1.Growth differentiation factor 5 induces adipose-derived stem cells differentiating into chondrocytes

BACKGROUND:Growth differentiation factor 5 (GDF5) has been shown to play a crucial role in the development of chondrocytes via different regulatory mechanisms.
OBJECTIVE:To explore the effect of GDF5 on the chondrogenic differentiation of adipose-derived stem cel s (ADSCs).
METHODS:ADSCs in passage 4 isolated from Japanese White rabbits were cultured in the medium containing 0 (blank control group), 10, 50, 100, 150 and 200μg/L, respectively. The morphological changes were observed, and the optimal concentration of GDF-5 was screened, and its round coverslips at 1, 2 and 3 weeks of culture were selected to undergo immunocytochemistry, toluidine blue staining and immunofluorescent staining.
RESULTS AND CONCLUSION:The growth of ADSCs was stable in the medium containing 10 and 50μg/L GDF5, while apoptotic cel s appeared after induction with 200μg/L GDF5. In the medium containing 100 and 150μg/L GDF5, some spindle-shaped cel s changed into irregular shape, and became round or oval after 7-10 days of culture. The optimal concentration of GDF5 was 100μg/L. The cel s transfected by 100μg/L GDF5 were positive for type II col agen obviously and with blue-stained nucleus. The transfected cel s were positive for toluidine blue, and metachromatic granules were visible in the cytoplasm. The proteoglycan mRNA expression of the transfected cel s was significantly increased, and reached the highest at 3 weeks. These results suggest that GDF5 promotes the chondrocytic differentiation of ADSCs.
2.Genetic polymorphism of GSTM1 to lung cancer patients in north Sichuan of China

Objective To analyze the genetic polymorphism of GSTM1 to lung cancer patients in north Sichuan of China and compare with race from other district.Methods PCR-based technique was used to detect the genotypes of GSTM1 in lung cancer patients.Results In local lung cancer patients,the frequency of homozygous deletions(null genotype) for GSTM1 was 58.4 % (73/125).Among the patients,the frequencys of null genotype for GSTM1 were 62.5 % (20/32) in female,56.9 % (53/93) in male,56.1% (32/57)in patients with squamous cell carcinoma and 54.8 % (17/31) in patients with adenocarcinoma,respectively.The frequency of deletions of GSTM1 in lung cancer patients from north Sichuan of China is slightly exceeding those of Europe and Americas (P <0.05) and similar to the domestic result (P >0.05).Conclusion The genetic polymorphism of GSTM1 to lung cancer patients in north Sichuan of China dosen' t show distinguished feature for this district and race.
3.Radioprotective effect of E0703 on AHH-1 cells and the related mechanism

Objective E0703 is derived from estradiol and has anti-radiation effects on irradiated mice, the effects of E0703 on cell viability,cell cycle and the related mechanism in irradiated AHH-1 cells were investigated in this study.Methods Cell viability,cell apoptosis and cell cycle were determined by Cell Counting Kit-8.Annexin V-FITC double staining kit and Flow Cytometry,respectively.Level of mRNA and protein were detected by RT-PCR and Western blot,respectively.Results 3.0 Gy 60 Co γ-rays induced significant cell cycle arrest,necrosis,apoptosis and reduction of viability.Pretreatment with E0703 for 12 h before irradiation could increase cell viability and regulate cell cycle.but had no obvious effect on cell necrosis and apoptosis.The expression of cell cycle arrest related molecules Rb and MDM2 were increased after 3.0 Gy irradiation,but decreased significantly with pretreatment of E0703.Moreover.the phosphorylation of Rb was also increased.Conclusions The radioprotective function of E0703 might be exerted through influencing on cell cycle and promoting cell proliferation instead of apoptosis or necrosis.
4.Study on the correlation between the early phase insulin secretion index and 72 hours continuous glucose levels in patients of impaired glucose tolerance

Objective To investigate the correlation between the early phase insulin secretion index and 72 h continuous glucose levels in patients of impaired glucose tolerance (IGT). Methods According to repeated 75 g oral glucose tolerance test (75 g OGTT) ,62 cases were divided into 2 groups: normal glucose tolerance group (NGT group, 30 cases) and isolated impaired glucose tolerance group (IGT group, 32 cases). Insulin levels were detected and HOMA-IR,HOMA-β , ΔI30/ΔG30,AUCI were calculated. The blood glucose levels were monitored by continuous glucose monitoring system for 72 h. The characteristics of postprandial glucose excursion were studied based on peak postprandial glucose (PPC) concentration, time to PPG (Δt) , postprandial glucose excursion (PPGE) and duration of postprandial glucose excursion (DPE). They were statistically analyzed by SPSS12.0. Results The levels of PPG and PPGE were significantly higher in IGT group (P < 0.05). Δt and DPE delayed obviously in IGT group (P < 0.05). HOM A-IR in IGT group was higher than that in NGT group (1.68 ± 1.03 vs 1.15 ± 0.90, P < 0.01), Δ I30/ΔG30 and HOMA- β was significantly lower in IGT group than that in NGT group (3.85 ± 1.04 vs 6.42 ±1.05,52.97 ± 2.02 vs 55.68 ± 12.45, P < 0.01 or < 0.05). Conclusions Higher postprandial glucose levels are characteristics of IGT patients,and the function of islet β cell after glucose load is impaired more severely. The levels of FPG and 2hPG are positively correlated with insulin resistance, and negatively correlated with islet β cell function.
5.Rapid diagnosis of venous catheterization complications: an ultrasound imaging study

Objective To explore the value of ultrasound imaging in complications associated with venous catheterization.Methods Patients (n =853) underwent ultrasound imaging examination after venous catheterization.Ultrasonography manifestations of venous catheterization complications and clinical curative effect were analyzed and summarized.Results Phlebitis,thrombus and mechanical complications were found by ultrasound imaging examination.Pneumothorax,hemothorax,arterial puncture and hematoma were among the main mechanical complications.Among the 853 patients,85 cases (9.96%) were found to have complications by ultrasound examination,with 35 cases (4.10%) of phlebitis,26 cases (3.05%) of catheter-related thrombus and 24 cases (2.81%) of mechanical complications.Conclusions In summary,a variety of venous catheterization complications can be diagnosed using ultrasound imaging non-invasively,effectively and efficiently.Real-time ultrasound imaging examination can dynamically monitor the develop ment and evolution of venous catheterization complications and effectively evaluate outcomes.

Abstract
As a common public health problem in society, it can seriously affect sleep, and lead to sleep structure disorder, sleep pattern changes, and sleep disturbance. Poor sleep can adversely affect the normal development of physiological, psychological, cognitive, and behavioral functions, and reduce quality of life. On the basis of summarizing the definition, classification, and epidemic status of child abuse, this paper summarizes the sleep characteristics and influencing factors of childhood abuse victims. It aims to provide a scientific basis for developing prevention and intervention measures to address sleep problems among childhood abuse victims, so as to improve sleep quality, and promote their physical and mental health.
7.Noninvasive evaluation of renal allograft fibrosis by virtual touch tissues quantification.

OBJECTIVE:
To explore the value of virtual touch tissue quantification (VTQ) in the diagnosis of renal allograft fibrosis and analyze its advantages and limitations.
METHODS:
The renal allografts of 54 patients with renal allograft biopsies or nephrectomy were assessed by Virtual Touch quantification of Siemens-Acuson S2000.
RESULTS:
Stiffness of renal allograft was significantly correlated to the value of VTQ (Spearman r=0.796, P<0.05).
CONCLUSION
VTQ is a new technique in the assessment of renal allograft fibrosis.

8.Mammogram texture analysis in prediction of axillary lymph node metastasis for breast carcinoma

Objective To explore the value of mammogram texture analysis in prediction of metastasis of axillary lymph nodes for breast carcinoma.Methods Mammograms and clinical data of 171 patients with breast carcinoma confirmed by pathology were retrospectively analyzed,and all patients underwent axillary lymph node dissection (ALND).Then the patients were divided into axillary lymph node metastasis group and non-metastasis group according to the result of ALND.The texture features of these lesions were statistically analyzed,including gray-level histogram texture parameters (mean value,standard deviation,skewness,kurtosis and variance) and gray-level co-occurrence matrix texture parameters (energy,entropy,correlation,inertia,inverse difference moment and contrast).Results In all of 171 breast cancer patients,96patients had axillary lymph node metastasis,while 75 patients had no metastasis.Mammograms showed negative axillary lymph nodes in 119 patients and positive axillary lymph nodes in 52 patients,and the sensitivity and specificity of mammograms in the diagnosis of positive axillary lymph nodes was 48.96% (47/96) and 93.33% (70/75),respectively.Mammogram texture analysis showed the values of energy,entropy,inverse difference moment and correlation in axillary lymph node metastasis group were higher than those in non-metastasis group,while the values of inertia and contrast in the axillary lymph node metastasis group were lower than those in non-metastasis group (all P<0.05).The rest texture parameters had no significant differences between two groups (all P>0.05).Area under curve (AUC) for texture parameters of energy,entropy,inertia,inverse difference moment,correlation and contrast was 0.610,0.610,0.374,0.599,0.612 and 0.421 (all P<0.05),respectively.AUC of mammography,mammogram texture features,and the combination of mammography and texture features was 0.711,0.676 and 0.787 (all P<0.05),respectively.The sensitivity and specificity of mammogram texture features,the combination of mammography and texture features in diagnosis of axillary lymph nodes metastasis was 62.5% and 64.6%,66.7% and 82.7%,respectively.Conclusion Mammogram texture parameters are helpful for predicting axillary lymph node metastasis,and the combination of mammography and texture features can improve diagnostic efficiency of axillary lymph node metastasis.

OBJECTIVETo study the effects of 18beta-glycyrrhizic acid and 18alpha-glycyrrhizic acid on mRNA and protein expression of cytochrome P450 3A in cultured rat primary hepatocyte.
METHODRat heaptocyte were isolated by two-step in situ collagenase perfusion method; the hepatocytes were seeded in dishes coated with type I rat tail collagen, culture medium was added and changed daily after gelation, the effect of 18beta-glycyrrhizic acid and 18alpha-glycyrrhizic acid on the expression of CYP3A was determined by reverse transcriptase-polymerase chain reaction (RT-PCR) and western blot respectively.
RESULTTreatment with 18beta-glycyrrhizic acid of primary rat hepatocytes resulted in marked up-regulation of CYP3A1 expression at both mRNA and protein levels in a concentration dependent manner, while exposure to 18alpha-glycyrrhizic acid of primary rat hepatocytes resulted in marked down-regulation of CYP3A1 expression at both mRNA and protein levels in a concentration dependent manmer.
CONCLUSION18beta-glycyrrhizic acid and 18alpha-glycyrrhizic acid up-regulate and down-regulate CYP3A1 expression at the transcriptive levels.
