1.Determination of 11 Phthalic Acid Esters in Soil by Accelerated Solvent Extraction-Liquid Chromatography Tandem Mass Spectrometry

A sensitive and convenient method based on accelerated solvent extraction ( ASE )-liquid chromatography tandem mass spectrometry (LC-MS / MS) was established for the simultaneous determination of 11 phthalic acid esters(PAEs) in soil. The optimized conditions were as follows: By using n-hexane as the extraction solvent, spiked sample was extracted by ASE at 160 ℃ for 4 times, 12 min for each time. The extract was concentrated by evaporation. Qualitative and quantitative analysis was carried out by the multiple reaction monitoring mode after the chromatographic separation with atmospheric pressure chemical ionization (APCI), using acetonitrile -0. 1% formic acid water as mobile phase. The limits of detection(LODs) for 11 PAEs were between 0. 03 - 13. 0 μg / kg. The recoveries and relative standard deviations were 72. 8% -101. 8% and 1. 7-6. 7% , respectively. This method is rapid, sensitive and suitable for the determination of PAEs in soil.

Aim Toinvestigatethepro-angiogenic effects of Danshensu derivative ADTM and explore its underlying possible signaling pathway using zebrafish embryosasinvivomodels.Methods Theangiogenesis activities of ADTM were determined in experimental models of normal and VEGFR tyrosine kinase inhibitorⅡ(VRI )-induced vascular defective zebrafish embry-os.Embryos were treated with various concentrations (50,100,200 μmol · L-1 ) of ADTM for indicated time.The diameter and the numbers of endothelial cells of zebrafish SIVs were evaluated,respectively.In VRI model,the number of intact and defective ISVs in each zebrafish embryo was counted.The total RNA of zebrafish embryos was extracted and transcriptional profiling was analyzed by deep sequencing.Quantita-tive real-time PCR(qPCR)was performed to 4 genes selected from transcriptional profiling to validate the data collected from transcriptome analysis.Results ADTMsignificantlyincreasedsubintestinalvessels (SIVs)diameter in a concentration-dependent manner in normal zebrafish as well as restored VRI-induced blood vessels defect in VRI-exposed zebrafish. The transcriptome data analysis demonstrated that 19 signif-icantly changed genes were mapped to insulin signaling pathway.The qPCR data are in good agreement with those obtained by deep sequencing and support the consistency between the two methods for determining relative expression levels in the zebrafish model.Con-clusion Inzebrafishmodel,ADTMexhibitsthe effects of angiogenesis and blood vessel restoration. The underlying mechanism may be involved in the acti-vation of insulin signaling pathway.
3.RELN Gene Single Nucleotide Polymorphism Related with Clinical Features of Autism

Objective To investigate the relationship between RELN gene single nucleotide polymorphism (SNP) and childhood autism in Jiamusi, Heilongjiang. Methods Polymerase chain reaction-restricted fragment length polymorphism (PCR-RFLP) was used to determine allele and genotype of SNP (exon 6) of RELN in 30 children with autism and 30 normal children. Autism Behavior Checklist (ABC) was used to evaluate the children. Results There was a significant difference in the distribution of the allelic frequencies and genotype in exon 6 between these groups (P<0.05). There was a significant difference in the communication factors between patients with genotype of A/A and A/G or A/A and G/G (P<0.05), as well as in the total scores of ABC between A/G and G/G (P<0.05). Conclusion The SNP of RELN (exon 6) associated with the childhood autism. There is a more serious communication disorder in children with genotype of G/G, A/G than that of A/A.

5.Application of metagenomic next generation sequencing in infectious diseases

Almost all pathogens have DNA or RNA genomes.Theoretically, genomes of pathogens can be effectively detected by sequencing technology , it would be of value for etiological diagnosis of infectious diseases.The next generation sequencing ( NGS) emerged in 2005, the advantages of low cost , high efficiency and high accuracy make it a favorable technical platform for clinical identification of microorganisms.As a new sequencing technique , metagenomic next generation sequencing ( mNGS) has great potential in improving the accuracy and efficiency of pathogenic diagnosis , especially in the areas where conventional diagnostic methods have limitations.This article introduces the common platforms and principle of the high throughput sequencing , reviews the clinical applications and the advantages of mNGS in identification of different pathogenic microorganisms , and reflects on its limitations and seeks possible solutions.
6.Discussion on the pathogenesis and treatment of epilepsy from the theory of "brain's qi collateral-abnormal collateral"

Epilepsy is a disease of the central nervous system caused by excessive neuronal discharges in the brain,characterized by sudden,recurrent and self-limited onset. The brain's qi collateral and the brain neural network are highly correlated and internally consistent in terms of structure and function. The theory of "brain's qi collateral-abnormal collateral",which is centered on the structural disorder and dysfunction of brain's qi collateral leading to the poor circulation of brain's qi collateral,can comprehensively explain the related pathogenesis of epilepsy and the law of disease evolution,so it has important clinical value. Taking the pathogenic characteristics as an entry point and based on the theory of "brain's qi collateral-abnormal collateral",this paper argues that phlegm and qi stagnation,wind in the brain's qi collateral,and phlegm and blood stagnation damaging the brain's collaterals,as well as the structural and functional characteristics of brain's qi collateral that circulate bi-directionally are the key factors for epilepsy to present sudden,recurrent,and self-limited characteristics. According to the therapeutic principle of "Collaterals need to be unobstructed to function normally",it is proposed that the method of regulating the qi and collaterals should be used as the basic treatment principle throughout the treatment. In addition,the method of resolving phlegm and eliminating blood stasis is supplemented for different pathological changes,while combining the syndrome differentiation of zang-fu viscera and attaching importance to the accompanying symptoms of epileptic seizures,to regulate the brain's qi collateral to achieve the effects of wind quenching and epileptic arrest. This is to provide reference for the treatment of epilepsy in traditional Chinese medicine.

Tourette syndrome(TS)is a chronic neurodevelopmental disorder.Acupuncture can effectively improve the clinical symptoms of TS patients.This article systematically summarized the clinical research status of acupuncture for the treatment of TS in recent years from the aspects of characteristic acupuncture methods,characteristic needles and comprehensive therapies,and put forward suggestions and prospects for systematically elaborating the peripheral-central mechanism of acupuncture for TS around the intestinal immunity and brain network mechanism in the future,so as to provide reference for optimizing clinical research and treatment.
8.Treatment of Minimally Conscious State with Musk Based on "Phlegm,Fire,Blood Stasis,and Deficiency"

Minimally conscious state (MCS) is at the edge between closed and open consciousness, but it still belongs to the category of "wind-strike block" syndrome. The basic pathogenesis of MCS is the obstruction of pathogenic qi, orifices closed and spirit hidden, with pathological factors including phlegm, fire, and blood stasis. Wind movement and water retention may also be present, and often leading to deficiency syndrome due to the exhaustion of qi, blood, yin, and yang at later stages. Treatment chooses Shexiang (Moschus) as the chief medicinal, emphasizing combination of medicinals and urgency of medication administration; the key therapeutic method is to open the orifices, with focuses on expelling pathogens and reinforcing healthy qi. For patients with severe phlegm or fire, use Xiaochengqi Decoction (小承气汤) to open the lower orifices, discharge heat and unblock the bowels, combined with Shexiang (Moschus) and Niuhuang (Bovis Calculus) to open the upper orifices, awaken the spirit and guide qi. For patients with turbid phlegm as the predominant, temporarily replace Shexiang (Moschus) with Baizhi (Angelicae dahuricae radix), using Ditan Decoction (涤痰汤) to eliminate phlegm to open the orifices, when turbid phlegm gradually subsided, Shexiang (Moschus) could be added. For patients with blood stasis as the predominant, Tongqiao Huoxue Decoction (通窍活血汤) will be used to activate blood and open orifice, if the blood circulates, the endogenous wind will be calmed, the water will be induced, the orifices will open and the consciousness will restore. For patients with closed orifices and body deficiency, the treatment should open the orifices and reinforce healthy qi, and consider the root and branch simultaneously; qi deficiency syndrome can be addressed with Buyang Huanwu Decoction (补阳还五汤) to boost qi and reinforce healthy qi; yin deficiency syndrome can be treated with Shaoyao Gancao Decoction (芍药甘草汤) combined with Fengsui Pill (封髓丹) to nourish yin, soften sinews, and secure kidney essence; yang deficiency can be managed by using Dihuang Yinzi Decoction (地黄饮子) to enrich yin, supplement yang, and open the orifices.

In the middle of December in 2019, a pneumonia outbreak caused by a new coronavirus, 2019 novel coronavirus (2019-nCoV), emerged in the populations in Wuhan city of China. The epidemic spreads rapidly and has been disseminated throughout the country and to 13 other counties in Asia, Europe, Oceania and North America. To accurately and deeply understand the biological characteristics, epidemiological features and pathogenicity of 2019-nCoV and related immunological characteristics, microbiological examinations and public protection measure, this study reviewed 2019-nCoV and 2019-nCoV pneumonia based on the newest relevant literatures and the newest version of National Diagnosis and Treatment Scheme of 2019-nCoV pneumonia.
10.Expression of PD-1hiCXCR5-CD4+T cells in patients with systemic lupus erythematosus

Objective To investigate the expression of peripheral programmed death (PD)-1hiCXCR5-CD4+T cells and its clinical significance in systemic lupus erythematosus (SLE). Methods Peripheral blood PD-1hiCXCR5-CD4+ T cells from 21 SLE patients and 16 healthy controls were examined by flow cytometry. The levels of serum anti-double-stranded deoxyribonucleic acid (dsDNA) antibodies were determined using immunoradiometric as-say. Data were analyzed with t test and Pearson's correlation test. Results The per-centages of PD-1hiCXCR5- cells within CD4+ T cell were significantly higher in SLE patients [(2.1 ±2.0)%] compared to normal controls [(0.3±0.3)%] (t=2.959, P<0.01). The percentages of PD-1hiCXCR5-cells within CD4+T cells in moderate to severe active SLE patients (3.0 ±2.0)% was significantly increased compared to patients with mild or inactive (1.0±1.4)%(t=2.574, P<0.05) and normal controls (0.3±0.3)% (t=5.149, P<0.01). The percentages of PD-1hiCXCR5- cells within CD4+ T cells from SLE patients were positively related with systemic lupus erythematosus disease activity index (SLEDAI) (r=0.475, P=0.0297). SLE patients in serum anti-dsDNA antibodies positive group (2.7±2.1)%displayed a higher percentage of PD-1hiCXCR5-cells within CD4+T cells than patients in serum anti-dsDNA antibodies negative group (0.6 ±0.5)% (t=2.303, P<0.05). The percentages of PD-1hiCXCR5-cells within CD4+T cells from SLE patients were positively correlated with anti-dsDNA antibody titers. Conclusion The percentages of PD-1hiCXCR5- cells within CD4+ T cells from SLE patients are increased and are positively correlated with SLEDAI and anti-dsDNA antibody levels. Increased percentage of PD-1hiCXCR5-cells within CD4+T cells might play an important role in the pathogenesis of SLE.
