1.Clinical Study on the Relationship Between the p53 Gene Examination and Colorectal Neoplasms

Objectives To study the clinical significance of the early diagnosis on colorectal cancer and the value of monitoring precancerous lesions of p53 gene examination. Methods Histological specimens of tumor from 110 patients with colorectal neoplasms were collected under the endoscopy, another 10 normal intestinal mucosa tissues with benign enteropathy as a control group. By PCR-SSCP methods, examined p53 exon 5~8 of mutation circumstance, made pathological section at the same time, and combine to track the observation. Results 24 patients p53 gene mutation were detected out in 78 cases colorectal cancers,counted up 32 4%, among 17 patients were moderate,low differentiation, 3 patients of them were mucinous adenocarcinoma.There are 6 patients p53 gene mutation in 36 cases colorectal adenoma,counted up 16 7%, among them an patient of Ⅱ~Ⅲ class atypical hyperplasia adenoma occurrence canceration in the next 10 months.There was no p53 gene mutation in normal intestinal mucosa tissue.Conclusions p53 gene examination combinated with pathological examination could monitor precancerous lesions and early diagnosis of the colorectal cancer.
2.Clinical significance of dynamic morphological changes of circle cistern observed by computed tomography scanning in patients with severe traumatic brain injury

Objective To investigate the clinical significance of observing the dynamic morphological changes of circle cistern on CT scanning in the patients with acute severe traumatic brain injtuy (TBI).Methods The CT imaging data of circle cistern of 135 patients with acute severe TBI were analyzed retrospectively.According to the morphological changes of circle cisterns on the initial CT images,135 cases were divided into 5 groups,and 135 cases carried on prognostic evaluation with Glasgow outcome score (GOS) 6 months later.The relationship between the dynamic morphological changes of circle cistern and the prognosis of patients were analyzed statistically.Results There was a significant relationship between the state of circle cistern and the prognosis of patients.The more obvious the dynamic morphological changes of circle cistern were,the worse prognosis and the higher the fatality was,especially higher mortality in patients with exacerbation of the dynamic morphological changes of circle cistern (79.5%),and the difference was statistically significant among 5 groups (P < 0.01).Conclusions There was a significant relationship between the state of circle cistern and the prognosis of the patients with acute severe TBI.The dynamic morphological changes of circle cistern on CT scanning might be used as a promising marker to evaluate the severity of severe TBI,especially in the patients at earlier posttraumatic interval.It is necessary to observe the dynamic change of circle cistern.
3.Collagen-gelatin scaffolds for the repair of peripheral nerve defects

BACKGROUND:Col agen-gelatin composite scaffolds have been reported to be able to promote the early recovery of peripheral nerve defects. However, this conclusion has not been further confirmed. OBJECTIVE:To investigate the biocompatibility of the col agen-gelatin scaffold and its treatment outcomes in the repair of peripheral nerve defects. METHODS:The col agen-gelatin scaffold was co-cultured with bone marrow mesenchymal stem cel s (BMSCs) of Sprague-Dawley rats for 5 days, and then the cel growth was observed. Twenty Sprague-Dawley rats were enrol ed, modeled into a left 30-mm peroneal nerve defect and randomized into experimental and control groups. The col agen-gelatin scaffold composited with BMSCs was implanted into the experimental group, and autograft nerve implanted into the control group. Morphology of the middle bridge was observed, and electrophysiology was conducted at 16 weeks after implantation. RESULTS AND CONCLUSION:BMSCs grew and adhered wel onto the scaffold. The myelinated nerve fiber density did not significantly differ between groups (P>0.05). The myelinated nerve fiber diameter, myelin sheaththickness and percentage of nerve tissues in the experimental group were significantly lower than those in the control group (P<0.05). There were no significant differences in the conduction velocity, latency of motor nerves and the conduction velocity and amplitude of sensory nerves between groups (P>0.05). The amplitude of motor nerves and the latency of sensory nerves in the experimental group were significantly lower than those in the control group (P<0.05). To conclude, the col agen-gelatin scaffold holds a good cytocompatibity and is ideal for the repair of peripheral nerve defects.
4.The rapetic analysis of 48 cases with subcutanous fistula excision to long-path low anus fistula

Objective To investigate the clinical effects of subeutanous fistula excision for the long-path low anus fistula.Methods Operation for long-path low anus fistula,fistula length>5cm,divided the pafients into two groups.The therapeutic group:using subcutanous fistula excision;the control group:using fistula icision.Comparing the healing time,arIus malformation,recurrence rate.Results Therapeutic group 47 cases heal,the healing time was 12 days;no anus malformation;no arms ineontenience.In the control group,the healing time was 23 days;anus malformation 23 cases;anus gas incontenience 12 cases.Conclusion To use subcutanous fistula excision for the longpath low anus fistula has the advantages of short healing time,no anus malformation,less pain,and low reccurrent rate.
5.Hypnotic interaction between midazolam and emulsified isoflurane in rats

Objective To examine the effects of different doses of midazelam on ED50 of emulsified isoflurane and to determine the type of interaction between them for hypnosis by isobelographic analysis in rats. Methods One hundred and twenty-five adult male SD rats weighing 240-300 g were randomized into 5 groups (n=25 each): group Ⅰmidazolam (M); group Ⅱ emulsified isnflurane (Ⅰ); group Ⅲ ,Ⅳ ,Ⅴ, 1/4, 1/2 and 3/4 ED50 of midazelam for hypnosis + emulsified isoflurane (MI1 , MI2, MI3). Up-and-down sequential experiment was used to determine ED50of midazolam and emulsified isoflurane for loss of righting reflex in group Ⅰ -Ⅴ . The intial dose of midazolm was 17.3 mg/kg in group M. The initial doses of emulsified isoflurane (120 mg/ml) were 0.55 (in group Ⅰ, 0.22 (MI1), 0.19 (MI2) and 0.12 ml/kg (MI3) respectively. In group MI1-3 midazolam was injected over 15 seconds and after an interval of 2.5 min emulsified isoflurane was injected over 10 s. ED50 was calculated using Dixon-Mood method. Isebolographic and algebraic analyses were used to determine the type of interaction between midazolam and emulsified isoflurane. Results The five groups were comparable with respect to M/F sex ratio and body weight. The Edso of midazolam was 26.0 mg/kg in group M. Midazolam 6.5, 13.0 and 19.5 mg/kg were given in group MI1 , MI2 and MI3 respectively. The ED50 of emulsified isoflurane was 0.67 (ingroup Ⅰ), 0.30 (MI1), 0.22 (MI2) and 0.18 ml/kg (MI3) respectively. The isobolographic analysis indicated that with increasing doses of midazolam, the Edw of emulsified isoflurane decreased progressively in a non-linear fashion. The isobolographic and algebraic analyses demonstrated that the interaction between midazolam and emulsified isoflurane was synergistic for hypnosis. Conclusion The hypnosis is synergistic when midazolam 6.5,13 mg/kg are combined with emulsified isoflurane and additive when midazolam 19.5 mg/kg is combined with emulsified isoflurane.
6.Synthesis and in vitro antibacterial activity of 7-(4-alkoxyimino-3-methyl-3-methylaminopiperidin-1-yl)quinolones.

To explore new agents of quinolone derivatives with high antibacterial activity, 7-(4-alkoxyimino-3-methyl-3-methylaminopiperidin-1-yl)quinolones were designed and synthesized, and their activity against gram-positive and gram-negative strains was tested in vitro. Sixteen target compounds were obtained. Their structures were established by 1H NMR, HRMS and X-ray crystallographic analysis. Compounds 14k and 14m-14o show good antibacterial activity against the tested five gram-positive strains and five gram-negative strains (MIC: 0.25-16 micromg x mL(-1)), of which the most active compound 14o is 8-fold more potent than levofloxacin against S. pneumoniae (MIC: 4 microg x mL(-1)), and comparable to levofloxacin against S. aureus, S. epidermidis, E. faecalis and E. coli (MIC: 0.25-1 microg x mL(-1)), but generally less potent than gemifloxacin.
7.Clinical significance of IL-17 in peripheral blood of patients with rheumatoid ar-thritis

Objective:To explore the role and significance of IL-17 on the pathogenesis of rheumatoid arthritis ( RA).Methods:The levels of IL-17 mRNA in peripheral blood mononuclear cell ( PBMC) of 66 RA patients (32 active,34 inactive) and 44 healthy controls were detected by reverse transcription-polymerase chain reaction ( RT-PCR ).The levels of serum IL-17 were detected by enzyme-linked immunosorbent assay ( ELISA).The levels of RF were detected by immunoturbidimetric assay.The correlations of serum IL-17 and RF were analyzed.Results: In RA patients the levels of IL-17 mRNA in PBMC were significantly higher than those in healthy controls ( P<0.05 ) , but there was no difference between the active and inactive RA ( P>0.05 ).The serum IL-17 levels in active RA patients were significantly higher than those in inactive RA patients and healthy controls ( P<0.05 ).The levels of serum IL-17 were positively correlated with RF in RA patients.Conclusion: IL-17 and RF play an important role in the pathogenesis and pathology process of RA ,and IL-17 may be associated with RA activity.
8.Reversal of Cerebral Vasospasm and Neuroprotection by Injecting Magnesium Sulfate into Cisterna Magna in Rabbit with Subarachnoid Hemorrhage

OBJECTIVE:To investigate whether the magnesium sulfate injection(MSI)into cisterna magna in a rabbit with subarachnoid hemorrhage(SAH)can reverse the cerebral vasospasm and damage of brain tissues.METHODS:The single-hemorrhage SAH rabbit model was used.Thirty New Zealand white rabbits were randomly divided into the following three groups(n=10,in the each group):sham group,model group and MSI group.At 24 hours after SAH,the rabbits were injected with 0.1 mL?kg-1 of saline into cisterna magna in sham and model groups,while 0.1 mL?kg-1 of 4% magnesium sulfate was injected into cisterna magna of rabbits in MSI group.All animals were sacrificed at 48 hours after SAH.Basilar arteries and hippocampus were then removed for measurement of the cross-sectional areas of basilar arteries and the hippocampus normal neuron density of CA1 regions.RESULTS:The cross-sectional areas of basilar artery and the hippocampus normal neuron density of CA1 regions were smaller in model group than in the other two groups(P0.05).CONCLUSIONS:This study results indicate that injecting magnesium sulfate into cisterna magna can reverse the cerebral vasospasm and the following hippocampal neuron damage in rabbits with subarachnoid hemorrhage.
9.Therapeutic effect of autologous stem cell transplantation of patients with cerebral hemorrhage sequelae under the stereotactic

Objective To investigate the therapeutic effect of autologous stem cell transplantation research for patients with cerebral hemorrhage sequelae under the stereotactic.Methods One hundred patients with cerebral hemorrhage from Jan.2011 to Sep.2013 in our hospital were selected and randomly divided into the experimental group (n =50) and the control group (n =50).The patients of experimental group were given autologous stem cell transplantation under the stereotactic in 6 months after cerebral hemorrhage,while the patients in control group were just given traditional treatment.At 6,7 and 12 months after cerebral hemorrhage,rate with neural function defect scale and functional independence measure(FIM) scores of the two groups were compared.Results FIM scores in the experimental group was 102.08 ± 8.28,significant higher than that in control group(95.28±8.75,P<0.05).Functional independence measure scores in the experimental group at 7 months after cerebral hemorrhage was 13.12±4.00,significant lower than that in control group(20.40±4.33,P <0.05).While,there was no statistical difference at 6 months and 12 months after cerebral hemorrhage between the two groups(P>0.05).Conclusion The therapeutic of autologous stem cell transplantation on patients with cerebral hemorrhage sequelae under the stereotactic is benefit at short term,but the long term therapy effective still needs further study.
10.Am80 inhibits neointima hyperplasia by promoting interaction of KLF4 with RARα

[ ABSTRACT] AIM:To investigate the inhibitory effect of Am80 on neointima hyperplasia in carotid arteries after balloon injury and to observe the interaction between Krüppel-like factor 4 (KLF4) and retinoic acid receptorα(RARα). METHODS:Neointima hyperplasia in carotid arteries was observed by hemotoxylin and eosin staining.The expression of KLF4 and cyclin D1 was examined by immunostaining and Western blotting analysis.To detect the interaction between KLF4 and RARαin the vascular tissue, the injured arteries were harvested, and the protein extracts were prepared and subjected to co-immunoprecipitation assay.RESULTS:Compared with injured group, Am80 significantly reduced neointi-mal hyperplasia and the thickness ratio of intima to media.Am80 not only up-regulated KLF4 or RARαexpression in caro-tid arteries, but also increased the interaction between KLF4 and RARαat tissue levels.CONCLUSION:Am80 inhibits neointima hyperplasia in carotid arteries after balloon injury by promoting the interaction between KLF4 and RARα.
