1.Study on the effect of neutrophil and interleukin-8 on exercise-induced asthma.

Objective To explore the relationship of neutrophil and interleukin-8(IL-8)with exercise-induced asthma(EIA).Methods From May 15,2001 to Dec 19.2005 the levels of peripheral neutrophil superoxide anion and IL-8 in the blood and the induced sputum from patients with EIA,non EIA and normal control subjects were measured respectively before and after exercises,meanwhile a comparison was made among them.Results Before exercise:the level of neutrophil superoxide anion was slightly higher in EIA group and non EIA group than that in control group;however,the differences were insignificant;the IL-8 and MDA levels in blood and induced sputum were higher in EIA group and non EIA group than those in control group,but there was no significant difference between EIA group and non EIA group;however,they were significantly different between EIA group and control group,non EIA group and control group respectively( P 0.05).Conclusion Oxyradical produced by neutrophil and IL-8 has significant effect on the production of EIA.
2.Influence of Jianpizhidong decoction on the behavioral record an bilateral striatal amino acid neurotransmitters in mice with Tourette syndrome disease

Objective To study the influences of Jianpizhidong decoction(JPZDD) on bilateral striatal amino acid neurotransmitters in mice with Tourette Syndrome (TS) disease and discuss the mechanism of JPZDD.Methods Thirty mice were injected 3,3-iminodipropionitrile (IDPN) with 350 mg· kg-1 once a day for 7 consecutive days except ten mice of the blank control group which were injected with saline.TS mice were randomly divided into three groups:TS model group,JPZDD group (the Chinese herbal group),Tiapride group (the western medicine group).The TS model group and the blank control group were gavaged with saline(0.9%)at 20 ml ·Kg-1,the Tiapride group with Tiapride at 50 mg · kg-1,and JPZDD group with JPZDD at 20 g · kg-1,respectively,once a day for six weeks.Then behavioral recordings were conducted,and the content of glumatic acid (Glu) and γ-aminobutyric acid (GABA) in the bilateral striatum of the mice were detected by using HPLC.Results Behavioral recording indicated that there were significant differences in the Chinese herbal group and the Tiapride group before(1.90 ± 1.80,2.05 ± 1.40) and after(1.15 ± 1.67,1.00 ± 1.08) treatment (P < 0.05).The content of Glu and GABA in TS model group (1858.33 ± 203.00,261.24 ± 47.31) and Tiapride group (1744.53 ± 234.01,258.57 ± 57.64) were higher compared with the blank control group (P < 0.05),and the content of Glu in JPZDD was significantly higher than that in TS model group.There was no statistical difference between other groups(P>0.05).The ratio of Glu and GABA were lower in the JPZDD,the Tiapride and the model groups than that in the blank control group,but there were no statistical differences between these groups (P> 0.05).Conclusion JPZDD can regulate the content of Glu and GABA in the striatum,and keep the dynamic equilibrium,which might be its pharmacodynamic mechanisms for treating TS.
3.Application of comprehensive evaluation of quality in the nursing management of cardiovascular intensive care unit

Objective To discuss the nursing management method in the cardiovascular intensive care unit. Methods The management evaluation standard was set up and the comprehensive evaluation of quality (CEQ) was introduced according to factors influencing nursing quality. Results The nursing quality control data and complications after operation before and after the application of CEQ were statistically different. Conclusions Nursing management should innovate unceasingly and comprehensive quality evaluation is one of the effective nursing management method.
4.Effect of ribosomal protein L34 gene knockdown on a human cutaneous squamous cell carcinoma cell line SCL-1

Objective:To evaluate the effect of ribosomal protein L34 (RPL34) gene knockdown on the proliferation and apoptosis of human cutaneous squamous cell carcinoma (cSCC) cells.Methods:From January 2016 to January 2017, 14 paraffin-embedded skin samples of cSCC and 16 paraffin-embedded normal skin tissue samples were collected from Department of Dermatology and Venereology, the Affiliated Hospital of Inner Mongolia Medical University, and RPL34 expression in the skin tissues was analyzed by immunohistochemical study. A lentivirus vector containing short hairpin RNA targeting RPL34 gene was constructed and used to transfect a human cSCC cell line SCL-1 (shRNA group) , SCL-1 cells transfected with an empty lentivirus vector served as control group, and the knockdown efficiency was verified by real-time quantitative PCR (RT-PCR) and Western blot analysis. At 72 hours after the transfection, flow cytometry was performed to analyze the cell cycle and detect apoptosis of SCL-1 cells, and methyl thiazolyl tetrazolium (MTT) assay to evaluate the cellular proliferative activity of SCL-1 cells. Comparisons between 2 groups were performed by using t test or rank sum test. Results:Immunohistochemical study showed that the cytoplasmic expression score of RPL34 was significantly higher in the cSCC tissues (2.143±1.956) than in the normal control tissues (0.500±0.516, z=3.53, P< 0.05) . RT-PCR showed that the relative mRNA expression of RPL34 in the SCL-1 cells was significantly lower in the shRNA group (0.149±0.016) than in the control group (1±0.018, t=36.95, P< 0.05) ; Western blot analysis revealed that the relative protein expression of RPL34 in the SCL-1 cells was significantly lower in the shRNA group than in the control group. Compared with the control group, the shRNA group showed a significantly increased proportion of S-phase cells ( t=13.76, P< 0.05) , but a significantly decreased proportion of G1-phase cells ( t=36.62, P< 0.05) ; the apoptosis rate was significantly higher in the shRNA group (9.42%±0.16%) than in the control group (4.58%±0.41%, t=19.02, P< 0.05) . MTT assay showed that the cell viability was significantly decreased in the shRNA group (0.815±0.005) than in the control group (1.886±0.005, t=265.91, P< 0.05) after additional 120-hour culture. Conclusion:The RPL34 gene was overexpressed in the cSCC tissues, and knockdown of the RPL34 gene in SCL-1 cells could interfere with cell cycle, decrease their proliferative activity, and promote their apoptosis.
5.Data sharing and privacy protection of colorectal cancer from the perspective of medical ethics

In the process of colorectal cancer(CRC)data sharing in the medical field,it is necessary to protect the privacy of patients and avoid data information leakage based on the medical ethics requirements,to achieve a balanced development of data sharing and privacy protection.This paper introduced the development status of CRC data sharing and analyzed its ethical issues,mainly including privacy protection,rights ownership,and interest imbalance.It is suggested that the ethical issues of CRC data sharing can be solved by following ethical principles,establishing ethics committees,and creating a safe platform.The aim is to provide a reference for relevant people and obtain good results of data sharing and privacy protection of CRC based on medical ethics.
6.Changes of C-reactive protein level and inflammatory factors in patients with obstructive sleep apnea syndrome

Objective To observe and analyze the changes of C-reactive protein and inflam-matory factors in patients with obstructive sleep apnea syndrome.Methods 205 patients with ob-structive sleep apnea syndrome were divided into observation group Ⅰ (n =72),group Ⅱ (n =68) and observation group Ⅲ (n =65)according to apnea-hypopnea index (AHI),and the other 50 pa-tients were designed as control group.Levels of C-reactive protein,interleukin-6 and interleukin-18 were detected.Results The sicker,apnea-hypopnea index gradually increased (AHI),oxygen sat-uration decreased,C-reactive protein,interleukin-6 and interleukin-18 levels increased gradually. Logistic regression analysis showed that C-reactive protein,interleukin-6 and interleukin-18 and AHI,oxygen saturation were positively correlated with AHI and negatively correlated with oxygen saturation.Conclusion C-reactive protein level,interleukin-6 and interleukin-18 and other related inflammatory factors are the important indicators for prognosis judgment of patients with obstructive sleep apnea syndrome,so it is worthy of clinical application.

8.Localization and validation of Abdominal Surgery Impact Scale-Revised

Objective? To introduce Abdominal Surgery Impact Scale-Revised (ASIS-R) and to verify its reliability and validity in acute pancreatitis patients. Methods? ASIS-R were translated into Chinese by 2 nursing postgraduates who passed College English Test-6 (CET-6) in accordance with Brislin's translation principle. The Chinese version of ASIS-R were then translated back into English by two nursing specialists who graduated abroad and were proficient both in English and Chinese. The scale was culturally adjusted through pre-survey. Totally 182 acute pancreatitis patients attending the Pancreas Center of a Class Ⅲ Grade A hospital in Jiangsu Province from October 2017 to April 2018 were selected by convenient sampling, and a total of 173 valid questionnaires were included to analyze the scale's reliability and validity in acute pancreatitis patients. Results? The Chinese version of ASIS-R had 14 items in total, including limitation of motion, function impairment, visceral function, sleep and psychological function. Its Cronbach's α coefficient was 0.719, and that of different dimensions was 0.966, 0.964, 0.972, 0.976 and 0.968, respectively. The retest reliability was 0.660. Evaluated by the committee of experts, the content validity of various items in the Chinese version of ASIS-R (S-CVI/Ave) averaged 0.95, and their content validity index (I-CVI) ranged 0.80-1.00. Totally 5 common factors were extracted by exploratory factor analysis, which could account for 94.93% of the total variance. The factor loading of various items on their common factors were>0.4. Conclusions? The Chinese version of ASIS-R has sound reliability and validity, which may serve as a tool for acute pancreatitis patients to evaluate the short-term quality of life in hospital by themselves.
9.Norcantharidin induces apoptosis through autophagosome accumulation in breast cancer MDA-MB-231 cells

The aim of this study was to investigate the effect of norcantharidin (NCTD) on the proliferation and apoptosis of triple-negative breast cancer cell line MDA-MB-231.Western blot was used to detect the effect of NCTD on the expression levels of apoptosis-related proteins Bax/Bcl-2, cleaved-PARP/PARP/PARP, cleved-caspase-9, cleaved-caspase-3 and MCL-1 in MDA-MB-231 cells.Also, the expression levels of autophagy-related proteins LC3-II/LC3-I, Parkin and PINK1 in MDA-MB-231 cells were measured by Western blot.Flow cytometry was used to measure the effect of NCTD on the changes of mitochondrial membrane potential and mitochondrial reactive oxygen species (ROS).The effect of NCTD on autophagy flow in cells expressing mCherry-EGFP-LC3 was detected by a confocal microscope.Moreover, the effects of NCTD combined with chloroquine (CQ) or 3-methyladenine (3-MA) on the apoptosis of MDA-MB-231 cells were detected by flow cytometry.The results showed that NCTD significantly increased the expression levels of Bax/Bcl-2, cleaved-PARP/PARP, cleaved-caspase-9, cleasved-caspase-3 and LC3-II/LC3-I proteins, and promoted the mitochondrial translocation of Parkin, and blocked the autophagic flow in MDA-MB-231 cells. Moreover, NCTD combined with CQ accelerated apoptosis, while NCTD combined with 3-MA decreased apoptosis.These results suggest that NCTD can induce autophagy accumulation and lead to apoptosis of MDA-MB-231 cells.
10.Study on the relationship between intestinal flora analysis and CD4 +T lymphocyte subsets in patients with systemic lupus erythematosus

Objective:To explore the characteristics of intestinal microbiota in patients with systemic lupus erythematosus (SLE), and further explore the relationship between microbiota and CD4 +T lymphocyte subsets and disease activity. Methods:Fecal samples were collected from 96 patients with SLE, and 96 sex- and age-matched healthy controls (HCs). The gut microbiota were investigated via 16s rRNA sequencing. Flow cytometry was used to detect peripheral CD4 +T lymphocyte subsets of Th1, Th2, Th17 and Treg cells. Indicators of disease activity such as erythrocyte sedimentation rate (ESR), C-reactive protein (CRP), complement C3 and C4, Systemic lupus erythematosus disease activity index(SLEDAI) for each patient were recorded. Differential abundance analysis was carried out using the edgeR algorithm. The Wilcoxon rank-sum test was used to compare alpha diversity indices, bacterial abundances, and the F/B ratio between groups. R (version 4.0.1) was used for comparative statistics, and Pearson′s correlation analysis was used to assess the correlations between the relative abundances of bacterial genera and serum levels of ESR, CRP, C3 and C4 in the samples. Results:The alpha estimators of richness (ACE and Chao 1) were significantly reduced in SLE feces samples compared with those of HCs ( P<0.01). Bacterial diversity estimators, including the Shannon ( P<0.01) and Simpson′s ( P<0.01) indices, were also significantly lower in SLE. Significant differences in gut microbiota composition between SLE and HCs were found using the edgeR algorithm. Compared with HC, 24 species of bacteria were significantly different in SLE patients at the genus level ( P<0.05). Moreover, there was a significant positive correlation between CRP and Coprococcus ( r=0.30, P=0.014), C4 and Corynebacterium ( r=0.31, P=0.013) and Faecalibacterium( r=0.25, P=0.048), Hemoglobin and Morganella( r=0.41, P=0.001), as well as SLIDA and Corynebacterium( r=0.25, P=0.047). In terms of lymphocyte subsets, there was significant positive correlation between B cells, Treg cells and Eubacterium eligens group, as well as CD8 +T, CD4 +T, NK cells and Corynebacterium. In additional, Th1 was positively correlated with Shigella Escherichia coli ( r=0.52, P=0.008), and Th2 was positively correlated with Dielma ( r=0.51, P<0.001). Conclusion:The abundance and diversity of intestinal flora in SLE patients were significantly reduced, and the differentially expressed bacteria were closely related to the CD4 +T lymphocyte subsets and disease activity indicators of patients.
