1.Breast Cancer Stem Cells and Genotyping

Objective To summarize the advancement of breast cancer stem cells and genotyping and analyze the correlation between the two.Methods Relevant literatures about breast cancer stem cells and genotyping,which were published recently were collected and reviewed.Results Cancer stem cell origin theory was supported by researches of correlation between breast cancer stem cells and genotyping,which also explained the complexity of intrinsic subtypes and heterogeneity of breast cancer.Conclusions A new way can be detected to study the formation mechanism and biological characteristics of breast cancer at the cellular and molecular level by researches of correlation between breast cancer stem cells and genotyping,which are expected to provide new strategies and tools for diagnosis and treatment of breast cancer.
2.Effect of Du-moxibustion on Cognitive Impairment after Stroke

Objective To observe the effect of Du-moxibustion on cognitive impairment after stroke. Methods 60 patients with cognitive impairment after stroke were randomly divided into control group (n=30) and treatment group (n=30). They were treated with conventional medical treatment and rehabilitation training, the treatment group received Du-moxibustion treatment in addition with once a day, 6 days per week for 8 weeks. They were evaluated with modified Barthel Index (MBI) and Montreal Cognitive Assessment (MoCA) before and after treatment. Results The scores of MBI and MoCA increased after treatment (P<0.05) in both groups and the scores were higher in the treatment group than in the control group (P<0.05). Conclusion Du-moxibustion can further improve the cognitive function and activities of daily living in stroke patients.

AIM:To investigate the effects of high-fat diet on the level of intracellular free calcium ([Ca2+]i) and the activity of angiotensinⅠconverting enzyme (ACE) in alveolar macrophages (AMs) of rabbits. The association between asthma and high-fat diet was also observed. METHODS: Twelve male New Zealand rabbits were medially divided into normal diet group and 1.2% high-cholesterol diet group randomly. 8 weeks later, bronchial alveolar lavage was performed in vitro. 4.Amyloid-? protein deposition in area of axon degeneration in Goll’s column of mouse

Objective To investigate the relationship between axon degeneration of Goll’s column system and Amyloid ? protein (A?) deposit sludge. Methods A? protein deposition in area of axon degeneration of Goll’s columns was measured by using immunohistochemical method.Results There was found strong positive round like A? deposit in the area of axon degeneration of Goll’s column system of GAD mouse. Spheroid appeared in the area of medullar nucleus of the Goll's column system at an age of 4 weeks. However the A? was negative and 9 weeks later, it became positive in partial spheroid and formed a strong round like material and became more and more standing up with age increase. The material mentioned above appeared consistently progressive in degeneration of axon in sequence.Conclusion It was suggested that the deposition of A? should be the result secondary to degeneration of nervous axon and be closely correlated with axon degeneration.
6.Effect of mulberry leaves extracts on glucose uptake of insulin-resistant HepG2 cells and the mechanism.

The effect and mechanism of mulberry leaves extracts (MLE) on glucose uptake of insulin-resistant HepG2 cells in vitro was explored. The insulin resistant models of HepG2 were induced by high concentration of insulin for 24 h. The models were incubated in a buffer containing mulberry leaves extracts. The glucose consumption was detected by glucose assay kits and the AMP-activated protein kinase (AMPK), Akt activation was examined by Western blotting. Mulberry leaves polysaccharides, mulberry leaves flavonoids and mulberry leaves extracts advanced glucose uptake of insulin-resistant HepG2 cells; Mulberry leaves extracts enhance phosphorylation of AMPK. Mulberry leaves extracts do not change the phosphorylation status of Akt. The glucose consumptions of insulin resistant model of HepG2 were promoted by mulberry leaves extracts. MLE stimulates HepG2 cell AMPK activity acutely without changing the Akt activity.

OBJECTIVE:To study the protective effect of shuanghuanglian sterilized powder for injection on hepatic in?jury of model mouse induced by human cytomegalovirus(HCMV)infection.METHODS:The mice were randomized into nor?mal control group,infection model group,ganciclovir group and shuanghuanglian sterilized powder for injection group(high,middle and low dosage),all of which except the normal control were vaccinated with HCMV5d to establish the hepatic injury model,each group was administered with different drugs;10days later,the enzyme activity of ALT and AST in the eye sockets blood of each group was determined;The mice were put to death by cervical vertebra dislocation and the necrosis degree of livers from the dead mice was evaluated and the pathological observation was conducted.RESULTS:The activity of ALT and AST were normal in the normal control,and no significant changes were found in the livers of this group;Compared with the normal control,the plasma ALT and AST levels have been increased significantly and there was a significant pathological hepatic injury in the infection model group;Compared with the infection model group,significant decreases were found in the activity of ALT and AST in the high and middle dosage groups of shuanghuanglian sterilized powder for injection,and there were obvious alleviated pathologic hepatic injury while no significant differences were found in the plasma ALT and AST ac?tivity in the low dosage group;Compared with the infection model group,the ALT and AST activity of the ganciclovir group were lowed significantly;Compared with the ganciclovir group,no significant differences were found in the ALT and AST ac?tivity in the high and middle dosage groups of shuanghuanglian sterilized powder for injection.CONCLUSION:Shuanghuan?glian sterilized powder for injection does have protective effect on hepatic injury of model mouse induced by HCMV.
10.Establishment and evaluation of detection method of bifidobacteria in human fecal using real-time fluorescence quantitative PCR

Objective To establish the real-time fluorescence quantitative PCR method for the detection of bifidobacteria in human fecal samples, and to provide an effective means for measuring intestinal bacteria. Methods Total DNA of bacteria was extracted from 60 cases of children's fecal samples. Three primers of bifidobacteria based on the 16S ribosomal RNA (16SrRNA)which possessed specialities of bacteria as amplified region were designed.The part of amplified 16SrRNA gene sequences was used as standard production.The serial dilution of standard was analyzed to build an absolute quantitative standard curve with SYBR GreenⅠ dye method, and the bifidobacterium contents in sixty human fecal samples were calculated. The sensitivity of the reaction was calculated by detecting the lowest detectable standard which determined the sensitivity of the reaction. The PCR products’melting curve was used to evaluate the specificity.The coefficient of variation (CV)of different batches of standard with the same concentration was used to evaluate the stability of reaction.Results The length of PCR product fragment which was used to build the standard curve was about 6 1 3 bp, the sequencing result was consist with the goals, and the standard sample of bifidobacteria was successfully established in real-time fluorescence quantitative PCR.The standard curve showed a good linear relationship with R2=0.999.The minimum detection value was 1.48×102 copies per reaction.The melting curve of real-time fluorescence quantitative PCR was a single peak.The test samples were batched and then examined by fluorescence quantitative PCR.The CV of standards’ Ct values which calculated from the standard (1.48 × 103 -1.48 × 107 copies · μL-1 )were 2.94%, 3.39%, 3.54%,3.08%,and 3.34%,respectively.The contents of bifidobacteria in fecal from 60 children was 7.77± 0.86(copies · g-1 wet fecal)transformed by logarithmic.Conclusion The established real-time fluorescence quantitative PCR method has high sensitivity, strong specificity and good repeatability, which is suitable for detection of human fecal bifidobacteria content.
