Objective To investigate the effect and mechanism of liver X receptor ( LXR ) agonist on expression of fatty acid synthase( FAS) in diabetic kidney. Methods In the part of in vivo study, immunostaining was used to detect the FAS protein expression in kidney. 16-week-old male db/db mice on C57BL/6 background were administered via gavage a LXR synthetic agonist, TO901317, at a dose of 3 mg · kg-1 · d-1 or vehicle ( 0. 5%Carboxymethyl Cellulose Sodium, CMC-Na) alone for 7 d;Quantitative RT-PCR and Western blot were used to detect mRNA and protein levels of FAS and SREBP-1. In the part of in vitro study, MCT cell(a mouse murine proximal tubule cell line)was treated with 10μmol/L TO901317 for 24 h or transfected with active SREBP-1c expression vector (SREBP-1cN). HEK293 cells(a human renal tubule cell line)were transfected with mFAS-(1. 7 kb)-luc, LXR expression vector or SREBP-1cN for 12 h. Quantitative RT-PCR and luciferase reproter assay were utilized to examine FAS mRNA level and FAS promoter activity. Results FAS was abundantly expressed in renal cortex, with low expresson in renal glomeruli. The mRNA and protein expressious of FAS in kidney of db/db mice were lowered compared with db/m mice. TO90137 treatment increased FAS mRNA expression by 1. 3-fold. TO901317 increased expression of SREBP-1 in kidneys of db/m and db/db mice by 5. 1-fold and 17-fold, respectively. TO901317 and overexpression of SREBP-1c increased expression of FAS in MCT cells by 1. 5-fold and 1. 8-fold. Transcription activity of FAS were induced by TO901317, LXR, and SREBP-1cN overexpressions in HEK293 cells. Conclusions Both direct(LXRE)and indirect(SREBP-1c)mechanisms may contribute to the up-regulation of FAS expression by LXR in renal proximal tubule cells.

Objective To explore the effect of a multimodal electroencephalogram ( EEG) data visualiza-tion system on the motor imagery ability of stroke survivors. Methods Twenty stroke patients were randomly di-vided into an experimental group and a control group, each of 10. Both groups were provided with brain-computer interface-based motor imagery ( MI) training. At the same time, the experimental group was monitored and guided using an online, multimodal EEG data visualization system developed in our department. The classification accuracy ( CA) and event-related desynchronization ( ERD) of the 2 groups′ motor imagery were compared before and after the treatment. Results Before the treatment, no significant differences in the average CA of MI were found be-tween the experiment group (50.92±2.08) and the control group (49.35±4.20)(P>0.05). After the treatment, however, the experimental group′s average CA had increased to (64.52±5.27), significantly higher than that of the control group (51.18±5.02). When the stroke patients imaged affected upper extremity movements, obvious ERD was observed in the α frequency around the bilateral central motor regions of both groups, especially in the experi-mental group, but without significant differences between the two groups. However, no significant changes were found in the ERD of theβwaves of the two groups( P>0.05) . Conclusion The proposed online multimodal elec-troencephalogram data visualization system can help stroke patients imagine movements actively. It is worth sprea-ding in clinical practice.

We reviewed the fermentation for dihydroxyacetone production. Microbial fermentation is better for dihydroxyacetone production as compared to chemical methods. Gluconobacter oxydans was recognized as the most important strain for industrial production of dihydroxyacetone. The dihydroxyacetone yield is associated with many factors such as substrate, product, oxygen and biomass concentration. Repeated fed-batch fermentation and immobilization fermentation were recognized as the most potential process in various fermentation mode. Construction of recombinant microorganism and optimization of process are future directions of dihydroxyacetone production.

Objective To analyze the global research status, hotspots, and frontiers of proprotein convertase subtilisin/Kexin type 9(PCSK9)monoclonal antibodies, and to provide a reference for related scientific research and the rational drug use in clinical practice in China. Methods The research literature related to PCSK9 monoclonal antibody included in the Web of Science database was searched for the period from January 2011 to February 2022,and the literature included in the study was visually analyzed by the CiteSpace software. Results A total of 723 articles were included, and the annual number of publica-tions showed an overall upward trend. The top three countries were the United States,France, and the United Kingdom.Sanofi was the organization with the largest number of articles, and the organization with the highest citation of articles was Brigham and Women's Hospital. The hotspots of research mainly included the use of PCSK9 monoclonal antibody in the treatment of patients with hypercholesterolemia, patients who do not tolerate statins, patients with high cardiovascular risk, and the efficacy and safety of PCSK9 monoclonal antibody in lipid-lowering therapy combined statins; The frontiers of research in recent two years is the appli-cation of PCSK9 monoclonal antibodies in patients with acute coronary syndrome and the clinical benefits after reducing the level of lipoprotein(a).Conclusion A large number of studies have confirmed the efficacy and safety of PCSK9 monoclonal anti-bodies in reducing blood lipids, but there is still a lack of research on its economics and application in special populations, which should be the focus of future research.

OBJECTIVE To explore the hotspots of aging adaptation of drug package inserts, and to provide evidence for the development of aging adaptation of drug package inserts in China. METHODS The relevant English literature on drug package inserts for the elderly published from 2012 to 2022 was retrieved from Web of Science Core Collection; bibliometric analysis was performed by using VOSviewer and CiteSpace software, to explore research hotspots in this field, and summarize obstacles and solutions for the development of this field. RESULTS & CONCLUSIONS This study collected a total of 335 literature related to the aging adaption of drug package inserts, from 819 research institutions in 51 countries (regions), involving 2 174 authors. The research development of drug package insert adaptation for the elderly has slowed down in the past decade, and developed countries such as the United States and Japan dominate this field. Authors such as Wolf from Northwestern University in the United States, have the largest number of publications(12 literature). The research focuses in this field include the risk management of medication for the elderly, the updating of medication information for the elderly in drug package inserts, and the understanding and compliance of the elderly with drug package inserts and their influencing factors. The solutions to related obstacles in the development of aging adaption in drug package inserts include improving the visibility and readability of drug package inserts, filling in the information on elderly medication in drug package inserts, and so on. China can learn from the experiences and methods of other countries, conduct investigations into the influencing factors of elderly package inserts and pharmacokinetic studies based on the characteristics of the Chinese population, and improve the safety of medication for elderly patients in multiple dimensions.
