1.Design of intelligent monitoring system for medical equipment

Objective: Intelligent monitoring system of medical equipment can realize the intelligent dynamic management to equipment in the hospital wireless or wired network environment. Methods: Use of communication network, intelligent call, system monitoring and other means of science and technology to design the intelligent micro device state sensor. Results:System is mainly for hospital equipment, which can capture and management of medical equipment data. When the elderly have abnormal condition, it can send information through intelligent system, through identification and location, to realize the construction of equipment state information. Conclusion: Intelligent sensor in the system application can achieve the sharing of resources; realize the whole hand or full coverage of the intelligent health network system, providing an effective guarantee for the health of the masses.
2.Ring Artifact Location Algorithm Based on Edge Detection

Objective To investigate the Ring Artifact Location Algorithm based on edge detection to eliminate the ring artifact.Methods The Sobel operator was used to extract the ring edge,and then an algorithm,which combined scan line searching with line fitting,was applied to determining the ring center and its size.Finally the linear interpolation algorithm was used to get rid of the ring artifact.Results This method was very effective to eliminate the ring artifact of both phantom and clinical images.Conclusion This algorithm is faster and more effective than the usual one and its speed and accuracy are improved.
3.Rapid detection of extrinsic harmful contaminants using quantum dots-coated probes and their application prospects in traditional Chinese medicine.

The contamination of extrinsic harmful contaminants including mycotoxins, heavy metals and pesticides, etc, brings serious risks to traditional Chinese medicines (TCMs), further to human health. Due to their unique photoluminescence, chemiluminescence, electrochemical and electrochemiluminescence properties, semiconductor quantum dots (QDs) nanoparticles are widely used to immobilize bioprobes and biosensors, etc. In this review, the luminescence characteristics and specific ligands of QDs probles which are used to determine contaminants were summed up. Then, the applications of QDs-coated novel probes in the determination of mycotoxins, heavy metals and pesticides were discussed in detail. In addition, the contamination levels and characteristics of extrinsic harmful residues in TCMs were investigated. Further, the maximum levels of those contaminants in TCMs were compared with those set by various countries. Finally, the future development trends and problems of QDs-coated probes in the determination of those extrinsic residues in TCMs were prospected.

4.Construction of recombinant adeno-associated virus vector expressing human tissue kallikrein gene and its effect on human umbilical vein endothelial cells

Objective To construct the recombinant adeno-associated virus vector(rAAV) expressing the human tissue kallikrein(HK)gene and to detect the expression of interested gene in human umbilical vein endothelial cells(HUVEC)which were infected with different titers of rAAV. Methods The HK gene was cloned into the pAAV-MCS and co-transfected AAV-293 cells with other two plasmids(the pAAV-RC,and pHelper)by lipofectamine.The recombinant AAV(rAAV HK)particles was harvested and its viral titer was measured.HUVEC were infected with different titers of rAAV-HK particles.Seventy-two hours later,the expression of the interested gene was detected by RT-PCR and ELISA.Results The AAV expression system of human tissue kallikrein gene was successfully constructed.The viral titer of recombinant AAV reached 6.2?10~7 particles/ml. When compared with the control group,the transcription of HK gene in the group which was infected with rAAV-HK increased significantly[(0.59?0.12)vs(0.26?0.05)(P<0.05)],and the expression of HK in the HUVEC was three times more than that in the control[(120.1?40.9)vs (30.8?12.8)](P<0.001).The transcription of eNOSmRNA in the infected HUVEC was higher than that of the control[(1.19?0.28)vs(0.66?0.11)](P<0.05).The transcription of caspase-3 mRNA was lower than that of the control[(0.30?0.25)vs(0.67?0.27)](P<0.05).And there was no obvious variation happened in the transcription of VEGF,ET-1,TR-B,bradykinin B1 receptor and Bradykinin B2 receptor.Conclusions When co-transfected AAV-293 cells with three plasmids (pAAV-HK,pAAV-RC,pHelper),the HK gene expression is significantly and stably increased. Introducing HK gene into HUVEC can improve endothelial transfection efficiency.
5.Effect of hydrogen-rich saline on neuropathic pain in rats

Objective To evaluate the effect of hydrogen-rich saline on neuropathic pain in rats.Methods One hundred and twenty male Sprague-Dawley rats,weighing 200-250 g,were randomly divided into 4 groups (n =30 each) using a random number table:sham operation group (S group); hydrogen-rich saline group (H group); chronic constrictive injury (CCI) group; CCI + hydrogen-rich saline group (CH group).Neuropathic pain was induced by CCI.The animals were anesthetized with intraperitoneal 10% chloral hydrate 300 mg/kg.The left sciatic nerve was exposed and 4 ligatures were placed on the sciatic nerve at 1 mm intervals.In H and CH groups,hydrogen-rich saline 10 ml/kg was injected intraperitoneally twice a day for 3 consecutive days starting from 1 day after CCI.The paw withdrawal threshold to mechanical stimulation with yon Frey filament (PWT) and paw withdrawal latency to thermal stimulation (PWL) were measured on the day before CCI (T0) and on 3,5 and 7 days after CCI (T1-3).The rats were sacrificed after the last measurement of pain threshold at T4.The left lumbar segments (L4-6) of the spinal cord were removed for determination of the contents of tumor necrosis factor-alpha (TNF-α),interleukin-1β (IL-1β),interleukin-6(IL-6) and high-mobility group box-1 (HMGB1) (by ELISA),and malondialdehyde (MDA) content and superoxide dismutase (SOD) and catalase (CAT) activities (using visible spectrophotometer).Results Compared with S group,PWT was significantly decreased,PWL was shortened,the contents of TNF-α,IL-1β,IL-6,HMGB1 and MDA were increased,and the activities of SOD and CAT were decreased at each time point after CCI in CCI and CH groups.Compared with CCI group,PWT was significantly increased,PWL was prolonged,the contents of TNF-α,IL-1β,IL-6,HMGB1 and MDA were decreased,and the activities of SOD and CAT were increased at all the time points after CCI in group CH.Conclusion Hydrogen-rich saline can alleviate neuropathic pain in rats via reducing the inflammatory response and lipid peroxidation response.
6.The study of clinical manifestations of osteogenesis imperfecta in a Chinese family.

Objective To study the clinical characters,the mode of inheritance of osteogenesis Imperfecta in a Chinese Family and effect of bisphosphonate on Osteogenesis Imperfecta.Methods Clinical data of proband and their family members were collected.The family patterns were mapped.clinical features were summarized and analyzed.Results(1)Clinical features:There are sixty members of four generations in the family.20 cases including proband's mother and cousin were diagnosed as having OI type Ⅰ based on clinical manifestations.15 cases of blue sclera,16 cases of dentinogenesis imperfecta,5 cases of hearing loss and 3 cases of fracture.Thyroid cancer and Turner's syndrome was found in Proband's mother and cousin respectively.(2)The genetic map showed that the disease was autosomal dominant inheritance.(3)Treatment:The proband,her mother and her cousin were treated with alendronate for two years.Bone pain relieved and bone mineral density increased significantly,and no fracture occurred so far.Conclusion(1)This OI family was diagnosed as having OI type Ⅰ.The mode of inheritance is autosomal dominant inheritance.(2)Bisphosphonates may be an effective drug for treatment of OI.
7.Studies on the Isolation and Antibiotic Activities of Endophytes in Paris polyphylla var. Chinensis franch

One endophytic stain SS02 was isolated from the underground stems of Paris polyphylla var. Chinensis franch. The ferments of SS02 showed antibiosis activities against 13 kinds of the crop causes germs. The characteristics of morphology,physiological and biochemical showed that SS02 belonged to Bacillus sp. The 16S rDNA of SS02 was PCR and sequenced. The accession of GenBank is AY842144. The one 16S rDNA phylogenetic tree was constructed by comparing with the published 16S rDNA sequences of the relative bacteria species. In the phylogenetic tree SS02 and Paenibacillus daejeonensis was the closest relative with 97.7% sequence similarity. According to the phylogenetic analysis it was identified as Paenibacillus daejeonensis SS02.
8.Percutaneous Discectomy Combined with Radiofrequency Ablation with Disc-FX for Contained Lumbar Disc Herniation: 36 Cases Report and 1-year Follow-up

Objective To investigate short-term effect of percutaneous discectomy combined with radiofrequency ablation with Disc-FXon contained lumbar disc herniation. Methods 36 patients were reviewed and followed up with Japanese Orthopaedic Association score(JOA score), the Visual Analogue Score (VAS) and Oswestry score for 12 months. Results The scores of JOA score, VAS and Oswestry improvedsignificantly (P<0.01) after operation. Conclusion Percutaneous discectomy combined with radiofrequency ablation with Disc-FXis effective on contained lumbar disc herniation.
9.Identification and expression analysis of a full-length cDNA encoding a Kandelia candel tonoplast intrinsic protein.

Soil salinity is an important issue, as most crop plants are low in salt tolerance. Salt tolerance, a complex, multifactorial, and multigenic process, has been known to be a quantitative trait. The identification of the salt stress responsive genes or salt tolerance genes is essential for the breeding programs. Most recent efforts have been focused on the products of structural genes (transport proteins, ion channels, enzymes of solute synthesis) while little attention were paid to the regulatory aspects of these proteins. Since the first aquaporin gene from plants was cloned and functionally expressed in 1993, there has been a growing interest in the molecular biology of MIPs (membrane intrinsic proteins) and their bearing on the biophysics of water flow across plant membranes. In the last decades, studies on Mangroves, a special kind of wood plants, grow in high-salt and flooding conditions have been concentrated almost exclusively on their physiological and ecological characteristics. Kandelia candel, one of the dominant species of mangroves along the Chinese coast, lacks salt glands or salt hairs used for removal of excess salt in other mangroves. This makes K. candel a perfect model to study the molecular mechanism of salt tolerance in mangrove plants. Using cDNA RDA, a cDNA-specific modification of genomic representational difference analysis, a series of salt responsive genes of Kandelia candel were cloned. Among these gene fragments, a 183 bp fragment (termed as SRGKC1) encoding a tonoplast intrinsic protein (TIP) in Kandelia candel (KCTIP1) was identified. Based on the sequence of SRGKC1, two gene specific primers were designed, and the 3' and 5' end of the KCTIP1 gene were obtained using the SMART RACE cDNA Amplification Kit. RACE products were purified from low-melting agarose, and sequenced directly with GSPs as the sequencing primers. A 500-bp fragment corresponding to the 3'end of this gene was obtained using the GSP1 primer, and a 690 bp fragment corresponding to the 5' end of this gene was obtained using the GSP2 primer. Two primers that flank the putative open reading frame (ORF) were designed to obtain the cDNA containing the complete ORF by RACE PCR reaction. The full-length cDNA of KCTIP1, containing a 756 bp open reading frame (ORF), was approximately 1.1 kb; the start codon was located at the nucleotides of 99-101 and stop codon at the nucleotides of 855-857 followed by a poly (A) tail. The KCTIP1 cDNA sequence in this research was released in GenBank with accession number AF521135. Using ExPASy Proteomics tools provided by EMBL, the isoelectric point and MWt of KCTIP1 are estimated as 5.77 and 26.3 kD respectively. Transmembrane prediction analysis revealed the deduced KCTIP1 protein sequence contains six transmembrane regions at amino acid residues of 20 - 42, 57 - 79, 86 - 108, 113 - 135, 142 - 164 and 217 - 239. Two highly conserved asparagine-proline-alanine (NPA) motifs were located at 85 - 87 and 199 - 201 amino acid residues respectively. KCTIP1 is also predicted to contain the Cys residue (Cys 118) that are shown to confer Hg-sensitivity in Arabidopsis gamma-TIP and delta-TIP. Similarity analysis showed that KCTIP1 shared 77% - 79% amino acid sequence identity with the TIPs from Vitis berlandieri, Brassica oleracea and Arabidopsis thaliana. Expression analyses indicated that KCTIP1 had different expression among species of Mangroves. Expressions of KCTIP1 in Kandelia candel, Rhizophora apoculata and Ceriops tagal were suppressed by salt, and were insensitive to salt stress in unknown species of Mangroves. Previous studied showed that salt conditions might result in large and rapid changes in extracellular water potential and serious disturbance to the cytoplasm. In order to compensate for this imbalance, the relative contribution of water channels to flow across the root could thus vary. K. candel is a species that is native to intertial zone of tropical and subtropical coast and is well-adapted to salt conditions. The coordinated down-regulation of aquaporins in this plant may decrease membrane water permeability and thus increase the cellular water conserva- tion during periods of salt stress. The results reported here are consistent with the postulated roles for tonoplast water channels in regulating the hydraulic permeability of the vacuolar membranes and in adjusting the water homeostasis of the protoplasm under various physiological conditions. The identification of KCTIP1 as one of salt-responsive genes implies that intracellular osmotic equilibration is a part of salt-tolerant mechanisms in Mangroves.

10.Application of Ivor-Lewis esophagectomy in elderly patients with carcinoma in middle and lower esophagus.

OBJECTIVETo evaluate the safety of Ivor-Lewis procedure for middle and lower esophageal carcinoma in the elderly.
METHODSFrom June 2009 to June 2010, 232 cases aged over 60 years were diagnosed as esophageal carcinoma. These cases were randomly divided into two groups using table of random digits. One group underwent abdominal and right chest approaches for middle and lower esophageal carcinoma (Ivor-Lewis procedure, n=116). The other group underwent posterolateral left thoracal incisions(Sweet procedure, n=116). Intraoperative and postoperative parameters were compared.
RESULTSThe radical resection rates in Ivor-Lewis and Sweet procedure were 95.7% and 92.2% respectively(P>0.05). The time required for opening the thorax was(47.2 ± 5.2) min and (105.4 ± 9.3) min(P=0.000), respectively. The respiratory failure rates were 1.7% and 6.9%(P=0.049). The incidences of supraventricular tachyarrhythmia were 3.4% and 10.3%, respectively. The overall complication rates were 22.4% and 34.5%(P=0.004). The perioperative mortalities were 1.7% and 3.4%(P>0.05). The postoperative ambulation time was (4.0 ± 2.0)d and (4.8 ± 3.7)d(P=0.046). The postoperative time in hospital was (11.5 ± 4.7)d and (13.7 ± 7.8)d(P=0.008).
CONCLUSIONSIvor-Lewis procedure is associated with little damage to diaphragm, shorter intrathoracic operative time, minimal influence on cardiopulmonary function, less postoperative complications, and quicker recovery. This procedure should be considered as the first choice for middle and lower esophageal carcinoma in the elderly.
