1.Effects of intermittent hypoxia on the growth of SW480 cell lines and the expression of HIF-1?

Objective: To determine the effects of intermittent hypoxia (IH )on the growth of SW480 cell lines and the expression of hypoxia-inducible factor(HIF)-1?,the phenomenon of IH in tumors and the relation between IH and HIF-1? were discussed. Methods: The hypoxia condition was simulated by CoCl2. Effect of IH on the growth of SW480 cell lines was observed by MTT assay. Effects of IH on the expression of HIF-1? were analyzed by Reverse Transcriptase Polymerase Chain Reaction (RT-PCR) and Western blot assay. Cells were divided into four groups: group A: normoxia group,group B: IH for 2 hours group,group C: IH for 3 hours group,and group D: continuous hypoxia (CH) for 3 hours group. Results: MTT assay indicated that in the 3 hypoxia groups,the growth of SW480 cell lines was inhibited,when compared with normoxia group. The growth of SW480 cell lines was also inhibited in group C and group D when compared with group B. No significant differences were observed between group C and group D.RT-PCR showed that the level of HIF-1? mRNA in group C was higher than that in group A,group B and group D. No significant differences were observed among the other 3 groups. Western blot found that the level of HIF-1? protein in group C was higher than that in the other 3 groups.The level of HIF-1? protein in group D was higher than that in group B and group A. No significant differences were observed between group B and group A. Conclusion: Same hypoxia time but different hypoxia manner inhibit the growth of SW480 cell lines to the same degree. A certain degree of IH can up-regulate the mRNA and protein level of HIF-1?.
2.RNAi-mediated silencing of HIF-1? inhibits growth and metastasis of colon cancer in tumor-bearing mice

Objective To explore the influence of RNAi-mediated hypoxia-inducible factor 1? 3.STUDY ON COMPARTMENTALIZATION, DIS-TRIBUTION AND CROSS-SECTIONAL AREAS OF FIBER TYPES IN HUMAN DELTOID MUSCLE

The deltoid muscles on both sides of 8 male cadavers were studied within 24 hours after death. According to its fiber architecture, origins and insertions, and nerve distribution, the deltoid muscle was divided into three compartments: an anterior, a middle and a posterior. The muscle samples were taken respectively from the superficial and the deep portion of each compartment of the deltoid muscle. Frozen transverse sections were stained fer myosin ATPase. Muscle fibers were identified as type Ⅰ and type Ⅱ. The proportion of fiber types in each portion was calculated on microscopic photographs and mean cross-sectional areas of both fiber types were measured by imaging analytical system. The results showed that the proportion of type Ⅰ fiber in the superficial and deep portion of the middle compartment is significantly higher than that of the corresponding portion of the anterior and posterior compartments, and the proportion in the deep portion of each compartment is higher than that of the superficial one, however no difference was found between the left and right sides. The fiber size among all portions is not obviously different, but the diameter of type Ⅱ fiber in the deep portion of the middle compartment is significantly smaller than that of other portions. Except the deep portion of the middle compartment, the diameter of both fiber types of the right muscle is larger than those of the left one, although statistical analysis showed that the difference is not significant. The authors suggest that the difference of fiber type distribution within the deltoid muscle is closely related with the functional differentiation of the muscle and the characteristic higher typeⅠ fiber proportion and smaller type Ⅱ fiber diamiter in the deep portion of the middle compartment probably indicate that the main function of this portion is to keep the shoulder joint stability.
4.Factors for the recurrence after endoscopic resection of early gastric cancer

Objective To investigate the related factors of recurrence of early gastric cancer (EGC) after endoscopic resection. Methods Clinicopathologic data of 169 patients with EGC who underwent endoscopic resection and periodically followed up by the Chinese PLA General hospital were analyzed retrospectively. Results During a follow-up of 13-57 months (median time 24. 5 months), 12patients had gastric cancer again and the recurrence rate was 7. 1% (12/169). The recurrence time varied from 3 to 36 (28 ± 23)months and the median time was 18 months. The recurrence rates of 0.5 year, 1st year, 2nd year and 3rd year were 1.18% (2/169), 3.55% (6/169), 9.91% (11/111) and 12.24%(12/98), respectively. Eleven patients had gastric cancer again within 2 years after resection.Undifferentiated histology (including poorly differentiated carcinoma and signet ring cell carcinoma),submucosal infiltration and lymphovascular invasion of the primary lesion of EGC were related to thepostsurgical recurrence ( all P < 0. 05). Conclusion Most recurrence of EGC occurred within 2 years afterendoscopic resecton and is related with undifferentiated histology, submucosal infiltration andlymphovascular invasion. It is important for these patients to receive endoscopy follow up.

objective To investigate the relationship between serum iron,ferritin(SF),transferrin(TRF)and coronary artery stenosis asd well to find the possible cause of coronary heart disease,Methods All the 315 pailents underwent coronary angiography.Serum iron,SF and TBF were measured.All patients were divlded into three groups according to the Gensini score.The distribution of serum iron,SF,TRF in the three groups underwent analysis of variance and the relationship between serum iron,SF,TRF and the Gensini score were further investigated by means of multiple linear regression analysis.Results The analysis of one-way variance showed that serum iron,SF and TRF/SF Were associated with the Gensini score(coefficient correlation=6.162,7.191 and 4.380,P<0.05).Muhiple linear regression analysis adjusted for many risk factors for coronary heart disease showed that SF had independent association with the Gensini score.(r=0.240.P<0.01).Conclusion Ferritin is independ-ently associated with coronary artery stenosis.

OBJECTIVE:To carry out individualized pharmotherapy and pharmaceutical care for patients.METHODS:With the aid of windows 98 operating platform,the Access 2000 database software was adopted to develop the patients'drug administration records on the basis of the results of the blood drug level monitoring and clinical information.RESULTS:The non-paper drug administration records built by Access had the advantages of convenient data process,huge information store,rapid inquiry system and so on.CONCLUSION:Access database software is worth recommending for clinical pharmacy and further studying.
8.Cloning and expression analysis of leucoanthocyanidin reductase gene in Fagopyrum dibotrys.

The leucoanthocyantin reducase (LAR) gene, an important functional gene of catechins biosynthesis pathway, was cloned from Fagopyrum dibotrys (D.Don) Hara by degenerate PCR and rapid amplification of cDNA ends (RACE). The full-length cDNA of FdLAR is 1 581 bp (GenBank accession: JN793953), containing a 1 176 bp ORF encoding a 391 amino acids protein, and its 3'-untranslated region has an obvious polyadenylation signal. The recombinant plasmid containing FdLAR completed ORF was transformed into E. coli BL21 (DE3). The target fusion peptide with molecular weight of 66 kD was expressed under the condition of 16 degrees C and induced by IPTG at final concentration of 1.0 mmol x L(-1). Bioinformation analysis indicated that the amino acid sequence of FdLAR showed great homology to other LAR with the NADB-Rossmann conversed domain in the N-terminus. Real-time quantitative PCR was used to detect the expression levels of FdLAR gene during different development periods. The determination of flavonoids contents in appropriate rhizomes showed that the relationship between FdLAR gene expression and the accumulation of flavonoids displayed different trends during vegetative growth and reproductive growth stages, suggesting that the FdLAR gene may be involved in the pathway of flavonoid metabolisms in Fagopyrum dibotrys.
9.Analysis of complications during and post to interventional therapy of common congenital heart disease in children

Objective To analyze the incidence of complications during and after interventional therapy for common con-genital heart disease (CHD) in children. Methods From January 2011 to December 2013, interventional therapy of common congenital heart disease which include ventricular septal defect (VSD), atrial septal defect (ASD), patent ductus arteriosus (PDA) and pulmonary valve stenosis (PS) were performed in 2356 patients. Among them, 159 patients who developed complications during and post to interventional therapy were retrospectively analyzed. Results The overall complication rate was 6.75%(159/2356) (11.40% post VSD occlusion, 7.50% post ASD occlusion, 3.09% post PDA occlusion, 1.63% post percutaneous balloon pulmonary valvuloplasty (PBPV) ).The rate of arrhythmia was 4.41%(102/2356). The severe complication rate was 2.71%(64/2356) (3.62%post VSD occlusion, 2.21%post ASD occlusion, 2.53%post PDA occlusion, 1.63%post PBPV). The intraoperative severe complication rate was 0.51%(12/2356);the early severe complication rate was 1.99%(47/2356);the late severe complication rate was 0.21%(5/2356). Interventional therapy rate was 0.13%(3/2356); cardiovascular surgery rate was 0.64%(15/2356);conservative treatment rate was 1.95%(46/2356). The mortality rate was 0.08%(2/2356). Conclusions The complications and mortality rate of interventional therapy for CHD in children are relatively low, but cannot be ignored. The complication could be reduced by choosing proper indications, following the operational procedures and careful operative follow-up.
10.The diagnostic value of confocal laser endomicroscopy in colorectal polyps

Objective To analyze endoscopic histological characters and establish diagnostic standards of colorectal polyps with confocal laser endomicroscopy (CLE) examination and to explore the diagnostic value of the CLE in adenomatous polyps and non-adenomatous polyps. Methods From June to December in 2009, 90 patients were recruited in this study, which included 40 pathologically confirmed colorectal polyps patients (total 48 colon and rectal polyps) and 50 patients for prospective study (total 106 colon rectal polyps). At same time 10 spots of normal mucosa was taken for comparison. Firstly the CLE images of 48 pathologically confirmed colorectal polyps (22 adenomatous polyps and 26 non-adenomatous polyps) were analyzed, and the diagnosis criteria for distinguishing adenomatous polyps and non-adenomatous polyps was established with CLE. Then according to the criteria, 106 colorectal polyps underwent prospective diagnosis with CLE. Finally, the CLE diagnosis result was compared with pathologically result to evaluate the diagnosis efficiency of CLE. Results In the 48 colorectal polyps of 40 pathologically confirmed colorectal polyps patients, there were 22 adenomatous polyps and 26 non-adenomatous polyps. The sensitivity, specificity and accuracy of CLE in adenomatous polyps diagnosis were 94. 0%, 92. 9%, and 93. 4%, respectively. The positive predictive value was 92.2% and the negative predictive value was 94.5%. The sensitivity, specificity and accuracy of CLE in non-adenomatous polyps diagnosis were 92. 9%, 94.0%, and 93. 4%respectively, the positive predictive value was 94.5% and the negative predictive value was 92.2%.The coherence of CLE and histopathology in adenomatous polyps diagnosis was pretty good (Kappa=0. 893). Conclusion The accuracy of CLE in adenomatous polyps and non-adenomatous polyps differential diagnosis was high, and the coherence with histopathology diagnosis was good, which provided experience for further detection of early rectal precancerous.
