1.Application of antibody drugs in the field of anti-infection.

In recent years, with the rapid development of antibody drugs, the antibody-based therapies have gradually expanded from the cancer and autoimmune diseases to metabolic and infectious diseases and so on. However, the development of antibody-based anti-infective drugs is much slower as there are only two kinds of drugs in the market. This is due to the complex infective mechanism of viruses, bacteria and other pathogens, and the monovalent character of monoclonal antibodies that greatly limit the anti-infection effect of antibody drugs. The development and application of novel technologies, such as recombinant polyclonal antibody technology, will greatly accelerate the development of antibody-based anti-infection drugs. This article will introduce the application and trends in the development of antibody-based drugs in the field of anti-infection therapy.

2.Improvement of Body Function with Physical Exercise in People with Disabilities (review)

Based on the theory and approach of International Classification of Functioning, Disability and Health (ICF), the literatures on the improvement of physical function by physical exercise were reviewed. People with disabilities can improve their body functions, in-cluding mental function, cardiovascular, blood, immune and respiratory function and neuromuscular skeletal and motor related functions via sport and exercise rehabilitation. Some advice was suggested about sports rehabilitation for the disabled.
5.Characteristics of growth and secretion in primarily cultured human pheochromocytoma cells

ObjectiveTo investigate the characteristics of growth and secretion in primarily cultured human pheochromocytoma cells.MethodsThe human pheochromocytoma tissues were digested successively,and the tumor cells were cultured.The growth curves of three tumors were plotted by cell count.At the same time,the catecholomine in the medium was measured with high performance liquid chromatography.ResultsThe number of primarily cultured human pheochromocytoma cells increased gradually from first to sixth day after plating,and then the cell number began to decrease from seventh day.The levels of norepinephrine(NE) and epinephrine(E) in the medium of cultured cells significantly decreased on first and second day,and the levels of NE and E had no significant difference on the second to eighth day.The levels of dopamine in the medium had no significant difference on the first to eighth day.ConclusionThe primarily cultured pheochromocytoma cells differentiate obviously after plating,but have no significant proliferation.The levels of catecholomine in the medium are decreased significantly on the first and second day.
6.In vitro inhibitory effect of CD_8~+ cells from patients with aplastic anemia on normal CFU-Mk growth is blocked by cimetidine

AIM: To investigate the effect of CD_8+ cells from aplastic anemia (AA) patients and its histamine type II (H_2) receptors on the growth of normal CFU-Mk. METHODS: The effects of CD_8+ cells and/or cimetidine, on normal human CFU-Mk growth were studied by using CFU-Mk assay. RESULTS: The CD_8+ cells from the perpheral blood of AA patients significantly suppressed the growth of normal allogeneic CFU-Mk. This inhibitory effect was blocked by cimetidine at concentration of 1.0?10~-5 mol/L. 1.0?10~-5 mol/L cimitidine alone didn't inhibit the growth of normal CFU-Mk. CONCLUSION: H_2 receptor antagonist cimitidine abolishes the suppressive effect of AA patients CD_8+ cells on the growth of normal CFU-Mk.
7.Effect of thrombopoietin on platelet activation in vitro

AIM:To study the effects of thrombopoietin (TPO) and other hematopoietic cytokines on platelet activation. METHODS: Using fluorescent-labeled monoclonal antibodies and flow cytometry.RESULTS: The per- centage range of platelets activated by 120 ng/mL TPO was 8. 89% -39. 92%,mean value was 17.43%; However, there were no effects of TPO 40 ng/mL and GM - CSF 100 ng/mL and IL- 3 100 ng/mL on platelet activation. CONCLUSION: Higher concentration of TPO directly stimulated platelet activation.
8.Urinary diary in evaluation of nocturia in benign prostatic hyperplasia patients over 60 years old

score, prostate volume and peak urinary flow rate, correlate with BPH, but not with nocturia. Keeping a detailed urinary diary for the patients is beneficial for elucidating their urinary function and in decision-making of their treatment regimen.
9.Cloning of D-hydantoinase Gene from Pseudomonas and Its Expression in E.coli

AIM The purpose is to construct D-hydantoinase genetic engineering strain for the purpose of the industrial production of D-p-hydroxyphenylglycine. METHODS D-hydantoinase gene was created from Pseudomonas putida 9801 by PCR technique and inserted into pMD18-T vector. The recombinant plasmid was transformed into several Escherichia coli strains. The positive transformants with D-hydantoinase activity were obtained by the two step screening, digoxigenin DNA labeling in situ hybridization and D-hydantoinase activity assay. RESULTS The D-hydantoinase activity of the genetic engineering strain E.coli BL21/pMD-dht was 1700 U*L-1 and increased as high as 8 times compared with those of wild-type strain Pseudomonas putida 9801. The subunit molecular weight of recombinant D-hydantoinase was about 53 kDa measured by SDS-PAGE. The amount of the recombinant D-hydantoinase was about 20 percent of total bacterial soluble proteins. CONCLUSION The genetic engineering strain E.coli BL21/pMD-dht possesses the initial industrial production prospects.
10.Correlation of tacrolimus blood concentration and the levels of blood lipid and fasting glucose after renal transplantation

Objective To study the implact of tacrolimus blood concentration (Tac) after renal transplantation on blood lipid and fasting glucose (FBG) in recipients.Method The recipients who regularly given Tac + mycophenolate mofetil + prednisone after renal transplantation were chosen and those had normal preoperative levels of blood lipid and fasting glucose as subjects.The recipients were classied into high concentration group,normal concentration group and low concentration group by comparing Tac blood concentration of different postoperative periods with corresponding normal concentration range.The changes of biochemical indexes such as steady Tac valley blood concentration,blood lipid and FBG were monitored,and the differences in blood lipid and FBG levels at different periods among three concentration groups were compared.Result TG level was significantly higher (P<0.05),and HDL-C level was significantly lower in high concentration group than in normal and low concentration groups one month after operation (P<0.05).Three months after operation,TG and FBG levels were significantly higher in high concentration group than in normal and low concentration groups (P<0.05 and 0.01,respectively).Six months after operation,TC,TG and FBG levels were significantly higher in high concentration group than in normal and low concentration groups (P<0.05,0.01 and 0.05,respectively).There was no significant difference between normal and low concentration groups in various biochemical indexes at any time point (P>0.05).Conclusion The higher blood concentration of Tac and the longer used after renal transplantation,the easier it might cause drug-induced hyperlipidemia and diabetes.
