1.Prevalence of anxiety and depression before idiopathic benign paroxysmal positional vertigo episode

Objective To investigate the prevalence of anxiety and depression one month before idiopathic benign paroxysmal positional vertigo (BPPV) episode. Methods Patients from Vertigo Clinic of Department of Neurology, West Campus, Chaoyang Hospital, were enrolled from January 2014 to June 2016. For patients with first visiting idiopathic BPPV, Hamilton Anxiety Scale and Hamilton Depression Scale were used at the consulting room by an experienced neurologist to reflect the emotional state one month before idiopathic BPPV episode. The occurrence rate of mood disorders was compared with usual model. Results One hundred and eighty five patients with idiopathic BPPV, aged 28 to 82 (57.6±12.0) years, 19.5% (36/185) being male, 80.5% (149/185) being female, were enrolled into this research. No anxiety, probable anxiety, definite anxiety and severe anxiety accounted for 15.1% (28/185), 33.0% (61/185), 47.6% (88/185) and 4.3% (8/185), respectively;no depression, probable depression, definite depression and severe depression accounted for 18.4% (34/185), 48.6% (90/185), 29.2% (54/185) and 3.8% (7/185), respectively;no mood disorders, probable mood disorders, definite mood disorders and severe mood disorders accounted for 11.4% (21/185), 33.5% (62/185), 49.2% (91/185) and 5.9% (11/185), respectively;total definite mood disorders and severe mood disorders accounted for 55.1% (102/185), higher than usual model (14.4%)Conclusion Anxiety and depression is common before idiopathic BPPV episode.

In this paper, the origins, characteristics and poisoning mechanisms of shellfish poisons including the paralytic shellfish poison(PSP),the diarrheic shellfish poison(DSP), the neurotoxic shellfish poison(NSP) and the amnestic shellfish poison(ASP) were introduced briefly. Methods of assay and detoxification of shellfish poisons were also discussed in detail.
3.Possibility of three-dimensional time-of-flight magnetic resonance angiography as a reliable screening tool for evaluation of intracranial vascular stenosis and occlusive disease prior to stent implantation

AIM: To investigate whether three-dimensional time-of-flight magnetic resonance angiography (3D TOF MRA) can be used as a reliable screening tool for evaluation of intracranial vascular stenosis and occlusive disease before stent implantation. METHODS: Thirty-three patients with suspected intracranial arterial stenosis received 3D TOF MRA and digital subtraction angiography (DSA) examinations in Chaoyang Hospital Affiliated to Capital Medical University between March 2007 and April 2008,and were included for this study. Two physicians blindly estimated stenosis,patient history,and clinical information of 363 vascular segments from 33 patients,including bilateral internal carotid artery (ICA),anterior cerebral artery (ACA),middle cerebral artery (MCA),posterior cerebral artery (PCA),vertebral artery,and basilar artery (BA). Stenosis was categorized as 30%-49%,50%-69%,70%-99%,and 100%. For each kind of stenosis,sensitivity,specificity,positive predictive value,negative predictive value,K and P values of MRA were calculated,respectively,as compared to DSA. RESULTS: A total of 42 diseased vascular segments were identified. Compared to DSA,for intracranial stenosis 50%-69%,3D TOF MRA showed sensitivity 100%,specificity 96.8%,positive predictive value 62.1%,negative predictive value 100%,K value 0.751,and P value 0.000; For intracranial stenosis 70%-99%,the corresponding value was 100%,98.6%,70.6%,100%,0.821,and 0.000,respectively; For intracranial stenosis 30%-49%,it was 25.0%,99.7%,66.7%,98.3%,0.356,and 0.000,respectively.CONCLUSION: For high sensitivity and specificity to intracranial stenosis 100%,70%-99%,or 50%-69%,compared to DSA,3D TOF MRA is a reliable screening tool for preoperational evaluation of intracranial vascular stenosis and occlusive disease.
4.A Study on IMRT Beam Weight and Orientation Optimization based on Genetic Algorithm with Two Populations

Objective To study the application of the genetic algorithm with two populations in optimization of both IMRT beam weight and orientation.Methods The genetic algorithm with two populations was used to optimize IMRT beam weight and orientations with three dimensional pencil beam dose calculation model. The algorithm was implemented in Visual c#. Net. And the results were analyzed. Results The modest change of the beam orientation resulted in more conformal dose distribution in target volume, and large high dose area was included. Further more, genetic algorithm with two populations gave such a clinically acceptable computation time. Conclusion Little change had distinct influence on dose distribution when there were few beam orientations. The genetic algorithm with two populations was an effective and global optimization method because of its parallel, robust and global. It provided satisfying results when used to optimize IMRT beam weight and orientations

AIM: To investigate the expression profile of peripheral-type benzodiazepine receptor(PBR) involved in mitochondrial permeability transition(PT) regulation,and to observe the binding dynamic of the mitochondrial PBR with specificity ligand during rat live regeneration.METHODS: Liver regeneration model was produced by 70% partial hepatectomy(PH) performed in male SD rats.The animals of sham groups underwent the same surgical operations as PH groups did,but the liver lobes were not resected.The animals in the PH groups and corresponding sham groups were sacrificed at 3,6,12,24,48,72,120 and 168 hours after the operation.The livers were removed,weighted and processed for isolation of mitochondria.Semi-quantitative RT-PCR was performed to examine the expression level of PBR in 70% hepatectomized rat livers during the whole regeneration process and compared to that in the sham and normal groups.Compared with healthy rats,the kinetic parameters of PBR was evaluated by using a specific radioligand -PK11195.RESULTS: Compared with healthy rats,the expression of PBR was unchanged.Meanwhile,the results obtained in the present experiments by scatchard analysis,Bmax of PK11195 for PBR significantly decreased,returned to normal level in 168 h after PH.Kd of PK11195 for PBR significantly decreased at 72 h and 168 h after PH of rat liver regeneration(P

Objective To conduct the amplification,cloning,bioinformatics analysis,prokaryotic expression and purification of enterovirus 71 VP1 gene segment and to initially confirm the biological activity of the recombinant expression product.Methods A pair of specific primers was designed according to GenBank EV71 sequence,viral RNA as a template was extracted from the throat swab specimens in the EV71 patients.EV71 VP1 gene was amplified by RT-PCR.After enzyme digestion,the expression vector pET28a was inserted.The prokaryotic expression vector of pET28a-EV71 VP1 was constructed.Then the E.coli DH5a transforma-tion was performed.IPTG was adopted for induction expression.The expression results were analyzed by using SDS-PAGE and Western blot.The bioinformatics analysis of the sequenced results was performed by the software.Expressed protein was purified and the plates were coated,ELISA was used to test the VP1 specific IgG antibody in serum samples of EV71 positive and COX A16-positive patients.Results The BLAST alignment showed that the homology of the objective gene EV71 VP1 was 99% com-pared with other strains(JQ766207.1)in GenBank.EV71 VP1 protein was about 32×103 ,which mainly existed in the form of in-clusion body.The bioinformatics analysis showed that EV71 VP1 protein was a hydrophilic protein,without transmembrane region and N-terminal signal peptide sequence,the tertiary structure existed.The ELISA results showed that the specific IgG OD value in EV71-positive patients was(2.425±0.521),OD value in COX A16 positive patients was(1.205 ±0.314),the normal control OD value was(0.353±0.128).The sensitivity and specificity of EV71 VP1 protein detection were 84% and 88% respectively.Conclu-sion The pET28a-EV71 VP1 expression vector is successfully constructed;the preliminary analysis on the serum of the infected patients by ELISA shows that the obtained objective protein has higher sensitivity and specificity,which is initially confirmed to have biological activity and can be further used for the related study on EV71 diagnosis and vaccine.
7.Effect of quercetin exposure during the prepubertal period on ovarian development and reproductive endocrinology of mice.

This study is to explore the effects of quercetin (QUE) on the 3 week-old mice ovarian development and relative hormone levels. The 3 week-old mice were exposed to QUE (45, 25, and 5 mg x kg(-1) x hd(-1)) by gavage for 50 days. The estrous cycle during 50 days and the changes of hormone level such as FSH, LH, etc were monitored. Moreover, the ovaries were removed after sacrifice. The organ index was measured, and the ratios of different stages of follicles were analyzed by HE staining. Furthermore, the proportion of PCNA positive cells during all stages was detected by immunohistochemistry. The results showed that QUE could increase body weight of mice and reduce the anogenital distance (AGD) to some extent, and was able to disrupt mice's estrous cycle, but it could not extend or reduce the cycle regularity. It increased ovarian organ index with a dose-dependent manner. The proportion of the primordial follicle and secondary follicles rose obviously, and that of mature follicles', atretic follicles' and corpus luteums' reduced, while primordial follicle had no change. Immunohistochemistry analysis showed that QUE could effectively increase the percentage of proliferating cells in all kinds of follicles. Serum hormone assay showed that there were significant changes of FSH and LH levels. In summary, QUE showed an estrogen-like effect on mice's ovarian development. The weight of ovary, the proportion of all kinds of follicles, the development of ovarian cells and the level of plasma hormone in mice were altered obviously by oral administration of QUE.
8.Analysis of magnetic susceptibility of normal brain of young adults by quantitative susceptibility mapping

Objective To explore the changes of susceptibility of different sides and gender in healthy young adults with quantitative susceptibility mapping (QSM).Methods Totally 41 healthy young right-handed adults underwent conventional brain MRI and QSM scan,and the susceptibility maps were obtained by the image post-processing software.Then the ROI of the bilateral frontal grey matter (FGM),frontal white matter (FWM),caudate (CA),globus pallidus (GP),putamen (PU),thalamus (TH),substantia nigra (SN),red nucleus (RN),dentate nucleus (DN),pons (PO),corpus callosum (CC) were manually drawn to obtain magnetic susceptibility on the susceptibility map.The magnetic susceptibility of each ROI was compare between both sides,as well as gender by Mann-Whitney test.Results The magnetic susceptibility of the bilateral ROI of GP was the highest,and SN was followed,FWM was minimum.The susceptibility of bilateral FGM,FWM,CA,GP,PU,TH,SN,RN,DN,PO,CC had no statistically significant differences (all P>0.05).The magnetic susceptibility in CA of different gender had statistically significant difference (P<0.05).Conclusion The brain magnetic susceptibility.can be measured by QSM,and it can assess brain iron content quantitatively.
9.The role of mitogen-activated protein kinase (MAPK) pathway in the action mechanism of PYM-induced KB cells apoptosis.

OBJECTIVETo study the role of mitogen-activated protein kinase (MAPK) pathway in the action mechanism of PYM-induced KB cell apoptosis.
METHODSWestern blot analysis was used to investigate the expression of the mitogen-activated protein kinase.
RESULTSWhen treated with PYM in cultured KB cells, the extracellular signal-regulated kinase (ERKl/2) showed a dose-and time-dependent decreasing in phosphorylation status of these proteins through a western blot analysis, whereas protein levels of p38 MAPK remained unchanged.
CONCLUSIONSThe mitogen-activated protein kinase (MAPK) pathway may play an important ro1e in PYM-induced apoptosis of KB cells.

10.Efficacy of postural restriction in preventing recurrence of Benign Paroxysmal Positional Vertigo:a randomized controlled clinical study

Objective To evaluate the effect of postural restriction in preventing recurrence of Benign Paroxysmal Positional Vertigo(BPPV). Methods A total of 217 unilateral posterior semicircular canal BPPV patients were diagnosed clearly and divided into two groups randomly:104 caes(observation group) were demand postural restriction(head up 45 degrees for 72 hours,but did not restricted sleeping posture or departure from bed) after Epley maneuver,while 114 case(control group) could freely sleep. Demographic data,concomitant diseases and betahistine mesylate treatment were recorded. All patients were follow-uped at 72 hours after the Epley maneuver at outpatients of our hospital. A induced vertigo by position change or positional nystagmus were defined as recurrence of BPPV. Results There was no statics defferrence between two groups for base line data. After 72 hours of Epley maneuver,BPPV recurrence rates was 21.2% and 41.6% for observation group and control group,respectively(P<0.01). Odds ratio of postural restriction for BPPV recurrence was 0.377(95%CI 0.207~0.687). Conclusion Head up 45 degrees for 72 hours after Epley maneuver was helpful for preventing BPPV early recurrence.
