1.Characteristics and significances of ECG and clinical laboratory examination results in 43 patients with acute aconitine poisoning

Objective To study the characteristics and significances of electrocardiogram(ECG)in the patients with acute aconi-tine poisoning.Methods Collected and analysed the data of 43 aconitine poisoning patients,summarize their clinic feature,ECG changes,laboratory test and treatment methords.Results All 43 cases showed arrhythmia under their ECGs,most of which were ventricular premature beats.Frequent multifocal ventricular arrhythmia,torsade de pointes,ventricular fibrillation occurred in severe aconitine poisoning patients.2 severe cases were treated with blood perfusion and purification in emergency and their arrhythmias were corrected rapidly.Serum K+ ,Mg2 + coucentrations decreased while myocardial enzymes increased in patients with severe and moderate poisoning.After active treatment,the ECG,cardiac creatase and electrolytes in all cases turned to normal.All 43 patients were cured.Conclusion The study shows the diversity of arrhythmia in acute aconitine poisoning patients and the deadly arrhyth-mia occurred in severe patients.Actively support therapy,early use of anticholinergic and antiarrhythmic drug,blood perfusion and purification in severe poisoning patients can rapidly cure arrhythmia and improve the rescue success rate.
2.The effect and mechanism of Sorcin silencing on drug resistance of human ovarian cancer SKOV3/CDDP cell lines

Background and purpose:Multi-drug resistance is a major reason for the chemotherapy failure of human ovarian cancer. Sorcin was found overexpression in drug resistance tumors and it may be the target of multi-drug resistance reversal. The present article was aimed to study the effect and mechanism of Sorcin silencing on drug resistance of human ovarian cancer SKOV3/CDDP cell lines. Methods:The stable Sorcin silencing SKOV3/CDDP cell lines were established. MTS assay, flow cytometry was used to analyze the intra-cellular Rh-123 content, cells apoptosis and cycle. Real-time, Western blot and report gene assay were used to analyze the expression changes of genes, including MDR1, MPR1, Survivin, Bcl-2, p-Akt and NF-κB. Results:Sorcin inhibition enhanced the drug sensitivity of SKOV3/CDDP cells, increased the intra-cellular Rh-123 content and cell apoptosis, and arrested cell cycle in G2/M. The protein levels of MDR1, MPR1, Survivin, Bcl-2, p-Akt were down-regulated, the mRNA levels of MDR1 of Sh-Sorcin-1 and Sh-Corcin-2 group were decreased to 42.3%and 26.5%of untransfected control, MRP1 were decreased to 33.2%and 18.9%of untransfected control, Survivin were decreased to 36.2%and 29.6%of untransfected control, Bcl-2 were decreased to 54.6%and 46.7%of untransfected control, transcriptional activity of NF-κB were decreased to 56.3%and 38.4%of untransfected control respectively inSKOV3/CDDP cell lines after Sorcin silence. Conclusion: Sorcin silencing could reverse SKOV3/CDDP drug resistance and enhance drug sensitivity, which involves the decreased phosphorylation level of Akt and transcriptional activity of NF-κB.

The extraction is the key operation step in the production of Chinese materia medica or the natural products.On the basis of brief description of traditional extracting technologies,extracting techniques appeared in recent five years are particularly reviewed in the principles,features,latest research and application results,and existing problems as well.These new techniques include supercritical fluid extraction(SFE),continuous countercurrent extraction(CCE),microwave-assisted extraction(MAE),ultrasound-assisted extraction(UAE),enzymatic treatment extraction(ETE),semi-bionic extraction(SBE),flooding extraction(FE),press extraction(PE),smashing tissue extraction(STE),heating-free extraction(HFE),air explosion extraction(AEE),ultra-high pressure extraction(UHPE),etc.Further research topics and application prospects of these extracting techniques are also suggested.

OBJECTIVE:To establish a method for contents determination of rutin,quercetin and total flavonoids in Buck-wheat,and compare the difference of flavone in buckwheat in different varieties and areas. METHODS:HPLC was adopted for content determination of rutin and quercetin:the column was Aglient C18 with mobile phase of methanol-0.4%phosphoric acid(gra-dient elution)at a flow rate of 1 ml/min,detection wavelength was 360 nm,column temperature was 40 ℃,and injection volume was 10 μl;UV spectrophotometry was adopted for content determination of total flavonoids:reference solution was methanol,de-tection wavelength was 360 nm,the standard was rutin. Statistical method was used to analyze the differences among the Fagopy-rum tataricum,F. esculentum and flavonoids of F. tataricum from different areas. RESULTS:The linear range was 0.400 9-16.03 μg for rutin (r=0.999 4) and 0.009 9-0.396 0 μg for quercetin (r=0.999 7),RSDs of precision,stability and reproducibility tests were lower than 2%,recoveries were 99.33%-104.00%(RSD=1.32%,n=9) and 96.92%-101.66%(RSD=1.60%,n=9),re-spectively. The linear range of total flavonoids(recovded by rutin)was 4.14-41.4 mg/L(r=0.999 9),RSDs of precision,stability and reproducibility tests were lower than 2,and recovery was 96.07%-101.96%(RSD=2.63%,n=9). The content of flavonoids on F. esculentum is significantly higher than in F. tataricum from the same area,and F. tataricum in Bijie area is better than other places. CONCLUSIONS:The method is simple,stable,reproducible,and can be used for the contents determination of rutin, quercetin and total flavonoids in F. esculentum. The study can provide theoretical basis for developing and using buckwheat.

8.The clinical significance of serum PARP-1 protein high expression in gastric cancer

Background and purpose:PARP-1 is closely related to malignant tumors. This study aimed to ex-plore the clinical significance of serum level of PARP-1 protein in onset and progression of gastric cancer.Methods:The serum samples from 145 patients with gastric cancer and 112 healthy check-up cases were collected. The serumHP spec-ificity IgA and PARP-1 protein levels were detected using enzyme-linked immunosorbent assay method. The correlation of serum PARP-1 protein levels with clinical characteristics of gastric cancer was analyzed.Results:Compared with healthy people, serum PARP-1 protein levels were significantly higher in gastric cancer patients [(407±139) pg/mLvs(258±120) pg/mL,P=0.014). Serum PARP-1 protein levels were significantly higher inHp(+) gastric cancer patients than those in patients withHp (-) (P<0.001). Serum PARP-1 protein levels were positively correlated with family gastric cancer history (P=0.033) and alcohol intake history (P=0.015) in gastric cancer patients. Compared with serum protein PARP-1 negative patients, PARP-1 protein positive patients had a significantly shorter cancer-free survival (P=0.011). However serum PARP-1 protein level was not found to be an independent risk factor for the overall survival of gastric cancer patients using multivariate COX regression.Conclusion:High expression of serum PARP-1 protein may be involved in the pathogenesis and progression of gastric cancer. Inhibition of PARP-1 may be potential new target for the treatment of gastric cancer.
9.Semicircular Canal Plugging Based on Endolymphatic Sac Decompression:The Surgical Technique Discussion for Intractable Meniere's Disease

Objective To study the effects of endolymphatic sac decompression and on semicircular canal plugging in treating intractable Meniere's disease.Methods A total of 14 cases of intractable Meniere's disease were included in this study.The age was from 33~67 years old and the history was from 1 to 40 years.The hearing level of bone was from 35 to 65 dB for the speech frequencies.All cases received the treatments according to the pre-op-erative design.The endolymphatic sac was decompressed and the three semicircular canal bones were drilled to cre-ate a fenestra followed by soft tissue plugging into the canal.The vertigo,hearing level and tinnitus were included in the following up.Results All cases had no facial palsy,no cerebrospinal leak,and no vertical after surgery.In the period of 3 to 18 months following up,the attack of Meniere's disease was completely controlled for 13 cases excep-tion of one lost case.For five cases,the hearing level descended 10 to 15 dB compared to pre-operation.Conclusion Although the endolymphatic sac decompression can relieve endolymphatic pressure,this may not adequately pro-tect the hearing caused by the semicircular canal plugging.The surgical technique is reliable and safe;however fur-ther clinical data should be gathered.
10.Comparative analysis on blood glucose results detected by portable glucose meter and Olympus AU2700 automatic biochemical analyzer

Objective To perform the comparative analysis on the blood glucose results detected by the portable glucose meter and the Olympus AU2700 biochemical analyzer.Methods 20 fresh whole blood samples with sodium fluoride and potassium oxalate anticoagulation were the Olympus AU2700 biochemical analyzer biochemical analyzer.Results In these 27 portable glucose meters, the comparison results in only 1 portable glucose meter was unqualified,which in the other 26 portable glucose meters were passed, the passing rate was 96.3%,which in 2 glucose meters were clinically acceptable and which in the other 24 glucose meters were completely acceptable and the clinical completely acceptable rate was 88.9%.Conclusion There are some biases of blood glucose results detected by the portable glucose meter and the fully automatic biochemical analyzer,but the overall correlation is well.The comparison and calibration of the blood glucose meters should be performed at the regular intervals for improving the consistency and accuracy of blood glucose detection results.
