1.Inhibition effect of PPARγ agonist on proliferation of human pterygium fibroblasts

Background Recurrence of pterygium is a common complication after the surgical excision of pterygium,and this procedure is related to cell proliferation,inflammation and neovascularization.Researches determined that rosiglitazone can suppress inflammation and neovaseularization and inhibit proliferation,hut few studies concerning the effect of rosiglitazone on pterygium were performed. Objective The aim of this study was to investigate the effect of peroxisome proliferator-activated receptor γ agonist on the proliferation and apoptosis of human pterygium fibroblasts(HPFs)in culture and search for a new drug to prevent and cure the recurrence after pterygium surgery. Methods Human pterygium samples were obtained during surgery and HPFs were cultured and purified using an explant method and 0.25%trypsin digestion,respectively.The identity of cultured HPFs was confirmed by immunohistochemistry using anti-vimentin and keratin antibodies.Rosiglitazone with the concentrations of 0(control),5,10,25,50,75,100,150,200,400μmol/L was then added in the culture medium for 12,24 or 72 hours.1%DMSO was used as blank control.The MTT method was used to assay the biologic effects of rosiglitazone on HPFs.Cell cycle distribution and apoptosis of HPFs after rosiglitazone treatment were studied by flow cytometic analysis.The expression of proliferating cell nuclear antigen(PCNA)mRNA in HPFs was detected by real-time PCR. Result Cultured HPFs radially migrated outward from the pterygium block,and then grew in long fusiform shape,showing positive response for vimentin and negative for keratin.The HPFs became round and thin with loose distribution after the addition of rosiglitazone.Following 25-125 μmol/L rosiglitazone administration for 12,48 or 72 hours,the A490 value of HPFs significantly declined with the increase of dosage(F=158.312,P=0.006)and lapse of time(F=1.924,P=0.135).Following the treatment of 25,75 or 125 μmol/L rosiglitazone for 24 hours,the number of HPFs in G0/G1 phase was markedly elevated;while the cell numbers in S phase decreased significantly in comparison with the control group(P<0.05).The apoptotic rate of HPFs in the 25,75 and 125 μmol/L rosiglitazone groups significantly increased with the increase of rosiglitazone concentration(P<0.05).Real-time PCR revealed that after 24 hours of rosiglitazone treatment,the expression of PCNA mRNA in HPFs was suppressed in a dose-dependent manner(F=3244.329,P<0.05). Conclusion Rosiglitazone inhibits HPFs proliferation,arrests their cell cycle progression in G0/G1 phase,induces apoptosis of HPFs and depresses the synthesis of PCNA in a dose-and time-dependent manner.
2.Retroperitoneoscopic pyelolithotomy and ureterolithotomy (report of 81 cases)

Objective To assess the clinical value of retroperitoneoscopic pyelolithotomy (RPPL) and retroperitoneoscopic ureterolithotomy (RPUL) in treating renal pelvis or ureteral calculus. Methods A total of 81 patients with renal pelvis or ureteral calculi underwent RPPL and RPUL on 88 sides. Of them 30 were women and 51 were men. Their age ranged from 12 to 65 years (mean,36 years).The calculi were found on left side in 41 cases,on right side in 33,and on both sides in 7.The calculi were 0.8 to 3.0 cm in diameters.Pyelolithiasis occurred in 11 cases,upper ureteral calculi in 68 cases and middle in 2 cases.Of them 3 had radioparent calculi in ureter.In these patients,21 had experienced unsuccessful ESWL,or ureterorenolithotripsy (URL),or both;2 had undergone conversion to retroperitoneoscopic surgery when the procedure failed in minimally invasive percutaneous nephrolithotomy (MPCNL) or perforation occurred during URL. Results All the stones were completely removed by single procedure.The operation time was between 30 and 210 min with a mean of 65 min.The intra-operative mean bleeding volume was 25 ml (range,10 to 60 ml).30 cases with other urinary tract diseases were cured by retroperitoneoscopic surgery at the same time.The hospital stay was from 4 to 10 days.During the follow-up (1 to 16 months) for the 60 cases,no recurrent calculus was found. Conclusions In selected patients with large,hard,radioparent,chronically impacted pelvis or upper ureteral stones,especially in solitary kidney,RPPL and RPUL may be considered as the first-line treatment and as a salvage procedure for failed ESWL and endoscopy.With these procedures the urinary tract complications can be treated concomitantly.
3.Establishment of a miniature porcine model for controlled cardiac deceased donor

Objective To establish a type of porcine model for controlled Cardiac Deceased Donor.Method Using the wuzhishan miniature pig 2 ~ 4 months of age.After intravenous general anesthesia and respiratory,after open heart surgery to produce myocardial infarction model,to heartbeat stop completely,stop breathing machine and drug support,so we established a miniature pig cardiac death donor model.Record during the heart rate,systolic pressure,diastolic blood pressure,central venous blood pressure,blood oxygen saturation and regularly take on blood gas analysis.Before cardiac arrest,monitoring hemodynamic,blood gas analysis,and the time of death before the circulatory failure.After cardiac arrest respectively 0 min,15 min and 30 min,perfusion for donor organs (liver/kidney),get the pig's liver/kidney in the different time of the groups,observed the pathological changes of liver/kidney tissues by HE staining.Result The heartbeat stop completely occurs 7 ± 0.17 minutes after left descending coronary artery ligation and cease of assisted respiration in the different groups,systolic pressure,diastolic blood pressure,central venous pressure,blood oxygen saturation,CO2 partial pressure changed significantly;Immediately after cardiac arrest for compared group (0 min),schemia-reperfusion that group of 15 min after cardiac arrest injury is obvious,ischemia-reperfusion that group of 30 min after cardiac arrest injury is further.Conclusion Miniature pig donor model obtained in this method respiratory cycle failure stability,can be controlled,no adverse drug reactions,the organ ischemia-reperfusion injury caused by repetitive is better.
4.Survey on individual occupational health protection behaviors of welding workers using theory of reasoned action.

OBJECTIVETo apply theory of reasoned action at survey on welding workers occupational health protection behaviors and explore related influencing factors.
METHODSnine companies were randomly selected from areas with many welding works in Zhejiang Province. All welding workers were surveyed using a questionnaire based on theory of reasoned action.
RESULTS10.06%, 26.80% and 37.50% of the respondents never or seldom used eyeshade, mask and earplug, respectively. After controlling the socio-demographic factors, welding workers' behavioral belief was correlated with the behaviors of eyeshade-mask and earplug use (χ(2) = 31.88, 18.77 and 37.77, P < 0.01). the subjective norm of company was correlated with all protection behaviors (χ(2) = 20.60, 10.98 and 19.86, P < 0.01), the subjective norm of colleague was correlated with mask and earplug use, (χ(2) = 27.43, 36.39, P < 0.01), and the subjective norm of family was correlated with mask use (χ(2) = 5.73, P < 0.05).
CONCLUSIONTheory of reasoned action is suitable for welding worker occupational health related behaviors. It is useful to improve occupational health education, to effectively select health education objective and to tailor health education contents.

Objective To investigate the metabolic alterations in the brain of neonates with HIE and correlate those alterations with clinical grading and prognosis of HIE.Methods Fourty-six eases of full-term neonates diagnosed as HIE clinically were performed MRI and 1~H-MRS,9 healthy neonates without the evidence of asphyxia were studied as controls,1~H-MRS techniques included single voxel proton MRS and two dimensional muhi-voxel chemical shift spectroscopy imaging,point resolved spectroscopy sequence was used for 1~H-MRS.Metabolic changes in the spectroscopy were analyzed in neonates with HIE,and study the relationgship between MRS findings and prognosis.Results(1)The typical 1~H-MRS manifestations of full- term neonates suffering from HIE were as follows:the peaks of Lac were elevated,GLx-? were elevated and NAA were decreased.(2)GLx-?/Cr ratio in control,mild,moderate and severe HIE group was 0.16, 0.21,0.64,and 1.31,respectively.Lac/Cr ratio in control,mild,moderate and severe HIE group was 0.12,0.14,0.19,and 0.26,respectively.There was a significant difference in the ratio of GLx-? and Lac/Cr between HIE group and control group(t=5.01,P
6.Correlation study between MR spectroscopic findings and cell proliferation in astrocytoma

Objective To investigate the relationship between findings of proton magnetic resonance spectroscopy(~1H-MRS)and tumor proliferative potential in patents with astrocytoma.Methods Thirty four patients of astrocytomas proved by surgery and pathology were examined by MRI and ~1H-MRS,among them 26 were of grade Ⅰ or Ⅱ(low grade)and 8 of grade Ⅲ or Ⅳ(high grade).Twenty one eases of astrocytomas(17 low-grade and 4 high grade respectively)were homogeneous type.Expression of proliferating cell nuclear antigen(PCNA)was detected by immunohistochemical method of streptavidin- biotin-peroxidase(SP).Results The Cho/NAA and Cho/Cr ratios of high-grade astrocytomas(M=4.895, 4.845 respectively)were significantly higher than those of low-grade astrocytomas(M=2.920, 2.000 respectively)in the 21 homogeneous astrocytomas(Z=-2.597,P=0.009;Z=-2.687,P= 0.007 respectively),and the ratios were found to be significantly correlated to the expression of PCNA(A= 7.880)(r=0.607,P=0.003;r=0.457,P=0.038 respectively).However,the differences of Cho/NAA、 Cho/Cr ratios between the high-grade and low-grade astrocytomas were not significant when 34 cases were included,and no relationship was found between Cho/NAA(M=3.965,2.890 respectively)、Cho/Cr(M= 3.080、1.960 respectively)ratios and the expression of PCNA(A=8.100)(r=0.060,P=0.738;r= 0.125,P=0.480 respectively).Conclusion By measuring the Cho/NAA and Cho/Cr ratios,~1 H-MRS may be a valuable method for presurgical in invo prediction of the malignant degree,tumor proliferative potential and prognosis of homogeneous-type astrocytomas.
7.Expression of wnt5a gene in hematologic diseases and leukemic cell lines.

This study was aimed to investigate the expression level of Wnt5a gene in some hematologic diseases and leukemic cell lines so as to provide a basis for further research of Wnt5a role and its mechanism in hematologic malignancies. The mononuclear cells of peripheral blood and bone marrow were isolated by human lymphocytic isolation solution. The expression of Wnt5a gene in specimen of 31 cases and three leukemic cell lines (Jurkat, K562, HL-60) were detected by RT-PCR. The results showed that in four out of five AML cases, negative or weak positive expressions were observed and negative expressions were observed also in K562 and HL-60 cells. Only in one AML case with complete remission and Jurkat cells the strong positive expressions were observed. The negative expression was observed in all six CML cases. In three out of four ALL cases, the expression was positive or weak positive and one negative. The expressions in two CLL cases were negative. Out of two MM cases, the expression in one was weak positive and in other was negative. Out of three lymphoma cases, the expression in one case was weak positive and in other two cases were negative. There were positive or weak positive expressions in two cases of AA, two cases of IDA, three cases of ITP, one cases of PV and ET cases. It is concluded that there have obvious down-regulated or lost expression of Wnt5a gene in 31 cases of hematologic disease and myelocytic leukemic cell lines except ALL samples. Nevertheless there have general positive expression of Wnt5a in cases of non-malignant hematologic diseases. These results suggest that the genesis of myelocytic leukemia is related to the down-regulated expression of Wnt5a.

8.High-resolution melting: a analysis for genotyping of MDR1 C3435T in benzene-exposed workers.

OBJECTIVEUsing high resolution melting (HRM) to analysis MDR1 C3435T in people exposed to benzene.
METHODSRestriction fragment length polymorphism (RFLP) was utilized to detect the polymorphism of MDR1 3435 in 121 benzene-exposed workers, and the results were compared with the HRM in 10% samples and were confirmed with direct sequencing for six people in them.
RESULTSBy direct sequencing, consistent results of benzene-exposed workers with RFLP or HRM were got. The new high resolution melting curve analysis is more efficient, more convenient, and cheaper than RFLP.
CONCLUSIONHigh-resolution melting analysis provides a valid approach to efficiently detect DNA genetic diagnosis, which is suitable for detect susceptible genes in occupational surveillance.

9.Influence of MDR1 gene C3435T on peripheral white blood cell counts in workers exposed to benzene.

OBJECTIVETo explore the effects of MDR1 C3435T on the peripheral white blood cell counts in workers exposed to benzene.
METHODSOne hundred and twenty-one benzene-exposed workers and 110 healthy controls without benzene exposure were enrolled in this study. White blood cell counts influenced by the polymorphism of MDR1 gene were analyzed.
RESULTSThe frequency of MDR1 3435 C/C, C/T, T/T in healthy controls was 37.27%, 46.36%, 16.37%, respectively, and it was 38.84%, 41.33%, 19.83% in the benzene-exposed workers, respectively. The frequency of the MDR1 gene was also not significantly different between benzene exposed workers and controls. Subjects exposed to benzene with MDR1 3435 mutation genotype (T/T) had the significantly lower WBC [(5.46 ± 1.51) × 10(9)/L] than those carrying wild type (C/C) and heterozygous (C/T), whose WBC were (6.08 ± 1.28) × 10(9)/L (P = 0.044).
CONCLUSIONP-glycoprotein encoded by MDR1 gene may be implicated into the hematotoxicity of benzene. Subjects carrying MDR1 3435 T/T genotype may have a higher risk of benzene poisoning.

10.The Establishment of Evaluation Index System for Emergency Response Capability among Health Supervision Institution

Objective:To construct an evaluation indicator system for emergency response capability in health supervision institutions.Methods:It made an initial indicator system by using brain storming,literature method and expert consulting,and constructed an evaluation indicator system by Delphi method.Results:Both the response rates of two round questionnaires survey were 100%.The means of authoritative coefficient for the first-dimension indicators was 0.82.For the second round of expert consulting,the coordination coefficients of the first,second and third-dimension indicators were 0.954,0.820 and 0.565 respectively (P<0.05).It constructed an evaluation indicator system for emergency response capability among health supervision institutions.There were 4 first-dimension indicators,10 second-dimension indicators and 37 third-dimension indicators in the indicators system.Conclusion:Experts opinions had high reliability.The process of screening indicators was normative.The indicator system was comprehensive,which could be used and examined in practice.
