1.AN EXPERIMENTAL STUDY OF LIVINGBCG'S EFFECT ON THE KILLING AND DIGESTIVE ABILITIES OF RABBIT ALVEOLAR MACROPHAGE LYSOSOME TO LIVING-YEAST
Acta Anatomica Sinica 1953;0(01):-
By means of the observation of acridine orange-stained lysosomes with fluorescence microscope, we studied the intracellular killing and digestive process after alveolar macrophage (AM) ingested living-yeast. Based on the morphological changes of the cells, we divided the process into three stages: the pre-phagosome-lysosome fusion stage, the phagosome-lysosome fusion and killing stage, and the digestive stage. The results revealed that all the values of phagocytic index, phagocytic rate, fusion index and fusion rate of the isolated AM of rabbits infected with living-BCG were higher than those of normal rabbits (P

Objective To investigate the application and significance of human leukocyte antigen (HLA) and cross reactive groups(CREGs) matching in clinical renal transplantation. Methods A total of 312 cases of kidney transplantation were divided into two groups. In one group of 149 cases of kidney transplantations,ClassⅠCREGs matching criteria were applied instead of conventional HLA A,B matching which had two A,B mismatches(MM).In the other group of 163 cases of kidney transplantation there were A,B 0~2MM by the conventional criteria. The graft survival rate at 1 year and incidence of acute rejection within 1 month after transplantation were compared between the 2 groups. Results The percentages of HLA Ⅰ antigens 0,1,2MM were 16.7%,41.6% and 34.2% by CREGs matching criteria, and were 6.7 %,21.5% and 71.8% by conventional matching criteria. The matching rates in CREGs 0,1MM group were significantly higher than those in corresponding conventional matching group ( P 0.05);however,the incidence rate of CREGs 0MM group was obviously lower than that of A,B 2MM group ( P 0.05 ),but the rate of CREGs 0MM group was obviously higher than that of A,B 2MM group ( P

AIM: To investigate the effect of advanced glycosylation end products(AGEs) modified protein on morphological changes of actin cytoskeleton in primary endothelial cells and the role of MAPK signaling pathways in this pathological procedure.METHODS: Human umbilical vein endothelial cells(HUVECs) were incubated with AGEs modified human serum albumin(AGE-HSA) at concentrations of 12.5,25,50,and 100 mg/L,respectively,for 2,4,8,12 and 24 hours. The cells were treated with different chemical compounds of inhibitors,or adenoviral deliver approach(either dominant positive or negative adenoviral constructs) of MAP kinases to specifically block or activate certain signal transduction pathways under above situations. As control,HSA of the same concentration was administered to cells at the same time. The treated cells were incubated with FITC-phalloidin to stain F-actin.RESULTS: F-actin in HUVECs was rearranged greatly by AGEs in a concentration and time-dependent manners. The unmodified HSA did not influence Factin distributions. The AGEs-induced changes were blocked by pretreatment with SB203580,PD98059 for 30 min,or pre-infection with recombinant virus of dominant negative form of MKK 6b [MKK6b(A)],MEK1 [MEK1(A)] for 24 h,while SP600125 and dominant negative form of MKK7 [MKK7(A)] failed to inhibit the effects of AGEs.Furthermore, the infection of recombinant virus of constitutive active form of MKK6b [MKK6b(E)] or MEK1 [MEK1(E)] could also induced re-arrangement of F-actin,respectively,while the effect elicited by [MKK6b(E)] was abolished by co-infection with recombinant adeno-virus of dominant negative p38?. CONCLUSION: AGEs-induced morphological changes of F-actin in endothelial cells are mediated by p38-and ERK MAPK signal pathways.
