1.The effect of 50 Hz sinusoidal electromagnetic fields on the proliferation of the osteosarcoma cell line MG-63

Objective To investigate the biological effect of 50 Hz sinusoidal electromagnetic fields at 1 mT on the proliferation of the human osteosarcoma cell line MG-63.Methods Osteosarcrma MG-63 cells were divided into control and experimental groups.The control group was incubated without an electromagnetic field; the experimental group was incubated in a 50 Hz,1.0 mT sinusoidal electromagnetic field.On the 2nd,4th and 6th day,their proliferation was determined using a cell counting kit-8(CCK-8)assay.Variations in the cell cycle were detected with flow cytometry(FCM).Semi-quantitative reverse transcription-polymerase chain reaction(RT-PCR)was used to measure cyclin B1 and cyclin D1 mRNA.Results Compared with the control group,proliferation of the experimental group cells was reduced significantly.The percentage of cells at G0-G1 phase increased,and the mRNA expression of cyclin B1 and cyclin D1 was significantly reduced.Conclusions A 50 Hz sinusoidal electromagnetic field at 1.0 mT can inhibit the proliferation of osteosarcoma cell line MG-63 significantly.

Objective: To investigate the relation between proliferation, apoptosis and Bcl-2 in human tongue squamous cell carcinoma(TSCC). Methods: Apoptosis, prolifexation and Bcl-2 protein expression were examined in 7 cases of normal tongue mucosa, 20(10 of Han and 10 of Uygur people) of tongue squamous cell papilloma (TSCP) and 42 of TSCC (30 of well-differentiated and 12 of middle-differentiated ) with immunohistochemical and in situ cell death detection technique. Objective To explore in vitro the best time window for using sinusoidal electromagnetic fields to promote the osteogenic differentiation of bone marrow mesenchymal stem cells (BMSCs).Methods BMSCs were isolated and cultured from 4-week-old Sprague-Dawley rats (male and female,80-120g).The BMSCs (from passage 3) were exposed 0,1,4 or 8h/d for 7d,14,or 28d,respectively,to 15Hz sinusoidal electromagnetic fields with a maximum amplitude of lmT.Those exposed 0h/d served as the control.The relative expressions of runt related gene-2 (RUNX2),bone sialoprotein (BSP) and osteopontin (OPN) were determined using real-time,quantitative reverse transcription-polymerase chain reactions (RT-PCRs).The level of RUNX2 protein was determined by Western blotting after 14d.Alizarin red staining was used to compare calcium distribution in each group.Results Obvious promotion of differentiation to osteoblasts was observed after 7 days of exposure to the15 Hz sinusoidal electromagnetic fields,most obviously manifested by an outstanding increase of the early osteogenic index RUNX2 in those exposed 4h/d.After 14 days of intervention,the 1h/d exposure showed to be most effective,especially in inducing the changes of the late osteogenic index OPN.The trends of changes in RUNX2 protein were similar in all groups.After stimulating 1h/d for 14 and 28days,calcium deposition increased to the greatest extent.Conclusions Exposure to sinusoidal electromagnetic fields induces osteogenic differentiation to osteoblasts in rat BMSCs in vitro.There is an apparent window effect.The best results are observed with more days of exposure and shorter exposure time (1h) every day.
4.Investigation on peripheral blood TSHR mRNA for differentiation of benign and malignant thyroid nodules

Objective To assess the value of peripheral blood thyroid stimulating hormone receptor(TSHR) mRNA determination in differential diagnosis of benign and malignant thyroid nodules.Methods Fine needle aspiration cytology (FNAC) and (or) postoperative histopathology as the gold standard were carried out,the expression of circulating TSHR mRNA was determined by RT-PCR in 33 patients with benign thyroid nodules,39 patients with thyroid cancer,and 20 normal controls.Results TSHR mRNA signals were not detected in normal controls,the positive rate of TSHR mRNA was higher in the group with malignant nodules than the group of benign nodules (91.2% vs 48.5%,P<0.01).TSHR mRNA level in the preoperative malignant group was significantly higher than that in the normal,benign,and postoperative cancer groups (all P < 0.01).Using peripheral blood TSHR mRNA for differentiating benign or malignant of thyroid nodule had a sensitivity,specificity,and accuracy of 91.2%,51.5% and 71.6%,respectively.The sensitivities of TSHR mRNA,FNAC,and these two methods combined in detecting malignant nodules were 91.3%,86.9%,and 100.0% respectively,while diagnostic accuracies were respectively 84.0%,80.0%,and 92.0%.TSHR mRNA expression showed no significant relationship with sex,age,size,and number of nodule in these patients (all P > 0.05),but it did exhibit significant difference between benign and malignant nodules(P<0.01).Conclusion The peripheral blood TSHR mRNA could be used as a molecular marker for thyroid cancer,and it would help enhance the preoperative differentiation of benign and malignant thyroid nodules.
5.Determination of magnoflorine in Coptidis Rhizoma and Phellodendri Chinensis Cortex by LC-MS.

A rapid and specific high performance liquid chromatography-mass spectrometric method was developed for determination of magnoflorine in Rhizoma Coptidis and Cortex Phellodendri Chinensis. Samples were extracted by methanol. Agilent Eclipse XDB-C18 ODS column (4.6 mm x 150 mm, 5 microm) was used, and mobile phase was methanol-water (60:40) at a flow rate of 0.8 mL x min(-1). Electrospray ionization model (ESI), and MRM model were used for quantification. The linear range of magnoflorine was 4.352-2720 microg x L(-1). The average recovery was above 98%. The method is simple, sensitive and accurate, it can be used for determination of magnoflorine in Rhizoma Coptidis and Cortex Phellodendri Chinensis.

6.Epidemiological characteristics of newly diagnosed HIV/AIDS cases aged50 years or over in Zhejiang Province from 2015 to 2019

Objective:
To analyze the epidemiological characteristics of newly diagnosed HIV/AIDS cases aged 50 years or over in Zhejiang Province from 2015 to 2019, so as to provide reference for the development of prevention and control strategies.
Methods :
The data of newly diagnosed HIV/AIDS cases in Zhejiang Province were collected through the China HIV/AIDS Comprehensive Response Information Management System, and the demographic characteristics, infection routes,regional distribution and time distribution of the cases were analyzed.
Results:
A total of 6 726 HIV/AIDS patients were recruited in this study, and the number of new patients from 2015 to 2019 showed an increasing trend ( P <0.05 ) . The number of participants diagnosed at the age of 50-59, 60-69 and 70-91 years old were 3 433( 51.04% ), 2 242 ( 33.33% ) and 1 051 ( 15.63% ). The majority of them were males ( 5 180, 77.01% ) , married ( 4 286, 63.72% ) , Zhejiang residents ( 5 304, 78.86% ) , and lived in rural areas ( 4 095, 60.88% ) . In terms of exposure history,6 586 cases ( 97.92% ) were infected by sexual contact. Among the 5 083 males infected by sexual contact, 82.63% were through heterosexual contact, 94.79% had extramarital sex partners, among whom 76.61% were commercial sex partners. The married women patients who confirmed HIV positive accounted for 75.44%. There were increasing trends in the proportion of the cases living in rural areas, male cases infected through heterosexual contact and those with extramarital and commercial sex partners, and female cases with their husbands positive along with age ( P <0.05 ) .
Conclusions
The number of newly diagnosed HIV/AIDS cases aged ≥50 years is increasing in Zhejiang Province. Many of them live in rural areas. Commercial sexual contact is the main route of HIV transmission among males and further lead to HIV transmission within couples.
7.Mediating effect of rumination between affiliate stigma and post-traumatic growth in parents of autistic children

Objective:To explore the relationship among ruminating,affiliate stigma and post-traumatic growth in parents of autistic children.Methods:Totally 339 parents of autistic children were selected.The Affiliate Stigma Scale(ASS),Post-traumatic Growth Inventory(PTGI),ERRI Intrusive Subscale(ERRI-I)and ERRI De-liberate Subscale(ERRI-D)of Event Related Rumination Inventory(ERRI)were used to measure affiliate stigma,post-traumatic growth,intrusive ruminating and deliberate ruminating.SPSS macro program PROCESS was used to test the mediating role.Results:There were positive correlations among the scores of ASS,ERRI-I and ERRI-D(r=0.39-0.72,Ps<0.01).The PTGl scores were negatively correlated with ASS scores(r=0.26,Ps<0.01)and positively correlated with ERRI-D scores(r=0.10,Ps<0.05).The ERRI-D scores played a partial mediating role in the relationship between ASS and PTGl scores(95%CI:0.01-0.07).The ERRI-I scores and ERRI-D scores played a serial mediating role in the relationship between ASS and PTGI scores(95%CI:0.03-0.14).Both me-diation paths had suppression effect.Conclusion:In the negative correlation between affiliate stigma and post-trau-matic growth in parents of autistic children,the deliberate ruminating plays a partial mediating role,while intrusive ruminating and deliberate ruminating play a serial mediating role.
8.Key Environment Factors and Regionalization of the Ecological Suitability of the Original Species of Chinese Medicine Cremastrae Pseudobulbus/Pleiones Pseudobulbus

Objective In order to offer a theoretical foundation for Cremastae Pseudobulbus/Pleiones Pseudobulbus original species scientific introduction and site selection for expansion.Methods We gathered sample distribution information for the original species of Cremastae Pseudobulbus/Pleiones Pseudobulbus nationwide by consulting various databases and conducting field investigations.A total of 257 effective distribution data were gathered.Altitude,temperature,solar radiation,precipitation,and water vapor pressure were among the 68 environment factors that were chosen from 5 categories.The ecological suitability of Cremastae Pseudobulbus/Pleiones Pseudobulbus original species was investigated using Maxent and ArcGIS.Results The investigation revealed that precipitation,solar radiation,and altitude were the core environment factors influencing the ecological suitability of Cremastae Pseudobulbus/Pleiones Pseudobulbus.The most important environment factors varied significantly between diverse original species.The most suitable potential areas for the growth of Cremastra appendicutata,Pleione bulbocodioides,and Pleione yunnanensis were concentrated in Taiwan,South China,and the adjacent areas of Yunnan and Sichuan,respectively.Conclusion Further ecological suitability levels were classified for their potential distribution areas,providing a scientific basis for the rational introduction,cultivation,and artificial stanhbzyydxdardized expansion of the Cremastae Pseudobulbus/Pleiones Pseudobulbus.
9.Study Actuality of Medicinal Pteridophyes Literature via Visualized Bibliometrics

Objective The current article aimed to explore the previous focuses,highlight the latest progresses and analyze the future trends on the research of medicinal Pteridophyes.Methods 794 scientific names and 414 Chinese names of medical used ferns and lycopods were established as search term from five public documentary libraries(i.e.,Web of Science,PubMed,CNKI,WangFang and Cqvip),respectively.After data cleaning,5465 publications in English and 8918 publications in Chinese were refined as effective datasets.Subsequently,the up-to-date bibliometric methods and visualization software packages were employed to fulfill the graphical bibliometric analyses.Results The yearly publication numbers in both English and Chinese showed fluctuating upward patterns,and each of them has undergone three phases with distinct rates of rise(i.e.,embryonic stage,smoothly-rising stage and sharply-rising stage).Significant national and territorial discrepancies exist in the various types of literature.The topics in English literature on medicinal Pteridophyes mainly included ontogenetic process and phylogeny,plant-environment interactions,and the pharmacological activities of various metabolites(e.g.alkaloids).The Chinese literature is not only concerned with individual development and pharmacological effects,but also focuses on the application of new technologies,emphasizes the application of them in traditional Chinese medicine and traditional Chinese and Western medicine.Moreover,our results pointed out rising hotspot medicinal Pteridophyes species such as Drynaria roosii and Huperzia serrata.Conclusion Our present study implied that the medicinal Pteridophyes have aroused wide attention in domestic and foreign scholar and will be continuous concerned for a long time in the future.Currently,4 different aspects(i.e.,phytoremediation,phylogenetic analysis,pharmacological activity and clinical application)are the main research focal points towards this clade of medical plant species,and it is an important emerging tendency that we should apply the multi-omics technology to investigate the Pteridophyes species and unravel their medical values.
10.Previous HIV testing behavior and related factors in men who have sex with men in gay bathroom in Hangzhou, Zhejiang province

Objective To understand the related factors on previous HIV testing behavior among MSM who visit gay bathroom in Hangzhou, Zhejiang province. Methods Through time-location sampling (TLS) survey, men who had oral sex or anal sex with men in past 6 months were interviewed in the gay bathroom in Hangzhou from October 2015 to January 2016. Software SPSS 22.0 was used to analyze the HIV testing acceptance, sexual behaviors and condom usage of MSM during past 6 months, and χ2 test and logistic regression model were used for univariate and multivariate analyses. Results Our study planned to recruit 480 gay bathroom MSM, due to the unwilling or other factors, 465 MSM were surveyed. A total of 47.3%(220/465) gay bathroom MSM received HIV testing during past 6 months. Multivariate analysis indicated that being married (OR=0.35, 95%CI:0.18-0.69), peer education (OR=4.31, 95%CI:1.32-14.09), homosexual behavior during past 6 months (OR=1.85, 95%CI: 1.08-3.14), extramarital heterosexual behavior during past 6 months (OR=4.12, 95%CI: 2.45-6.91) might be positive factors related with HIV testing acceptation in MSM. Conclusion Being married, homosexual behavior during past 6 months, extramarital heterosexual behavior, partner education might be the positive factors related with acceptance of HIV testing among gay bathroom MSM. Therefore, peer education needs to be strengthened. HIV testing after high risk sexual behavior needs to be promoted in MSM.
