1.Development of dot immunogold filtration assay kit for rapid detection of antibody to schistosome in human sera

Objective To develop a rapid kit applied to the field for detection of antibody to schistosome in human sera. Methods A new kit for rapid detection of antibody to schistosome was developed through improving the dot immunogold filtration assay (DIGFA). A total of 100 cases of sera from chronic schistosomiasis patients and 140 from healthy people, HBV patients and the people infected with other parasites were detected by the kit. The sensitivity, specificity, Youden's index and Kappa value were utilized as the evaluation standard. Results The sensitivity of detecting antibody to schistosome, specificity, Youden's index and Kappa value were 92% , 95.08% , 0.87 and 0.87, respectively. The cross reaction to patients with clonorchiasis was 5%. Conclusion DICFA kit is practical for antibody to schistosome detection in the field because of its advantages such as smaller serum needed and faster in reaction.
2. Evaluation of causes and clinical effects of revision surgery after artificial hip replacement

Objective:
To investigate the clinical effects of primary revision after artificial hip replacement and causes of revision surgery.
Methods:
A total of 344 patients who underwent revision surgery after total hip arthroplasty (THA) and artificial femoral head replacement from January 2010 to December 2016 were retrospectively analyzed. There were 141 males and 203 fe-males, with a mean age 65.64±10.81 years (28-87 years). A total of 351 hip revisions were performed in 344 patients with 7 pa-tients in bilateral revisions. All patients were followed up for 60.38±22.75 months (24-105 months). All patients with periprosthet-ic infection underwent two-stage revision after prosthesis placement, and all others underwent one-stage revision. The clinical out-comes of revision surgery were assessed with the Harris hip score and the survival rate of the revision prosthesis. According to the duration from artificial hip replacement to revision surgery, all patients were divided into early revision (less than 5 years) and late revision (more than 5 years) groups. They were also divided into two groups according to the age of the patient in revision sur-gery: >55-year-old and ≤55-year-old groups. The causes of revision, hip reoperation and re-revision were recorded.
Results:
In the 351 hip revisions, a total of 238 (67.8%) had aseptic loosening, 41 (11.7%) periprosthetic fractures, 30 (8.5%) periprosthetic infection, 23 (6.6%) recurrent dislocation and 19 (5.4%) the eccentric liner wear. 3.Literature Research on Herbal Medicine of Fenugreek

Fenugreek is a kind of Chinese herbal medicine with great development prospects.At present,there are many modern reports on its components extraction,quality standards,processing technology and pharmacological effects.However,there is still a lack of research on sorting out and analyzing the relevant ancient records,and people's herbological cognition of fenugreek is still controversial and incomplete.In this study,through systematic review and analysis of ancient records,the names,original plant,processing methods and properties(including nature,flavor,meridian tropism,action,application,toxicity and contraindication)of fenugreek were comprehensively verified for the first time,which provided a herbological reference for further development,utilization and in-depth study of Fenugreek.It is found that fenugreek has several names such as Kudou,Luba,Huba and Jidou.Based on analyzing the descriptions and pictures of fenugreek original plant in ancient records,there is a strong likelihood that the ancient and modern medicinal fenugreek belong to the same species.In addition to the current common method of stir-frying with salt solution,the ancient books also recorded eight processing methods of fenugreek,such as stir-frying,steaming,baking,and calcining.According to the research of properties,it is found that fenugreek is warm-hot in nature,bitter,sweet,and pungent in flavor,and attributive to the kidney,stomach,liver and bladder meridians.In addition to the efficacy recorded in pharmacopoeia,this study also complemented fenugreek's actions of"improving hearing and eyesight,guiding fire to origin"and application of"fullness and discomfort in abdomen and hypochondrium".According to the textual research of toxicity and contraindication,fenugreek has been recorded as non-toxic in the past dynasties,but it should be used cautiously in the excess-heat syndrome,Yin deficiency syndrome,and pregnant women.
4.Mutation analysis for a family affected with Charcot-Marie-Tooth disease type 4C.

OBJECTIVETo identify potential mutation in a Chinese family affected with Charcot-Marie-Tooth disease(CMT).
METHODSClinical data of the family was collected, and genomic DNA was extracted from peripheral blood samples of the family members. Seventy-two candidate genes of the proband were captured and sequenced by targeted next-generation sequencing, and the results were confirmed by Sanger sequencing. The protein structure was predicted with PyMOL-1 software.
RESULTSA homozygous missense mutation c.1894G>A(p.E632K) was identified in the exon 11 of the SH3TC2 gene in the proband. Heterozygous c.1894G>A mutation was also detected in the proband's father, mother and daughter, but not in the healthy family members and 300 normal controls. Retrieval of the NCBI, HGMD and 1000 genome databases has verified the c.1894G>A to be as a novel mutation. Computer simulation has suggested that the mutation has altered the 3D structure of the SH3TC2 protein.
CONCLUSIONThe proband was diagnosed as CMT4C, for which the underlying gene was SH3TC2. This finding has expanded the spectrum of SH3TC2 mutation in association with CMT4C.

5.Establishment and application of a nomogram model for prognostic risk prediction in patients with epithelial ovarian cancer

Objective:To explore the prognostic factors of epithelial ovarian carcinoma (EOC), construct a nomogram model, and evaluate the prognosis of EOC patients.Methods:A retrospective analysis was performed on clinicopathological data of 208 cases of EOC patients who received initial treatment in the First Affiliated Hospital of Army Medical University from August 11, 2016 to July 11, 2018, including age, preoperative ascites, preoperative neoadjuvant chemotherapy, surgical method, pathological type, pathological differentiation degree, surgical pathology stage, preoperative and post-chemotherapy serum cancer antigen 125 (CA 125) level, human epididymal protein 4 (HE4) level, platelet count and platelet/lymphocyte number ratio (PLR). The univariate and multivariate Cox risk ratio models were used to analyze the related factors affecting progression free survival (PFS) in EOC patients, and the prediction nomogram of PFS in EOC patients was established to evaluate its efficacy in predicting PFS. Results:Univariate analysis showed that preoperative neoadjuvant chemotherapy, pathological type, pathological differentiation degree, surgical pathology stage, serum CA 125 and HE4 level before operation and after chemotherapy, platelet count and PLR before operation and after chemotherapy were significantly correlated with PFS in EOC patients (all P<0.05). Multivariate analysis showed that surgical pathology stage, preoperative PLR, serum CA 125 and HE4 level after chemotherapy were independent prognostic factors affecting PFS of EOC patients (all P<0.01). The index coefficient of the prediction model for the prognosis of EOC patients established by this method was 0.749 (95% CI: 0.699-0.798), which had good prediction ability, and could help clinicians to more accurately evaluate the prognosis of EOC patients. Conclusion:The nomogram model constructed based on surgical pathology stage, preoperative PLR, serum CA 125 and HE4 level after chemotherapy could effectively predict the PFS of EOC patients after initial treatment, could help clinicians to screen high-risk patients, provide individualized treatment, and improve the prognosis of EOC patients.

Mass spectrometry and database searching are necessary to identify proteins and peptides. With the rapid development of mass spectrometry technology, mass spectrometry data in proteomics are acquired very quickly, providing a powerful method to identify large-scale proteins and peptides, making mass spectrometry data-based proteomics research more and more into the mainstream. The traditional database searching method has many limitations to identify post-translational modifications of peptides. This paper systematically reviews the development, theoretical concept and applications of spectral network method, and the advantages of spectral network library to identify peptides.
7.Effects of salvianolic acid on expression levels of serum GFAP,PGP9.5,IL-5,IL-6,IL-8 and IL-10 and prognosis in patients with acute cerebral infarction

Objective To investigate the effect of Salvianolic acid on the expression of glial fibrillary acidic protein (GFAP),protein gene product (PGP9.5),IL-5,IL-6,IL-8,IL-10 and prognosis in patients with acute ischemic stroke.Methods 84 patients with acute ischemic stroke were divided into salvia miltiorrhiza group (n=42) and control group (n=42).Barthel index and NIHSS score were performed on the day of admission and 14 days.The post-infarction volume was calculated and compared after head MRI.The expression levels of serum GFAP,PGP9.5,IL-5,IL-6,IL-8 and IL-10 were detected and compared by magnetic particle chemiluminescence method.The prognosis was determined by modified Rankin score (mRS) 90 days after infarction.Results After treatment,NIHSS score,Barthel index and infarct volume in salvia miltiorrhiza group were significantly decreased (P<0.05),significantly increased (P<0.05) and significantly decreased (P<0.05).GFAP,IL-6 and IL-8 decreased significantly in salvia miltiorrhiza group after treatment compared with the control group (all P<0.05).After treatment,IL-10 expression level in salvia miltiorrhiza group increased more significantly than that in control group (P<0.05).The prognosis of Salvia miltiorrhiza group was better than that of control group (P<0.05).Conclusion Salvianolic acid in the treatment of acute ischemic stroke can effectively inhibit inflammatory reaction,reduce brain injury,improve neurological impairment,improve daily behavior and improve prognosis.
