MiR-494 involves in cell cycle regulation,differentiation and apoptosis processes of normal ceils.Recent study shows that abnormal expression of miR-494 is associated with oncogenesis closely,and it participates in the invasion and metastasis of tumor cells and so on.MiR-494 is not only a tumor suppressor gene,but also can be considered as a cancer-promoting gene.MiR-494 can regulate the oncogenesis and development of the tumor through a variety of target genes and signaling pathways.
2.Application of parecoxib sodium for preemptive analgesia in patients undergoing cerebral angiography

Objective To evaluate the effects of parecoxib sodium for preemptive analgesia in patients undergoing cerebral angiography. Methods Sixty patients undergoing cerebral angiography were divided into two groups by random digits table with 30 cases in each : group P and group F. Patients in group P received parecoxib sodium 40 mg 30 min before operation, while group F received fentanyl 1 μ g/kg 2 min before operation. The changes of hemodynamics were observed before operation (To), immediately right internal carotid artery angiography during operation (T1), immediately right vertebral artery angiography (T2),immediately left internal carotid artery angiography(T3), immediately left vertebral artery angiography(T4), 10min after operation (T5). The mean arterial pressure (MAP), heart rate (HR), pulse oxygen saturation (SpO2)and visual analogue score (VAS) were recorded at different times during the whole operation. Results There were no significantly differences on MAP, HR and VAS between group P and group F (P > 0.05 ). But the SpO2 at T1 ,T2,T3 in group F (0.94±0.03,0.95±0.02,0.95±0.02) were significantly lower than those in group P (0.98 ± 0.01,0.98 ± 0.02,0.98 ± 0.02 )(P<0.05 ), and 2 cases SpO2 < 0.90 at Ti. Conclusion Parecoxib sodium administered preemptively provides a nice analgesic effect in patients undergoing cerebral angiography, and is more safe than fentanyl.
3.Effect of Ganoderma polysacchride on intracellular free calcium and intracellular pH in murine T cells

AIM To extend the approach of the act ion of ganoderma polysacchride on intracellular signal events in T cells. METHODS Laser scanning confocal microscope imaging of the calcium and p H fluorescent indicator dye Fluo-3/AM and SNARF-1/AM were used to determine th e kinetic changes of [Ca2+]i and [pH]i in murine T cells induced b y a ganoderma polysacchride,designated GLB7. RESULTS It was fou nd that GLB7(20 mg*L-1) could increase [Ca2+]i and [pH]i at 1 min were 334.7%±16.4%(n=3)、171.6%±10.4%(n=3) respectively. T he increase in [Ca2+]i induced by GLB7 was due to the influx of extr acellular Ca2+ and intracellular Ca2+ release through both IP3-se nsitive and IP3-insensitive Ca2+ stores, and increase in [pH]i indu ced by GLB7 was relative to Na+/H+ exchange systems and [Ca2+]i. GLB 7 did not influence [Ca2+]i and [pH]i in murine T cells induced by Con A(3 mg*L-1). CONCLUSION Stimulation of the increase in [Ca2+]i and [pH]i may be an important channel for gano derma polysacchrides to achieve their pharmacological actions.
4.Effects of Ganoderma Polysacchrides on PKC and PKA Activities in Murine T Cells

AIM:To investigated the effects of Ganoderma polysaccthride(GLB7)on protein kinase A(PKA) and protein kinase C(PKC) activitives in murine T cells.METHODS:A new ion-pair reversed-phase high liquid chromatography method was used to determinate the activities of PKA and PKC in T cells.RESULTS:GLB7 could markedly increase the activities of PKA and PKC in murine T cells in a dose-dependent manner.The peak time was at 5 min and 20 min and the activities of PKA and PKC returned to basic level at 20 min and 1.5h respectively.GLB7 could induce translocation of PKC and antagonize the inhibition effect of staurosporine(10μ mol· L-1) on PKC in T cells.CONCLUSION:The immunopotentiating and antitumour effects of Ganoderma polysacchride may be associated with its activation on PKA and PKC in murine T cells.
5.The antimicrobial activity of luteolin against four bacteria in vitro

Broth microdilution method was hired to measure the minimum inhibitory concentrations (MICs) of luteolin against Trueperella pyogenes,Escherichia coli,Salmonella and Streptococcus in order to evaluate the antimicrobial activity of luteolin in vitro.Meanwhile,the bacteria growth curves in medium containing sub-inhibitory concentration of luteolin were measured in this test.The results indicated that Tureperella pyogenes was the most sensitive to luteolin with a MIC of 0.078 g/L than that of these strains;and Salmonella was also sensitive to luteolin (MIC:1.25 g/L).However,The inhibitory effect of luteolin on Escherichia coli and Streptococcus is relatively weak,and shared the same MIC with 2.5 g/L.Luteolin showed inhibitory effects on the growth curves of all the strains in this test at sub-inhibitory concentration,and the inhibitory effects on the growth curves increased with the concentration of luteolin(P<0.05).

OBJECTIVE:To provide the scientific evidence and reference for improving medical reform. METHODS:Based on background ofSanming Medical Reform,self-designed questionnaires were used to investigate the cognition and satisfaction of local patients. The results of investigation were analyzed statistically. RESULTS:A total of 275 paper questionnaires were sent out and all were collected;345 electronic questionnaires were sent out by Wechat and all collected. Among 620 questionnaires,372 valid questionnaires were collected with effective rate of 60.00%. More patients(31.45%)thought the cost of treatment decreased, and less (22.31%) thought the cost increased. The proportion of high-income people who thought cost reduced was significantly higher than the proportion of low-income people (P=0.018). 27.42% of patients believed shortage of drugs,and tertiary hospi-tals were more prone to drug shortage than other medical institutions(P=0.062). The patients'satisfaction with medical staff and healthcare reform were both high(86.56% and 79.03%),but the satisfaction of rural patients was lower than that of urban patients (P=0.051). Understanding of patients onSanming Medical Reformwas in low level. Only 20.70% of the patients fully under-stood or generally understood ofSanming Medical Reform. The patients who thought the skills of medical staff were improved (40.32%)were more than those who thought the skills of medical staff were reduced(5.91%). CONCLUSIONS:Most patients are satisfied withSanming Medical Reform,and agree the achievements;but some problems are not solved,such as the shortage of some drugs,lack of grass-roots medical personnel,not understand medical policy.
7.Radiographic Evaluation of Chest After Open Heart Surgery

Objective To study the radiographic findings of the chest in patients after open heart surgery and its evaluating methods.Methods 236 cases after open heart surgery were divided into 3 groups according to the date of the first postoperative chest films:0~3 d,4~7 d and 8 d or more.The findings on pre and postoperative chest films were compared in 7 aspects respectively.Results 8 typses of complications were detected including severe ones such as acute pulmonary edema,the low cardiac output syndrome and ARDS.The incidences of widening of the mediastinum,pleural effusion and pulmonary interstitial edema were lower in over 8 d group than those in the other two groups(?
8.Preventive Effect and Underlying Mechanism of Tetramethylpyrazine on Hypobaric and Hypoxic Pulmonary Hypertension in Rats

Objective:To investigate the preventive effect of tetramethylpyrazine (TMP) on hypoxic pulmonary hypertension in rats and the underlying mechanism.Methods:Thirty-six male Sprague-Dawley rats were randomly divided into three groups:the normal control group,hypobaric and hypoxic group and TMP group (100 mg·kg-1·d-1).After the animal models of hypobaric and hypoxic pulmonary hypertension were established,mean pulmonary artery pressure (mPAP),mean carotid artery pressure (mCAP),ratio of right ventricular/(lift ventricular + interventricular septum) (RVHI) and morphological changes of pulmonary vessels were observed and the rates of wall thickness/external diameter (WT%) and wall area/total vascular area (WA%) were calculated.The contents of nitric oxide (NO),endothelin-1 (ET-1),hypoxic-inducible factor-1 (HIF-1) and vascular endothelial growth factor (VEGF) were detected in serum after the 21-day treatment with TMP.Results:The values of mPAP,RVHI,WT% and WA% of hypobaric and hypoxic group were significant higher than those of the normal control group (P<0.05 or P<0.01),and those of TMP group were obviously lower than those of hypobaric and hypoxic group (P<0.05 or P<0.01).The condition of hypobaric and hypoxic had no significant effect on mCAP,and there were no significant differences among the groups (P>0.05).The values of NO in serum of hypobaric and hypoxic group was lower than those of the normal control group (P<0.05) and those of TMP group were obviously higher than those of hypobaric and hypoxic group (P<0.05).The values of ET-1,HIF-1α and VEGF in serum of hypobaric and hypoxic group were higher than those of the normal control group (P<0.05) and those of TMP group were obviously lower than those of hypobaric and hypoxic group (P<0.05).Conclusion:TMP can effectively prevent pulmonary hypertension induced by hypobaric and hypoxic and structural remodeling of pulmonary arterioles.The mechanism may be related to the content up-regulation of NO and activity down-regulation of ET-1,HIF-1α and VEGF in serum of rats.
9.Leading effect of PETCO2 on mechanical ventilation in New Zealand white rabbits

Objective To observe the leading effect of end-tidal pressure of carbon dioxide in artery ( PET CO2 ) on mechanical ventila-tion in New Zealand white rabbits, and to establish parameters for medical animal experiments in terms of hemodynamics, blood gas, blood glucose, electrolyte. Methods 31 anesthetized New Zealand rabbits were practiced tracheostomy tube and mechanical ventilation. Respira-tion rate was 40 breaths/min and tidal volume was adjusted so that PET CO2 was 29 mmHg. Invasive blood pressure, electrocardiogram and PET CO2 were monitored. Blood gas analysis, electrolyte, hemoglobin and blood glucose were tested. Results When PET CO2 was maintained at 29 mmHg, the results were as follows:PH (7.42 ±0.07), 95% confidenceinterval (7.40~7.45);PaCO2(38.5 ±5.8) mmHg, 95%confidenceinterval (36. 4~40. 6) mmHg;BE (1. 45 ± 2. 80) mmol/L,95% confidenceinterval (0. 43~2. 48) mmHg. Conclusion Moni-toring of PET CO2 is good to guide mechanical ventilation in New Zealand white rabbits.
10.In vitro inhibitory effect of CD+8 cells from patients with aplastic anemia on normal CFU-GM growth was blocked by cimetidine

Objective To investigate the effect of histamine type Ⅱ (H2) receptors of the CD+8 cells from aplastic anemia (AA) patients on the inhibition of hematopoiesis.Methods Employing the modification of the panning method with monoclonal antibody the CD+8 cells from peri-pheral blood mononulcear cells of 17 aplastic anemia patients were obtained. The ribs moved during thoracic operations from 17 patients with non-hematologic diseases were as normal bone marrow resource. According to different group 2×104/ml CD+8 cells from AA patients and/or 1.0×10-5 mol/L or 0.5×10-5 mol/L cimetidine alone or in combination were added in the normal CFU-GM cultures.Reusults The normal CFU-GM from the rib bone marrows was 147.69±24.75/105 cells (±s), if 2×104/ml CD+8 cells from AA patients were added in thecultures, the CFU-GM formation significantly decreased (89.49±20.49/105 cells, P<0.01). If CD+8 cells from AA patients and 1.0×10-5 mol/L or 0.5×10-5 mol/L cimetidine were in combination added in the cultures, the CFU-GM were 144.96±25.34/105 cells or 156.28±32.81/105 cells respectively, compared with normal group P>0.05. If 1.0×10-5 mol/L or 0.5×10-5 mol/L cimetidine was added alone in the cultures, the CFU-GM number were 110.47±27.46/105 cells (compared with control P<0.01) or 160.06±32.22/105 cells (compared with control P>0.05) respectively.Conclusion 1.0×10-5 mol/L and 0.5×10-5 mol/L H2 receptor antagonist cimetidine completely abolished the suppressive effect of CD+8 cells from AA patients on the growth of normal CFU-GM and lower concentration cimetidine (0.5×10-5 mol/L) didn't suppress the growth of normal CFU-GM.
