1.Observation on the effect of percutaneous endoscopic lumbar discectomy in treatment of lumbar disc hernition

Objective To investigate the efficacy of percutaneous endoscopic lumbar discectomy for the treatment of lumbar disc herniation. Methods Under local anesthesia,48 patients(oberserve group) with single lumbar disc herniation were treated by percutaneous endoscopic lumbar discectomy, and the spinal cord and the nerve root were decompressed under the guidance of the C-arm X-ray machine. The visual analogue scale (VAS) of preoperative and postoperative 1 month and final follow up of patients was used for the pain assessment, and at the same time the modified MacNab criteria was used for efficacy assessment, compared with 40 cases of percutaneous endoscopic discectomy patients( control group). Results Observe the operation time( 62.7 ± 12. 3) min and hospital stay (4.6 ± 1.2) d was significantly lower than the control group (87. 2 ± 16. 7) min, (8. 9 ± 3. 7) d, after 1 month (3.13 ±0.76) and the final follow-up( 1.35 ±0.23) VAS scores were lower than the control group(4.85 ± 1. 24) (2.64 ±0. 81) ( P < 0. 05) , using modified Macnab criteria to evaluate the clinical efficacy, excellent observation group was 77.1 % , significantly higher than 62.5 % (P < 0.05 ). Conclusion The efficacy of percutaneous endoscopic lumbar discectomy for the treatment of lumbar disc herniation was positive,but it should be strictly to master surgical indications.
2.Comparison of the construct validity of anxiety animal models in Kunming mice between young and old

Objective To explore the effect of different ages on behavioral dimensions of anxiety animal models in Kunming mice.Methods Young/Old,male,Kunming mice were recorded twice,for five minutes,with a one-week inter-trial interval in open field test (OFT),elevated plus maze (EPM) and light/dark box (LDB).The following parameters were evaluated:percentage of time spent in the OFT central area (Ctime%),percentage of number of squares visited in the OFT central area (Ccross%),the total number of horizontal squares in OFT (Cross),the total number of rears in OFT (Rear),percentage of time spent in the EPM open arms (Otime%),percentage of number of entries into the EPM open arms (Oentries%),the total number of entries into the EPM open and closed arms (Entries),percentage of time exploring in the LDB light area (Ltime%),percentage of number of squares crossing in the LDB light area (Lcross%),percentage of rears in the LDB light area (Lrear%),the total number of transition in LDB (Transition),the total number of horizontal squares in LDB (Cross) and the total number of rears in LDB (Rear).Results OFT-Ctime%,OFT-Ccross%,EPM-Otime%,EPM-Oentries%,LDB-Ltime%,LDB-Lcross% and LDB-Lrear% had significant loadings on anxiety factor; OFTCross,EPM-Entries,LDB-Transition and LDB-Cross loaded on locomotion-exploration factor.Old mice had higher anxiety emotion (Ctime%,t =3.12,P< 0.01; Ccross%,t =2.37,P< 0.01; Otime%,t =1.31,P< 0.05; Oentries%,t=1.57,P<0.05; Ltime%,t=4.15,P<0.01 ; Lcross%,t=3.28,P<0.01 ; Lrear%,t=2.73,P<0.01) and lower locomotion and exploratory activity (Cross,t =2.05,P < 0.01 ; Entries,t =2.09,P < 0.05 ;Transition,t=2.51,P<0.01; Cross,t=3.02,P<0.01),compared with Young.Conclusion Ages have no effect on structural dimensions of anxiety animal models.With the growth of ages,old mice have higher anxiety emotion but lower locomotion and exploratory activity.
3.Simultaneous determination of tryptophan and kynurenine in plasma by HPLC with UV detection

ObjectiveTo establish an accurate method for simultaneous determination of plasma Kyn and Trp by HPLC-UV detection.Methods Kyn and Trp were separated on Agilent Hypersil ODS column using 3-nitrotyrosine as internal standard.The mobile phase consisted of 15 mmol/L sodium acetateacetic acid (pH 5.5):acetonitrile 94∶ 6(v/v) at a rate of 0.8 ml/min.The chromatographic separation was performed at 25 ℃.The eluate was monitored with programmed wavelength setting at 360 nm from 0 to 4 min for Kyn and at 302 nm from 4 to 5 min for Trp.The method was applied to determination of plasma Kyn and Trp in 8 chronic glomerulonephritis,10 idiopathic thrombocytopenic purpura,15 chronic hepatitis B virus patients and 15 healthy controls from September to December in 2010.The differences were compared using ANOVA and SNK methods.Results The retention time of Kyn and Trp were 2.9 min and 4.4 min,respectively.For Kyn,the assay was linear from 0.44 μmol/L to 18.30 μmol/L.For Trp,the linearity was from 3.67 μmol/L to 470.00 μmol/L.The detection limits were 0.014 μmol/L for Kyn and 0.122 μmol/L for Trp,respectively.The within-day CVs were < 3% and the between-day CVs were < 4%.The mean recoveries yield were in the range of 92.29 to 104.40.The plasma concentrations of Kyn were ( 1.59 ± 0.28),(2.73 ± 0.56),(2.69 ± 0.44) and ( 1.54 ± 0.48 ) μmol/L,the plasma concentrations of Trp were (59.8 ± 10.0),(46.1 ± 11.7),(58.5 ±8.0) and (41.4±13.1) μmol/L,the Kyn/Trp were (0.027 4±0.007 5),(0.061 6 ±0.016 5),(0.046 7 ±0.009 1) and (0.038 3 ±0.007 5)in controls,chronic glomerulonephritis patients,idiopathic thrombocytopenic purpura patients and chronic hepatitis B virus patients,respectively.There were significance difference of Kyn,Trp and Kyn/Trp amony the four groups (F=23.734,8.463,20.921,all P<0.01).Conclusion The method is simple,fast,and suitable for applicability to clinical measurement.
4.Clinical observation of posterior pedicle screw fixation indirect decompression in treatment of thoracolumbar burst fractures

Objective To compare the clinical effect of the two surgical methods of posterior pedicle screw fixation decompression in treatment of thoracolumbar burst fractures,and in order to provide the basis for choosing the rational treatment.Methods Retrospectively analyzed the clinical data of 98 patients with thoracolumbar burst fractures underwent surgery of posterior pedicle screw fixation decompression from January 2007 to January 2011.Fifty-six patients were given posterior fixed indirect decompression surgery (indirect decompression group) and 42 patients were given posterior fixed direct decompression (direct decompression group).The patients were followed up for (1.03 ± 0.36) years after surgery,the image,recovery of neurological function,postoperative complications and capacity for independent living of the two groups were compared and evaluated.Results After surgery,the vertebral height ratio,Cobb angle,canal compromise rate in indirect decompression group were (91.67 ± 26.19)%,(10.10 ± 2.89)°,(18.61 ±5.32)%,in direct decompression group were(86.23 ± 24.64)%,(11.98 ± 3.42)°,(22.37 ± 6.39)%.There was significant difference compared with before surgery (P < 0.05) and no significant difference between two groups (P >0.05).After surgery,the neurological function of the two groups were improved,and the improvement in indirect decompression group was better than that in direct decompression group (P < 0.05).The postoperative complications ratio in indirect decompression group was 23.2% (13/56),significantly lower than that in direct decompression group[83.3%(35/42)](x2 =10.370,P< 0.01).There was 60.7%(34/56)patients with capacity for independent living in indirect decompression group,significantly higher than that in direct decompression group [40.5% (17/42)] (x2 =4.329,P < 0.05).Conclusion The posterior pedicle screw fixation indirect decompression in treatment of thoracolumbar burst fractures is a feasible operation method,and is worth to utilize in clinic.
5.The correlation analysis between dielectric parameters and haematological parameters in whole blood cell.

We have measured the AC impedances of blood samples from 30 healthy adults with the Agilent 4294A impedance analyzer at frequency ranging 0.01-100MHz. At the same time, we measured the erythrocyte sedimentation rate (ESR), hematocrit (HCT), plasma fibrinogen (FIB), blood glucose (BG) and D-dimer. Then we analyzed the correlation between ESR and dielectric parameters of blood by linear correlation analysis, and used multiple regression analysis to reveal the influence of these hematological data on dielectric spectral characteristics of whole blood cell when these data were available. Statistical analysis showed that ESR had a linear correlation with the part of dielectric parameters in whole blood cell. The Multiple linear regression analysis indicated that HCT and BG had an influence on dielectric spectral characteristics of human whole blood cell, and HCT was the major factor in haematological parameters that influenced dielectric spectral characteristics of whole blood cell.

7.Multi-slice spiral CT perfusion imaging for the evaluation of angiogenesis and cell cycle protein D1 expression in peripheral lung cancer

Objective To investigate the relationship between multi-slice spiral CT perfusion imaging and tumor angiogenesis and cell cycle protein D1 (cyclinD1) expression in patients with peripheral lung cancer. Methods Seventy-three patients with peripheral lung cancer underwent 16-slice spiral CTperfusion imaging. The CT perfusion imaging were analyzed for time density curve (TDC), perfusion parametric maps and the respective perfusion parameters (BF, BV, PS, PH, and PHpm/PHa). Correlation between the respective perfusion parameters and immunohistochemical findings of MVD measurement and cyclinD1 expression were evaluated. Statistical method used one factor analysis of variance (One-wayANOVA) and Pearson correlation analysis. Results The shape of TDC of three groups of peripheral lung cancers (adenocarcinoma, squamous cell carcinoma, and other type of cancer) was similar. They showed a steeper slope, was obvious increase of nodular HU value [ peak height of TDC is respectively (44. 87±6. 83), (34. 91 ± 8.05), (40. 66±5. 87) HU ], changed little after reaching peak height, became flat at peak, had a platform. Forty-four patients were cyclinD1 positive expression, the peripheral lung cancers with cyclinD1 positive expression showed significantly higher MVD value than that of the peripheral lung cancers with cyclinD1 negative expression [ respectively (33.88± 14. 81), (23. 17±11.66) band/high magnification, P < 0. 01]. The cancers with cyclinD1 positive expression showed significantly higher PH,PHpm/PHa, BF,BV, PS value than those of the cancers with cyelinD1 negative expression[respectively P <0. 05]. MVD was positively correlated with PH, PHpm/PHa, BF, BV, and PS of the three groups ofcancers with cyclinD1 positive expression. Among, the correlation coefficient (r value) of BV, PS, BF was0. 409, 0. 517,0. 503 respectively(all P < 0. 01). MVD was significantly correlated with them. The r valueof PH, PHpm/PHa is 0. 319,0. 324 respectively(all P < 0. 05). MVD was less correlated with them. MVDwas not correlated with PH, PHpm/PHa, BF, BV, and PS of the three groups of cancers with cyclinD1 negative expression. Conclusions Multi-slice spiral CT perfusion imaging moderately correlated with tumorangiogenesis and reflected MVD measurement and cyclinD1 expression. It provided not only a noninvasive method of quantitative assessment for blood flow patterns of peripheral lung cancer but also an applicable diagnostic method for peripheral lung cancer.
8.hARD1 antiserum preparation and primary immunohistochemical analysis of hARD1 in tumor tissues.

Human arrest defective 1 (hARD1) is an acetyltransferase; its physiological significance remains unclear. To explore the relationship between ARD1 protein and tumors, we detected the hARD1 protein in tumor tissues in vivo. We cloned hARD1 gene from Hela cell and construct recombinant plasmid pET28b-hARD1. The recombinant plasmid was transformed into E. coli BL21 (DE3)plysS. hARD1 protein was expressed by inducing with IPTG(1 mmol/L) and purified up to 95% through Ni2+ chelation affinity chromatography. We used the purified hARD1 protein as antigen immunized the Balb/c mice and obtained the hARD1 specific polyclonal antiserum. Through immunohistochemical analysis of different tumor tissues in vivo, we found that hARD1 expressed at high frequency in breast cancer, prostate cancer and lung cancer, especially, hARD1 expression frequency in breast cancer was up to 70%, which is higher than in the other tumors. These results indicate that the high expression level of hARD1 could be an indicator of the breast cancer. This new finding would be a foundation to further explore the relationship between breast tumor and hARD1.

9.Anticoagulant Ability and Heparinization of Decellularized Biomaterial Scaffolds.

In order to enhance the anticoagulant properties of decellularized biological materials as scaffolds for tissue engineering research via heparinized process, the decellularized porcine liver scaffolds were respectively immobilized with heparin through layer-by-layer self-assembly technique (LBL), multi-point attachment (MPA) or end-point attachment (EPA). The effects of heparinization and anticoagulant ability were tested. The results showed that the three different scaffolds had different contents of heparin. All the three kinds of heparinized scaffolds gained better performance of anticoagulant than that of the control scaffold. The thrombin time (TT), prothrombin time (PT) and activated partial thromboplastin time (APTT) of EPA scaffold group were longest in all the groups, and all the three times exceeded the measurement limit of the instrument. In addition, EPA scaffolds group showed the shortest prepared time, the slowest speed for heparin release and the longest recalcification time among all the groups. The decellularized biological materials for tissue engineering acquire the best effect of anticoagulant ability in vitro via EPA heparinized technique.

10.Analysis of test results of urinary hydroxyproline and c-terminal telopeptide of collagen type Ⅱ concentrations among Kashin-Beck disease patients in Qinghai plateau

Objective To detect urinary bio-markers of hydroxyproline (HYP) and c-terminal telopeptide of collagen type Ⅱ (CTX-Ⅱ) among population from Kashin-Beck disease (KBD) regions in Qinghai Province,and to provide the scientific data for prevention and control of adult KBD.Methods According to the "Diagnosis of Kashin-Beck Disease" (WS/T 207-2010),using case-control study,120 KBD patients (males 55,females 65) and 89 healthy controls (males 41,females 48) in Qinghai KBD regions were divided into case group and control group.Morning urine samples were collected.HYP and CTX-Ⅱ contents were analyzed by enzyme-linked immunosorbent assay (ELISA),then these results were corrected with creatinine.All the data were analyzed by SPSS 17.0 software.Results There was no significant difference in the age of male and female between case group and control group (t =1.813,1.131,P > 0.05).The medians of urinary HYP and CTX-Ⅱ contents among male patients were 74.91 μg/μmol Cr and 630.77 ng/μmol Cr,respectively,which were higher than those of control groups (51.38 μg/μ mol Cr,401.32 ng/μmol Cr,Z =3.068,3.246,P < 0.01).The medians of urinary HYP and CTX-Ⅱ contents among female patients were 91.07 μg/μmol Cr and 637.17 ng/μmol Cr,respectively,compared with those of control groups (88.37μg/μmol Cr,546.47 ng/μmol Cr),there was no significant difference in HYP content (Z =0.273,P > 0.05),however,the difference in CTX-Ⅱ content was statistically significant (Z =2.002,P < 0.05).Conclusion The urinary HYP contents of male patients with KBD change significantly,while the degradation of type Ⅱ collagen in male and female patients increases,and CTX-Ⅱ could reflect the metabolic changes of collagen in KBD.
