Objective To study the influence of high-fluoride on thyroid function and brain damage. Methods Thirty-six Wistar rats were randondy divided, according to weight and gender into 3 groups(12 rats each), i.e. control group, high fluoride group, and high fluoride plus thyroid tablet treatment group. The rats were fed with normal tap water containing no more than 5 mg/L NaF and the tap water added 100,100 mg/L NaF, respectively. After 7 months of experiment, the rats in high fluoride plus thyroid tablet treatment group were given with 0.04% thyroid tablet( 1.8 ml·kg~(-1)·d~(-1)) by gastric perfusion for three weeks. The contents of TT_3 and TT_4 in serum were detected by radio-immunological assay; the histomorphology in thyroids and brains were observed under microscopy; and the protein level of NMDAR2B subunit of glutamate receptor in the hippocampal CA1 and CA3 was measured by immunohistochemistry. Results As compared to the values of TT_3 and TT_4 in serum of rats in control group[ (0.97 ± 0.15), (84.03 ± 12.45)nmol/L], TT_3 and TT_4 in high fluoride group were obviously lower [(0.24 ± 0.07), (15.16 ± 2.08)nmol/L, all P < 0.01]; while no changes in TT_3 and TT_4 were detected in high fluoride plus thyroid tablet treatment group[ (1.02 ± 0.19), (85.63 ± 9.55)nmol/L, all P < 0.05] as compared to controls, but higher than those in high fluoride group(all P < 0.01 ). The pathological changes including partial hyperplasy, arrangement disorder, atrophy, and decreased colloid of the thyroid follicular epithelial cells in high fluoride group were observed under microscopy. In high fluoride plus thyroid tablet treatment group, the degree of the thyroid cellular hyperplasy was relatively slight as compared to high fluoride group. The swelling and disarrangement of neurons in the hippocampus were observed in high fluoride group, whereas the changes of the neurons were not so obvious in high fluoride plus thyroid tablet treatment group. The grey values of NMDAR2B positive cells in the hippocampal CA1 and CA3 in high fluoride group(167.05 ± 7.31 ) were significantly increased as compared to controls (92.53 ± 9.67 ) or high fluoride plus thyroid tablet treatment group( 101.66 ± 12.21, all P < 0.01 ). Conclusions High fluoride can induce the decreased function and changed histomorphology in thyroid and result in pathological damages in the brains of rats. However, treated with thyroid tablet to those having damages induced by high fluoride, the thyroid function and morphology can be normal, and the brain damages can be alleviated. The results indicate that hypothyroidism caused by high fluoride might be an important participating factor in brain damages caused by fluorosis.

BACKGROUND:Skin grafting is crucial for patients with deep burns, but limited source of autologous skin grafts is an existing difficulty.
OBJECTIVE:To investigate the effect of xenogeneic (porcine) acelular dermal matrix in the treatment of deep burn wounds and the feasibility of its application.
METHODS:Forty-one patients with deep burn were divided into two groups according to the intention of the patients, 21 cases in control group and 20 cases in observational group, folowed by autologous split-thickness skin grafting alone or combined with different (porcine) acelular dermal matrix, respectively. After 12 months of folow-up, the graft success rate at postoperative 1, 2, 3, 4 weeks and skin graft contraction rate and wound repair at postoperative 3, 6, 9, 12 months were observed and compared between two groups. Moreover, levels of inflammatory factors were detected and compared between two groups at postoperative 1, 2, 3 months.
RESULTS AND CONCLUSION: The skin graft success rates showed no difference between two groups at postoperative 1, 2, 3, 4 weeks (P > 0.05). The skin graft contraction rates also showed no difference between two groups at postoperative 3, 6, 9, 12 months (P > 0.05). After 12 months of folow-up, no serious scar hyperplasia, but soft texture appeared in the control group. In the observational group, three cases presented with local pigmentation at the early stage, but it gradualy subsided with time; no obvious scar, but only smal, point-like scar, was visible, and the repaired wound exhibited soft touch. No adverse events and death occurred in both two groups. Experimental results show that the treatment of deep burns with autologous split-thickness skin grafting combined with xenogeneic (porcine) acelular dermal matrix is safe and effective, which can improve the quality of wound healing.

Objective To evaluate whether three chronic kidney disease epidemiology collaboration (CKD-EPI) equations (CKD-EPI2009Scr,CKD-EPI2012SCysC and CKD-EPI2012Scr-SCysC) are applicable in the prediction of glomerular filtration rate (GFR) in Chinese patients with diabetic nephropathy (DN).Methods One hundred and eight patients with DN who were hospitalized in the First Affiliated Hospital of Guangzhou Medical University with GFR being measured by dynamic renal imaging with 99mTc-DTPA from June 2012 to April 2014 were enrolled in this study.GFR measured by dynamic renal imaging with 99mTe-DTPA was used as the reference value (rGFR).GFR was estimated by the CKD-EPI2009Scr equation, the CKD-EPI2012SCySC equation, and the CKD-EPI2012Scr-SCysC equation (labeled as eGFR1,eGFR2,eGFR3).The correlation,30% accuracy, staging consistency, deviation and diagnostic accuracy were compared among the three CKD-EPI equations. Results The rGFR in 108 DN patients was (61.78±26.51) ml· min-1· (1.73 m2)-1.The correlation between three eGFRs and rGFR was significant (all P < 0.01),the correlation coefficients were 0.738,0.708,0.782.The 30% accuracy were 74.07%,52.78%,67.59%,The 30% accuracy of eGFR1 and eGFR3 were higher than eGFR2 (all P < 0.05),but there was no significant difference between eGFR3 and eGFR1 (x2=0.874,P=0.436).The staging consistency was not ideal,Kappa values were 0.391,0.180 and 0.422.For the deviations between three eGFRs and rGFR,there was no significant difference between eGFR3 and rGFR (P > 0.05),eGFR1 underestimated rGFR,eGFR2 overestimated rGFR (all P < 0.01).The results of the Bland-Altman chart showed that consistencies between three eGFRs and rGFR were poor, the degree of deviation of eGFR3 was the smallest. The area under the ROC curve (AUC) of three eGFRs was 0.878,0.883 and 0.915.The AUC,sensitivity, specificity, overall compliance rate and Youden index of eGFR3 were the highest. Conclusions The eGFRs predicted by the three CKD-EPI equations showed good relevance, accuracy and diagnostic accuracy with the rGFR,but poor in consistencies.Comparatively,CKD-EPI2012Scr-SCysC may be better than others, but its consistency limits exceeds the acceptable limits. Therefore, the applicability of using the three CKD-EPI equations to predict the GFR in Chinese DN patients requires a larger sample and multiple verifications as well as further improvement.

AIM: To investigate the efficacy of N-acetylcysteine (NAC) and taurine (Tau) in preventing hyperoxia-induced the lens opacification and the changes of biochemical parameters on organ cultured rabbit lenses.METHODS: Twenty-four lenses from adult rabbits were divided into the control group, the hyperoxia-exposed group, the hyperoxia-exposed group containing 20mmol/L of NAC, the hyperoxia-exposed group containing 80mmol/L of Tau, respectively. The treated groups incubated with hyperoxia ( pO2>80%) for 4 hours per day throughout a 7-day period. Lens transparency, histology and enzymatic activities measurements were determined after this incubation.RESULTS: Gross morphological examination of these lenses revealed some severe cortical opacification in the hyperoxia-exposed group, moderate cortical opacification in the control group and the Tau treated group. There was minimal cortical opacification in the NAC treated group. The glutathione (GSH) content and the activity of Na, K-ATPase were significantly decreased in the hyperoxia-exposed group than that of the control group, by 37.8% (P<0.05) and 53.5% (P<0.05), respectively. However, they were increased in the two treated groups, especially in the NAC treated group. There were no significant differences in the water-soluble protein content and the catalase and GSH reductase activities in all group lenses.CONCLUSION: Hyperoxia can induce the cortical opaci-fication in the lens. The role of NAC in the prevention of hyperoxia-induced cataract is superior to Tau.
