1.Effect of sodium cantharidinate on the growth of human gastric cancer grafted onto nude mice

Objective To study the effect of sodium cantharidinate on the growth of human gastric cancer grafted onto nude mice. Methods Nude mice xenograft models of human gastric cancer were established.BGC823was injected peritoneal and the mice were weighed. The proliferating and apoptosis rates of xenografta was evaluated by TUNEL assay and immunohistochemical staining respectively. Results The xenografts were obviously inhibited with various dosage of sodium cantharidinate(P<0.01 ), the proliferating rate of turnout cells after using sodium cantharidinate was lower than that before using sodium cantharidinate(P<0.01 ), but apoptosis rate of tumour ceils after using sodium cantharidinate was higher than that before using sodium cantharidinate(P<0.01 ). Conclusion Sodium cantharidinate can inhibit gastric cancer growth by inhibiting tumour cell proliferating or inducing cell apoptosls.
2.The Application of MR Total Spine Mobitrak Scan in Fast Diagnosis of Acute Spinal Trauma

Objective To study the values of MR total spine mobitrack(MTSM) technique in acute spinal trauma and spinal cord injury.Methods 71 patients with trauma in cervical vertebra,thoracic vertebra and lumber vertebra underwent MTSM scan.Results All 71cases obtained clear images of total spine and spinal cord,of which,2 cases were normal,69 cases showed fractures of vertebrae(including 36 cases showed fracture of single vertebrae,33 cases showed fractures of multiple vertebraes).69 cases with fracture of vertebrae,accompanied with 12 articles of vertebral body slippage,the 15 segments of spinal cord injury and the 19 places of the ligaments bruise.Conclusion MTSM technique is good to show the location and quantity of vertebral trauma and will greatly improve the diagnostic accuracy.
3.Effects of Qingluo Tongbi Granula on Proliferation of Fibroblast-like Synoviocytes and RANKL Expression in Rheumatoid Arthritis Patients

Objective To observe the effect of serum containing Qingluo Tongbi Granula (QTG) on proliferation of fibroblast-like synoviocytes(FLS) and receptor activator of nuclear factor-?B ligand (RANKL) expression in FLS from patients with rheumatoid arthritis.Methods Synovial specimens obtained from the joint of patients were minced into small pieces for the culture of FLS.The morphological features of FLS subcultrued for 3~5 generations were observed under inverted microscope and electron microscope.The effect of QTG-containing serum on the proliferation of FLS was examined by MTT assay.The expression of RANKL in FLS was assessed by RT-PCR.Results After cocultured by serum containing large-dose QTG for 72 hours,proliferation of FLS and expression of RANKL were inhibited.Conclusion QTG can inhibit FLS proliferation,and reduce the expression of RANKL,which may be one of the therapeutic mechanisms of QTG for rheumatoid arthritis.
4.Effect of nucleos (t)ide analog antiviral treatment on the pathological differentiation and prognosis of ;hepatitis B virus-related hepatocellular carcinoma

Objective To explore the effect of nucleos(t)ide analog (NA)antiviral treatment on the pathological differentiation of hepatitis B virus (HBV)-related hepatocellular carcinoma (HCC)and the prognostic factors of HCC.Methods Totally 127 patients with HBV-related HCC who were hospitalized and received partial hepatectomy in First Affiliated Hospital of Harbin Medical University from March 2007 to November 2013 were included in this study.Sixteen cases received antiviral treatment before operation and the remaining 111 cases had no history of NA treatment.The differences of histopathological grading were compared between the two groups.Twenty-nine patients received antiviral treatment for the first time after surgery,and the rest 82 patients did not.All these patients were followed up for survival and recurrence.Multivariate analysis was used to explore the prognostic factors for HCC.The categorical variables were analyzed byχ2 test or Fisher exact test.Survival rate was compared with Log-rank test. Univariate or multivariate Cox regression analysis was used to explore the related factors of survival. Results The proportions of well-,moderately- or poorly-differentiated HCC in patients with antiviral treatment before surgery were 18.75 %,68.75 % and 12.5 %,respectively.Whereas the proportions in those without treatment were 16.22%,66.67% and 17.11 %,respectively.There was no significant difference in histopathological grading of HCC between the two groups (χ2=0.224,P =0.885 ).The overall median survival time was 39 months.The 6-month,1-and 2-year survival rates were 91 .7%, 77.5 % and 59.3%,respectively.The 6-month,1- and 2-year survival rate of postoperative antiviral treatment were 96.3%,92.4% and 78.5 %,respectively,which were significantly higher than those of no antiviral treatment group (85 .9%,70.0% and 48.5 %,respectively;χ2= 6.967,P = 0.008 ). Univariate analysis showed that tumor number,size,portal vein transfer,AFP level,postoperative antiviral treatment,histopathological grading,TNM staging,BCLC staging,γ-GT and PTA were prognostic factors for postoperative HCC survival.Multivariate analysis showed that AFP level (HR=1 , 95 %CI :1 .0004—1 .002,P =0.004),postoperative antiviral treatment (HR =0.38,95 %CI :0.38—0.15 ,P =0.04)and BCLC stage (B vs A:HR=1 .55 ,95 %CI :0.76—3.18;C vs A:HR=3.63,95 %CI :1 .31 —10.09,P =0.04)were independent prognostic factors.Conclusions Preoperative antiviral treatment has no impact on the histopathological grading of HCC. BCLC stage, AFP level and postoperative antiviral treatment are independent prognostic factors for HBV-related HCC.
5.Biomechanical and histological analysis of alveolar distraction osteogenesis in a canine model

BACKGROUND:Alveolar distraction osteogenesis is an important method for treating alveolar bone atrophy, the osteogenesis process and biomechanics play a crucial role in the fol owing implantation and repair. At present, no related experimental studies are found.
OBJECTIVE:To analyze the biomechanical and histological characteristics of alveolar distraction osteogenesis in a canine model.
METHODS:Twelve adult mongrel canines received premolars extraction and alveoloplasty in mandible to establish an atrophy alveolar model. After 3 months, a segmental alveolar osteotomy was performed in the randomly selected unilateral atrophy alveolar and two intra-osseous distractors were placed. After a 7-days latency period, the alveolar ridge was augmented at a rate of 1.0 mm/d for 5 days. After a consolidation of 1, 2, and 3 months, the canines were sacrificed and the specimens of the distracted alveolar bone were harvested for clinical, radiographic, histological and biomechanical analysis.
RESULTS AND CONCLUSION:The alveolar distractors obtained good healing with surrounding tissue. The atrophy alveolar bones were augmented for (4.80±0.50) mm and (5.12±0.47) mm by clinical and radiographic findings immediately after distraction, respectively. The bone trabeculae in the distracted chamber matured from 1 to 3 months of consolidation by histological analysis. The shearing force of alveolar distraction chamber increased from 1 to 3 months. After 3 months’ consolidation, the shearing force of distracted chamber was comparable to host bone. The histological and biomechanical property of distracted alveolar chamber is comparable to host bone after 3 months’ consolidation.
6.Molecular Cloning and Sequence Analysis of Rab11 GTPase in Trichomonas vaginalis

Objective Rab11 GTPases play an essential role in regulating membrane trafficking pathways in eukaryotic cells. Nonetheless, there has been little work done on characterizing the transport machinery of Trichomonas. The aim of this study is to clone and characterize a Rab11 gene of Trichomonas vaginalis.Methods A cDNA expression library was constructed with T. vaginalis total RNA. A cDNA clone, which showed a high degree of homology with Rab proteins of different species, was isolated and sequenced. Sequence analysis was performed using BLASTP, RPS-BLAST and ClustalW programs. The genomic DNA corresponding to the cDNA sequence was amplified using PCR techniques and following by sequencing. Results cDNA with a length of 710 base pairs and an open reading frame of 636 bp was obtained. The deduced amino acid sequence from the open reading frame was found to possess 211 residuals. Sequence analysis demonstrated that this cDNA clone was homologous to the Rab11 subfamily of different species (60% identity and 79% similarity with Arabidopsis thaliana Rab11c, 58% identity and 78% similarity with human Rab11b), and that the amino acid sequence contains all the well known conserved sequence elements of Rab family. Specific Rab motifs were also detected in the deduced amino acid sequence. Phylogenetic analysis showed that its closest homologues are Rab11 proteins from other species. Sequencing of the PCR product of genomic DNA revealed that the genomic DNA sequence encompassing the putative 5'-ATG and 3'-stop codon is identical to the cDNA sequence.Conclusion A cDNA clone corresponding to the T. vaginalis Rab11 gene was obtained.The function of this gene in regulating membrane trafficking pathways of the parasitic protist is still under investigation.
7. Analysis of 833 times measured hepatic venous pressure gradient

Objective:
To summarize and analyze the clinical data of hepatic venous pressure gradient (HVPG) and to explore the application value of HVPG in the diagnosis, evaluation and clinical treatment of portal hypertension in cirrhosis.
Methods:
The patient data of HVPG measurement performed in Shandong Provincial Hospital from April 2010 to November 2017 were collected.
Results:
A total of 633 patients with 833 times of HVPG measurements were included. There was significant difference in HVPG between patients with different etiologies, different Child-pugh grades and different degrees of decompensated cirrhosis.
Conclusion
The HVPG test is suitable for the diagnosis and evaluation of portal hypertension. The HVPG of patients with different severity of liver cirrhosis can guide the choice of the treatment plan, and the HVPG measurement should also be strictly standardized and quality control.
8.Progress of immunotherapy strategies for uveal melanoma

Uveal melanoma (UM) is the most common intraocular malignancy in adults. Recently, great progresses have been made in the diagnosis, treatment and prognosis of UM, however, nearly 50% of patients still develop liver metastases, which severely affects on the survival of UM patients. Whether UM patients will benefit from the immune checkpoint blockade similarly as the cutaneous melanoma (CM)? Whether the specific gene mutations targeting UM could improve the anti-tumor efficacy? Whether chimeric antigen receptor T cell or T cell receptor T cell immunotherapy is effective to UM patients with liver metastases? How about the combinational therapies in UM and the clinical effects? This review summarizes the anti-tumor research and novel treatment options of UM, analyzes the current achievements and problems.
9.Efficacy comparison between endoscopic resection and tradition surgery in the treatment of early gastric cancer.

Endoscopic resection (ER) has been a preferred treatment for early gastric cancer (EGC) based on the guidelines. In recent years the technique is developing rapidly. Surgical operation, a traditional treatment, still has its advantages such as complete tumor removal and low recurrence rate. In this review, a long-term outcome comparison of these two methods in EGC is discussed.

OBJECTIVETo investigate the neuroprotective effects of the constituents in Herba Erigerontis on neuroblastoma SH-SY5Y cells, and find out its possible material foundation in treating Alzheimer's disease (AD).
METHODDifferent combinations of the three constituents in Herba Erigerontis were prepared according to the orthogonality experiment, and the indexes (MTT reduction assay, lipid peroxidation and expressions of nAChR alpha7 protein)were observed upon the SH-SY5Y cells followed by treatment of these combinations and beta-amyloid peptide (AP). The pharmacology data thus obtained and peak data in UPLC fingerprint were analyzed through ANOVA and correlationship by SPSS to give the information of active possible material foundation.
RESULTConstituents B and C showed clear activity and peaks of 4, 7-12 did positive correlationship according to the correlation of fingerprints and pharmacology.
CONCLUSIONThis study makes a valid approach for deducing the active constituents even the exact compounds against neurotoxicity induced by Abeta by correlation of fingerprints and pharmacology.
