1.The study of the correlation between the IFN-γ mRNA expression in heart allograft and postoperative time

Objective: This study was designed to determine the expression of IFN-γ mRNA in heart allograft and its correlation to postoperative, cell infiltrating and allograft survival time. Methods: The heterotopic rat heart transplantation model was established with a simplified method, and the allografts and isografts were harvested at 1, 3,5,7,9 and 11 postoperative days. After the total RNA was prepared, the standard primer and target primer were amplified in the same tube, the average ratio of the target products pixel volume to primer's was calculated and a coordinates chart was made. Results: The expression level of IFN-γ mRNA in heart allograft was elevated and its peak was on the fifth postoperative day, and prior to the heart death has a positive correlation with cell infiltrating. Conclusion: IFN-γ may play on important role in postoperative rejection.

4.Relationship between tissue factor and coagulation in acute vascular rejection of heart xenotransplantation

Objective To investigate the role of tissue factor in the occurrence and development of co agulation in acute vascular rejection of heart xenotransplantation. Methods An animal model for acute vascular rejection of heart xenotransplantation was es tablished by using rat as recipient and guinea pig as donor. The xenografts were removed at 4, 8, 12, 16, and 24 h after the operation for fibrin sedimentation by immunohistochemical technique to evaluate coagulation. Meanwhile, the express ion of tissue factor mRNA was detected at different time points by RT-PCR. Hear ts of normal guinea pigs were used as controls. Results Immunohistochemical examination showed that coagu lation developed at the 8 h when interstitial fibrin deposition and fibrinous th rombus appeared and aggravated with time. High expression of guinea pig tissue factor mRNA appeared at 4 h after the transplantation and then steadily d eclined and totally terminated at the 16 h. The expression of rat tissue factor mRNA appeared 16 h post operation and then kept steadily enhanced. Conclusions The tissue factor acts as an important role in the acute vascular rejection of h eart xenotransplantation. The high expression of donor tissue factor mRNA might be related to the trigger of coagulation and the high expression of the receptor ’s tissue factor may be related to the further development of coagulation.
5.Clinical observation on 32 cases of hypothyroidism of thyroid cancer after operation treated with Shengxian decoction combined with levothyroxine sodium tablets

Objective To observe the clinical effects of modified Shengxian decoction in the treatment of thyroid cancer postoperative hypothyroidism patients.Methods 60 patients of thyroid cancer postoperative hypothyroidism due to deficiency of spleen and kidney were randomly divided into a control group and a treatment group.The control group (28 cases) was by levothyroxine sodium tablets conventional treatment,while the treatment group (32 cases) was treated with Shengxian decoction based on thecontrol group.The changes of the symptom integral and HAMD-17 were observed between two groups before and after treatment.Results After treatment,the total effective rate was 87.5% in the treatment group and 64.3% in the control group,the clinical effect of treatment group was significantly better than the control group (x2=4.499,P< 0.05).After treatment,the total score of HAMD-17 was 6.8±2.3 in the treatment group and 15.8±2.1 in the control group,both significantly improved than that before the treatment (which were 19.4±1.45 and 18.9± 1.32 respectively).Compared with the control group,the treatment group improved significantly (P< 0.01).Conclusion Shengxian decoction plus western medicine has remarkable curative effect in the treatment of thyroid cancer postoperative hypothyroidism,it can improve the depressive state of patients and improve the quality of life.
6.Analysis of factors related to breastfeeding of premature infants discharged from neonatal intensive care unit

Objective:To retrospectively analyze the influencing factors during the hospitalization of premature infants in the neonatal intensive careunit (NICU) based on the breast-feeding choices of the primary family caregivers of premature infants discharged from NICU.Methods:According to the breast-feeding status of 342 premature infants discharged from the NICU from The First hospital of Jilin University from June 1st, 2019 to December 31th, 2019, a self-designed data collection form for premature infants was used to investigate them, and the influencing factors of NICU hospitalization on breast-feeding selection were retrospectively analyzed.Results:The factors influencing breastfeeding included gestational age ( tvalue was-2.177, P=0.029), birth weight ( tvalue was-2.036, P=0.043), feeding mode during hospital stay ( χ2value was 6.582, P=0.010), length of hospital stay ( Zvalue was-2.205, P=0.027), maternal age ( Zvalue was-2.975, P=0.003), maternal education level ( χ2value was 8.350, P=0.04) and twin pregnancy ( χ2value was 7.367, P=0.007). The results of multivariate Logistic regression analysis showed that breast-feeding during hospitalization ( P=0.003) and older age of the mother ( P<0.001) were favorable factors to promote breast-feeding. Twin pregnancy ( P=0.006), low maternal education ( P=0.001) and gestational age ( P=0.006) were the risk factors that were not conducive to the implementation of breastfeeding. Conclusion:During the hospital period, the implementation of breastfeeding and the old age of the mother are conducive to the choice of breastfeeding after discharge, while twin pregnancy, low education of the mother and premature infants of large gestational age are not conducive to the implementation of breastfeeding.
7.Effects of Tangtong Formula on RSC96 Schwann Cells Apoptosis and Proliferation Induced by High Glucose

Objective To screen the optimal dose of Tangtong Formula in vitro. Methods RSC96 Schwann Cells were cultivate by DMEM mediums which contains different concentrations of glucose (5–125 mmol/L). The prevention effects of Tangtong Formula at different concentrations (0.25, 0.5, 0.75, 1.0, 1.5, 2.0, 3.0, 5.0 mg/mL) on the proliferation of RSC96 Schwann Cells induced by high glucose were detected. After the RSC96 Schwann Cells were cultivated in 100 mmol/L and 125 mmol/L high glucose mediums for 72 h, the apoptosis of RSC96 Schwann Cells was detected by flow cytometry Annexin V/PI, and the apoptosis rate was calculate; the proliferation situation of RSC96 Schwann cells in different times was detected by CCK-8 method. Results RSC96 Schwann cells were in apoptosis after being intervened by 100 mmol/L and 125 mmol/L high glucose mediums. The apoptosis rates were respectively(7.46±0.96)% and(16.53±1.01)%, with statistical significance compared with control group (P<0.01). Different concentrations of Tangtong Formula could alleviate the inhibitory effect of high glucose on the proliferation of RSC96 Schwann cells, and the threshold concentration of Tangtong Formula in 24 h was 0.25 mg/mL. The concentrations of Tangtong Formula in 0.5 mg/mL, 1.0 mg/mL, and 1.5 mg/mL could inhibit the apoptosis of RSC96 Schwann cells induced by high glucos, compared with 125 mmol/L high glucose group, the apoptosis rate of RSC96 Schwann cells decreased significantly (P<0.05, P<0.01). Conclusion Among three different doses, when the dose of Tangtong Formula is in 1.5 mg/mL, the effects on inhibiting apoptosis are the best.
8.Review of autophagic mechanism on the TCM benefiting qi and promoting blood circulation for diabetes

The literatures were searched and summarized about the autophagic operating mechanism on the TCM benefiting Qi and promoting blood circulation for the diabetes and diabetic complications. The study found that Chinese herbs with the function of benefiting Qi and promoting blood circulation could motivate the tissues's autophagy, which had good the rapeutic effect on diabetes and diabetic complications.
9.Clinical Study of Chinese Herb Formula Tangtongfang by Bathing the Affected Feet with Diabetic Peripheral Neuropathy

Objective To investigate the effects of Chinese herb formula Tangtongfang by bathing the affected feet of patients with diabetic peripheral neuropathy (DPN) in the pattern of Qi deficiency and blood stasis. Methods A parallel, randomized, controlled clinical research was performed. 70 diabetics in the inpatient and outpatient divisions of the Department of Endocrinology, Guanganmen Hospital, from January 2005 to October 2007 were investigated. They were divided randomly into two groups, treatment group and control group, 35 cases in each group. Before study, there were no statistic differences between the two groups, including gender, age, course of diabetes mellitus(DM), course of DPN, blood glucose (including fasting blood glucose and postprandial blood glucose), traditional Chinese medicine(TCM) symptom scores and nerve conduction velocity (NCV). The treatment group were treated with Tangtongfang bathing and the control group were treated with warm water bathing. Each group was also administered Vitamin B1 20 mg, t.i.d and Vitamin B6 20 mg, t.i.d respectively. All patients were keeping their routine treatment and diabetic care to control their blood glucose, such as diets, practices and drugs. Clinical symptoms and electrophysiological tests in diabetic peripheral neuropathy syndrome were investigated. The period of treatment was 8 weeks.Results After study, 9 cases retreated from this clinical trial totally, 4 cases in the treatment group whereas 5 in the control group. Among 4 common single symptoms, coolness feeling of the feet of diabetic patients was significantly improved (P<0.05) after bathed with Tangtongfang. The change of TCM symptom scores and tibialis NCV was significantly improved (P<0.001, P<0.05) in the treatment group. The improvement ratio was 45.2% and 61.3% in the treatment group, while was 10% and 3.3% in the control group. In each group there was no statistical difference between pre-and post-study with blood test, urine test, blood ALT, blood BUN and Cr, ECG examination (P>0.05). No adverse event was observed in each group.Conclusion Tangtongfang bathing improved several symptoms of patients suffering from DPN, especially the symptom of coolness feeling of the patients' feet. No adverse event was observed. This result showed that Tangtongfang bathing was worthy of using as a safe agent in treatment of DPN.
10.Risk factors of thyroid nodule in diabetic patients and the correlation with Traditional Chinese Medicine constitution

Objective:To explore the risk factors of thyroid nodules in diabetic patients and its correlation with Traditional Chinese Medicine (TCM) constitution.Methods:A Total of 213 cases of diabetic patients in Guang’anmen Hospital and Tangshan Hospital from January 2019 to August 2020 were choosen to do the questionnaire, with containly symptom and constitution. The patients were divided into diabetes with thyroid nodules group and diabetes without thyroid nodules group according to whether thyroid nodules were combined. We compared the clinical data characteristics of 2 groups, and used multi-factor logistic regression model to analyze the risk factors of diabetic patients with thyroid nodules and their correlation with TCM constitutions. Results:Diabetes patients aged from 50-80 years old [ OR=2.949, 95% CI (1.266-6.714)], females [ OR=3.736, 95% CI (1.823-1.541)], diabetes duration≥15 years [ OR=1.558, 95% CI (1.623-1.585)], elevated HbA1c [ OR=5.862, 95% CI (1.418-23.629)], elevated VLDL [ OR=2.851, 95% CI (1.597-6.824)], frequent insomnia [ OR=1.970, 95% CI (1.315-3.395)], Qi stagnation [ OR=4.357, 95% CI (2.634-8.377)], blood stasis [ OR=4.420, 95% CI (1.874-15.258)] are more likely to suffer from thyroid nodules ( P<0.05). Conclusion:Diabetic patients aged from 50-80 years old, females, diabetes duration≥15 years, elevated HbA1c, family history of thyroid nodules, frequent insomnia, and mood swings are more likely to develop thyroid nodules; qi stagnation and blood stasis are dangerous constitutions for diabetic patients with thyroid nodules.
