1.Accuracy of BIS and CSI for monitoring levels of sedation induced by different effect-site concentrations of propofol during TCI of propofol combined with sufentanil

Objective To evaluate the accuracy of BIS and anesthetic depth index (CSI) for monitoring levels of sedation induced by different target effect-site concentrations (CT) during TCI of propofol combined with sufentanil. Methods Ninety ASA Ⅰ or Ⅱ patients of both sexes aged 20-49 yr weighing 45-70 kg undergoing elective surgery under general anesthesia were randomly divided into 6 groups (n=15 each): group Ⅰ, Ⅱ, Ⅲ TCI of propofol with CT set at 2, 4 and 6 μg/ml respectively (P1-3);groupⅣ, Ⅴ,Ⅵ sufentanil 0.7 μg/kg + propefol TCI with CT set at 2, 4 and 6 μg/ml (SP1-3). Anesthesia was induced with propefol TCI with CT set at 4 μg/ml in all 6 groups. As soon as the patients lost consciousness, tracheal intubation was facilitated with vecuronium 0.1 mg/kg. The patients were mechanically ventilated. PET CO2 was maintained at 35-45 mm Hg. Anesthesia was maintained with propofol TCI with CT set at 2 μg/ml(in group P1, SP1), 4 μg/ml(in group P2, SP2) and 6 μg/ml(in group P3,SP3) immediately after intubation respectively. Sufentanil 0.7 μg/kg was given iv at 20 min after propofol TCI was started in group SP<1-3. MAP, HR, BIS (Aspect) and CSI (Danmeter Denmark) were continuously monitored and recorded before induction of anesthesia (T0, baseline), at 1 min before tracheal intubation (T1), and at 30 s(T2), 15 min(T3), 30 min(T4), 35 min(T5) and 40 min (T6) after tracheal intubation. Results BIS and CSI values were gradually decreasing at T3-6 in group P1-3 and SP1-3. BIS and CSI values were significantly lower at T4-6 in group SP1 and SP2 than in group P1 and P2. CSI values were significantly lower at T4-6 in group SP3 than in group P3, but there was no significant difference in BIS values at T4-6 between SP3 and P3. Conclusion CSI and BIS can monitor the levels of sedation indueed with TCI of propofol with CT set at 2 and 4 μg/ml when combined with sufentanil 0.7 μg/kg but only CSI can monitor the level of sedation induced by propofol TCI with CT set at 6 μg/ml when combined with sufentanil 0.7 μg/kg.
3.Effects of Dexmedetomidine on the Blood Glucose, TNF-α and IL-6 in Perioperative Patients with Neurosurgery

Objective:To explore the effects of dexmedetomidine on the blood glucose,tumor necrosis factor-α (TNF-α) and interleukin-6(IL-6) in perioperative patients with neurosurgery.Methods:50 patients with meningioma who were ready to accept craniotomy in department of neurosurgery of our hospital from February 2016 to November 2016 were enrolled,they were randomly divided into research group and control group,with 25 patients in each group.The research group was given DEX 1.0 μg/kg at 10 min before anesthesia,which was added in 0.9% sodium chloride injection 50 mL,the injection was finished in 10 min,then the DEX change to the rate of 0.5 μg/kg/h.Control group was given 0.9% sodium chloride injection with constant rate and volume.Record heart rate (HR),mean arterial pressure (MAP) and blood glucose,serum TNF-α,IL-6 levels of the two groups at 10min before anesthesia(T1),after the anesthesia (T2),the beginning of surgery(T3),1 hour after the surgery beginning(T4),the end of surgery(T5).Results:Compared with T1,HR of the two groups at T2 and T3 raised significantly,but research group was lower than control group(P<0.05);In research group,compared with T1,MAP at T3 raised significantly;in control group,compared with T1,MAP at T3 and T4 raised significantly,but MAP of research group at T3 and T4 were lower than control group (P<0.05).Compared with T1,blood glucose of the two groups at T2,T3,T4 and T5 raised sig-nificantly,but the research group at same points were lower than control group (P<0.05).Serum TNF-α,IL-6 levels of control group at T2,T3 and T4 were significantly higher than T1 (P<0.05),which in research group at different time points had no significant change (P>0.05),and were lower than control group at T2,T3 and T4 (P<0.05).Conclusion:DEX could maintain stable hemodynamics,antiinflammation,inhibit elevated blood glucose,so as to reduce the inflammatory response and stress response in patients with neurosurgery,then it could promote these patients postoperative recovery.
4.Investigation of application and administration of hyperbaric oxygen chambers in Beijing

Objective To investigate the application and administration of hyperbaric oxygen chambers in Beijing.Methods Forty-eight hospitals with hyperbaric oxygen chambers in Beijing were investigated by field investigation,letter investigation or telephone investigation.The investigation includes grades of hospital,personnel,equipment,therapy of severe cases,time of therapy and treatment plans in questionnaire.Results Hyperbaric oxygen was popular in Beijing,which ensured emergency treatments.But the scale was little relatively and the number of full-time technicians was less,besides,the therapeutic system and plans were diversified,which should be standardized.Conclusions Something must be done to improve the conditions of hyperbaric oxygen medicine in Beijing,such as cultivating personnel,standardizing therapeutic system,standardizing treatment plans,realizing computer control and so on.
5.Study on the mechanism of proliferative inhibition of arsenic trioxide and rhizoma curcumae on lens epithelial cell by MTT assay

Objective To investigate the proliferative inhibition of lens epithelial cell (LEC) by arsenic trioxide (ATO) and rhizoma curcumae (RC), in order to provide scientific basis for pursuing safe and effective natural drugs to prevent and cure after cataract. Design Experimental study. Participants Cultured Bovine LEC in vitro. Methods Changes of cellular form were observed with microscope. Different concentration of ATO (2.5, 5, 10?mol/L) and RC(5, 10, 20mg/ml) were added to the proliferative LEC separately. The effects of proliferative inhibition by ATO and RC on the LEC were measured with methyl thia-zolyl tetrazolium (MTT) assay method after 72 hours incubation. Main Outcome Measures Cellular form, Absorbance. Results Under the microscope, good growth and quantity increase were found in proliferative control group. Slowing proliferation,poor growth, and few disintegration were observed in ATO group and RC group. In the magnetic induction tomography(MIT) system,the electrical conductivity of biological tissue is direct proportion to the phase difference between the excitation signal and the detection signal.To obtain the image of the contribution of tissue's electrical conductivity,the system must have the function of phase detection with high accuracy.The paper focuses on the means of digital phase detection,including FFT method,the correlation method and the classic method,which are ultimately compared with analogue phase detection method.The experimental results show that FFT method and the correlation method,with low error level and high linearity,can better detect the phase difference with the level of 0.1?.The digital phase difference detection provides a kind of effective method for MIT system.
7.Signal transduction mechanism of curcumin in inhibiting the proliferation of bovine lens epithelial cell induced by recombinant human epidermal growth factor

OBJECTIVE: To investigate the signal transduction mechanism of curcumin in inhibiting the proliferation of bovine lens epithelial cell (LEC) induced by recombinant human epidermal growth factor (rhEGF). METHODS: There were three groups in this experiment, which were normal control group, untreated group and curcumin-treated group. Proliferation of LEC was induced by rhEGF (50 microg/L). The concentration of intracellular Ca(2+) ([Ca(2+)](i)) in LEC was measured with Fura-2/AM by spectrofluorimetry. The contents of intracellular cAMP and cGMP were assayed by radioimmunoassay. RESULTS: The [Ca(2+)]i in LEC was obviously increased in the untrated group as compared with that in the normal control group (P<0.01), and the [Ca(2+)](i) in LEC in the curcumin-treated group was highest among three groups (P<0.01). The content of intracellular cAMP in LEC was decreased while the content of intracellular cGMP was obviously increased in the untreated group as compared with those in the normal control group (P<0.01). The content of intracellular cAMP in LEC was higher in the curcumin-treated group than that in the untreated group, while the content of intracellular cGMP was lower than that in the untreated group (P<0.01). CONCLUSION: The antiproliferation effects of curcumin on LEC may relate to the regulations of multiple processes of signal transduction.
8.Development and application of simple gas dynamic exposure device

Objective To develop a gas dynamic exposure device with a simple structure.Methods The device was composed of a water tank, an gas chest, an exposure kit and an gas-out bottle. The chest body was made of organic glass pane, drainage method was used to distribute the gas properly, and airflow was regulated with the gravity and valve.Results The device was proved to have simple structure and high effects.Conclusion The device has low cost, simple structure and properly distributed gas, and can be reused for exposure test.
9.Deficiency of rpoS is the major factor leading to attenuation of Salmonella enterica serovar Choleraesuis vaccine strain C500

Salmonella enterica serovar Choleraesuis strain C500 is a live attenuated vaccine that has been widely used in Chi‐na for over 50 years to prevent piglet paratyphoid .However ,as C500 is obtained by chemical methods ,the genetic background of this strain remained unclear .In this study ,we compared the genomic differences between the virulent reference strain C 78‐2 and C500 by suppression subtractive hybridization combined with the mirror orientation selection method (MOS‐SSH ) .Six genes (asr ,ydgF ,ydgD ,ydgE ,rpoS ,and ptsG) were lost in C500 strain .Using real‐time PCR analysis ,we demonstrated that the genes regulated by rpoS ,a vital transcriptional regulator playing an important role in Salmonella infection ,were downregulated in C500 .Additionally ,the virulence of the rpoS mutant strain C78‐2ΔrpoS was 100 000 times lower than the parental strain in BALB/c mice .So loss of rpoS gene is the major factor leading to the attenuation of C500 strain .
10.Changes of protein kinase regulatory pathway in inhibition of curcumin on human lens epithelial cells proliferation

Background Our previous study showed that curcumin suppresses the proliferation of human lens epithelial cells (LECs) in vitro and has an influence on the signal transduction of cyclic adenosine monophosphate (cAMP) and cyclic guanosinc monophosphate (cGMP).Actually,the regulation for biological behavior of LECs in vivo is complex.Objective This study was to investigate the changes of signal transduction of protein kinase (PK) in inhibition of curcumin on human LECs proliferation.Methods HLE-B3 was cultured and then divided into the blank control group,recombinant human basic fibroblast growth factor (rhbFGF) group and rhbFGF+ curcumin group.rhbFGF of 10 ng/ml was added in the medium in the rhbFGF group,and 20 mg/L curcumin was added into rhbFGF-induced cell medium for 24 hours in the rhbFGF+ curcumin group.The expression rates of PKA,PKC,PKG and calmodulin (CaM) in the cells were assayed using flow cytometry.Results The expression rates of PKA protein were (46.847± 1.673) %,(33.250 ± 2.242) % and (71.645 ±2.432) % in the blank control group,rhbFGF group and the rhbFGF+ curcumin group,respectively,and the expression rate of PKA protein was significantly reduced in the rhbFGF group compared with the blank control group (t =11.904,P<0.01),but the expression rate of PKA protein in the rhbFGF+ curcumin group was significnatly higher than that in the rhbFGF group (t=28.430,P<0.01).In the blank control group,rhbFGF group and the rhbFGF+ curcumin group,the expression rates of PKC protein in the cells were (35.575± 1.937) %,(50.652±2.068) % and (27.662t4.481) %,those of PKG protein were (63.838±0.486) %,(86.562 ± 1.325) % and (40.733 ± 2.175) %,while those of CaM protein were (67.408± 1.391)%,(83.887±3.499)% and (53.785 ± 1.942)%,the expression rates of PKC,PKG and CaM were significantly lower in the rhbFGF group in comparison with the blank control group (all at P<0.01),and those in the rhbFGF+ curcumin group were significantly declined in comparison with the rhbFGF group (all at P<0.01).Conclusions Suppression of curcumin on HLE-B3 proliferation probably is associated with the up-regulation of PKA expression and down-regulation of PKC,PKG and CaM expression in the cells.
