Objective: To develop a new quantitative determination method for the biological activity of recombinant human ciliary neurotrophic factor. Methods: Dorsal root ganglions were derived from the chick embryo and dispersed into single neuron cell,The rhCNTF was added to neuron cells and incubated for 64 hours,The activity of acid phosphatase in neuron cells was determined and the biological activity of rhCNTF was analyzed quantificationally. Result: rhCNTF could promote original era dorsal root neuron cells of chick embryo surviving,the livability of neuron cells was positively related to the amount of rhCNTF added to the culture. Conclusion: A quantitative determination method for the biological activity of rhCNTF was developed by testing the activity of acid phosphatase in neuron cells. Compared with the typical ways,this method was quantificational easily,repeatable better and with much fewer disturbance factors.

Objective To investigate the role of IL-28B gene on the antiviral curative effect in the patients with chronic hepatitis B.Methods 205 HBeAg-positive patients treated with the antiviral therapy of pegylation interferon(PEG-IFN)were detected the IL-28B rs12980275 and rs12979860 genotypes by TapManSNP genotyping assay and the relationship between the IL-28B genotypes and HbeAg seroconversion at end of treatment was analyzed.Results Among 205 cases,6 cases of genotype were uncertainty,the frequencies of IL-28B genotypes in other 199 cases were 77%,19%,5% for AA,AG,GG genotypes at rs12980275 and 76%,20%, 4%,for CC/CT/TT genotypes at rs12979860 respectively.The IL-28B polymorphisms were significantly correlated with the HbeAg seroconversion at the end of PEG-IFN treatment (P =0.001).In rs12979860 genotype,by the HBV genotyp,age,HBV lev-el,ALT and the combined therapy,the odds ratio (OR)of AA versus AG/GG was adjusted to 3.16 by HbeAg seroconversion.The similar result also obtained for the rs12980275 genotype.The IL-28B genotype was also associated with the HBsAg clearance.Con-clusion The IL-28B gene polymorphism is associated with the antiviral curative effect in the patients with HBeAg positive chronic hepatitis B.

Objective To study the influence of cell loss in the CA3 area on hippocampal neurogenesis in mice with temporal lobe epilepsy .Methods Newly born cells in the subgranular zone (SGZ ) of dentate gyrus were labeled by the proliferation marker bromodeoxyuridine (BrdU ) at 2 months after pilocarpine‐induced status epilepticus .Double labeling of BrdU+ NeuN was carried out to compare neuronal differentiation between type Ⅰand Ⅱ mice ,which represented partial cell loss and almost complete cell loss in the CA 3 area ,respectively .DCX and BrdU single staining were made to analyze the proliferation of progenitor cells in SGZ and the survival of the newly born cells .Results When compared with that of the type Ⅱ mice ,the number of double labeled cells of BrdU+NeuN in the subgranular zone‐granule cell layer (SGZ‐GCL) in the type Ⅰ mice increased significantly ( P<0 .05) .However ,the percentages of double labeled cells in all the BrdU + cells were found to be similar between the two groups (P>0 .05) .No significant difference in the number of DCX + cells in SGZ was found between the typeⅠ and Ⅱ mice (P>0 .05) .At 6 weeks after BrdU labeling ,more remaining BrdU+ cells were found in the SGZ‐GCL in the type Ⅰ mice when compared with their counterparts in the type Ⅱ mice (P<0 .05) .Conclusion The cell loss in the CA3 area of pilocarpine‐induced epileptic mice contributes to the declined hippocampal neurogenesis by exerting negative effects on the survival of newly born cells .

Objective To study the diagnostic value of serum α-L-fueosidase (AFU), transforming growth factor alpha(TGF-α)and ferritin (Fer) in primary hepatic carcinoma (PHC). Methods AFU and Fer in the serum were studied in 36 patients with benign hepatism and 56 patients with low concentration AFP of PHC hepatic carcinoma by automatic biochemistry analyzer Roche Modular. ELISA was used to assay the degree of TGF-α The sensitivity and specificity of AFU, TGF-α and Fer for low expression AFP of PHC were evaluated by ROC curve. Results The serum AFU, Fer and TGF-α levels were all significant difference in the patients than those in controls (P <0.05). The area under ROC curve of these indexes in descending order was AFU (0.707), TGF-α (0.677) and Fer (0.592). The diagnostic sensitivity of Fer was lower than AFU and TGF-α in diagnosis of AFP lowly expression PHC. The ratio showed AFU better than TGF-α The diagnostic specificity of AFU (64 %) was higher than that of TGF-α (61%) when TGF-α and AFU was at the same diagnostic sensitivity (64 %). Conclusion AFU is more valuable than Fer and TGF-α for negative or AFP lowly expression PHC, meanwhile it has much more accuracy.

Objective To evaluate the security and efficacy of combination of epristeride and terazosin for treatment of benign prostatic hyperplasia (BPH). Method One hundred and eighty-four patients with BPH were treated by epristeride for 6 months and terazosin for 1 month, the efficacy and complication were observed. Results After 6 months treated, compared with before treated, the common symptoms improved, the residual urine decreased 20.74ml, maximum flow rate increased 3.76 ml/s, prostate volume grown downwards 6.70cm3 and the quality of life raised apparently(Pall＜0.05 ). Condusion Combinationof epristeride and terazosin for treatment of BPH is safe and effective.

Objective To investigate the levels of pro-inflammatory cytokine interferon (IFN)-γand anti-inflammatory cytokine interlenkin (IL)-10 in expressed prostatic secretions (EPS) of chronic prostatitis/chronic pelvic pain syndrome (CP/CPPS) and their clinical significance. Methods The levels of IFN-γ and IL-10 in EPS of 20 patients with CP (type Ⅲ A, group A ), 20 patients with CPPS (type Ⅲ B,group B) and 10 healthy men (control group) were measured using enzyme-linked immunosorbent assay (ELISA). The results were analyzed comparatively with NIH-chronic prostatitis symptom index (NIH-CPSI).Results The levels of IFN-γ in EPS of group A and group B were ( 14.92 ± 7.85 ), ( 13.74 ± 5.96) ng/L respectively, which were higher than that in control group [(7.47 ± 1.40) ng/L] (P ＜ 0.05 ). And the levels of IL-10 in EPS of group A and group B were (216.40 ± 33.65 ), (223.70 ± 45.21 ) ng/L respectively, which were higher than that in control group [( 162.10 ± 43.81 ) ng/L] ( P ＜ 0.05 ). There was no statistically significant difference between group A and group B. No correlation between NIH-CPSI and IFN- γ,IL-10 levels in EPS(P ＞ 0.05 ). Conclusion IFN-γ and IL-10 have a very important role in etiology of CP/CPPS and they can be the objective parameters in the diagnosis of CP/CPPS.

OBJECTIVE：To set up a method to determine the content of hyoscymine alkaloides in belladonne tablet by spectrophotometry.METHOD:Hyoscymine alkaloides reacts with bromomethyl phenol to produce a colorful ionic couple at a certain pH solution.Product is extracted using CHCl3 solvent and determined by spectrophotmetry.The detection wavelength is 415nm.Atropine sulfuric acid is used as reference standard.RESULTS:The calibration curve is linear within the range of 0～17.5μg/ml for hyoscymine alkaloides.The average recovery is 101.1% and the related standard deviation is 1.0%.CONCLUSION:The method is simple,accurate and practical for the determination of hyoscymine alkaloides in belladonne tablet.

OBJECTIVETo observe blood uric acid levels and Goldstein grading, as well as their correlation in Wilson's disease (WD) patients with different Chinese medical syndrome types.

METHODSTotally 906 WD patients in line with inclusive criteria were assigned to 6 groups, i.e., the heart spirit confused by phlegm group (HSCP, 26 cases), the phlegm-fire disturbing heart group (PFDH, 90 cases), the retention of damp-heat group (RDH, 113 cases), deficiency of qi and blood group (DQB, 168 cases), the deficiency of Gan-yin and Shen-yin group (DGYSY, 327 cases), the deficiency of Gan and Shen group (DGS, 182 cases) due to different Chinese medical syndrome types. Recruited were another 160 healthy subjects having similar ages and diet structures, who came for medical examinations, as the healthy control group. Venous blood was collected from the medial cubital vein of each-patient on an empty stomach in early mornings to detect blood uric acid levels. Results Blood uric acid levels were lower in each syndrome type group than in the healthy control group (146.08 +/- 67.24 micromol/L in the HSCP group; 157.08 +/- 69.77 micromol/L in the PFDH group; 162.58 +/- 97.72 micromol/L in the RDH group; 156.20 +/- 62.63 micromol/L in the DQB group; 161.83 +/- 111.23 micromol/L in the DGYSY group; 194.41 +/- 90.01 micromol/L in the DGS group; 242.39 +/- 87.55 micromol/L in the healthy control group, P < 0.01). Blood uric acid levels were higher in the DGYSY group than in the other 5 syndrome groups (P < 0.01). Correlation analyses between Goldstein grading and blood uric acid showed that, along with increased Goldstein grade (that was aggravating disease conditions), WD patients' blood uric acid levels decreased (P < 0.01).

CONCLUSIONSWD patient's blood uric acid levels decreased more. Blood uric acid levels and Goldstein grading were different in various Chinese medical syndrome types. Blood uric acid levels had certain value in assessing the severity of WD.

Objective To explore the effects of action observation therapy on upper-extremity motor function after ischemic stroke and on the motor cortex using functional magnetic resonance imaging (fMRI).Methods Forty patients with ischemic stroke were randomly assigned to an observational group (n =20) or a control group (n =20).Both groups received conventional rehabilitation,while the observational group was additionally provided with action observation therapy for 8 weeks.Both groups were assessed using the Fugl-Meyer assessment (FMA) and the Barthel index (BI) before and after the 8 weeks of treatment and functional magnetic resonance imaging was performed before treatment.Two months after the treatment,nine patients of the experimental group and 8 of the control group who continued to receive their respective treatments after discharge were again assessed using functional magnetic resonance imaging.Results After the treatment the average FMA score and BI score of both the observational group and the control group had increased significantly.The increase in the average FMA score of the observational group was significantly greater than that of the control group.However,there was no significant difference between the two groups in the increases in BI score after 8 weeks of treatment.The fMRI results showed that there was a significantly greater rise in activity in the bilateral precentral gyrus,parietal lobe and the supplementary motor area of the patients in the observational group after the treatment compared with the control group.Conclusion Action observation therapy can improve upper extremity motor function and performance in the activities of daily living after ischemic stroke and induce changes in the excitability of the cerebral motor cortex.

Objective　To determine whether cold-stress stimulation could lead to pregnancy-induced hypertension syndrome (PIH) in rats. Methods　Female adult Wistar female rats were randomly divided into 4 groups: non-pregnant control group (NN), non-pregnant cold-stress group (NC), pregnant control group (PN) and pregnant cold-stress group (PC). The rats were kept in (4±2) ℃ for 4 h (cold-stress groups) every day or remained in 25 ℃ (control groups) from the 1 st day to 19 th day of pregnancy. The blood pressure, urine protein, body weight, haematocrit (HCT), weight of placenta, length and weight of fetus were all measured. The histological change of the placenta and kidneys were also observed. Results　After cold-stress stimulation for 2 weeks, the blood pressure, urine protein in NC and PC group increased significantly compared with that in control group, while the weight of placenta and fetus, the length of fetus in PC decreased significantly than that in PN. Obvious ischemic and anoxic histological changes in kidneys and all layers of placenta were found in PC group. Conclusion　Repeated cold-stress stimulation can induce PIH in rats, thus provide a new non-invasive method to establish an animal model of PIH.
