1.Genetic Association Research between Mutation in CYP1A2 Gene and Erxian Soup Response

Objective To investigate the genetic association between mutation in cytochrome P450 1A2 (CYP1A2) gene and Erxian suop response. Methods Forty-eight patients with male climacteric syndrome of both Yin and Yang deficiency received a treatment of erxian suop with three months. The severity of symptoms and responses to Erxian suop were quantizated by the method of Department of Psychology in Istanbul Bosphorus University and Clinical Research Guidance Principle on New Drug of Traditional Chinese Medicine published by China. A single nucleotide polymorphism (SNP) at position 2964 (G-A) of CYP1A2 gene were identified by polymerase chain reaction (PCR) in order to analyse the relationship between Erxian response and CYP1A2 single nucleotide polymorphism. Results Statistical analysis showed there was no significant difference among genetype and genetype distribution of CYP1A2 G2964A between the patients and control group. The genotype of the 16 patients among 32 effective patients was G/G (56.25%). The genotype of the 11 patients among 16 ineffective patients was G/A (68.75%). There was no significant difference in symptoms, physical sign between the two groups at the pre-treat and post-treat. Also the two groups had no significant difference among the score of major symptoms and hyposymptoms. Conclusion The patients with genotype of G/G have higher efficiency, while with genotype of G/A have lower efficiency among patients with CYP1A2 G2964A gene when treated with erxian soup.
2.Determination the Contents of Bruchine and Strychine in Pingxiao Tablet by RP-HPLC

Objective To establish the RP-HPLC method for determination of Bruchine and Strychine in Pingxiao Tablet. Methods The lichrospher C18 (4.6 mm?250 mm, 5 ?m) was Used as the stationary phase. The Mobile phase consisted of water-acetonitrile (60∶40) with 1.7 g sodium dodecylsulphate and 3.4 g KH2PO4 in 1 000 mL mixture. The Detecting wavelength was 254 nm and flow rate was 1.0 mL/min. The column temperature was 25 ℃. Result The linear ranage of Bruchine and Strychine were 0.102~0.918 ?g and 0.208 ~ 1.872 ?g respectively. The recovery rates of bruchine and strychnine were 101.07% (RSD=2.26%) and 99.29% (RSD=2.79%) respectively. Conclusion This method is realiable and accurate, the method can be applied to determination the contents of Bruchine and Strychine in Pingxiao Tablet.

4.Survey of prevalence of iodine deficiency disorders and iodine nutrition in high-risk areas of Inner Mongolia in 2007 and 2008

Objective To investigate new cases of endemic cretinism in high-risk areas of Inner Mongolia, the prevalence of endemic goiter, the implementation of comprehensive measures, and to provide scientific basis for developing control strategies against iodine deficiency disorders (IDD). Methods In 2007 and 2008, a search for new cretin cases was conducted among children under the age of 10 in 11 historical serious epidemic cretinism prevalent areas of Songshan district, Keshiketeng qi, Kalaqin qi, Zhalaite qi, Zhuozi county,Liangcheng county, Qingshuihe county, Helin county, Dongsheng district, Zhungeer qi and Yinjinhuoluo qi. Three to 5 towns were selected in each qi(county, district) and 3 to 5 village primary schools were selected in each town.One hundred and twenty to 200 children aged 8 to 10 were selected in each school to inspect thyroid by B ultrasound and palpation and to test intelligence and urinary iodine. Two villages were selected in each town and 30 households were selected to determine urinary iodine of housewives and salt iodine. Results A total of 56 cases of suspected cases of endemic cretinism were found from the 11 counties but no case was confirmed. The goiter rate of children aged 8 to 10 was 5.2% (309/5922) by palpation and 4.3% (252/5922) by B-ultrasound. The median urinary iodine of children aged 8 to 10 was 241.5 μg/L, and urinary iodine was 100 to 300 μg/L, accounted 65.9%(3901/5920). Median urinary iodine of the 9 Qis(counties, districts) was in the range of 200 and 300μg/L.Higher than 5% of the 11 Qis(counties, districts) with urinary iodine lower than 50 μg/L was zero. Higher than 10% of the 11 Qis(counties, districts) with urinary iodine lower than 100 μg/L was 1. The median urinary iodine of housewives was 225.6 μg/L, and lower than 50 μg/L accounted for 2.2%(35/1597). Higher than 10% of the 11 Qis(counties, districts) with urinary iodine lower than 100 μg/L was 4. A total of 2109 households were investigated and 97.45%(2055/2109) of them ate iodized salt and 2.55%(54/2109) of them ate non-iodized salt. Conclusions In the 11 investigated counties, goiter rate of children has decreased to less than 10%, and no new cretin was found. It could be concluded that the fulfillment of prevention and control of IDD is effective. The iodine nutrition of children and women of childbearing age is in an adequate level. The coverage rate of iodized salt has maintained at a higher level, the sales network is sound, the salt price is reasonable and salt is easy to get and the children's intelligence is protected effectively. But, the health education about IDD is still weak, need to be reinforced.
5.Comparative analysis of specific IFN-γproduction induced by Mycobacterium tuberculosis in two dif-ferent nonhuman primates

Objective To analyze the differences of immune responses against Mycobacterium tu-berculosis antigens induced in two different nonhuman primates and to provide rationales for the selection of suitable animal models for vaccine efficacy evaluation.Methods Expression of functional surface markers including CD69 and HLA-DR, the activation markers on CD4+and CD8+T cells from in rhesus macaques and cynomolgus monkeys were measured by flow cytometry analysis.PBMCs were isolated from rhesus ma-caques and cynomolgus monkeys with Mycobacterium tuberculosis infection and stimulated with PPD and pep-tide pools ( ESAT-6/CFP-10) .Enzyme-linked imunospot ( ELISPOT) assay was performed to detect IFN-γproducing lymphocytes.Results The CD4+and CD8+T cells isolated from rhesus macaques without Myco-bacterium tuberculosis infection expressed higher levels of CD69 and HLA-DR than those from healthy cyno-molgus monkeys (P<0.01).The numbers of IFN-γspot forming cells/106 PBMCs in rhesus macaques with Mycobacterium tuberculosis infection for 10 and 11 months were respectively 3 and 3.5 times higher than that of cynomolgus monkeys upon after the stimulation of PBMCs with PPD.The levels of IFN-γproduction by the cells from rhesus macaque group were also higher than those from cynomolgus monkey group upon after the stimulation of PBMCs with ESAT-6 or CFP-10 peptide pools.Conclusion More IFN-γproducing cells were induced in rhesus macaques than that in cynimolgus monkeys after stimulation with Mycobacterium tu-berculosis antigens.Therefore, the rhesus macaques might be a better animal model for evaluating immune responses induced by Mycobacterium tuberculosis vaccines.
6.The application of mask oxygen supply in propofol-sedated gastroscopy

7.Relationship between serum 25-hydroxyvitamin D and indicators of glucose metabolism in inpatients with type 2 diabetes mellitus

Objective To investigate the correlation between 25-hydroxyvitamin D [ 25 ( OH ) D ] and indicators of glucose metabolism in inpatients with type 2 diabetes mellitus .Method We retrospectively ana-lyzed the clinical records of 214 inpatients with type 2 diabetes mellitus , including age , body mass index (BMI), systolic blood pressure, diastolic blood pressure, and laboratory test results such as serum parathyroid hormone (PTH), 25(OH)D, creatinine, calcium, phosphorus, glycosylated hemoglobin A1c (HbA1c), fast-ing blood glucose , and fasting insulin .Results The prevalence of vitamin D deficiency was high in these pa-tients, with 47.2%of them having a serum 25(OH)D concentration lower than 10 ng/ml.Using 25(OH)D level less than 10 ng/ml as the cut-off point, the patients were divided into vitamin D severe deficiency (Vit-SD) group and vitamin D non-severe deficiency (Vit-NSD) group.Compared with the Vit-NSD group, the Vit-SD group had younger age [ (55.27 ±13.71) years vs.(60.76 ±12.32) years, P=0.001] and higher HbA1c level [ (9.00 ±2.01)% vs.(8.45 ±1.86)%, P=0.025].BMI [ (25.09 ±4.01) kg/m2 vs.(25.39 ± 3.53) kg/m2, P=0.523], fasting blood glucose [ (8.91 ±3.31) mmol/L vs.(8.16 ±3.02) mmol/L, P=0.063], fasting serum insulin [ (21.32 ±32.50) mIU/L vs.(21.92 ±26.95) mIU/L, P=0.873] and homeostasis model assessment of insulin resistance index (97.60 ±8.92 vs.7.53 ±9.39, P=0.954) in these two groups were with no statistically significant difference .The association analyses showed that HbA 1c was sig-nificantly negatively correlated with 25 ( OH ) D ( r =-0.190 , P=0.003 ) and not correlated with serum PTH, calcium, or phosphorus , after adjusting for age and creatinine .Multiple linear regression revealed that HbA1c had a significantly negative correlation with age (β=-0.220, P=0.000) and 25 (OH) D (β=-0.184 , P=0.000 ) .Conclusions There is high prevalence of vitamin D deficiency in inpatients with type 2 diabetes mellitus, which is even higher in relatively young patients .Serum 25 (OH) D may be negatively correlated with HbA1c in these patients independently from age , BMI, fasting serum insulin, and homeostasis model assessment of insulin resistance index .
8.Clinical effect of Kudiezi injection on renal function based on propensity score.

To explore the effect of Kudiezi injection on renal function in the real world, in order to provide the basis for the clinical medication safety. Patient aged between 18-80 were selected from 18 large hospitals information system (HIS) databases established by clinical research institute for basic traditional Chinese medicine of China academy of Chinese medical sciences. The patients who were treated with Kudiezi injection (24 225 cases) were defined as the exposed group, whereas those who were not treated with Kudiezi injection (14,191 cases) were defined as the non-exposed group. The propensity score method was used to balance the confounding factors. Classic logistic regression, GBM weighted propensity score logistic regression, GBM propensity score weighted logistic regression with covariate and sensitivity analysis were adopted to study the effect of Kudiezi injection on renal function. The results showed no significant difference in the possibility in abnormality in serum creatinine (Scr) (P = 0.940, 0.679, 0.834) and urea nitrogen (BUN) (P = 0, 0.045, 0.164) between both groups. Therefore, the existing data indicated no damage of Kudiezi injection on renal function. Because this study is a retrospective study based on the real world, there may be unknown confounding factors and potential bias. Therefore, further studies shall be conducted to monitor whether Kudiezi injection causes damage on renal function, in order to ensure the clinical medication safety.

9.Variations in surface protein genes of avian influenza virus before and after infecting mouse

Objective To investigate the variations of surface protein genes of avian influenza virus (AIV)before and after infecting mouse.Methods Mouse lung tissue was infected with A/Goose/Guangaong/NH/2003(H5N1)and the virus was isolated 12 hours and 9 days after replication in lung tissue of mouse.The isolated strains were amplified in embryonated chicken eggs,anti the virion RNA was transcribed into cDNA by reverse transeriptase.After amplification and purification,dideoxy-mediated chain termination was performed to detect synthetic oligonucleotide primers and DNA sequence was analyzed.Results The homology of nucleotide sequence for HA gene of three isolated strains was 99.6%-99.8%.and that of amino acid sequences was 99.3%-99.6%.The homology of nucleotide sequence for NA gene of three strains was 99.8%-99.9%.all of them were synonymous mutatinns.No variation was found in M gene.Conclusion After replication in mouse lung tissue,no significant mutation was found in the surface protein genes of AIV except some point mutations in HA genes.

The impact of hypofunction of kidney on evaluating of islets β cell function in patients with type 2 diabetes was investigated. 635 type 2 diabetic patients with normal liver function were grouped using Cockcroft-Gault. Following the decrease in kidney function, blood C-peptide concentration was increased with decreased urinary exeretion of C-peptide(P<0. 05). It is proposed to pay an attention to renal function while evaluating islets β cell function in the patients.
