1.Influencing factors of ankylosing spondylitis patients’life quality and medical coping styles

Objective To investigate the influencing factors of ankylosing spondylitis(AS)patients’life quality and medical coping styles.Method Two hundred patients visiting at the outpatient department of our hospital took part in the assessment on AS life quality and medical coping styles to look into the living quality of AS patients and its medical coping methods and influencing factors.Results The total score on life quality was(66.82±5.57),among whose dimensions the psychological function was scored the lowest by(10.65±4.05). Pearson correlation analysis showed that the scores on confrontation of coping styles were positively related with the dimension of social function in life quality(r=0.18,r=0.13,P<0.05),the scores on the avoidance of coping style were positively related to the dimension of physiological function in the life quality(r=0.12,r=0.14,P<0.05),the scores on yielding in coping styles was negatively correlated to physiological function of the life quality(r=-0.20,P<0.05).Regression analysis results showed that the AS patients’life quality was impacted by the course of the disease,coping styles and modes for paying medical expenses(P<0.05).Conclusions The AS patients’life quality is low and it is related to their coping style,the course of the disease and modes for paying medical expenses.Medical personnel should implement targeted interventions to guide patients into taking correct medical coping styles according to the specific condition of patients,in order to improve their quality of life.
4.Protection of hypothermic preserved isolated rat hearts by resveratrol and its underlying mechanism.

OBJECTIVETo investigate whether resveratrol (RES) plays a protective role in hypothermic preserved isolated rat hearts and whether it is mediated by regulation of silent information regulator protein-1 (Sirt-1) expression.
METHODSThe Langendorff model of isolated rat heart was used. After stored in different Celsior solution at 4 degrees C for 9 h, SD rat hearts were randomly divided into 7 groups: blank control group;9 h group (soley hypothermic preservation for 9 h); RES group (3, 10, 30 micromol/L RES treatment plus hypothermic preservation for 9 h ), niacinamide (NAM) group (40 micromol/L NAM added in Celsior solution plus hypothermic preservation for 9 h), RES + NAM group (30 micromol/L RES and 40 micromol/L NAM were added in Celsior solution plus hypothermic preservation for 9 h). The morphological changes of cardiomyocytes were detected by the HE staining with the light microscope. The mRNA and protein expression levels of Sirt-1 were detected by Real-Time PCR and Western blot respectively.
RESULTS(1) Compared with the blank control group, myocardiocytes were injured remarkably in the 9 h group and the Sirt-1 mRNA and protein expression levels were decreased significantly (P < 0.01); (2) Compared with the 9 h group, rat myocardial injury was alleviated gradually in 3, 10, 30 micromol/L RES group and the Sirt-1 mRNA and protein expression levels were increased in a dose-dependent manner (P < 0.05); (3) The above protective effects of RES were attenuated by Sirt-1 inhibitor NAM.
CONCLUSIONRES can protect myocardiocytes from injury caused by long range hypothermic preservation and this protective effect maybe mediated by upregulation of Sirt-1 expression.

7.Two-dimensional ultrasound imaging of atrial appendages in normal fetuses

Objective To understand demonstration methods and ultrasonographic features of left and right atrial appendages in normal fetuses.Methods 200 consecutive normal fetuses during gestation age 19-28 weeks from November 2011 through April 2012 entered the study.Ultrasonographic features and the demonstrated rates of atrial appendages at different incidence angle of ultrasonography and different gestational weeks were recorded and calculated,respectively.Results Atrial appendages of normal fetuses can be demonstrated on atrial appendages plane,which was little lower the view of parasternal great artery short axis.The display rate of left,right and left-right atrial appendages was 92%,68%,65.5%,respectively.When ultrasound beam entranced into chest from the right side,the display rate of left-right atrial appendages was highest(94.3%).The best time to observe atrial appendages was 22 to 24 week of pregnancy.The majority of left atrial appendages were fingerlike hook shapes,with narrower bases and longer bodys,and the endocardial surfaces were relatively smooth.Sometimes with incisures on external edges.Whereas the majority of right atrial appendages were obtuse-angled triangle shapes,with broad bases and shallower bodys.On endocardial surface the pectinate muscles were often seen,which looked like serrated echoes,sometimes prominent taenia sagittalis were noted in right atrial appendages.Conclusions Atrial appendages of normal fetuses can be demonstrated using two-dimensional ultrasonography on atrial appendages plane.The demonstrated rates of atrial appendages were different according to different incidence angle of ultrasonography and gestational weeks.There were some differences in ultrasonographic features between left and right atrial appendages,which is very helpful in determinating atrial situs.
8.Co-culturing cytokine-induced killer cells with dendritic cells enhances the cytotoxic activities of CIKs against hepatocarcinoma cells

Objective To explore in vitro cytotoxic activities of DCIKs against hepatocarcinoma cells by co-culturing cytokine-induced killer cells (CIKs) with dendritic cells (DCs) derived from peripheral blood of patients with hepatocellular carcinoma (HCC).Methods Peripheral blood mononuclear cells (PBMCs) were isolated from 23 patients with HCC and cultured with cytokines to induce DCs and CIKs.DCIKs were induced by co-culturing CIKs with DCs.After 14 days of co-culture,the phenotypes of DCIKs and CIKs were analyzed by flow cytometry,and their in vitro cytotoxic activities against SMCC-7721 and HepG2 hepatocarcinoma cells were measured by MTT assay.Levels of IL-12,IL-4 and IFN-γ in the supernatants of cell culture were detected by enzyme-linked immunosorbent assay (ELISA).Results High expressions of CD3+CD8+ and CD3+CD56+ were observed on DCIKs.The percentages of effector cells,cytotoxic activity and cytokine secretion were all significantly increased with DCIKs as compared with those CIKs without DC co-culture (P<0.05).Conclusion Co-culture of CIKs with DCs can enhance the differentiation of effector cells and the cytolytic activities of CIKs against hepatocarcinoma cells in vitro.Immunotherapy with DCIKs may be a promising strategy for the treatment of patients with HCC.
9.Study on effect of pH on rectum permeability of active ingredients in Reduning suppositories in vitro.

OBJECTIVETo investigate the effect of different pH on rectum permeability of chlorogenic acid and geniposide.
METHODFour kinds of Reduning suppositories of different pH were separated and put into the rectum to study the suppositories in vitro and the content of chlorogenic acid and geniposide samples was determined by HPLC to calculate the permeation in 24 hours.
RESULTWith increase of pH within 2.5-7.4, the steady state flux of chlorogenic acid was increased, but the steady state flux of geniposidesamples was steady.
CONCLUSIONAdjusted the pH can increase the rectum permeability of active ingredients in Reduning auppositories.

10.Local expressions of hypoxia-inducible factor-1α and vascular endothelial growth factor after photodynamic therapy in rats with glioma

Objective To observe local expressions of hypoxia-inducible factor-1α (HIF-1α) and vascular endothelial growth factor (VEGF) in Fischer 344 rats with glioma in the sub-acute phase after photodynamic therapy (PDT).Methods Thirty-two fischer 344 rats were implanted 9L cells into brain and randomly subjected to no treatment and PDT treatment (40 J/cm2,80 J/cm2,120 J/cm2).All animals received PDT treatment mediated with 5-Aminolevulinic acid (ALA) on the seventh day after implantation,respectively,and were then sacrificed at 7 days after PDT treatment.The tumor volume was measured by H&E staining,and the expressions of VEGF and HIF-1α in either glioma tissue or Brain adjacent to tumor(BAT) were detected by Western blot.Results The tumor volumes of control and 40,80,and 120 J/cm2 groups were (103.27 ± 8.83),(73.93 ± 7.55),(57.89 ± 7.53),and (48.57 ± 6.86) mm3,respectively.Tumor volumes decreased significantly after ALA-PDT treatment(F =216.183,P =0.000),and the decrease in tumor volume was PDT optical dose dependent(P < 0.001).Both VEGF and HIF-1 α expressions in glioma tissue were very high and no significant difference was found (VEGF:F =1.867,P =0.155 ; HIF-1 α:F =2.106,P =0.119) in all groups in the sub-acute phase after PDT,which were detected by Western blot.However,there were little VEGF and HIF-1α expressions in BAT and the expressions gradually increased with the increased dose of PDT(VEGF:F =37.065,P =0.000 ; HIF-1 α:F =39.775,P =0.000),among which PDT with 120 J/cm2 had the most obvious effect.Conclusion ALA-PDT effectively reduces glioma;however,it induces local expressions of HIF-1α and VEGF in BAT.
