1.The roles of hypoxia-inducible factor-1 alpha and vascular endothelial growth factor in the pathogenesis of hypoxia-induced pulmonary hypertension in neonatal rats

Objective To studytherolesof hypoxia-inducible factor-lalpha( HIF-1α )and vascular endothelial growth factor ( VEGF) in the pathogenesis of hypoxia-induced pulmonary hypertension ( HPH) in neonatal rats.Methods Wistar neonatal rats were assigned into HPH group and the control group using random number table method. Neonatal rats in HPH group were exposed to hypoxia according to HPH model. On day 3 , 7 ,14 and 210f hypoxia,the mean pulmonary artery pressure ( mPAP) , the level of mRNA and protein expression of HIF-1α and VEGF in lung tissue were examined using RT-PCR and Western blot methods respectively. The correlation of HIF-1,VEGF and mPAP were also analyzed. Results The mPAP ( mmHg)in HPH group on day 3 ,7 ,14 and 210f hypoxia were all higher than the control up[8.5±1.5)vs.5.2±1.0),(12.1±2.1)vs.9.6±0.8),(12.9±2.0)vs.(9.1±0.8),(21.0±2. 3) vs. (11. 2 + 1. 6) ,P < 0. 05]. 0n day 3, 7 and 140f hypoxia,the mRNA of HIF-1α in lung tissue of HPH group were significantly higher than the control group ( P <0. 05 ) . On day 70f hypoxia, the HIF-1αprotein in lung tissue of HPH group was significantly higher than the control group(P <0. 05 ) . On day 7 ,14 and 210f hypoxia,the mRNA and protein of VEGF in lung tissue of HPH group were also significantly higher than the control group( P <0. 05 ). Correlation analysis showed that HIF-1α protein were positively correlated with mPAP on day 3 ,7 . 14 and 210f hypoxia in HPH group ( r = 0. 504 .P = 0. 002) , and VEGF protein were positively correlated with mPAP in HPH group on day 7 , 14 and 210f hypoxia( r = 0. 782, P < 0. 001) .Conclusion Both HIF-1α and VEGF play roles in the occurrence and development of HPH in neonatal rats.
2.Effect of magnetic field simulating electroencephalogram rhythm on memory

BACKGROUND: Recently, many researches suggest that transcranial magnetic stimulation has effect on learning and memory. Moreover, the effects of magnetic fields simulating electroencephalogram (EEG) rhythm on memory have been investigated.OBJECTIVE: To explore the effect of magnetic field simulating EEG rhythm on memory retrieval ability and its mechanism.DESIGN: Randomized controlled study.SETTING: Department of Medical Imaging, Tianjin Medical University,and School of Precision Instrument & Photoelectronic Engineering, Tianjin University.MATERIALS: The experiment was conducted at the Electroscopic Center of TianjSn Medical University and Laboratory of Biomedical Engineering Department of Tianjin University from February 2002 to February 2004.Totally 25 Wistar rats and 5 domestic cats were selected from the Experimental Animal Center of Tianjin Medical University.INTERVENTIONS: All the 25 Wister rats were randomly divided into 5 group, 4 experimental groups and one control group with 5 rats in each group. Magnetic field simulating EEG rhythm was used to stimulate Wistar rats in the 4 experimental groups, but the time and frequency rate were different. Group 1 was stimulated by weak magnetic field with frequency in α rhythm (10 Hz); Group 2 was stimulated by weak magnetic field with frequency in β rhythm (20 Hz); Group 3 was stimulated by weak magnetic field with frequency changing from α to β rhythm; and Group 4 was stimulated by weak magnetic field with frequency changing from β to α rhythm. Memory retrieval ability and changes of neurotransmitters in the brain were recorded. Five cats were treated with weak magnetic field with frequency changing from α to β rhythm for 20 minutes, and the changes of EEG power spectra before and after stimulation were compared.information from long-term memory was tested with modified cross maze;hippocampus of the rats was measured with high performance liquid multiple-channel electrophysiological recording device.Comparison of memory retrieval ability: Memory retrieval ability in Group 2 and Group 4 was obviously higher than that in control group 5 hours after Content ofdopamine (DA) in rats' cerebral hippocampus: It was obviously higher in the 4 experimental groups than in control group [(192.72±18.66,percentage of power spectrum of β rhythm was increased after stimulation [(23.0±3.4, 14.2±1.3) %.].CONCLUSION: Most of the weak magnetic fields simulating EEG rhythm of human brain can decrease the retrieval of long-term memory, but magnetic field with special rhythm may increase memory retrieval ability.All the effects are related to the release of hippocampal neurotransmitters which is mediated by the change of intracellular calcium ([Ca2+]i) and synaptic vesicle.
3.Effect of modulated pulse magnetic fields on memory retention and retrieval ability of rats and its mechanism

Objective To explore the effect and the mechanism of modulated magnetic pulse fields on memory retention and retrieval ability of rats. Methods The influence of different pulsed magnetic fields on memory performance of rats was evaluated by using cross maze test, the morphological changes of neurons and nerve synapses in the hippocampus of rats were observed by the electronic microscopy. Results The 20 Hz modulated-pulse magnetic field impaired the memory retrieval ability and the memory retention significantly after 1 hour of exposure of the rats to it ( P 0.05). These influences on nerve tissues were also observed in rats underwent the 10Hz modulated magnetic stimulation, manifested as an initial increase in ability of memory in the first 24 hours after the stimulation, but no significant difference from the control was found. The influence of magnetic fields on memory depended on what the rats remembered. Conclusion Changes in the nerve synapses may be a main factor of mechanisms of the influence of pulse magnetic fields on memory ability of rats.
4.Effects of Different Extracts ofTangwang Mingmu Granules on High Glucose Induced VEGF and IL-1α Expressions in Vascular Endothelial Cells

Objective To observe the mechanism of action of different extracts ofTangwang Mingmu Granules on high glucose induced VEGF and IL-1α gene and protein expressions in vascular endothelial cells.Methods Human umbilical vein endothelial cells EA.hy926 were divided into six groups: blank, high glucose,Tangwang Mingmu Granules, extract 1 (glycoside and flavonoid), extract 2 (organic acid and polysaccharides) and extract 3 (alkaloids) groups. High concentration glucose was used to establish the high glucose model in EA.hy926 cells. The expressions of VEGF and IL-1α mRNA were detected by semi-quantitative RT-PCR. The contents of VEGF and IL-1α protein were tested by ELISA.Results The gene expression and protein levels of VEGF and IL-1α were significantly up-regulated under the high glucose condition (P<0.05). However, the above indicators were significantly reduced after the treatment ofTangwang Mingmu Granules. The activity of different parts ofTangwang Mingmu Granules was as follows: extract 1> extract 3> extract 2.Conclusion The action intensity of glycosides and flavonoids, alkaloids, organic acids and polysaccharides on VEGF and IL-1α expression inTangwang Mingmu Granules weakens in sequence.

ObjectiveTo observe the effect of different extracts ofTangwang Mingmu Granule on hypoxia induced gene expressions in vascular endothelial cells.Methods COCl2 intervention cells were used to copy hypoxia models. Human umbilical vein endothelial cells EA. Hy926 were divided into blank group, hypoxia model group,Tangwang Mingmu Granule group, extract 1 (glycosides and flavonoids) group, extract 2 (orgain acids and polysaccharides) group and extract 3 (alkaloids) group. The changes in gene expressions of VEGF, VEGFR-1, VEGFR-2, ICAM-1 and IL-1α mRNA were detected by semi-quantitative RT-PCR.ResultsThe gene expression levels of VEGF, VEGFR-1, VEGFR-2, ICAM-1 and IL-1α were significantly up-regulated under the hypoxic condition (P<0.05), and the ratio of VEGFR-1/VEGFR-2 was significantly reduced. However,Tangwang Mingmu Granule significantly reversed the expressions of these genes.Conclusion The function intensity of gene expressions weakens in the following sequence:glycosides and flavonoids>alkaloids>organic acids and polysaccharides inTangwang Mingmu Granule.
6.The distribution of gene polymorphism of methylenetetrahydrofolate reductase and methionine synthase among Han women in Zhangjiagang city

Objective To explore the 5,10-methylenetetrahydrofolate reductase (MTHFR) and 5-methyltetrahydro-folate-homocysteine methyltransferase reductase ( MTRR ) gene polymorphisms among the Han women in Zhangjiagang city .Methods A total of 4008 Han women were recruited .Their oral epithelial cells were collected to extract genome DNA in order to detect gene polymorphisms of MTHFR and MTRR using fluorescence quantitative PCR.Then the results were compared with those in other cities in China .Results The genotype frequencies of MTHFR C677T CC, CT and TT among Zhangjiagang women are 32.2%, 49.5%and 18.3%, respectively.The C allele frequency is 64.3%, T allele frequency is 35.7%.The genotype frequencies of MTHFR A1298 C AA, AC and CC are 68.7%,28.7% and 2.5%, respectively .The A allele frequency is 80.8%, C allele frequency is 19.2%.The genotype frequencies of MTRR A66 G AA, AG and GG are 54.4%, 38.5%and 7.1%, respectively . The A allele frequency is 76.3%, G allele frequency is 26.4%.Conclusions The MTHFR, MTRR polymorphism distribution of Han women in Zhangjiagang city is region specific.
7.Antimicrobial Resistance of Acinetobacter baumannii and Clinical Strategies

OBJECTIVE To analyze the antimicrobial resistance of Acinetobacter baumannii.METHODS A.baumannii was collected in our hospital from Jan 2003 to Dec 2006.Antimicrobial susceptibility testing was performed by disk-diffusion method.RESULTS A total of 548 A.baumannii were collected during 4 years.Of these isolates,130(23.7%) strains were from intensive care unit(ICU),190 strains(34.7%)from surgical,and others from medical and emergency departments.Most of strains(78.5%) were isolated from sputum,next from wound.The prevalence of A.baumannii was increasing during last 4 years.Results of susceptibility test showed that imipenem was the most active antibiotic against A.baumannii.The resistance rates were high to most antibiotics.Most of the resistant strains was from the ICU and the general ward differentand had significant difference.More than 50% of isolates were resistant to all antimicrobial agents tested except imipenem and ciprofloxacin in ICU.CONCLUSIONS The prevalence of A.baumannii is increasing.A.baumannii isolates show high resistance to multiple antibiotics,especially in ICU.
8.Correlation of Lactate Dehydrogenase and Neuron?specific Enolase with Prognosis of Extensive Small Cell Lung Cancer

Objective To determine whether neuron?specific enolase(NSE)affects the prognosis of extensive small cell lung cancer by analyz?ing the association between NSE before treatment and disease progression and overall survival of patients. Methods This study retrospectively an?alyzed 83 inpatients in the medical oncology department of the First Affiliated Hospital of China Medical University from March 2008 to September 2014. The Kaplan?Meier method and Cox proportional hazards models were used to analyze relevant factors affecting the prognosis of extensive small cell lung cancer;statistical significance was determined for a P value less than 0.05. Results The lactate dehydrogenase(LDH)level be?fore treatment was significantly associated with the progression?free survival(PFS)(P=0.001)and overall survival(OS)(P=0.036). The NSE level before treatment was also significantly associated with the PFS(P=0.007)and OS(P=0.013). Conclusion LDH and NSE affect progno?sis and survival of patients with extensive small cell lung cancer.
9.Correlation of Hemoglobin and Bilirubin with Prognosis of Extensive Stage Small Cell Lung Cancer

Objective To explore the factors that affect the prognosis of extensive small cell lung cancer by analyzing the association between lab-oratory indicators before treatment of extensive small cell lung cancer patients and the initial evaluation results with disease progression and overall survival. Methods This study retrospectively analyzed 96 cases of hospitalized patients in the medical oncology department of The First Hospital of China Medical University from March 2008 to September 2014. Kaplan-Meier method and Cox proportional hazards models were adopted to ana-lyze the relevant factors affecting the prognosis of extensive small cell lung cancer. P<0.05 was considered statistically significant. Results There was no obvious correlation between HB level before treatment with PFS of patients(P=0.179),but there was obvious significant correlation be-tween HB level and OS of patients(P=0.041). Our results showed that the TBIL level of patients before chemotherapy was significantly associated with the PFS(P=0.039)and OS(P=0.026)of patients. Conclusion HB and TBIL levels are the influencing factors that affect the prognosis and survival of patients with extensive small cell lung cancer.
10.Investigation on spoligotyping and phenotypes of drug resistance to four first-line drugs in 251 Mycobacterium tuberculosis isolates from Qinghai, China

This study aims to learn drug resistance situation of four first-line anti TB drugs among 251 Mycobacterium tuberculosis isolates from Qinghai and to explore their relationships with genotypes by Spoligotyping,so as to provide basis for effective prevention of tuberculosis.Isolates of 251 M.tuberculosis were tested susceptibilities of four first-line drugs including isoniazid (INH),rifampicin (RFP),streptomycin(SM) and ethambutol (EMB) by using conventional proportion method and genotyped by Spoligotyping.Relationship between drug resistance and genotypes was analyzed statistically.Results showed the total drug resistance rate was 56.2% (141/251) among 251 M.tuberculosis isolates.Resistance rates of four first-line drugs were 43.0% (108/251) for INH,37.1% (93/251) for RFP,39.0% (98/251) for SM,27.9% (70/251) for EMB respectively.Rate of multidrug-resistant TB (MDR TB) was 31.5% (79/251).All 251 isolates of M.tuberculosis were typed by spoligotyping.The 185 (73.7 %) were Beijing genotypes and 66 (26.3.%) were non-Beijing genotypes,and no statistical association was found with drug resistance.This paper concludes that isolates of M.tuberculosis prevail in Qinghai have both high rates of drug resistance and MDR and dominant isolates are Beijing genotypes by spoligotyping.
