1.Specific expression of nucleolin in cap-stage tooth germ of mouse

Objective To investigate the specific expression of nucleolin in the development of cap-stage tooth germ of mouse embryo,and detect the possible biological function of nucleolin. Methods In situ hybridization and immunohistochemistry were performed to detect the expression of nucleolin mRNA and protein in the development of tooth germ at embryo day 15(E15).Double staining for apoptotic cells and nucleolin was employed to explore the relationship between nucleoin and apoptosis in tooth germ. Results The expression of nucleolin mRNA and protein was mainly detected in the cervical loop,inner epithelium and the underlying dental papilla.Moreover,nucleolin protein was also located in the primary enamel knot and basement membrane.It was revealed by double staining that the fluorescence images of nucleolin and the location of apoptotic cells were not overlapped. Conclusion The expression pattern of nucleolin mRNA do not completely coincide with that of nucleolin protein at the cap stage of tooth germ.The location for nucleolin protein in the basement membrane suggests that nucleolin may be involved in the reciprocal interactions between the inner epithelium and dental papilla mesenchyme,subsequently affect the morphogenesis of tooth germ.
2.Extra-bone joint appearance of 18F-FDG PET/CT in patients with adult-onset Still's disease

Objective To summarize the extra-bone joint characteristics of 18F-FDG PET/CT images in patients with adult-onset Still's disease. Methods Twelve patients with adult-onset Still's disease as confirmed by clinical and laboratory tests were enrolled in this study. The characteristics of 18F-FDG PET/CT images were retrospectively analyzed. Results Among the 12 patients, no patients were found with solid malignant tumors. Various degrees of high uptake were found in liver, spleen, bone marrow or lymph node. The hepatic SUV was 2.74 ± 0.89, the spleen SUV was 3.68 ± 1.24, and the SUV of thoracic spine was 4.36 ± 1.35. For the lumbar vertebral, the value of SUV was 4.26 ± 1.25. Lymph node high uptakes were found in 8 patients at different sites , including retroperitoneal , pelvic cavity and pelvic wall , inguinal bilateral neck , hepatic portal , axillary lymph nodes. The value of SUV was 3.76 ± 1.41. Conclusions 18F-FDG of AOSD has certain characteristics, such as diffusely uptake of liver, spleen, bone marrow, or lymph node, with or without pleural effusion.
3.Systematic report on re-evaluating kudiezi injection.

There are few articles or reports collecting evidence about Kudiezi injection from premarketing and postmarketing research or studies systematically. This article is an exact miniature of a systematical report about Kudiezi injection. We analyzed information from four aspects, such as quality control reports, non-clinical premarketing safety experiments, postmarketing research (efficacy studies, hospital information system data and national spontaneous reporting system data), and literature analysis. All the four aspects build an evidence body for Kudiezi injection in order to inform its safety use in clinical practice and further study.

4.Comparative study between off-pump and on-pump of coronary artery bypass grafting

Objective To compare the clinical effect of off-pump coronary artery bypass grafting (eft-pump CABG)and on-pump coronary artery bypass grafting(on-pump CABG).Methods The clinical dates of 152 cases with coronary artery bypass grafting(off-pump CABG group 85 cases,on-pump CABG group 67 cases)were received from June 2005 to December 2008,the preoperative dates,postoperative mortality and the rate of complication were compared between two groups.Results The preoperative dates of two groups were no significant difference(P>0.05);2 cases(2.35%)were died in off-pump CABG group and 13 cases(15.29%)had complications in off-pump CABG group;9 cases(13.43%)were died in on-pump CABG group and 25 cases(37.31%)had complications in on-pump CABG group.The mortality and the rate of complication in off-pump CABG group was significantly less than that in on-pump CABG group.it was significant difference(P<0.01).Conclusion Off-pump CABG is safe therapy of mieosurgieal operation.
5.Establishment and evaluation of a modified plasma protamine paracoagulation test.

OBJECTIVETo establish a modified plasma protamine paracoagulation test.
METHODSPlasma protamine paracoagulation, modified plasma protamine paracoagulation and D-dimer (D-D) tests were performed for the plasma samples collected from 98 cases of disseminated intravascular coagulation (DIC) and 156 normal subjects. The sensitivity and specificity of the 3 tests were analyzed. The plasma samples from 8 cases of suspected myocardial infarction were detected using modified plasma protamine paracoagulation for diagnostic purpose.
RESULTSThe sensitivity of plasma protamine paracoagulation, modified plasma protamine paracoagulation and D-D tests was 16.33%, 88.76% and 77.56%, and the specificity was 100%, 88.46% and 97.44%, respectively. Positive results occurred earlier in modified plasma protamine paracoagulation test than in plasma protamine paracoagulation and D-D tests in 5 cases of myocardial infarction.
CONCLUSIONThe modified plasma protamine paracoagulation test has a higher sensitivity than plasma protamine paracoagulation test and a higher specificity than D-D test, and can be helpful in early diagnosis of thrombosis and fibrinolysis.

7.Comparison of the mechanisms of intralesional steroid, interferon or verapamil injection in the treatment of proliferative scars.

OBJECTIVETo investigate the effects of intralesional steroid, interferon alpha-2b or verapamil injection on proliferation, apoptosis and TGF-beta1 expression in keloid and hypertrophic scar in vivo.
METHODS6 patients with keloids and 6 patients with hypertrophic scar were treated with intralesional injection of triamcinolone acetonide (40 mg/ml) or IFN alpha-2b (15 x 10(5) U/ml) or verapamil (2.5 mg/ml). Samples were collected on the 7th day after intralesional injection. Samples of untreated keloid and hypertrophic scar and normal skin were used as control. Expression of PCNA and TGF-beta1 was detected in situ by immunohistochemical staining, and apoptosis was detected in situ by terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate-biotin nick end labeling (TUNEL).
RESULTS1) Triamcinolone acetonide could prohibit proliferative scars through inhibiting cell proliferation and TGF-beta1 expression, as well as inducing apoptosis. 2) IFN alpha-2b could prohibit proliferative scars through inhibiting cell proliferation and TGF-beta1 expression, but not inducing apoptosis; 3) Verapamil could also prohibit proliferative scars through inhibiting proliferation and TGF-beta1 expression in fibroblasts, as well as inducing apoptosis. While the effect of inducing apoptosis was stronger than that of triamcinolone acetonide, the effect of inhibiting TGF-beta1 expression was weaker than those of triamcinolone acetonide and IFN alpha-2b.
CONCLUSIONSAlthough intraleional injection of steroid, interferon alpha-2b or verapamil were all effective in the treatment of keloid and hypertrophic scar, their mechanisms are not similar.

8.Early transplantation of double rectus abdominis musculo-cutaneous flaps from upper abdomen for the repair of patients with electrical injury in the wrist.

OBJECTIVETo observe the effect of the early transplantation of double rectus abdominis musculo-cutaneous flaps on the repair of electrical injury of the wrist.
METHODSThe study involved six patients suffering from circumferential deep electrical burn with only small amount of normal skin left on the dorsal side. The wounds were covered with double rectus abdominis musculo-cutaneous flaps raised from the upper abdomen with pedicles in both proximal and distal ends at an early postburn stage. The postoperative recovery of wrist function and wound repair were evaluated.
RESULTSThe wrist wounds in all the 6 patients were primarily healed, with perfect function and appearance.
CONCLUSIONEarly application of double rectus abdominis musculocutaneous flaps on the electrically injured wrists can promote the wound healing processes and plays important roles in the preservation of wrist function.

9.Establishment of BGC-823/pBaBb-puro-MACC1 gastric cancer cell line stably expressing MACC1 and its tumor-related gene expression profiles.

OBJECTIVETo establish a gastric cancer cell line with stable expression of metastasis-associated in colon cancer 1 (MACC1) and detect the changes in tumor-related gene expression profiles for investigating the possible regulation mechanisms between MACC1 and the differentially expressed genes.
METHODSThe full-length MACC1 cDNA was amplified from human embryonic kidney 293FT cells and cloned into the pBaBb-puro vector. The recombinant pBaBb-puro-MACC1 expression vector, after identification with restriction enzyme digestion, was transfected into 293FT cells, and the expression of fluorescent reporter gene was observed. pBaBb-puro-MACC1 vector was transfected into human gastric cancer BGC-823 cell line to establish BGC-823/pBaBb-puro-MACC1 cell line stably expressing MACC1. Quantitative RT-PCR and Western blotting were used to detect MACC1 expression in both BGC-823/pBaBb-puro-MACC1 and control BGC-823 cells. High-throughout cDNA microarray was used to screen the effects of MACC1 on the gene expression profiles of gastric cancer cells.
RESULTSThe recombinant pBaBb-puro-MACC1 plasmid was successfully constructed and verified by PCR and sequencing. BGC-823/pBaBb-puro-MACC1 cells showed significantly increased MACC1 mRNA expression as compared with the control cells. The results of cDNA microarray identified 33 up-regulated and 24 down-regulated genes in the cells after MACC1 transfection involved were in various cellular functions.
CONCLUSIONThe established BGC-823/pBaBb-puro-MACC1 gastric cancer cell line show some important molecular changes caused by MACC1.

10.Analysis of related factors of psychic symptoms in patients with chronic prostatitis.

OBJECTIVETo investigate the incidence and related factors of psychic symptoms in patients with chronic prostatitis.
METHODSPatients diagnosed with chronic prostatitis were selected as research objects, their course of disease, therapeutic process and psychic symptoms were inquired and recorded carefully, clinical symptoms were evaluated, expressed prostatic secretions (EPS) were examined, and many related factors were compared with psychic symptoms.
RESULTSAmong the 315 patients selected, 51.1% (161/315) had psychic symptoms. Psychic symptoms in patients with chronic prostatitis had nothing to do with course of disease, the incidence of psychic symptoms in patients with course less than 1 year, 1 to 2 years, and more than 2 years were 49.1%, 48.0%, and 56.1%, respectively; had closed relationship with therapeutic process, the incidence of psychic symptoms in patients to see doctors for the first times, the 2 to 3 times and more than 3 times were 35.3%, 43.5%, and 62.6%, with significant statistical difference (P < 0.05); had no relationship with severity of clinical symptoms, the incidence of psychic symptoms in patients with mild, media and severe clinical symptoms were 46.2%, 52.4%, and 55.9%; no statistical difference was noticed in patients with different degrees of inflammation, the incidence of psychic symptoms in patients with inflammatory and non-inflammatory prostatitis were 53.8% and 47.3%.
CONCLUSIONPsychic symptoms in patients with chronic prostatitis had direct relationship with times patient seeking medical care, and prolonged therapeutic process aggravated the psychic symptoms.
