6.Establishment of a simple and convenient animal model of cardiac arrest for studying cerebral resuscitaion

Objective To establish an simple and convenient animal model of cardiac arrest for studying cerebral resuscitation.Method Clean male Sprague Dawley rats were randomly divided into sham group and experimental group.Cardiac arrest was induced by asphyxiation and ice-cold 0.5 mol KCl with blood flow cut off for 5 minutes.Animals were resuscitated with external cardiopulmonary resuscitation (CPR),mechanical ventilation,and epinephrine injection.Blood pressure,heart rate,successful ratio of resuscitation after 72 hours, time of cardiac arrest (T_(CA)) and return of spontaneous circulation (T_(ROSC)) were recorded.Neural deficit scores (NDS) and levels of maleic dialdehyde (MDA) in plasma were evaluated at 3,6,12,24,48,72 hours after ROSC.The damage score of cortex was measured by transmission electron microscope examination at 3 hours and 72 hours after ROSC.Results All the rats in experimental group had cardiac arrest rapidly.T_(CA) and T_(ROSC) were (137.3?10.2) seconds and (64.4?9.3) seconds,respectively,while the successful rate of resuscitation was 82.5%.The lowest NDS was at 3 hours after ROSC,while the NDS increased gradually.After CPR,the level of MDA in plasma increased significantly,slightly declined at 72 hours after ROSC,but still significantly higher than before the model.Electron microscope examination of cortex showed neuron slightly,organelle and astrocyte,but became better after 72 hours post ROSC.Conclusions The model of cardiac arrest was easy to establish,and the data provided was accurate,which is useful to study the mechanism of cerebral resuscitation.
7.Effect of Sciatic Nerve Compression with Different Pressures and Sites on Neural Blood Flow in Rats

Objective To observe the changes of neural blood flow when oppressed with different pressures and at different sites in rat's sciatic nerve.MethodsThe sciatic nerve compression model of rat with pressure and time controlled simultaneously was established.The changes of neural blood flow were observed by oppressing the sciatic nerve with five grades pressures using Gasbag.The rats were randomly divided into the distal compression group and proximal compression group,and the changes of neural blood flow were observed in each group.ResultsThe neural blood flow changed significantly(P<0.05) when the sciatic nerve was oppressed with different pressures and it had the decreasing tendency with the pressures increasing.The neural blood flow of each group decreased obviously(P<0.01).The effect of oppressing distal nerve on neural blood flow was more obvious than that of oppressing proximal nerve(P<0.01).ConclusionMechanical compression can influence the neural blood flow obviously.The distal vessels of sciatic nerve are the major sources of nerve blood supply.
8.Clinicopathological and related gene analysis in gastric adenocarcinoma and their correlation with prognosis

Scm in volume(42/60),multiple site involvement(44/60),blood type"O"(31/41),in comparison with those of survival group,and the difference was statistically significant.C-erbB-2,p16,p53,P-gp,CD_(44) and CD_(25)expression were not significantly different in these two groups. Conclusion The clinical stage, lymph node metastasis,lymphatic tumor emboli and/or neural involvement,infiltration depth,histological dif- ferentiation,tumor volume,involvement extension are important prognostic factors in patients with gastric can- cer,while the significance of cancer-related gene expression in gastric carcinomas needs to be studied further.
9.Analysis of monitoring results of occupational hazard factors in workplaces in Zhejiang Province, China in 2006 - 2010.

OBJECTIVETo analyze the monitoring results of the occupational hazard factors in workplaces in Zhejiang Province, China in 2006 - 2010.
METHODSDescriptive analysis was performed on the monitoring results of the occupational hazard factors in workplaces in Zhejiang Province from 2006 to 2010.
RESULTSFrom 2006 to 2012, the number of monitored objects for each occupational hazard factor increased gradually, and the qualified rate for each factor remained unchanged or rose slightly. The qualified rates for silica dust and asbestos dust were less than 60%. The qualified rates for benzene, toluene, and xylene, which were always the factors highlighted in the monitoring of toxic chemicals, were all above 85%. The numbers of monitored objects for hexane, hydrogen sulfide, and cyanide grew significantly in recent years. However, the qualified rates for physical factors, which mainly included high temperature and noise, were less than 70%, lower than those for chemical factors.
CONCLUSIONThe qualified rate is as important as the number of monitored objects in the monitoring of the occupational hazard factors in workplaces in Zhejiang Province, China. The acute and chronic factors, physical and chemical factors, and traditional and new factors should be balanced in monitoring.

10.Apoptosis of Lewis Lung Carcinoma Cells Induced by Microwave via p53 and Proapoptotic Proteins In vivo

Background:Microwave therapy is a minimal invasive procedure and has been employed in clinical practice for the treatment of various types of cancers.However,its therapeutic application in non-small-cell lung cancer and the underlying mechanism remains to be investigated.This study aimed to investigate its effect on Lewis lung carcinoma (LLC) tumor in vivo.Methods:Fifty LLC tumor-bearing C57BL/6 mice were adopted to assess the effect of microwave radiation on the growth and apoptosis of LLC tumor in vivo.These mice were randomly assigned to 10 groups with 5 mice in each group.Five groups were treated by single pulse microwave at different doses for different time,and the other five groups were radiated by multiple-pulse treatment of a single dose.Apoptosis of cancer cells was determined by terminal deoxynucleotidyl transferase dUTP nick-end labeling assay.Western blotting was applied to detect the expression of proteins.Results:Single pulse of microwave radiation for 5 min had little effect on the mice.Only 15-min microwave radiation at 30 mW/cm2 significantly increased the mice body temperature (2.20 ± 0.82)℃ as compared with the other groups (0.78 ± 0.29 ℃,1.24 ± 0.52 ℃,0.78 ± 0.42 ℃,respectively),but it did not affect the apoptosis of LLC tumor cells significantly.Continous microwave radiation exposure,single dose microwave radiation once per day for up to seven days,inhibited cell division and induced apoptosis of LLC tumor cells in a dose-and duration-dependent manner.It upregulated the protein levels of p53,Caspase 3,Bax and downregulated Bcl-2 protein.Conclusions:Multiple exposures of LLC-bearing mice to microwave radiation effectively induced tumor cell apoptosis at least partly by upregulating proapoptotic proteins and downregulating antiapoptotic proteins.Continuous radiation at low microwave intensity for a short time per day is promising in treating non-small-cell lung cancer.
