2.Clinical analysis and pathological characteristics of lymph nodes of cat-scratch disease

Objective To explore the histopathological features, immunohistochemical characteristics and clinical manifestation of lymph nodes in patients with cat-scratch disease (CSD). Methods Nineteen cases of lymph nodes of CSD were analyzed by histopathological and immunohistochemical techniques. Results All patients with CSD had a clear history of close contact with cats. The local lymph nodes were punctured swelling. The histopathological characteristics were granulomatous small abscess, lymphocytes and immunoblastic cells growing. CEA (-), EMA (-), CD_ 45 (+), CD_ 20 (+), CD_ 45 RO (+) and CD_ 68 (++) were observed in immunohistochemistry. Conclusion CSD is a self-limited infectious disease caused by cat scratching and biting, its pathogen is bartonella henselae. Its histopathological characteristics, immunohistochemical expressions and clinical history have very important meaning for the diagnosis and differential diagnosis of CSD.
3.Brachytherapy by CT-guided Percutaneous Paracentesis Implantation of Iodine-125 Seed for Lung Cancer in Old Patients

Objective To observe the efficiency and safety of brachytherapy by CT-guided percutaneous paracentesis implantationof iodine-125 seed for lung cancer in elder patients.Methods 46 elder patients with lung cancer(65~81 years)were undergonebrachytherapy by CT-guided percutanepus paracentesis implantation of iodine-125 seeds with implanting needle(18-gauge). The effect of treatment according to observe the changes of tumors in size was evaluated after operation 1,2,3 and 6 months,respectively. Results There were totally 57 lesions in 46 cases,7 cases were undergone two times of implantation,and 1 case was undergone three times of implantation,totally the puncture accounted for 76. The effective rate (CR+PR) of treatment after one ,two, three, and six months was 8.77%(5/57), 40.35%(23/57),89.47%(51/57) and 96.49%(55/57),respectively. 5 patients(6.58%) accompanied by pneumathorax.Conclusion Brachytherapy by CT-guided percutaneous paracentesis implantation of iodine-125 seed is a safe, reliable and effective treatment for lung cancer, especially for elder patients.

5.Chemical constituents of Euphorbia dracunculoides.

Sixteen compounds including daphnoretin (1), isofraxidin (2), scopoletin (3), kaempferol (4), quercetin (5), guaijaverin (6), astragalin (7), quercetin-3-O-β-D-glucopyranoside (8), naringenin-7-O-β-D-glucopyranoside (9), 5-O-methylapi- genin-7-O-β-D-glucopyranoside (10), methyl gallate (11), prionitiside A (12), (2S)-2,3-dihydroxypropyl-1,6,8-trihydroxy-3- methyl-9,10- dioxoanthracene-2-carboxylate (13), 3,3'-di-O-methyl ellagic acid (14), 3'-O-methyl-3,4-O,O-metheneellagic acid-4'-O-β-D- glucopyranoside (15) and 3,4-methylenedioxy-3'-O-methylellagic acid (16), were isolated from the 70% acetone extract of Euphorbia dracunculoides Lam. Among them, compounds 1-3, 6-9, 11, and 14 were isolated from E. dracunculoides for the first time, and compounds 10, 12, 13, 15, and 16 were firstly obtained from the genus Euphorbia. Their structures were elucidated by spectroscopic analysis, including 1H-NMR, 13C-NMR, and ESI-MS.

6. Improvement effect of berberine on acute lung injury and inflammation induced by lipopolysaccharide and its mechanism

Objective: To establish the acute lung injury (ALI) mouse models induced by lipopolysaccharide (LPS), and to investigate the protective effect of berberine on ALI and its mechanism in vivo. Methods; A total of 40 C57BL/6 mice were randomly divided into control group, LPS group, berberine + LPS group and berberine group (n=10). The mice in control group were treated with saline; the mice in LPS group were treated with 5 mg · kg-1 LPS for 6 h; the mice in berberine +LPS group were pretreated with 50 mg · kg-1 berberine for 5 d before LPS treatment, then treated by 5 mg · kg-1 LPS for 6 h; the mice in berberine group were pretreated with 50 mg · kg-1 berberine for 5 d. The morphology of lung tissue of the mice in various groups were detected by HE staining. The ratios of wet to dry weights of lung tissue of the mice in various groups were detected. The concentrations of total protein and the number of effusion cells in bronchoalveolar lavage fluid (BALF) of the mice in various groups were detected; the levels of tumor necrosis factor (TNF-α) and interleukin-6 (IL-6) in BALF of the mice in various groups were detected by ELISA method; the reactive oxygen species (ROS) levels in lung tissue of the mice in various groups were detected by dihydreethidium (DHE) method. The mitochondrial membrane potential in lung tissue of the mice in various groups were detected by JC-1 method. The expression levels of Bcl-2 and Bax proteins in lung tissue of the mice in various groups were detected by Western blotting method, and the ratios of Bcl-2/Bax were caculated. Results; Compared with control group, the inflammatory cell infiltration of lung interstitium and the alveolar space of the mice in LPS group were significantly increased, and the alveolar wall was thickened, the ratio of wet to dry weight of lung tissue was significantly increased (P<0. 01), the level of total protein (P<0. 05) and the number of exudation cells in BALF were increased (P<0. 01), the levels of TNF-a and IL-6 in BALF were significantly increased (P<0. 01), the ROS level in lung tissue was increased, the mitochondrial membrane potential in lung tissue was decreased (P<0.01), and the ratio of Bax/Bcl-2 in lung tissue was increased (P<0. 01). Compared with LPS group, the inflammatory changes of lung tissue of the mice in berberine + LPS group were improved, the ratio of wet to dry weight of lung tissue was decreased (P<0. 05), the level of total protein and the number of exudation cells in BALF were increased (P<0. 05), the levels of TNF-a and IL-6 in BALF were decreased (P<0. 01), the ROS level in lung tissue was decreased, the mitochondrial membrane potential in lung tissue was increased (P<0. 05), and the ratio of Bax/Bcl-2 in lung tissue was decreased (P< 0. 05). Conclusion; Berberine could improve the degrees of LPS-induced ALI and inflammation of the mice, and the mechanism might be related to reduction of the ROS level and protection of mitochondrial function in lung tissue.
7. Antidepressant effects of Jiaotai Pills based on NO-cGMP signal transduction pathway in chronic mild unpredictable stress depression rats

Objective To investigate the effect of Jiaotai Pills on antidepressants in chronic mildly unpredictable stress (CUMS) depression model rats based on nitric oxide-cyclic guanosine monophosphate (NO-cGMP) signal transduction pathway. Methods The depression model of rats was induced by CUMS. On day 21 of the experiment, the rats in each group were treated with continuous ig administration for 14 d. The concentrations of NO and cGMP in hippocampus and plasma of rats were detected by Elisa method. The mRNA expression of NO synthase (including iNOS and nNOS) and NMDA receptor subunits NR1, NR2A, and NR2B in rat hippocampus was detected by RT-PCR. Results Compared with the control group, the levels of NO and cGMP in the hippocampus and plasma in the model group were significantly increased. The expressions of iNOS mRNA, nNOS mRNA, NR1 mRNA, NR2A mRNA, and NR2B mRNA in the hippocampus were significantly increased in the model group. Compared with the model group, Jiaotai Pills high, medium, and low dose and positive drug administration reversed the above changes. Conclusion Jiaotai Pills have the antidepressant effect on depression rats with CUMS by regulating NO-cGMP signal transduction pathway.
8. Based on cAMP-CREB-BDNF pathway to explore mechanism of antidepressant effect of Bupleuri Radix and Paeoniae Alba Radix drug pair

Objective To investigate the antidepressant effect of Bupleuri Radix and Paeoniae Alba Radix drug pair based on the cAMP-CREB-BDNF pathway. Methods The rat depression model was established by CUMS. The contents of cAMP, p-CREB, BDNF, and PDE4 in rat hippocampal and cAMP levels in rat plasma were determined by ELISA. The expression of BDNF mRNA in hippocampus, hypothalamus, and cortex were measured by RT-PCR. Results Compared with the model group, the positive drug group and Bupleuri Radix and Paeoniae Alba Radix drug pair can reverse the cAMP content in the hippocampus and plasma and the decreased contents of CREB and BDNF in the rat hippocampus. At the same time, the positive drug group, Bupleuri Radix, and Paeoniae Alba Radix can increase the expression of BDNF mRNA in hippocampus, cortex, and hypothalamus of rats. Conclusion The Bupleuri Radix and Paeoniae Alba Radix drug pair has obviously antidepressant effect on CUMS rat model, which can achieve antidepressant effect by regulating cAMP-CREB-BDNF pathway.
9.Correlation between BRAF mutation and clinicopathological characteristics in pediatric papillary thyroid carcinoma

Objective:To investigate the BRAF(V600E)gene mutation of pediatric papillary thyroid carcinoma (PTC) and refine their clinicopathological correlates. Methods:Tumor tissue samples of pediatric PTCs (≤18 years old) were collected from tumor tissue bank of Tianjin Medical University Cancer Institute and Hospital from January 2012 to December 2016.The medical records of 22 patients with pediatric PTC were reviewed retrospectively.The frequencies of BRAF(V600E) mutation were evaluated and the correlation between BRAF(V600E) mutation and clinical characteristics were analyzed. Results:BRAF(V600E) mutations were present in 45.5% of cases (10 cases). BRAF(V600E) mutation in pediatric PTC was obviously lower than that in adults PTC(77.7%) ( χ2=11.250, P=0.001). BRAF(V600E) mutation in>12-year-old group (66.7%) was remarkably higher than that in ≤12-year-old group (20.0%) ( P<0.05). BRAF(V600E) mutation in female (69.2%) was greatly higher than that in male (11.1%) ( P<0.05). There was no significant correlation with BRAF(V600E) mutation and multiple tumor, tumor size, highly invasive subtype, extrathyroidal extension, lymph node metastasis and radiological history of infants (all P>0.05). The median follow-up time was 45 months.No patients died and BRAF(V600E) mutation was not associated with the increase of recurrence rate ( P>0.05). Conclusions:BRAF(V600E)gene mutation in pediatric PTC is lower than that in adults. BRAF(V600E) mutation does not portend a more aggressive clinical biological behavior in pediatric PTC.
10.The role of thyroglobulin in diagnosis of lateral cervical lymph node recurrence in papillary thyroid cancer after radioiodione therapy

Objective:To evaluate the role of Tg in diagnosis of lateral cervical lymph node recurrence in papillary thyoid cancer(PTC)after radioactive iodine(RAI) therapy.Methods:From Jan 2012 to Aug 2018, 22 PTC patients who received RAI therapy after operation were reoperated for lateral cervical lymph node recurrence. The clinical data was retrospectively analyzed.Results:The median recurrence time was 30.5 (5-86) months. All 22 patients received RAI therapy after the first operation, and the median dose of RAI was 250mCi(100-700 mCi) and the episode of RAI therapy ranged from 1 to 4. All 22 PTC patients underwent neck reoperation, among which 20 cases were identified to have lymph node metastasis. The median number of lymph nodes dissected was 31 (8-83) and median number of metastatic lymph nodes was 4 (1-19) . The diagnostic accuracy of ultrasonography in detecting lymph node metastasis was 90.9%. Before reoperation, the median Tg was 1.305 (0.10-99.51) μg/L, with the cutoff value of Tg being 0.2 μg/L, and its sensitivity and specificity were 80.0% and 100%, respectively. The median stimulated Tg was 5.89 (0.14-255.80) μg/L in the 10 patients, with the cutoff value of stimulated Tg of 2 μg/L, and its sensitivity and specificity were 88.9% and 100%, respectively.Conclusions:The serum Tg level is helpful for monitoring the recurence of PTC, but recurrence cannot be completely ruled out for those with low Tg.
