1.Effect of acrylamide on the degeneration and regeneration of rat myelinated fiber after sciatic nerve crush injury

BACKGROUND:Ola rats is a kind of rats with genovariation, who displays Wallerian degeneration after peripheral neuroaxonal damage that is slower than that normal 6J rats, thereby additional damage factor may help fully understand the property of Ola rats.OBJECTIVE: To observe the effect of acrylamide on the degeneration and regeneration of sciatic nerve medullated fibers following crush injury of C57BL/Ola (Ola) rat and C57BL/6J (6J) rat.DESIGN: Randomized controlled experiment.SETTING: Department of Neurology,Second People's hospital of Shenzhen; Department of Neurology of First hospital of Jilin University MATERIALS: This experiment was carried out in the neurological department in the University of Occupational and Environmental, Japan from January to June 1996. Twelve adult Ola rats and 6J rats were adopted and evenly randomly divided into experimental group and comparison group.METHODS: Rats were subjected to general anaesthesia, and then the proximal section of sciatic nerve was exposed and frustrated with hemostatic forceps for 10 s before suture. Rats in the experimental group were given intraperitoneal injection of acrylamide in a total dosage of 350 mg, which replaced by the same volume of physiological saline in comparison group.At 14 days after sciatic nerve torsion injury, all rats were anaesthetized again and the distal section of sciatic nerve was obtained and cut into slices, meanwhile the cross sectional area, the density and size frequency distribution of medullated fibers, as well as the number of medullated fibers in each nerve were determined.MAIN OUTCOME MEASURES: The density and size frequency distribution of sciatic nerve medullated fibers, as well as the number, the maximum diameter and the mean diameter of medullated fibers in two group of 0la rats and 6J rats.RESULTS: Totally 12 Ola rats and 6J rats entered the result analysis.① No Ola rat displayed Wallerian degeneration; But medullated fiber degeneration and following neonatal small diameter medullated fibers could be observed in 6J rats. ②In the experimental group, the total density of sciatic nerve medullated fibers in 6J rats was lower than that of Ola rat (P < 0.05) ;with the total number of medullated fibers in 6J rats also less than that of Ola rat (P < 0.01 ), which predominated by obviously reduced big diameter fibers (P < 0.01); The mean diameter of medullated fiber in 6J rats was also obviously smaller than that of 0la rat (P < 0.01 ).CONCLUSION: The Wallerian degeneration is extremely slow in Ola rat after torsion injury, which cannot be affected by acrylamide; while acrylamide has obvious inhibition on the axonal neogenesis in 6J rat after torsion injury.
2.Experimental pathology study on the effect of ACR on axon of both Ola mice and 6J mice

Objective :To observate the axon changes in pathology of Ola mice,compared with those of 6Jmice. Methods:The peroneal nerve and sural nerve were studied by light-microscope and electronmi-croscope. Results :In light-microscope,the total transverse fascicular area was significantly large ;density ofmyelinated fibers was significantly less;the maximal diameter of myelinated fibers was significantly less;minimal diameter of myelinated fibers had no changens in 6J mice. The Ola mice were nomal. In electronmi-croscope observation, the neurofilament was accumulated within axons. Conclusion: In Ola mice treatedwith ACR,the like-Wallerian degeneration wes delayed. However,in 6J mice the neurofilament and mito-chondria accumulation was found within axons.

Objective To explore the change of opioid peptide content such as ?-endorphin(?-EP), dynorphinA_ 1-13(DynA_ 1-13) in plasma and cerebrospinal fluid(CSF) of patients with cerebral hemorrhage, and the interference effect of Naloxone on them.Methods 60 patients with cerebral hemorrhage were randomly divided into Naloxone-treated group and control group. Conventional treatment was used in both groups, then Naloxone-treated group intravenous drip of Naloxone, each 3.0 mg, once a day, for 14 d. The concentrations of ?-EP, DynA_ 1-13 of the patients were measured with radioimmunoassay (RIA) during the acute stage before treatment and at 7th, 14th day after treatment compared with those of normal group (the normal people and the patients were treated by operations). Changes of ?-EP and DynA_ 1-13 in bleeding part and amount of bleeding were analyzed. Observation, comparison and analysis of the degree of symptom relief, the size of the side effect, the prognosis of patients and the satisfaction of patients with medical treatment.Discussion on Chinese medicine in the treatment of subacute thyroiditis the high compliance of advantages, the advantage of fewer side effects,therapeutic advantage and the advantage of improving the prognosis and reducing the sequela of disease. [Result] Through clinical observation and comparison, traditional Chinese medicine to western medicine in the treatment of subacute thyroiditis, good treatment compliance, curative effect are exact, side effects rarely, high patient satisfaction,good prognosis show obvious advantage. 7.Radiologic Analysis of Non-bacteria Infectious Pneumonia in Children(A Report of 218 Cases)

Objective To study the X-ray characteristic of non-bacterial infectious pneumonia in children.Methods In 218 cases of non-bacteria infectious pneumonia, the pathogenic agents included influenza virus,parainfluenza virus,respiratory syncytial virus(RSV),adenovirus,mycoplasma(MYCO) and chlamydia.Of them,mono-pathogenic infection were 152,mixed infection were 66.All the cases had chest conventional radiograph,129 cases were followed up for 2 times or more.The average period of the follow up were 7 days.The findings of chest conventional radiograph were analysed.Results The radiographic features included lung marking heavy 197,disorder 187,straight 105 and obscure 194;pulmonary hilum enlarged 91,hilum obscure 93,emphysema 82,point opacities 81,pulmonary radio-translucent diminution 34,patch opacities 33.In 129 cases followed up the lesions were perfectly absorbed 9(6.97%),partly absorbed 56(43.41%),non-absorbed 53(41.08%),progress 7(5.42%).Conclusion The absorbed period of non-bacteria infectious pneumonia is more 7 days.There are some radiographic characteristcs in RSV and MYCO pneumonia.
8.Relationship between microalbuminuria and severity of acute ischemic stroke and outcomes

Objective To investigate the relationship between microalbuminuria (MAU) and the risk factor for acute ischemic stroke,the severity of the disease and outcomes.Methods A total of 156 consecutive patients with acute ischemic stroke were enrolled prospectively.They were randomly divided into either an MAU positive group (≥ 30 mg/g) or an MAU negative group (< 30 mg/g) according to urinary albumin/creatinine ratio (UACR).They were also randomly divided into either a good outcome group (0-2) or a poor outcome group (>2) according to the modified Rankin scale (mRS) scores.The various demographic and clinical data were compared,and the poor outcome of acute ischemic stroke and the independent factors of positive MAU were analyzes.Results A total of 156 patients with acute ischemic stroke were enrolled,including 84 males and 72 female; aged 53 to 78 years (mean 65.4 ± 6.2); the time from onset to admission was 1.5 to 28 h; 94 patients had good outcomes,62 had poor outcomes,and no one died; MAU was positive in 76 patients and MAU was negative in 80 ones.Multivariate logistic regression analysis showed that advanced age (odds ratio [OR] 1.992,95% confidence interval [CI] 1.108-2.374; P =0.015),complicated with diabetes (OR 2.497,95% CI 1.177-5.298; P =0.017) and atrial fibrilhtion (OR 2.338,95% CI 1.062-5.148; P =0.035),high serum homocysteine (Hcy) level (OR 2.541,95% CI 1.073-6.02; P =0.047) and UACR (OR 2.130,95% CI 1.396-3.017; P =0.001),MAU positive (OR 3.291,95% CI 1.681-6.444; P =0.001),high baseline National Institutes of Health Stroke Scale (NIHSS) score (OR 9.196,95% CI 2.828-19.815; P< 0.001) were the independent risk factors for poor outcomes in patients with acute ischemic stroke.There were significant differences in the proportion of the patients complicated with diabetes (P =0.038) and fasting blood glucose level (P =0.025),serum Hey level (P =0.022),and carotid intima-media thickness (IMT) (P =0.019) between the MAU positive group and the MAU negative group.The proportion of anterior circulation infarction was lower (P =0.033),the rates of the baseline NIHSS score (P =0.003) and poor outcome were higher in the MAU positive group (P < 0.001).Multivariate logistic regression analysis showed that increased diabetes (OR 2.237,95% CI 1.036-4.829; P =0.040) and fasting blood glucose (OR 1.223,95% CI 1.145-1.673; P =0.027),the increased Hey level (OR 2.542,95% CI 1.047-6.612; P=0.025),carotid artery IMT (OR 1.295,95% CI 1.106-1.362; P =0.023) and baseline NIHSS score (OR1.206,95% CI 1.044-1.219; P =0.023) were correlated independently with the positive MAU in patients with acute ischemic stroke.Conclusions Positive MAU is one of the independent risk factors for poor outcomes of acute ischemic stroke,it is closely associated with some risk factors for acute ischemic stroke,and it has a significant impact on the severity of acute ischemic stroke and outcomes.
9.The study of heat shock protein 70 as a adjuvant carrier on HBV-specific immune response in vitro

Objective To investigate the effect of mycobacterium tuberculosis heat shock protein 70(TB .HSP70) as an adjuvant carrier on stimulating hepatitis B virus (HBV) core antigen(HBcAg)specific immune response to an accompanying cytotoxic T lym-phocytes epitope peptide from HBV core antigen in vitro .Methods Recombinant proteins HSP70(P1)、HSP70-HBcAg(18-27) (P2)、HSP70-PreS2B (18-24)-PreS2Th(37-53)-HBcAg(18-27)(P3) were expressed in methylotropic yeast Pichia pastoris GS115 . The expression of recombinant proteins was identified by SDS-PAGE and Western blot .The effect of recombinant proteins on den-dritic cell and lymphocytes of chronic HBV infection volunteers was investigated in vitro .The maturation of dendritic cell was meas-ured by flow cytometry ;the secretion of Th1 cytokines such as IL-12p70 ,IL-1β,TNF-αand IFN-γ was measured by ELISA ;the proliferation of lymphocytes was measured by TdR-3H ;the HBV-spesific cytotoxic activity was measured by the classic 51 Cr .Re-sults The recombinant proteins (P1 ,P2 ,P3) were constructed successfully .P1 ,P2 ,P3 could activate dendritic cell from chronic HBV infection volunteers by upregulation CD1a ,CD40 ,CD86 and production Th1 cytokines such as IL-12p70 ,IL-1β and TNF-α. Especially P3 could better induce autologous T cells to generate HBV specific cytotoxic T lymphocytes response ,activate the prolif-eration of lymphocytes and release IFN-γeffectively .However ,the recombinant HSP70 showed no target cell killing and could not induce immune response effectively .Conclusion TB .HSP70 can be used as an adjuvant carrier to stimulate HBV specific immune response to an accompanying cytotoxic T lymphocytes epitope peptide from HBV core antigen ,and enhance immunogenicity of the cytotoxic T lymphocytes epitope peptide .The P3 with B-and T-epitope can activate the HBV specific immune response effectively .
10.Diagnostic and clinical value of low-dose volumetric high-resolution CT in children with bronchiectasis

Objective To investigate the value of low-dose volumetric high-resolution CT (vHRCT)in children with bronchiectasis.Methods Forty-nine children with bronchiectasis were diagnosed by vHRCT.The CT manifestations and follow-up changes were observed. Results Among the 49 cases,there were 43 cases(85.7%)with cylindrical bronchiectasis,2 cases with cystic bronchiectasis(5.7%) and 4 cases with varicose bronchiectasis(8.6%).Forty-four cases with bronchiectasis were complicated with pneumonic illness,among which 9 cases with atelectasis,5 cases with pulmonary fibrotic changes and 3 cases with airway foreign body.And 5 cases were complicated with endobronchial inflammation only.The CT signs of cylindrical bronchiectasis were as follows:bronchial wall thickening in 38(88.4%), double-track sign in 29(67.4%),signet ring sign in 35(81.4%),mucus plugging in 6(14.0%),air bronchogram in 9(20.9%),and bronchi appeared within 1 cm distance from the pleura in 18 cases(41.9%).Following-up 10 cases,bronchial dilatation improved significantly in 4 cases,progressed in 2 cases,and there were 4 cases with no significant change.Conclusion vHRCT is an effective method for early diagnosis of bronchiectasis in children,which is valuable for improvement of bronchiectasis by prompt and effective treatment.
