1.Effect of Yiqi Fuzhi Granule in Improving Learning and Memory of Rats with Multi-infarct Dementia

Objective To investigate the eitect oi Yiqi Fuzhi Granule in improving learning and memory of rats with mul- ti-infarct dementia (MID). Methods MID rat models were established by embolus injection through the common carotid artery. Effects of Yiqi Fuzhi Granule(24. 84 g/kg, 12.42 g/kg, 6.21 g/kg, ig)on rat learning and memory and cerebral structure were observed. Results Yiqi Fuzhi Granule could obviously increase the scores of maze test and protect cerebral tissue from damage of multi-infarction rats. Conclusion Yiqi Fuzhi Granule has good effect in improving intellect and treating dementia.

3.Common characteristic of papillary microcarcinoma of thyroid in ultrasound and pathology

Objective To explore the pathological basic of some common characteristic of papillary microcarcinoma of thyroid in ultrasound. Methods Totally 117 thyroid papillary microcarcinomas that were completed all thyroid ultrasound examinations before the surgical excisions were collected. Shape, border, internal echogenicity, halo, posterior echogenicity and calciifcation were observed in ultrasound. While the degree of inifltration of the tumor, the tumor’s internal structure as well as calciifcation was observed in pathology. Results IIrregular shape (87.18%, 102/117), unclear border (80.34%, 94/117), hypoechoic (90.60%, 106/117) and microcalciifcations (65.81%, 77/117) were recognized by ultrasound while the main pathological features were invasive growth (93.16%, 109/117), cell components internally (46.15%, 54/117) and calciifcation (56.41%, 66/117). Of the 109 neoplasms which were invasive growth in pathology, 102 (93.58%) nodules presented irregular shape in ultrasound while 91 (77.78%) presented unclear border. All of the 8 neoplasms which were not invasive growth in pathology showed regular shape in ultrasound. Thirteen nodules showed halos in ultrasound, and 8 of them showed visible ifbrous capsule surrounding the mass. Whatever the tumor’s internal structure was, most nodules presented hypoechoic. And the hyperechoic nodules were all found cell components internally. Of the 66 neoplasms which were found calciifcation by microscope, 35 (53.03%) nodules showed calciifcation in ultrasound and 31 (46.97%) did not. And of the 51 neoplasms in which calciifcation were not found by microscope, 42 (82.35%) nodules showed calciifcation in ultrasound and 9 (17.65%%) did not. Conclusions There is no doubt that all the performances of papillary microcarcinoma in ultrasound have bases in pathology. It is helpful to the judgment of a thyroid nodule if we could understand the correlation of performances in ultrasound and pathology.

5.An epidemiological study of functional bowel disorders in Zhejiang college students and its relationship with psychological factors

Objective To study the epidemiologic features of functional bowel disorders (FBD)among medical students and science and engineering students in Zhejiang province,China,and to explore the relationship between psychological factors and FBD.Methods This study was a survey of random sampling based on the Rome Ⅲ Diagnostic Questionnaire ( Rome Ⅲ -DQ) conducted from November 2010 to March 2011.The survey was carried out among medical students and science and engineering students respectively in two universities in Zhejiang province.Symptom Check List 90 ( SCL-90 ) was used for psychological analysis.Results Among 1870 participants,1033 were diagnosed as FBD based on Rome Ⅲ criteria,with an overall incidence of 55.24%.The majority cases were unspecified functional bowel disorder,with an incidence of 26.58% (497/1870),others were functional constipation 16.95%(317/1870),irritable bowel syndrome 6.90% ( 129/1870),functional bloating 4.12% (77/1870) and functional diarrhea 0.70% (13/1870).The prevalence varied in different genders,majors and grades.In the multivariate analysis,women had a higher risk of any FBD than men [ 68.33% (617/903) vs 43.02%(416/967) ] except for functional diarrhea; and medical students had a higher risk of any FBD than science and engineering students [ 68.53% ( 734/1071 ) vs 37.42% ( 299/799 ) ].Generally higher grade undergraduates had relatively higher risks than lower grade students,especially in medical students.Among the participants with FBD,44.05% (455/1033) had other functional gastrointestinal disorders.In those overlapped parts,9.00% ( 93/1033 ) had belching disorders,and 8.23% ( 85/1033 ) had functional dyspepsia.Students with FBD got higher scores in SCL-90 than healthy students.Conclusions This study revealed a high rate of FBD among college students in Zhejiang province.The prevalence varied in different majors,and female higher than male.Psychological factors also had close relationships with FBD.
6.An epidemiological survey of functional dyspepsia and its relationship with psychological factors in college students of Zhejiang Province

Objective To investigate the prevalence of functional dyspepsia (FD) and its relationship with psychological factors in college students of Zhejiang Province. Methods The students of two colleges in Zhejiang Province were selected.The multi-stage stratified random cluster sampling method was conducted.The Rome Ⅲ adult FD diagnosis questionnaire (ROME Ⅲ-DQ) and psychological symptoms self rating scale questionnaire (SCL-90) was applied in the investigation.The data were analyzed by x2 test and t test.Results A total of 1870 college students of Zhejiang Province were surveyed.The prevalence of FD was 5.78%.The prevalence in female was higher than that in male (7.53 % vs 4.14 %,x2 =9.884,P<0.05).The prevalence in senior students was highest in all grade (x2 =13.83,P<0.05).In FD subtypes,postprandial distress syndrome was more than epigastric pain syndrome.The functional constipation and functional defecation disorders were most common in the overlap with other functional gastrointestinal disorders. In SCL-90 questionnaire survey,the scores of each factor of FD group were higher than those of the healthy control group.Conclusions The prevalence of FD was high in the college students of Zhejiang Province. The incidence was correlated with psychological factors.
7.Transplantation of bone marrow mesenchymal stem cells harboring fusing gene of enhanced green fluorescent protein and human insulin gene in treatment of diabetic mice

Objective:To construct a retroviral vector carrying both enhanced green fluorescent protein(EGFP) and human insulin gene for transfecting the bone marrow mesenchymal stem cells (BMSCs),and to observe the treatment effect of transfected BMSCs after transplanted into diabetes mice.Methods:Two gene segments of insulin-IRES-EGFP and IRES-EGFP were obtained by overlap extension PCR technology,and then the 2 segments were cloned into retroviral vector (pMSCV).The vectors were used to transfect BMSCs after packaged with PT67 cells.The expression of EGFP was observed by inverted fluorescence microscope and the expression of insulin gene was examined by RT-PCR in the infected BMSCs.The transfected BMSCs with 2 viruses were transplanted into diabetic mice separately.The blood glucose levels and body weights of mice were examined in 2 groups,and the results were compared with those of normal control group.Results:The recombinant retroviral vectors pMSCV-insulin-IRES-EGFP and pMSCV-IRES-EGFP were successfully constructed.The BMSCs infected by both vector stably gave out green fluorescence under microscope; the cells infected with pMSCV-insulin-IRES-EGFP also stably expressed human insulin gene.The blood glucose level of the mice transplanted with BMSCs transfected with pMSCV-insulin-IRES-EGFP was significantly lower than that transplanted with BMSCs transfected with pMSCV-IRES-EFGP (P
8.Effects of Qingluo Tongbi Granula on Proliferation of Fibroblast-like Synoviocytes and RANKL Expression in Rheumatoid Arthritis Patients

Objective To observe the effect of serum containing Qingluo Tongbi Granula (QTG) on proliferation of fibroblast-like synoviocytes(FLS) and receptor activator of nuclear factor-?B ligand (RANKL) expression in FLS from patients with rheumatoid arthritis.Methods Synovial specimens obtained from the joint of patients were minced into small pieces for the culture of FLS.The morphological features of FLS subcultrued for 3~5 generations were observed under inverted microscope and electron microscope.The effect of QTG-containing serum on the proliferation of FLS was examined by MTT assay.The expression of RANKL in FLS was assessed by RT-PCR.Results After cocultured by serum containing large-dose QTG for 72 hours,proliferation of FLS and expression of RANKL were inhibited.Conclusion QTG can inhibit FLS proliferation,and reduce the expression of RANKL,which may be one of the therapeutic mechanisms of QTG for rheumatoid arthritis.
9.Effects of cyclooxygenase-2 on immunologic function of rats with sepsis

Objective To study the expression of cyclooxygenase-2(COX-2)in hepatic inflammatory reaction and the effects of COX-2 on immunologic function of rats with sepsis.Methods Fifty-four Wistar rats were divided into sham group(n=6),sepsis group(n=24),and NS-398 intervention group(n=24).All rats were subjected to cecal ligation and puncture or sham operation.The expression of COX-2 mRNA in rat hepatic tissue was determined by RT-PCR,serum levels of IL-6,IL-10 and TNF-α were detected by ELISA,and percentage changes of CD4+,CD8+ cells by flow eytometry.The pathological changes of liver were observed at the same time.Results (1)Severe pathologic injuries of liver were observed in sepsis group,while not in NS-398 intervention group.(2)The expression of COX-2 mRNA was up-regulated in sepsis group and NS-398 intervention group,and the expression value was higher in sepsis group than that in NS-398 intervention group.The expression of COX-2 mRNA was the lowest in sham group.(3)The level of IL-6 was higher in sepsis group than that in sham group and NS-398 intervention group(F=125.582,134.712,54.760,121.441,P<0.05).(4)The level of IL-10 was higher in NS-398 intervention group than that in sham group and sepsis group(F=39.064,34.382,51.115,8.174,P<0.05).(5)The levels of TNF-α in sepsis group and NS-398 intervention group were increased,and the difference between the 2 groups had no statistical significance(x2=5.600,6.162,7.136,7.200,P>0.05).(6)The ratio of CD4+to CD8+ was higher in NS-398 group than that in sepsis group(F=17.448,15.055,30.068,64.210,P<0.05).Conclusions COX-2 plays an important role in the development of sepsis by changing the dynamic equilibrium between pro-inflammatory and anti-inflammatory cytokine and that between CD4+and CD8+.
10.Effect of chitooligosaccharides on cyclin D1, bcl-xl and bcl-2 mRNA expression in Hela cells

BACKGROUND: Studies have shown that chitooligosaccharides have antitumor effect. However, the influence of chitooligosaccharides on cyclin D1, bcl-2 and bcl-xl remains unclear.OBJECTIVE: To observe the JnhJbJtJon effect of chitooligosaccharides on the proliferation of Hela cells, and the influence on cyclin D1, bcl-xl and bcl-2 mRNA expression.METHODS: Hela cells were stimulated by different concentrations of chitooligosaccharides (0.1, 0.5, 1, 2, 5 g/L). The effects of chitooligosaccharides on Hela cells were detected by CCK8 kit. Using real-time PCR methods, the gene expression of cyclin D1, bcl-xl and bcl-2 mRNA were determined.RESULTS AND CONCLUSION: Chitooligosaccharides inhibited the proliferation of Hela cells. With the concentrations of chitoolJgosaccharides increased from 0.1 g/L to 2 g/L, the inhibition effects on the gene expressJon of cyclin D1, bcl-xl and bcl-2 were enhanced, peaked at 2 g/L, and decreased at high concentration (5 g/L). Antitumor activity of chitooligosaccharides may exert through two aspects: it inhibits cyclin D1 mRNA expression to suppress the proliferation of tumor cells; on the other hand, chitooligosaccharides inhibits the expression of bcl-xl and bci-2 to promote the apoptosis of tumor cells. Moreover, the effects of the former are stronger than the latter.
