1.Determination of Trace Nickel in Human Hair by Kinetic Fluorimetry

Objective To establish a new kinetic fluorimetry method for the determination of trace nickel in human hair samples. Methods The method was based on the catalysis of nickel on the oxidation of fluorescein(CF) by hydrogen peroxide in an almost neutral medium. As nickel is present, the fluorescence intensity of the solution decreases. Results The wavelength of excitation was 492 nm and the wavelength of emission was 516 nm. The reaction was optimized at 100 ℃ for 10 min. The linear range of determination was 4.0-80.0 ?g/L and the detection limit was 2.2 ?g/L . The RSDs and recovery rates of the method were 2.6%-3.2%, and 96.7%-102.8% respectively. Conclusion This method is simple, rapid and sensitive for the determination of trace nickel in human hair samples.
2.Curative Effect of Xiaocuo Decoction Combined with Vitamin A in the Treatment of Acne Vulgaris

Objective:To observe the clinical efficacy and safety of traditional Chinese medicine Xiaozuo decoction combined with 0. 025% vitamin A for external use in the treatment of acne vulgaris. Methods:Totally 86 patients with acne vulgaris were randomly di-vided into the treatment group and the control group with 43 ones in each. The treatment group was treated with Xiaocuo dcoction, 1 dose/day, and the control group was orally treated with erythromycin ethylsuccinate, 0. 25g per time, 3 times per day. The treatment course was 6 weeks, and both groups were treated with 0. 025% tretinoin cream once every night. The clinical efficacy and adverse drug reactions were compared between the two groups. Results:The obvious effective rate of the treatment group and the control group was 69. 77% and 41. 86%, respectively, and the total effective rate was 88. 37% and 69. 77%, respectively. There was significant difference in the obvious effective rate between the two groups (P<0. 01), while there was significant difference in the total effective rate between the two groups (P<0. 05). Recurrence rates of two groups were 10. 00%and 22. 22%,respectively,and there was no sig-nificant difference (P>0. 05). There were no serious adverse reactions in the two groups. Conclusion: Xiaocuo decoction combined with 0. 025% tretinoin in the treatment of acne vulgaris has promising clinical efficacy with high safety.

4.Distribution and antibacterial resistance of pathogens of ventilator-associated pneumonia in general intensive care unit

Objective To investigate the distribution and drug resistance of pathogens of ventilator-associated pneumonia(VAP) in general intensive care unit(ICU).Methods Statistical methods were used to analyze retrospectively the data of pathogens and drug resistance of VAP in general ICU from 2004 to 2006.Results Totally 347 pathogens were isolated from deep-part secretion of lower respiratory tracts of 302 VAP in general ICU.The main pathogens included Pseudomonas aeruginosa(32.3%),Acinetobacter baumanii(31.1%),Staphylococcus aurous(16.7%),Klebsiella(7.5%) and Coagulase-negative Staphylococci(6.1%).Gram-negative bacillus showed a high resistance to piperacillin and Cefotaxime but high susceptibility to carbapenems and the enzyme inhibitor antibiotics.Gram-positive coccobacteria showed a high resistance to Penicillin,Clindamycin and Erythromycin but high susceptibility to Vancomycin and Quinolone.Conclusion Gram-negative bacillus was the main pathogen of VAP in general ICU,multidrug resistance was serious.The rate of methicillin-resistant staphylococcus aurous(MRSA) and methicillin-resistant coagulase-negative staphylococci(MRCNS) was high,with serious drug resistance.According to the data of the bacteria pathogen and antibiotics susceptibity,we can select antibiotics reasonably to control the infection and delay the emergency of new drug-resistant bacteria.
5.The relationship between muscle strength, creatine kinase and electromyography in lipid storage myopathy

Objective To assess the relationship between muscle strength , creatine kinase ( CK) and electromyography ( EMG) in patients with lipid storage myopathy ( LSM).Methods Motor and sensory nerve conduction study, F wave, and quantity EMG were performed in 30 patients with LSM.Muscle strength and CK were studied before EMG.The relationship between EMG , muscle strength and CK were analyzed.Results EMG showed myopathic changes in 19 patients,myopathic coexistence with neuropathic changes in five , and neuropathic changes in two.Four patients had normal EMG.Motor nerve conduction studies showed decreased compound muscle action potential amplitude in seven nerves of two patients , while abnormal sensory nerve conduction in 16 nerves of five patients.Quantity EMG was performed in 78 muscles.Thirty-eight muscles of 21 patients had decreased amplitude and short duration motor unit potential ( MUP) , 6 muscles of 2 patients had increased amplitude and long duration MUP , and 30 muscles of 16 patients had normal MUP.Abnormal spontaneous activities ( SA) were detected in 16 muscles.The M( Q25 , Q75 ) of serum CK was 295.5 (201.0, 4 845.8) U/L in patients with abnormal SA in EMG , and 482.0 (292.8, 963.8) U/L in patients without abnormal SA in EMG (Z=0.281, P=0.778).Medical Research Council ( MRC) Scale in deltoid muscle was 4.3 ±0.7 in muscles with SA and 4.1 ±0.8 in muscles without SA (t=0.490, P=0.628), and that in quadriceps femoris was 3.9 ±0.6 for muscles with SA and 3.7 ± 0.6 for muscles without SA ( t =0.725 , P =0.474 ).MRC Scale in deltoid muscle was 4.1 ±0.7 in muscles with myogenic MUP and 4.1 ±0.9 in muscles without myogenic MUP ( t=0.101, P=0.920), which in quadriceps femoris was 3.8 ±0.6 for muscles with myogenic MUP and 3.7 ±0.7 for muscles without myogenic MUP ( t=0.368 , P=0.716 ).Conclusions Neurogenic or normal MUP and abnormal sensory nerve conduction can be presented in some patients with LSM , while SA is not common.The changes in MUP and SA are neither correlated with CK nor with muscle strength.

To investigate the efficacy and safety of minimal y invasive endovascular techniques treatment of hydronephrosis in pregnancy complicated by infection.
65 cases of pregnancy hydronephrosis coinfected patients with minimal y invasive endovascular treatment. Patients were fol owed up for 3-12 months, compared with 48 patients with the same period of conservative treatment .
Ureteral perforation and other surgery-related complications occurred in 65 patients in the observation group has not occurred. The patient's duration of hospitalization、the recurrence rate of postoperative infection, antibiotic duration of treatment, fever days were significantly lower than the control group(P<0.05).
Compared with conservative treatment, minimal y invasive endovascular technical treatment of gestational hydronephrosis co-infection with faster recovery, shorter hospital stay, can significantly reduce the number of days of fever patients and reduce the number of days of antibiotic use, can be used as conventional means of treatment of gestational hydronephrosis infection combined conventional.
7.Low-frequency electromagnetic fields enhance the recovery of spinal cord injured rats undergoing bone mesenchymal stem cell transplantation

BACKGROUND:Bone marrow mesenchymal stem cel transplantation is considered as a promising therapy for spinal cord injury. How to more effectively promote the survival of bone marrow mesenchymal stem cel s in the area of spinal cord injury and to accelerate the recovery of motor function after spinal cord injury is a current study focus. Previous studies have found that low-frequency electromagnetic fields can promote bone marrow mesenchymal stem cel proliferation and differentiation, but whether the low-frequency electromagnetic fields can be applied to bone marrow mesenchymal stem cel transplantation for treatment of spinal cord injury requires further studies. OBJECTIVE:To discuss the effects of low-frequency electromagnetic fields on motor function of spinal cord injury rats after transplantation of bone mesenchymal stem cel s. METHODS:Sixty-four rat models of incomplete spinal cord injury at T 10 were established by compression method and then randomized into control group, transplantation group (bone mesenchymal stem cel transplantation), electromagnetic field group and combination group (electromagnetic field+bone mesenchymal stem cel transplantation). After successful modeling, bone mesenchymal stem cel s labeled with 5-bromo-2'-deoxyuridine were injected into the original injured site in the transplantation group and combination group, which were isolated and purified with the fast adherence method;while alpha-minimum essential medium was injected into the electromagnetic field group and control group for instead. At 24 hours post-operation, the electromagnetic field group and combination group were explored to low-frequency electromagnetic fields (frequency 50 Hz, magnetic indaction intensity 5 mT) for 60 minutes per day. RESULTS AND CONCLUSION:After cel transplantation for 21 days, the Basso, Beattie, and Bresnahan scores in the combination group was higher than the other groups (P<0.05). 5-Bromo-2'-deoxyuridine positive cel s grew wel , and integrated into the normal spine;syringomyelia was reduced, and the number of spinal neural cel s was increased in the combination group. In addition, glial fibril ary acidic protein expression was decreased in the combination group, while matrix metal oproteinase 2 expression was increased. It indicates that low-frequency electromagnetic fields could promote recovery of motor function in the spinal cord injury rats transplanted with bone mesenchymal stem cel s, which could be associated that low-frequency electromagnetic fields facilitate the survival of transplanted bone mesenchymal stem cel s, up-regulate the expression of matrix metal oproteinase 2, and reduce glial scar formation in the spinal cord injured site.
8.Clinical significance of hepatic artery variation in hepatic portal lymphadenectomy

Objective:To investigate the recognition and injury prevention strategies of hepatic artery variations during hepatic portal lymphadenectomy. Methods:A retrospective analysis was performed, and 12 patients of hepatic arterial variation among 62 pa-tients with hepatic portal lymphadenectomy were the subjects. The study was conducted in the First Affiliated Hospital of Bengbu Medi-cal College between January 2013 and July 2014. The intraoperative treatment and postoperative complications were recorded. Results:Among 12 cases of hepatic artery variation, we found the following cases:3 cases (25.0%) of Michels' Type III, 2 cases (16.7%) of Mi-chels' Type VI, 1 case (8.3%) of Michels' Type IX, 1 case (8.3%) of Hiatt's Type 6, 2 cases (16.7%) of spatial location variation between right hepatic artery and hepatic duct, 2 cases (16.7%) of left and right hepatic artery originating from a common hepatic artery, and 1 case (8.3%) of right hepatic artery originating from the gastroduodenal artery. No injury of hepatic artery occurred. Two cases had post-operative complications, including 1 case of pancreatic leakage and 1 case of incision infection;postoperative hemorrhage, bile leakage, hepatic abscess did not occur in these two cases. Patients recovered well in general. Conclusion:Hepatic arterial injury can be signifi-cantly reduced by the following:increased familiarity with the various types of hepatic artery variations;complete imaging examina-tions for inspection and evaluation before surgery;and careful and meticulous operations in surgery.
9.The effects of low intensity electromagnetic fields on the reconstruction of full skin loss wounds grafted with human epidermal stem cells

Objective To observe the effects of low intensity electromagnetic fields (LIEMFs) in promoting the reconstruction of full skin loss wounds grafted with human epidermal stem cells (ESCs). Methods Fifty nude mice aged 7 to 8 weeks with full skin loss wounds were equally divided into 3 experimental subgroups ( 1 Hz, 10Hz and 50Hz) and two control groups (a cell suspension control group and a blank control group) , with ten mice each. In the 3 experimental subgroups and the cell suspension control group, ESCs separated from human foreskin and cultured in vitro were grafted to the wounds using collagen sponge scaffolds. The experimental subgroups were then stimulated with an LIEMF (magnetic field intensity 5mT) at the appropriate frequency for 30min/day for 15 days. The blank control group was put under the same conditions without the cell suspension and LIEMF. The healing rates of the wounds were observed, and tissue slices were stained and observed under a light microscope. The inner structure of the regenerating skin was observed using transmission electron microscopy. Results The ESCs were successfully grafted. A few human integrin β1 positive stained cells appeared in the regenerating skin. The average healing rates in the experimental subgroups were significantly superior to those of the control groups. Well differentiated epidermis and dermis could be seen in the regenerating skin in all of the experimental groups. The epidermis had more cell layers and was thicker than in the control groups. More desmosome, hemidesmosome and keratin filaments were seen among the epidemic cells of the experimental groups. Conclusions LIEMF promotes the healing of full skin loss wounds grafted with ESCs in nude mice, and can promote complete repair of skin defects and the regeneration of skin function.
10.Effects of low-frequency electromagnetic fields on skin wound healing after the transplantation of gene modified epidermal stem cells

BACKGROUND:The repair and management of ful-thickness skin defects resulting from burns and chronic wounds remain a significant unmet clinical chal enge. Using epidermal stem cel s and keratinocyte growth factor for ful-thickness wound repair is a promising approach. Low-frequency electromagnetic fields which are a non-invasive physical stimulation therapy have been recognized as a good method to enhance wound healing. OBJECTIVE:To develop a new strategy to accelerate wound healing by transplanting transfected epidermal stem cel s and keratinocyte growth factor and treating with low-frequency electromagnetic fields in a mouse model. METHODS:Epidermal stem cel s from Sprague-Dawley neonatal rats were isolated and cultured in vitro, then the cel s were labeled with 5-bromo-2-deoxyuridine and transfected by Ad-KGF, a recombinant adenovirus carrying the keratinocyte growth factor. Mice were given to create ful thickness skin wound on the dorsum and randomly assigned to four groups:control group, transplantation of epidermal stem cel s group, transplantation of keratinocyte growth factor gene modified epidermal stem cel s group, and transplantation of keratinocyte growth factor gene modified epidermal stem cel s plus low-frequency electromagnetic field exposure group. RESULTS AND CONCLUSION:The best healing pattern was observed in the keratinocyte growth factor gene modified epidermal stem cel s plus low-frequency electromagnetic field exposure group (P<0.05) at days 9 and 16. 5-Bromo-2-deoxyuridine labeled cel s existed in the wound in the treated groups at day 9. A significantly increased expression of endogenous keratinocyte growth factor was detected in the transplantation of Keratinocyte Growth Factor gene modified epidermal stem cel s group, and transplantation of keratinocyte growth factor gene modified epidermal stem cel s plus low-frequency electromagnetic field exposure group at day 16. A wel-advanced epithelialization was observed in transplantation of keratinocyte growth factor gene modified epidermal stem cel s plus low-frequency electromagnetic field exposure group at days 16 and 30. These results suggest that low-frequency electromagnetic fields enhanced wound healing fol owing the transplantation of keratinocyte growth factor gene modified epidermal stem cel s.
