1.Resistance reverse effect of anemoside B4 and tetrandrine on oxaliplatin-resistant human colon cancer cells and the mechanism

Background and purpose:Oxaliplatin (L-OHP) is one of the most commonly used chemotherapy drugs in colorectal cancer and L-OHP-resistance is very common in colorectal cancer therapy. This research was to discuss the reversal effect in L-OHP-resistant human colon cancer cell line LoVo/L-OHP by anemoside B4 (AB4) and tetrandrine (Tet) and to clarify their molecular mechanism. Methods:LoVo/L-OHP cells were treated for 48 h by AB4 and Tet at different concentrations to get non-toxic dose. Drug sensitivity was measured by MTT. After the treatment, the cell cycle and apoptosis of the cells were detected. Expression of P-gp mRNA, zDHHC9 mRNA and SMAD4 mRNA were detected by RT-PCR. Expression of P-gp, zDHHC9 and SMAD4 protein were detected by Western blot. Results:The IC50 of LoVo/L-OHP for L-OHP was (112.5±23.6) μg/mL, and the IC50 decreased to (62.8±21.4) μg/mL (P<0.05) and (58.9±26.3) μg/mL (P<0.05) after the treatment with 0.71 μg/mL AB4 and 0.45 μg/mL Tet (non-toxic dose) separately. The cell cycle experiment showed that the cells in G1 stage decreased and in S stage increased but with no statistical signiifcance (P>0.05). The cell apoptosis experiment showed that the apoptotic rate increased after the treatment with AB4 and Tet with non-toxic dose but with no statistical signiifcance (P>0.05). The RT-PCR experiment showed that the expressions of P-gp mRNA and zDHHC9 mRNA were increased and SMAD4 mRNA was decreased in LoVo/L-OHP cells compared with in LoVo cells (P<0.05), while it was found that P-gp mRNA in LoVo/L-OHP cells was decreased after the treatment with AB4 at non-toxic dose (P<0.05). Western blot experiment showed the protein of P-gp in LoVo/L-OHP cells was decreased after treatment with AB4 (P<0.05), which was accordant to the PCR result. The expression of zDHHC9 protein in LoVo/L-OHP cells was decreased in LoVo/L-OHP cells after treatment with Tet (P<0.05). Conclusion:AB4 and Tet have some reversal effect on resistant to L-OHP in LoVo/L-OHP cells. The molec-ular mechanism of the resistance reverse effect was related to down-regulation of P-gp for AB4 and down-regulation of zDHHC9 for Tet.
2.Analysis of eleven cases of sentinel lymph node biopsy in male breast cancer

Objective To evaluate the performance and feasibility of sentinel node biopsy(SNB) in male breast cancer patients using Methylthioninium Chloride Injection.Methods At the First Affiliated Hospital of Zhengzhou University,there are 11 patients in group from March 2010 to December 2014.The clinical stage was cT1-T2N0M0.All patients using Methylthioninium Chloride Injection as the tracer.11 patients are given with sentinel lymph node biopsy,while given the axillary lymph node dissection.Results In 11 cases of male breast cancer patients,10 cases obtained the sentinel lymph nodes,the detection rate was 90.9% (10/11).The sentinel lymph node is in 1-3,the average is 1.7 gold.Non sentinel lymph nodes are in 8-14,the average is 10.5 gold.The coincidence rate is 90.0% The sensitivity is 100% and The precision is 60%.Conclusions Sentinel lymph node biopsy can accurately predict the metastasis of axillary lymph node of breast cancer in male patients.
3.The correlation between the activity of GSP-? and intestinal metaplasia of gastric mucosa

Objective To study the relationship between the activity of GSP ?, intestinal metaplasia(IM) of gastric mucosa, and proliferation typing.Method Immunohistochemical method was used to detect GSP ? in 167 cases undergoing biopsy or surgery ,among them slices of 112 pathology proved chronic atrophic gastritis(CAG) were counter stained by mucohistochemistry.Results[WT5”BZ] The result showed that the positive rates of GSP ? in gastric adenocarcinoma, CAG with IM or gastric dysplasia were significantly higher than in chronic superficial gastritis (CSG).The positive rate of GSP ? in high proliferation typing (HPT) was significantly higher than in middle or low proliferation typing ( MPT or LPT).Conclusion GSP ? activity could be used as a marker of precancerous lesions and HPT highly correlates to the occurence of gastric cancer.
4.Epstein-Barr virus and lymphoma

More and more studies have found that EBV infection is closely related to the occurrence, development, treatment and prognosis of lymphoma. The treatment of EBV-positive lymphoma bases mainly on combined chemo-radiotherapy together with ganciclovir, acyclovir and other antiviral drugs. Also there are novel ways to treat EBV positive- lymphoma including CD70 monoclonal antibody, butyrate, etc. The only way to prevent EBV infection is to inoculate anti-viral vaccine. The criterion of treating EBV positive-lymphoma remains to be further investigated.
5.Prevention and treatment for chemotherapy-induced peripheral neuropathy: a review

Chemotherapy- induced peripheral neuropathy (CIPN) can be caused by many commonly used chemotherapeutic agents, such as taxanes, vinca alkaloids, platinum drugs, thalidomide,and also by newer agents such as bortezomib. Both animal experiments and clinical studies are being conducted to investigate strategies for preventing CIPN or ameliorating established CIPN without affecting the antitumor efficacy of chemotherapy. Treatments using calcium and magnesium infusions,glutathione,lipoid acid are being evaluated for their clinical application.
6.Clinical feasibility of 125Ⅰ combined、with high intensity focused ultrasound(HIFU)therapy for hepatocellular carcinoma

Objective To evaluate efficacy and clinical feasibility of 125Ⅰ combined with high intensity focused ultrasound(HIFU)therapy for hepatocellular carcinoma.Methods.The study included 10 cases of hepatocellular carcinoma(primary hepatic carcinoma,5 cases.Tumor recurrence of hepatocelluhr carcinoma after hepatecomy,5 cases).All the cases were treated with 125I combined with HIFU.Resultsl The operationWas successfully completed in all the cases.No severe post-operative complication occurred.CT acall found no displacement of 125Ⅰ seeds.CT 8can showed that reduced size of tmnor changed in different degree.The l month,3 month,6 month and l year survival cases were 8,7,5 and 2.During 3 to 18 months follow up,2 cases died within 1 month.Two cases survived more than 22 months.Conclusion Using 125Ⅰ combined with HIFU is safe,minimally invasive and effective for treatment of hepatocellular careinom.
7.Construction of three-dimensional finite element model with lateral mass screw fixation following lower cervical three-segment laminectomy

BACKGROUND:With the improvement of cervical posterior surgical techniques, lateral mass screw fixation technology has been widely used in the reconstruction surgery of the cervical spine for stability. However, currently, the finite element study on the lateral mass screw fixation reconstruction of the cervical spine is rare. OBJECTIVE:To establish a fine normal lower cervical spine (C3-C7 ) three-dimensional finite element model and a reconstructed finite element model with three-segment laminectomy with lateral mass screw fixation. Then, to do an initial biomechanical analysis of the lateral mass screw fixation reconstructed lower cervical finite element model. METHODS:We col ected a normal female volunteer aged 30 years old to do CT scan for the whole cervical spine. The Dicom data were obtained. Then, the CT scanning images were dealt with software Mimics 10.01, Geomagic Studio 12.0, Solidworks2012, HyperMesh 10.1 and Abaqus 6.12 software to build the normal lower cervical spine (C 3-C7 ) finite element model, the laminectomy finite element model and the rebuilt finite element model. At last, we analyzed the stress changes of reconstructed models under the state of flexion, extension, lateral bending and rotational motion. RESULTS AND CONCLUSION:The lower cervical spine finite element model contained 503 911 elements and 93 390 nodes with a fine realistic appearance. It successful y passed the validation. The surgical procedure was completed in the software, and the lateral mass screw fixation reconstruction finite element model has been established. Lateral mass screw fixation system provides good stability for the postoperative finite element model. The activity of rebuilt finite element model is much lower than the normal finite element model. In the extension condition, the stress of lateral mass screw fixation system becomes strong.

The phosphoinositide-3 kinase (PI3K)/protein kinase B (PKB/AKT)/mammalian target of rapamycin (mTOR) pathway is asso-ciated with cell growth, proliferation, differentiation, apoptosis and metabolism. The abnormalities of this signal pathway are found in various malignant tumors. This pathway has also been investigated as an anti-tumor target. Recently, novel inhibitors have been stud-ied in clinical trials of lymphoma. This review summarizes the activation status of the PI3K/AKT/mTOR pathway and its use for targeted therapy of lymphoma.
9.The expressions of IL-2, IL-6 and TNF-αin the serum of extranodal NK/T-cell lymphoma (ENKTL) patients and their clinical significances

Background and purpose:Extranodal NK/T-cell lymphoma, nasal type (ENKTL) belongs to a rare type of non-Hodgkin's lymphoma (NHL). Its incidence rate in Asian country is higher than that in Western country. This disease is highly invasive, the pathogenesis of it is still unclear. Resent research shows that epstein-barr virus (EBV) is closely related to the occurrence of it. There is still no standard treatment guidelines of ENKTL, and the prognosis is very bad. Therefore, it is imperative to explore the pathogenesis of ENKTL. This study aimed to investigate the expressions of interleukin-2 (IL-2), interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) in the serum of ENKTL patients and their clinical signiifcances. Methods: Luminex liquid chip technology was used to detect the expression levels of IL-2, IL-6 and TNF-αin the serum of 67 ENKTL patients and 26 normal persons. Results:The expression levels of IL-2, IL-6 and TNF-αin the serum of 67 ENKTL patients were (564.1±387.6), (293.3±191.6) and (181.3±91.8)pg/mL, while in the normal persons were (1 097.0±365.7), (417.5±289.6) and (291.3±89.4)pg/mL, respectively. Compared with normal persons, the expression levels of IL-2, IL-6 and TNF-αin ENKTL patients were signiifcantly lower (P<0.05). Further study showed that the expression level of TNF-αin 5 complete remission ENKTL patients [(162.7±10.3)pg/mL] was significantly higher than that in initial treatment patients [(125.2±7.3)pg/mL, P<0.05]. Conclusion:The expressions of IL-2, IL-6 and TNF-αare reduced in the serum of ENKTL patients, and the serum expression level of TNF-αis closely related to the effect of chemotherapy.
10.Risk of non-medullary thyroid cancer influenced by polymorphic variation in the thyroglobulin gene

Background and purpose: Germline variation in Tg (thyroglobulin) and TSHR (thyroid stimulating hormone receptor) confers an increased risk of benign thyroid disorders. Benign thyroid disorders are strong risk factors for non-medullary thyroid cancer (NMTC). To explore the hypothesis that polymorphic variation in these genes affects the risk of NMTC. Methods: Tg A7589G and TSHR C253A polymorphisms were determined by polymerase chain reaction-restriction fragment length polymorphism (FCR-RFLP) method, to analyze the relationship between the Tg and TSHR gene polymorphisms and NMTC in NMTC and control groups. Results: Among 360 cases, there was no statistic difference in the frequencies of genotype and allele of TSHR C253A between NMTC and control groups. There were Tg A7589G polymorphisms in the 360 cases. The frequencies ofAG+GG genotype in NMTC group were significantly higher than those in control groups (P<0.05). The frequencies of G allele in NMTC group were significantly higher than those in control groups (P<0.001). Conclusion: There were Tg A7589G gene polymorphisms in NMTC and control groups. G allele may be the predisposing gene of NMTC.
