5.Construction and identification of Stathmin gene Pichia pastoris expression system

Objective To provide the experimental basis for the further research of the interacting proteins with Stathmin ,the Stathmin gene Pichia pastoris expression system was constructed ,the expressed Stathmin product was purified and identified .Meth‐ods Stathmin gene was amplified from tumor cell line of SKBR3 by PCR method and cloned into the yeast expression vector pPIC3 .5K .The recombinant vector pPIC3 .5K‐Stathmin was constructed and transformed into Pichia pastoris GS115 .The positive clones were screened by YPD medium containing Geneticin 600 μg/mL .Expression was induced with 0 .5% methanol and expres‐sion products were identified by SDS‐PAGE and Western Blotting .Results DNA sequencing result showed that the gene fragment was consistent with Stathmin gene sequence .pPIC3 .5K‐Stathmin was selected from YPD culture medium containing Geneticin ,and the positive clones were identified by PCR .SDS‐PAGE showed that a 37 × 103 protein band could be seen on the PAGE gel after Coomassie Blue staining ,which was further confirmed and identified as Stathmin protein by Western Blotting .Conclusion Stathmin yeast expression vector is successfully constructed and expressed in Pichia pastoris ,which laid the foundation for the study of inter‐acting proteins with Stathmin ,and for the preparation of the biological treatment drugs of Stahtmin target .
6.The retrospective analysis in forensic of 135 killing familiar cases

Objective Summarize the information of 135 killing familiar cases and study the characteristics of these cases, in order to analysis the key points of the case investigation. Methods Collected the data of the 135 killing familiar homicides cases which have solved and happened in a province middle of China from 2011 to 2013. The paper analyzed the data accordding the basic information, the forensic professional, the forensic scene and so on. Results Most killing familiar cases occured between husband and wife or parents and children. The cases frequently occur in rural areas. Most of the cases occur between the adults. The killing familiar cases commonly occur in Decemember, January, Feburary, the cases commonly occur in evening(from 6 pm to 6 am). The motivate of the cases is unobvious, the murder suffer from the mental patient is commonly. In these cases, mostly tools of crime is the blunt tools and the sharp tools. Mechanical asphyxia and mechanical iniury were the most commonly causes of death in these cases. Conclusion The social characteristics of suspect, injury tool, cause of death, time of the incident in the killing familiar case showed similar general characteristics, but how collect evidence in the case is the difficulty of the case investigation.
7.Correlation of Breast Cancer Cells Invasive and Stathmin Gene Expression

Objective To explore the relationship between the Stathmin gene expression in breast cancer cells MDA-MB-231, MCF-7 and the biological behaviours such as cell growth,adhesion and invasion,and provide experimental basis of breast cancer metastasis for further study.Methods Used RT-PCR and Western Blot methods to detect the Stathmin gene expres-sion levels in MDA-MB-231 and MCF-7 cells,and in the mean while to test the MDA-MB-231 and MCF-7 cell growth,adhe-sion,invasion ability by CCK-8 cell proliferation experiments,cell adhesion experiments,cell invasion experiments,then, analyed the relationship of Stathmin gene expression and cell growth,adhesion,invasion ability.Results Over-expression levels of Stathmin gene were observed both in the MDA-MB-231 and MCF-7 cells (F=10.173,P<0.05),and furthermore, the expression levels of Stathmin gene in MDA-MB-231 cells was higher than in MCF-7 cells (t=4.562,P<0.05).While, the growth,adhesion and invasion ability of the MDA-MB-231 cells was higher than that of MCF-7 cells(P<0.05).Conclu-sion The higher level of Stathmin gene expression,the stronger breast cancer cells had ability of growth,invasion,and ad-hesive.The Stathmin gene expression levels was closely correlated with breast cancer cell invasive.
8.CT evaluation and classification of the Le Fort fracture

Objective To evaluate the CT findings and classification of the Le Fort fracture. Methods Sixty-two cases with Le Fort fractures were studied with thin-slice high-resolution CT scanning and analyzed with three-dimensional(3D)imaging reconstruction.Results Of the 62 patients,10 had Le Fort type Ⅰ fracture,9 had Le Fort type Ⅱ fracture,8 had Le Fort type Ⅲ fracture,and 35 had various combinations of the three types of Le Fort fractures,including 18 Le Fort Ⅰ+Ⅱ fracture,7 Le Fort Ⅰ+ Ⅱ+Ⅲ fracture and 10 Le Fort Ⅱ+Ⅲ fracture.Fifty-five cases had associated multiple fractures in the maxillofacial region.On 2D CT images,Le Fort fracture manifested as multiple and complex fractures. Though 2D image was better than 3D image in accurately defining tiny fractures and fractures of deep structures,the diagnosis of Le Fort fracture could not be correctly made solely on 2D image.3D CT clearly and stereoscopically demonstrated the entire shape and orientation of Le Fort fracture,thus facilitating the correct classification of Le Fort fracture.Conclusion 3D CT image is important in providing information about the space relationship of Le Fort fracture,thus very valuable for the preoperative planning.
9.Correlation of the expression of HIF-1α and CTSD with vasculogenic mimicry and its clinicopathological features in esophageal squamous cell carcinoma

Purpose To investigate the expression of vasculogenic mimicry (VM) and hypoxia inducible factor-1 α (HIF-1α),cathepsin D (CTSD) in esophageal squamous cell carcinoma (ESCC) and its relationship with clinicopathological parameters.Methods 120 specimens of primary ESCC and their clinical data were collected.Dual staining of CD34 and PAS were performed to validate the existence of VM in ESCC.Immunohistochemical of EnVision two-step method was used to detect the expression of VM-related markers vascular endothelial cadherin (VE-cadherin) and vascular endothelial grow factor (VEGF),HIF-1α and CTSD in 120 cases of ESCC.The relationship between HIF-1α,CTSD and clinical pathological parameters were analyzed.Results The positive rate of HIF-1α,CTSD was correlated with infilatrating,lymph node metastasis,clinical stage and cancer embolus.VM existed in 25 of 120 cases.The expression of HIF-1α and CTSD in cases with VM was obviously higher than that without VM,the expression of HIF-1 α and CTSD was positively correlated with VM in ESCC.The expression of CTSD in case with HIF-1 α was obviously lower than that without HIF-1α,the expression of CTSD was positively correlated with HIF-1α in ESCC.The expression of VE-cadherin and VEGF in cases with CTSD was obviously higher than that without CTSD,the level of VE-cadherin and VEGF expression was positively correlated with CTSD in ESCC.Conclusion CTSD may promote VM formation in ESCC via HIF-1α.High expression of HIF-1α and CTSD indicate poor prognosis of ESCC patients.
