2.Application of Perphenazine to Treat Psychotic Symptoms of Senile Dementia:31 Case Report

Objective To observe the effect of small dosage of perphenazine(2~6 mg)on psychotic symptoms of senile dementia.Methods 31 senile cases with psychotic symptoms after dementia(Alzheimer disease or vascular dementia)accepted small dosage of perphenazine in addition of the conventional therapy.They were assessed with Sadoz Clinical Assessment Geriatric(SCAG)and Treatment Emergent Symptom Scale(TESS).Results The score decreased 50%~75%(markedly improved)in 2 cases,25%~50%(improved)in 22 cases,lower than 25%(no improved)in 7 cases.Some mild emergent symptoms have been observed.Conclusion Small dosage of perphenazine is safe and effective in treating psychotic symptoms of senile dementia.
3.Relationship between aldose reductase gene expression in peripheral blood mononuclear cells and the risk of diabetic nephropathy in type 2 diabetic patients

Aldose reductase (AR) mRNA level in peripheral blood mononuclear cells was assayed by RT-PCR. There was a positive correlation between PBMC AR mRNA level and urinary albumin excretion rate in type 2 diabetic patients. The result suggests that AR activity is related to the pathogenesis of diabetic nephropathy.
4.Methamphetamine neurotoxicity and changes of nitration in rats striatum

Aim To investigate the neurotoxicity and the changes of nitration in the striatum of rats treated with methamphetamine(MA).Methods The rats were randomly divided into MA group and control group.The stereotyped behavior and body temperature of the rats were recorded.The differences of nitric oxide, nitric oxide synthase(NOS)and nitroprotein were compared between the two groups.Results The score of stereotyped behavior and the body temperature of the rats in MA group were significantly higher than those in the control group(P <0.01);Compared with the control group, the content of NO and the activity of NOS of striatum in MA group were significantly increased(P <0.01), and nitration of protein was also upregulated(P <0.01).Conclusion The increases of NOS activity, NO level and nitroprotein may play an important role in MA-induced neurutoxity in rat striaturn.
5.Astragalus polysaccharide induces apoptosis of human KG-1a cells through inhibiting PTEN-PI3K-Akt signaling pathway

Objective To investigate the effect of astragalus polysaccharide (AP) on proliferation and apoptosis of human acute myelocytic leukemia cell line KG-1a and the underlying mechanisms.Methods Human acute myelocytic leukemia cell line KG-la was cultured under 37 ℃ and 5 % C02,when appropriate cell passage and cryopreservation were performed.After treatment for 48 h with different concentrations of AP,the proliferative activity and apoptosis rate of KG-1a cells were examined by CCK-8 assay and flow cytometry,respectively.The expressions of p-Akt and bcl-2 protein in AP-treated KG-1a cells were evaluated by Western blot.The expression of PTEN mRNA by quantified real-time PCR.Results The proliferative activity of KG-la cell was obviously suppressed by AP treatment,and the inhibition rate increased in a dosedependent manner.The flow cytometry showed that,compared with the control group,the apoptosis rates of KG-1a cells were significantly increased after treatment with AP for 48 h.The early apoptosis rates were (1.98±0.16) %,(12.60±0.48) %,(16.31±0.73) %,the late apoptotic rates were (3.11±0.19) %,(17.17±1.40) %,(21.17 ± 0.81)%,the differences were statistically significant (P < 0.05).Western blot showed that the expressions of p-Akt and bcl-2 protein in KG-1a cells decreased significantly after treatment with AP (P < 0.05).In contrast,the mRNA level of PTEN increased (P <0.05),which was shown by real-time PCR.Conclusion AP could repress cellular proliferation activity of KG-1a cells,which could be attributed to inhibition of PTEN-PI3K-Akt signaling pathway.
6.Diffusing effect of DTT and 2-ME during SDS-PAGE electrophoresis

Objective: Dithiothreitol(DTT) and ?-Mercaptoethanol(2-ME) are important reducing agents for SDS-PAGE.This study is to observe the diffusing effect of DTT and 2-ME during electrophoresis,and to find a way of avoiding this effect.Methods: We placed protein samples containing reducing agents and non-reduced protein samples separately in the sample wells at intervals for SDS-PAGE electrophoresis,and determined whether or not the electrophoretic lanes of the non-reduced samples were interfered by the adjacent lanes.Results: DTT and 2-ME diffused to the neighboring lane,so that the non-reduced samples were reduced partially.The spreading effect was positively correlated with the content of the reducing agent.Conclusion: DTT and 2-ME have a diffusing effect during SDS-PAGE electrophoresis.In separating the reduced and non-reduced proteins in the same gel at the same time,at least a blank lane should be set up in between them in order to avoid the diffusing effect.
7.Synthesis, characterization and blood compatibility studies of biomedical aliphatic polyurethanes.

The one-step method was adopted in this study to synthesize aliphatic polyurethane with 4,4-methylene dicyclohexyl diisocyanate(HMDI), 1,4-butanediol (BDO) and poly (tetrahydrofuran) (PTMG). The tests conducted on this material were: FIR spectrum, mechanical properties test, water contact angles test, hemolysis test and platelet adhesion test. Results showed that this material has a good tensile strength up to 30 Mpa, similar to aromaphatic polyurethane. But its tensile elongation, tensile permanent change, hydrophility are better than those of aromaphatic polyurethane. The hemolysis test and platelet adhesion test showed that it has good blood compatibility.

8.Synthesis, characterization and blood compatibility studies of waterproof breathable polyurethanes.

Adopting the two-step method and changing the proportion between PEG (Polyethylene glycol) and PTMG (poly (tetrahydrofuran), we used the MDI (4,4'-diphenylmethane diisocyanate) and short chain extender BDO (1,4-butanediol) as hard segment, the PTMG and PEG as soft segment, and hence prepared a series of polyether-based thermoplastic polyurethanes. FTIR showed the structure character of these polyurethanes. The determination of mechanics property and water contact angles revealed their good mechanics properties and hydrophilicity. Blood compatibility was evaluated by hemolysis test and platelet adhesion test, which revealed their good hemocompatibility. So those polyurethanes may be of wide application in the future.

9.Diagnosis and treatment of traumatic carotid cavernous fistula.

OBJECTIVETo discuss the diagnosis and management of traumatic carotid cavernous fistula (TCCF).
METHODSIn all 15 patients with TCCF confirmed by angiography, 8 patients got early diagnosis and cure. With Seldinger technique adopted in the puncture of femoral artery, Magic 3 F-1.8 F BD catheters combining with balloon were used to embolize the fistula or the internal carotid artery.
RESULTSEarly diagnosis and cure were achieved in 8 patients within one week and no sequelae occurred. Seven patients with delayed diagnosis who were cured beyond one week had some sequelae such as hypopsia in 5 cases, incomplete oculomotor paralyses in 3 and incomplete abducent paralyses in 2. Among all the 15 cases, the internal carotid artery was preserved in 12 cases accounting for 80%. Occluding the fistula with sacrifice of the internal carotid artery was performed in 3 cases and no repatency of the fistula occurred by following up beyond three months.
CONCLUSIONSThe preferred therapy for TCCF is to occlude the fistula using detachable balloon. The diagnosis and treatment for TCCF can significantly reduce occurrence rate of the complications and sequelae.

10.A comparative study on the difference coefficients of DRG payment in various cities in Zhejiang province

In order to compare the setting of difference coefficients in DRG point payment in different cities in Zhejiang province, the implementation rules of DRG point payment issued by 11 cities in Zhejiang province were comprehensively analyzed. It was found that the difference coefficients in different cities could be divided into three categories, including hospital coefficients alone, hospital coefficients and grade coefficients weighted, and weighted by hospital coefficients, grade coefficients, personal burden levels, case mix indexes, and head-to-time ratio. Its setting differences included four aspects: connotation composition, weight distribution, threshold value, and classification of medical institutions. The authors suggested that the adjustment cycle should be set scientifically to dynamically adjust the difference coefficient, and the scientific setting of the difference coefficient should be promoted through provincial coordination.
