1.Evaluation and development of preoperative liver reserve function evaluation methods

Liver resection is one of the important treatments of liver diseases,especially hepatocellular carcinoma.In China,the vast majority of liver cancer patients suffer from generalized damage of the liver parenchyma such as cirrhosis,lead to liver reserve function reducing in various degrees.Liver dysfunction or even liver failure after liver resection becomes an important reason of perioperative death and influences the patients' long-term survival.Therefore,accurate preoperative evaluation of liver reserve function is very important.Though there are seveal kinds of assessment of liver reserve function in recent years,it still lack of a clinically recognized,comprehensive assessment method.This paper reviewed the clinical commonly used preoperative liver reserve function evaluation methods,summarizes and analyzes the value and the insufficiency of several important methods,and prospects the development of evaluation methods about preoperative liver reserve function.
2.A DYNAMIC FAULT TOLERANT ALGORITHM FOR IMPROVISING PERFORMANCE OF MULTIMEDIA SERVICES

Multimedia Services has drawn much attention from both industrial and academic researchers due to the emerging consumer market, how to provide High-Availability service is one of most important issues to take into account. In this paper, a dynamic fault tolerant algorithm is presented for highly available distributed multimedia service, then by introducing SLB(server load balancing) into fault tolerance and switching servers in different ways according to their functions, the proposed schema can preserve reliability and real-time of the system .The analysis and experiments indicate that resuming server's faulty by this method is smooth and transparent to the client The proposed algorithm is effectively improving the reliability of the multimedia service.
3.Genetic polymorphisms of 19 Y-STR loci in in the Han population in Northeast China and the cluster analysis of ethnic groups in northeast Asia region

Objective To investigate the polymorphisms of the 19 Y-STR loci in the Han population in Northeast China,and analyze the genetic relationships of 15 ethnic groups in northeast Asia region,and to evaluate their forensic value and population genetic value.Methods The 19 Y-STR unrelated Han males in 200 unrelated Han males in Northeast China were amplified with the Y-filer system,and the PCR products were analyzed by the 310 Genetic Analyzer.The AMOVA analysis,cluster analysis and MDS analysis were calculated by Arelquin3.11,Mega4.1 and SPSS17.0.Results The polymorphisms of 19 Y-STR loci in the Han population in Northeast China had generally higher gene diversity value which were ranged from 0.350 9 (DYS391) to 0.971 1 (DYS385a/b),and totally 200 haplotypes were observed.The 19 Y-STR loci displayed high genetic polymorphisms in the Han population in Northeast China,indicating that these 19 loci were useful genetic markers for forensic personal identification and paternity testing.There were distinctions among 15 ethnic groups.The genetic distance between 15 ethnic groups were ranged from 0.000 9 to 0.643 2,and the conclusion of cluster analysis and MDS analysis were similar to the ethnogeny research and ethnic migration history.Conclusion The 19 Y-STR loci displayed high genetic polymorphisms in the Han population in Northeast China,and these 19 loci were useful genetic markers for forensic personal identification and paternity testing.
4.Bone marrow mesenchymal stem cells transplanted into a rat model of Alzheimer’s disease:improvement in the learning and memory ability

BACKGROUND:Drug therapy can partly reduce and delay the progress of Alzheimer’s disease, but only 30%with the single drug treatment obtain clinical cure. OBJECTIVE:To study the therapeutic effect of bone marrow mesenchymal stem cel s transplantation for rats with Alzheimer’s disease. METHODS:Amyloidβ-protein was injected into the hippocampus of Sprague-Dawley rats to construct the model of Alzheimer’s disease. And bone marrow stromal stem cel s were transplanted into the hippocampus of the rat models. RESULTS AND CONCLUSION:At 2 weeks after modeling, compared with the control group, the escape latency in the model and experimental groups was significantly longer (P<0.05), which indicating that Alzheimer’s disease models were successful y established. At 4 weeks after cel transplantation, compared with the model group, the average escape latency in the experimental group was significantly decreased, but retention time on the platform quadrant was significantly prolonged (P<0.05). Besides, at 4 weeks after cel transplantation, expression of choline acetyltransferase in the experimental group was significantly higher than that in the model group (P<0.05). In conclusion, bone marrow mesenchymal stem cel s cannot only differentiate and survive in the hippocampus of rats with Alzheimer’s disease, but also improve the learning and memory ability.
5.N-acetylcysteine blocked hypoxia-reoxygenation induced apoptosis through ROS-p38 MAPK signaling pathway in neonatal rat cardiomyocytes

Objective Previous investigations have shown that N-aeetylcysteine (NAC) could regulate diverse cell type's apoptosis. The purpose of this study was to evaluate the mechanism of NAC reversed apoptosis ofcardiomyocytes induced by hypoxia-reoxygenation (H/R). Methods Cardiomyocytes were treated with hypoxia 6 h and reoxygenation 72 h in the absence and presence of NAC (100 μmol/ L). The ROS was assayed by using Image-iTTM LIVE green reactive oxygen species detection kit. The viability of cell was assayed with trypan blue. Early stages ofapoptosis were assessed by flow cytometry using Annexin V, and late stages ofapoptosis were assessed using TUNEL system. Bcl2 and bax mRNA levels were determincd by real-time quantitative PCR. Bcl2, bax, p38 and pp38 protein levels were determined by western blot. Results We found that H/R could markedly increase ROS generation and induce the apoptosis of cardiomyocytes (P<0.01). NAC (100 p tool/L) significantly reduced the generation of ROS and apoptosis (P all <0.01). NAC also significantly reduced the protein ratio of pp38 and p38 and increased the RNA and protein ratio of bcl2 and bax (P all <0.01). Conclusion The results showed that NAC significantly reduced apoptosis through inhibiting the phosphorylation of p38 signal pathway, which has potential value for clinical cardiac diseases.
6.Relationship between primary hypothyroidism and hyperuricemia:an investigation

Objective To explore the relationship between primary hypothyroidism and hyperuricemia and to speculate their possible mechanism .Methods The levels of FT3 ,FT4 ,TSH, Anti-TG ,Anti-TPO, TRAb, UA, TC, TG, LDL-C, HDL-C,Hcy, Cys,β2 MG and 24hUA were measured in 36 patients with hypothyroidism , compared with 20 healthy sub-jects.According to the absence or presenceof Anti-TG, Anti-TPO and TRAb, the patients were divided into two sub-groups:the Hashimo to(HT) hypothyroidism group (27 cases) and the non-HT hypothyroidism group (9 cases)before they were compared with each other .Results Compared with healthy subjects , patients with hypothyroidism had higher TSH , TC,TG, LDL-C, UA, Hcy,Cys,andβ2MG,and lower FT3,FT4,HDL-C,and 24hUA.The difference was significant (P<0.05);the thyroid function, autoimmune antibodies, TC, LDL-C, UA and 24hUA were also significantly differen between the two sub-groups.Conclusion Patients with hypothyroidism are more prone to hyperuricemia , which may be related to decreased thyroid hormone , lipid metabolism disorder and hyperhomocysteine .The level of UA is higher in HT hypothyroidism than in non-HT hypothyroidism , indicating that autoimmune factors may be involved in the occurrence of hyperuricemia .
7.Overexpressing 3-ketosteroid-Δ1-dehydrogenase for degrading phytosterols into androst-1,4-diene-3,17-dione.

We constructed plasmid pMTac to overexpress 3-ketosteroid-Δ1-dehydrogenase (KSDD) in Mycobacterium neoaurum JC-12 for improving androst-1,4-diene-3,17-dione (ADD) production. To construct pMTac, pACE promoter on pMF41 was replaced by tac promoter, and then four recombinants were constructed, which were M. neoaurum JC-12/pMF41-gfp, M. neoaurum JC-12/pMTac-gfp, M. neoaurum JC-12/pMF41-ksdd and M. neoaurum JC-12/pMTac-ksdd. Fluorescence detection results show that much more green fluorescent protein (GFP) was expressed in M. neoaurum JC-12/pMTac-ksdd than M. neoaurum JC-12/pMF41-ksdd. The activity of KSDD was 2.41 U/mg in M. neoaurum JC-12/pMTac-ksdd, 6.53-fold as that of M. neoaurum JC-12 and 4.36-fold as that of M. neoaurum JC-12/pMF41-ksdd. In shake flask fermentation, ADD production of M. neoaurum JC-12/pMTac-ksdd was 5.94 g/L, increased about 22.2% compared to the original strain M. neoaurum JC-12 and 12.7% to M. neoaurum JC-12/pMF41-ksdd. AD (4-androstene-3,17-dione) production of JC-12/pMTac-ksdd was 0.17 g/L, decreased 81.5% compared to M. neoaurum JC-12 and 71.2% to M neoaurum JC-12/pMF41-ksdd. In the 5 L fermenter, 20 g/L phytosterols was used as substrate, ADD production of M. neoaurum JC-12/pMTac-ksdd was improved to 10.28 g/L. pMTac is favorable for expressing KSDD in M. neoaurum JC-12, and overexpression of KSDD has beneficial effect on ADD producing, and it is the highest level ever reported using fermentation method in M. neoaurum.

8.Evaluation of liver function reserve with gadoxetate disodium-enhanced MR

Objective To determine the values of gadoxetate disodium-enhanced MRI for quantitatively evaluating the liver function reserve.Methods Forty consecutive liver cirrhosis patients were enrolled in this retrospective study.All the patients underwent complete routine laboratory tests and the gadoxetate disodium-enhanced MR imaging with the same parameters.Liver volume and the relative enhancement index of the whole liver in hepatocellular phase were used to calculate liver relative uptake value,which is supposed to represent the total liver function reserve.And the function reserve of all liver segments was evaluated respectively by the relative enhancement index of each liver segment.One-way ANOVA was used for statistical analyses.Results The patient number in Child-Pugh A,B and C group was 23,10 and 7,respectively.Significant differences was observed in liver relative uptake value among patients in different Child-Pugh classes (F =122.05,P < 0.01).Mean liver relative uptake value was highest in Child-Pugh A group(1212 ± 168),followed by B group and C group (695 ± 161,234 ±55).In Child-Pugh A group,the relative enhancement index of S1 to S8 was 1.13 ± 0.22,1.12 ± 0.50,0.81 ± 0.24,1.08 ± 0.32,1.41 ± 0.25,1.10 ± 0.30,1.16 ± 0.41 and 1.17 ± 0.23,respectively (F =5.93,P < 0.01).There was no significant difference in B and C group (F =1.95,1.83 ; P > 0.05).Conclusion Gadoxetate disodiumenhanced MRI can be used for evaluating whole liver function reserve quantitatively.This technique also has a potential value for evaluating of the segmental liver function reserve before partial hepatectomy.
9.Reverse dedifferentiation of atrial cardiomyocytes after restoration of sinus rhythm from atrial fibrillation in goats

Objective Chronic atrial fibrillation (AF) results in dedifferentiation of atrial cardiomyocytes that plays an important role in the perpetuation of AF.In this study,we aimed to investigate the changes of titin and α-smooth muscle actin (α-SMA) after long time of AF reversal.Methods Twenty-four goats were randomized into four groups:(1) sinus rhythm (SR),(2) 3 months AF (3-mo AF),(3) 3 months SR after 3 months AF (3-mo post AF),(4) 6 months SR after 3-mo AF (6-mo post AF),with 6 in each group.By pacing on the anterior bottom of left atria appendage (LAA),we established a goat model of chronic AF.Atria effective refractory period (AERP) was measured with electrophysiological methods.Ultra-structure was studied with echocardiography,light and electron microscopy.Titin and α-SMA protein expressions were determined by Western blot.Results The animals underwent high rate pacing on LAA for a mean of 42.23±21.70 days before presenting AF.Electrophysiological analysis revealed that AERP completely resumed in 3-mo post AF goats.Echocardiography displayed that the size of left atrium resumed almost in 6-mo post AF goats (P< 0.01).Pathological and electron microscopic examination revealed the disorder of myofibrils,augmentation of intercellular space,myolysis,accumulation of glycogen,and numerous bigger mitochondria among atrial cardiomyocytes in 3-mo AF goats.They recovered mostly in 6-mo post AF goats.Western blot showed that the band density of titin significantly reduced in 3-mo AF goats compared to SR ones [1826±319 vs 5012±854,P<0.01].In 3-and 6-mo post AF goats,titin increased gradually and it reversed completely in 6-mo post AF goats (3841±601 and 4523±833 respectively,P < 0.01).Conversely,the band density of α-SMA was significantly higher in 3-mo AF goats (5324±948) than in SR ones (1619± 271,P<0.01).In 3-and 6-mo post-AF goats,α-SMA decreased gradually,and it recovered mostly in 6mo post AF goats (4437± 792 and 2205±540 respectively,P<0.01,).Conclusions These data indicate that the reversal of dedifferentiation of atrial cardiomyocyts is a very slow process,and it is definitely essential for normal cardiac function.
10.Role of PLC/PIP2 signaling pathway in radiation-induced NRP1 + Treg cells

Objective To explore the role of PLC/PIP2 signal pathway in the changes of mouse thymus CD4 + CD25 + NRP1 + Treg and TGF-β1 after different doses of X-ray irradiation Methods 36 ICR mice were randomly divided into 6 groups according to the irradiation doses of 0,0.5,1.0,2.0,4.0 and 6.0 Gy,respectively.Flow cytometry was used to detect the expression of NRP1 + Treg,and ELISA was used to detect the expression of TGF-β1 in mouse thymocytes at 16 h post-irradiation.The EL-4 cells were irradiated by X-rays at the dose of 4.0 Gy after co-cultured with the PMA and TMB-8 for 2 hours.Flow cytometry was used to detect the expression of NRP1 + Treg,and ELISA was used to detect the changes of TGF-β1 at 48 h post-irradiation.Results The NRP1 + Treg appeared a transient decrease at both 0.5 and 1.0 Gy irradiation and reached its valley value at 1.0 Gy (t =6.96,P < 0.01),then showed a dosedependent increase and reached its peak at 6.0 Gy (t =6.70,P < 0.01).The TGF-β1 level decreased after 0.5 Gy X-rays (t =12.53,P <0.01),then increased at a dose-dependent manner and reached its peak at 4.0 Gy (t =10.40-19.56,P < 0.01).Compared with the sham-irradiation,NRP1 + Treg was decreased significantly after PMA treatment (t =3.06,P < 0.01),while it was up-regulated significantly after irradiation in the presence of PMA (t =8.27,P < 0.01).TGF-β1 was reduced in the presence of PMA with or without irradiation (t =10.46-39.69,P < 0.01).NRP1 + Treg and TGF-β1 were increased significantly after TMB-8 treatment (t =5.53-44.26,P < 0.01).Conclusions NRP1 + Treg cells and TGF-β1 were up-regulated after a high dose radiation,and the PLC/PIP2 signal pathway may participate in the regulation.
