3.Expression of Toll-like receptors signaling pathway in chronic rhinosinusitis with nasal polyps

Objective:To investigate the expression of Toll-like receptors(TLRs)signaling pathway in chronic rhinosinusitis(CRS)with nasal polyps.Method:Gene microarray analysis was used to detect the expression of TLRs signaling pathway in CRS with nasal polyps.Result:Of 19 differentially expressed(two-fold changes),4genes were upregulated and 15 genes were downregulated.Conclusion:The differentially expressed genes in TLRs signaling pathway may exert its effect in the pathogenesis of CRS.In addition,the roles of TLR9 and its agonists need further study.
4.Mechanism and Chemical Basis of the Effect of Bacteria on Urinary Stone Formation

There are two populations of bacteria to affect the formation of urinary stones in humanity. The first one can promote the formation of urinary stone by increasing urinary pH, decreasing concentration of urinary inhibitors , and damaging the protective urothelial glycosaminoglycan layer. The second inhibit the formation of urinary stones. These bacteria ( mainly the intestinal oxalate degrading bacteria such as Oxalobacter formigenes, lactic acid bacteria, Enterococcus faecalis etc) can decrease urinary oxalate concentration by regulating exogenous oxalate. The problems faced and the developing direction were also indicated.
5.Correlation of age with detection rate, grade and stage of prostate caner in patients with PSA 4-10 μg/L

Objective To analyze the impact of age on the detection rate,grade and stage of prostate cancer in patients with prostate specific antigen (PSA) 4-10 μg/L.Methods A retrospective analysis was made in 426 patients with PSA 4-10 μg/L who underwent prostate biopsy and divided into 4 group according to their age less than 60 (n=33),60-69 (n=163),70-79 (n=202) and more than 80 (n=28) to compare the correlation between age and the cancer detective rate.Among them,115 patients,diagnosed as prostate cancer,were further divided into 4 group according to their age less than 60 (n =3),60-69 (n=32),70-79 (n=69) and more than 80 (n=11).The correlation between age and the Gleason score or clinical stage was analyzed in 115 prostate cancer patients.Results The detective rate in each group was 9.1% (3/33),19.6% (32/163),34.2% (69/202) and 39.3% (11/28).There was significant difference in prostate cancer detective rate among those groups.The cancer detective rate became significantly higher as the age increased (P<0.001).In the patients with prostate cancer,the number of patients with Gleason score more than 7 in each group was 1 (50.0%),11 (37.9%),30 (44.8%),7 (63.6%).No significant correlation was found between age and grade (P=0.292 4).And each group had cases with localized prostate cancer was 2 (100.0%),14 (60.9%),39 (78.0%),4 (66.7%),with locally advanced prostate cancer 0,5 (21.7%),7 (14.0%),1 (16.7%),with metastatic prostate cancer 0,4 (17.4%),4(8.0%),1 (16.7%).No significant difference was found between each group and clinical stage (P =0.101 6,P=0.285 2,P=0.349 1).Conclusions The prostate cancer detective rate significantly increases with the age increasing.There is no significant correlation between age and grade or clinical stage in the patients with PSA 4-10 μg/L.
6.Cloning of D-hydantoinase Gene from Pseudomonas and Its Expression in E.coli

AIM The purpose is to construct D-hydantoinase genetic engineering strain for the purpose of the industrial production of D-p-hydroxyphenylglycine. METHODS D-hydantoinase gene was created from Pseudomonas putida 9801 by PCR technique and inserted into pMD18-T vector. The recombinant plasmid was transformed into several Escherichia coli strains. The positive transformants with D-hydantoinase activity were obtained by the two step screening, digoxigenin DNA labeling in situ hybridization and D-hydantoinase activity assay. RESULTS The D-hydantoinase activity of the genetic engineering strain E.coli BL21/pMD-dht was 1700 U*L-1 and increased as high as 8 times compared with those of wild-type strain Pseudomonas putida 9801. The subunit molecular weight of recombinant D-hydantoinase was about 53 kDa measured by SDS-PAGE. The amount of the recombinant D-hydantoinase was about 20 percent of total bacterial soluble proteins. CONCLUSION The genetic engineering strain E.coli BL21/pMD-dht possesses the initial industrial production prospects.
7.Urinary diary in evaluation of nocturia in benign prostatic hyperplasia patients over 60 years old

score, prostate volume and peak urinary flow rate, correlate with BPH, but not with nocturia. Keeping a detailed urinary diary for the patients is beneficial for elucidating their urinary function and in decision-making of their treatment regimen.

CardiovascuIar disease is the number one cause for morbidity and mortaIity worIdwide. Possi-biIities for new therapies in the emerging fieId of cardiac signaIIing prompted extensive research on myocardiaI re-modeIIing over the past decades. In this review,we as-sembIe an overview of the recent findings on the muIti-functionaI enzyme,p21-activated kinase 1( Pak1),a member of a serine/ threonine protein kinase famiIy in the heart,particuIarIy its cardiac protective effects. We pres-ent a modeI for Pak1 signaIing that provides a mecha-nism for specificaIIy affecting cardiac ceIIuIar processes. We discuss its cardiac protective effects such as anti-hy-pertrophy,anti-ischaemic injury and roIe in maintaining ventricuIar Ca2+ homeostasis and eIectrophysioIogicaI stabiIity under physioIogicaI, β-adrenergic and hyper-trophic stress conditions.We aIso discuss the potentiaIs of Pak1 activation by naturaIIy occurring sphingosine and its anaIogues FTY720,and bioactive peptides designed to diminish Pak1 auto-inhibition as noveI thera-peutics for major cardiovascuIar diseases.
9.Imatinib combined with hematopoietic stem cell transplantation or chemotherapy for bcr-abl positive acute lymphoblastic leukemia

Objective To investigate the efficacy of imatinib combining with allogeneic hematopioetic stem cell transplantation or chemotherapy for bcr-abl positive acute lymphoblastic leukemia (ALL). Methods 12 cases were diagnosed on morphology, cytochemistry, immunophenotype and bcr-abl fusion gene. The induction is imatinib (400 mg/d) combining chemotherapy. 8 cases accepted allogeneic hematopoietic stem cell transplantation after complete remission (CR). If bcr-abl became positive, the patient was treated with imatinib (400~600 mg/d). 3 cases were tested with imatinib alternating chemotherapy after cr. Results 11 patients gained CR, CR rate 91.7 %; 5 patients (41.7 %) became bcr-abl negative through 2 courses induction. 3 cases relapsed after transplantation. 2 cases relapsed in imatinib combining chemotherapy group. The median remission interval is 16 months (imatinib combining transplantation group) and 10 months (imatinib combining chemotherapy group) (P <0.01) respectively. The median survival time is 18 months (imatinib combining transplantation group), and the other group (imatinib combining chemotherapy) is 12 months (P <0.01). Conclusion Imatinib combining chemotherapy achieved high CR rate for the bcr-abl positive ALL. Imatinib combining allogeneic hematopoietic stem cell transplantation is superior to imatinib combining chemotherapy for CR patients.
