1.IN SITU BY BRIDIZATION DETECTING L- FORM DNA OF STAPHYLOCOCCUS AUREUS OF LARYNGOCARCINOMA

Aim To study whetger there is positive express in situ hybridization of L-form DNA or Staphylococcus aureus in laryngocarcinoma cell. Methods 20 cases of expressnon of L-form DNA of Staphylococcus aureus in laryngocarcinoma karyon were detected with nucleic acid in situ hybridization. Results 60% cancer karyon, cytoplasm display positive signal of Staphylococcus aureus L-form DNA. Positive rate of Staphylococcus aureus L-form antigen is 75% with immunohischemical (s-p) stainipg. The rate is 70% with gram staining. Conclusion Staphylococcus aureus L-form DNA had get into laryngocarcinoma ceells, is great possibly to integrate into host ce11, and influence hyperplasis and canceration of cell from gene level. Staphylococcus aureus and its L-form infection have possibly close relation to happen and development of laryngocarcinoma.
2.Acupuncture at "Siguan" combined with Gallbladder Meridian acupoints for migraine: a randomized controlled trial.

OBJECTIVETo observe and evaluate the clinical effect of acupuncture at "Siguan" combined with the Gallbladder Meridian acupoints on migraine.
METHODSWith randomized controlled trial design, 87 patients were randomly divided into an observation group (42 cases) and a control group (45 cases). The observation group was treated with acupuncture at "Siguan" including Hegu (LI 4) and Taichong (LR 3), combined with the Gallbladder Meridian acupoints including Fengchi (GB 20), Shuaigu (GB 8), Qiuxu (GB 40) and Zulinqi (GB 41), while the control group was treated with acupuncture at the Gallbladder Meridian acupoints, including Fengchi (GB 20), Shuaigu (GB 8), Qiuxu (GB 40), Zulinqi (GB 41), Yanglingquan (GB 34) and Hanyan (GB 4). The treatment was given every other day in two groups, 3 times per week for totally 6 weeks. The migraine intensity, migraine frequency and lasting time of each migraine were observed before and after treatment, and 3-month follow-up was performed after treatment. The clinical efficacy in two groups was evaluated.
RESULTSCompared with those before treatment, the migraine intensity, frequency and lasting time were all reduced significantly after treatment in the two groups (all P<0.01), which were more obvious in the observation group (all P<0.05). At the end of the treatment, the total effective rate was 95.2% (40/42) in the observation group and 80.0% (36/45) in the control group, which had statistical difference (P<0.05). After 3-month follow-up, the total effective rate was 88.1% (37/42) in the observation group and 75.6% (34/45) in the control group, which had no statistical difference (P>0.05).
CONCLUSIONThe "Siguan" combined with the Gallbladder Meridian acupoints could significantly relieve the migraine intensity, frequency and lasting time, and its improvement of symptoms and short-term efficacy are superior to those of the Gallbladder Meridian acupoints alone.

3.Experimental study of ballon-dilatation in an establishment of restenosis model in carotid artery of rabbit

Objective To study the mechanism of restenosis for the prevention and treatment by PTA (percutaneous transcather angioplasty) through establishing an model.Methods Under median cervical incision, to eplore right rabbit carotid artery and then clip the segments of ICA at the origination and CCA near the cardiac side of bifucation, balloon catheter with guided wire were finally inserted from proximal cardiac side of the ligated ECA to perform TA after withdrawing the clipper. Histological and morphological analysis with the controll of normal left CCA were carried out during different follow up periods. Results Histopathological and morphometric analysis indicating thrombosis was the main changes in early stage and followed by VSMC from media to intima and irregular proliferation leading to obviously intimal thicking and restenosis at 15 days after PTA.Conclusions This experimental study show low mortality, high practicability and good reproducibility of the model as an ideal presentation for study the mechanism and the prevention of restenosis.
4.Development of X-ray image guided radiofrequency catheter ablation system

Objective To investigave the X-ray guided radiofrequency ablation system. Methods The ablation catheter was guided using the X-ray image so the location of electrode on top of the catheter can be fixed on. Alternating current was carried out at the focus to make the ablation. The multi-channel electrophysiology recorder was combined here to record the electrophysiology status before, during and after the surgery to ensure safety of the patient. Results One case was reviewed, which was treated with the X-ray guided radiofrequency ablation system. After the ablation, the A waves and the V wave were separated and the ablation surgery was successful. Conclusion Some simple and fast arrhythmia can be treated effectively by the above system.

In this framework, nanotechnology-based solutions, and in particular silver nanoparticles ( AgNPs) , have recently emerged as promising candidates in the market as new antibacterial agents because of the enhanced broad-range antibacterial/antiviral properties and low cost.Here we analyze the experimental conclusions on the bactericidal effects of AgNPs, and discuss the safety issues.

Poly(ADP-ribose) polymerase-1 (PARP-1) has emerged as a promising anticancer drug target due to its key role in the DNA repair process. It can polymerize ADP-ribose units on its substrate proteins which are involved in the regulation of DNA repair. In this work, a novel series of para-substituted 1-benzyl-quinazoline-2, 4 (1H, 3H)-diones was designed and synthesized, and the inhibitory activities against PARP-1 of compounds 7a-7e, 8a-8f, 9a-9c and 10a-10c were evaluated. Of all the tested compounds, nine compounds displayed inhibitory activities with IC50 values ranging from 4.6 to 39.2 micromol x L(-1). In order to predict the binding modes of the potent molecules, molecular docking was performed using CDOCKER algorithm, and that will facilitate to further develop more potent PARP-1 inhibitors with a quinazolinedione scaffold.

8.Advances in high mobility group box-1 protein mediated multiple organ dysfunction and its potential interventional strategies.

High mobility group box-1 protein (HMGB1) has recently been shown as a crucial late mediator of inflammation and sepsis, and is involved in mediating multi-organ functional lesions, including acute lung, liver, and intestine injuries. As a delayed inflammatory cytokine, HMGB1 provides a wider therapeutic time window for clinical intervention. HMGB1 has been proven to be a promising therapeutic target to prevent the development of multiple organ dysfunction syndrome in experimental models of severe sepsis. The pharmacological strategies include neutralization of antibodies or specific HMGB1 antagonists, suppression of HMGB1 secretion (ethyl pyruvate, agonists for alpha7-nicotinic acetylcholine receptors), and down-regulation of HMGB1 expression (sodium butyrate, signaling inhibitors for Janus kinase/signal transducer and activator of transcription).
