflammation caused by APAP, and the mechanism may be related to the Keap1/Nrf2 signaling pathway and the signal transduction factors on the TLR4/NF-κB p65 pathway.

The present study investigated the mechanism of the Tibetan patent medicine Ershiwuwei Shanhu Pills(ESP) in alleviating Alzheimer's disease in mice via Akt/mTOR/GSK-3β signaling pathway. BALB/c mice were randomly assigned into a blank control group, a model group, low(200 mg·kg~(-1)), medium(400 mg·kg~(-1)) and high(800 mg·kg~(-1)) dose groups of ESP, and donepezil hydrochloride group. Except the blank control group, the other groups were given 20 mg·kg~(-1) aluminum chloride by gavage and 120 mg·kg~(-1) D-galactose by intraperitoneal injection for 56 days to establish Alzheimer's disease model. Morris water maze was used to detect the learning and memory ability of mice. The level of p-tau protein in mouse hippocampus and the levels of superoxide dismutase(SOD), malondialdehyde(MDA), catalase(CAT), and total antioxidant capacity(T-AOC) in hippocampus and serum were detected. Hematoxylin-eosin staining and Nissl staining were performed for the pathological observation of whole brain in mice. TdT-mediated dUTP nick-end labeling(TUNEL) staining was employed for the observation of apoptosis in mouse cortex. Western blot was adopted to detect the protein levels of p-mTOR, p-Akt, and GSK-3β in the hippocampus. Compared with the model group, the ESP groups showcased alleviated pathological damage of the whole brain, decreased TUNEL positive cells, reduced level of p-tau protein in hippocampus, and risen SOD, CAT, and T-AOC levels and declined MDA level in hippocampus and serum. Furthermore, the ESP groups had up-regulated protein levels of p-mTOR and p-Akt while down-regulated protein level of GSK-3β in hippocampus. Therefore, ESP can alleviate the learning and memory decline and oxidative damage in mice with Alzheimer's disease induced by D-galactose combined with aluminum chloride, which may be related to Akt/mTOR/GSK-3β signaling pathway.
