2.The effect of pigment epithelium-derived factor on apoptosis of glioma cells

Objective To assess the impact of pigment epithelium-derived factor(PEDF)on the proliferation and apoptosis of the glioma cells by detecting expression of apoptosis related proteins.Methods U87 cells were treated with PEDF(1000μg/ml,U87PEDF),or without PEDF(U87com0),cell proliferation assays were performed by MTT assay to test the effect of PEDF on proliferation of glioma cells;Apoptosis assays were performed by flow-cytometric analysis;Western-blot Was used for evaluating the expression of p16 protein.Results The induced inhibitony rates of glioma cells by PEDF were(54.29±0.62)% Compaxed with the control(t=2.63,P<0.05).The apoptosis assay showed that(21.84±0.36)% of PI- negative/annexin V-positive Was present in the U87 PEDF cells.The appoptosis was associated with the incteases of p16 protein(0.82±0.09)compared with tlle control(0.43±0.03,P<0.05).Conciusion PEDF may play a significant role in apoptosis regulation and proliferation of glioma cell accompanied with the increase of the p16 protein.
3.The inhibitory effect of pigment epithelium-derived factor on migration of gliomas

Objective To assess the impact of pigment epithelium-derived factor(PEDF) on the migration of the glioma cells.Methods PEDF(100 ng/ml)was added to U87 cells(U87PFDF),and VEGF of 0.25μg/ml was added to U87PEDF+VEGF while U87 cells as control(U87con).The cell migratory assav was used tbr evaluating its inhibitory migration rate of PEDF.Real time RT-PCR was used for evaluating the expression of Laminin-8 gene.Results The number of migratory cells was higher than those with added PEDF,and the inhibitory rate of migratory was 38% even in the presence of VEGF.Real time RT-PCR revealed that the mRNA expression levels of α4,β1,γl were(1.043±0.090),(0.823±0.012).(0.762±0.05) copy/μl,which were higher than those treated by PEDF(0.633±0.004),(0.442±0.005).(0.424±0.002)copy/μl,respectively (P<0.05).Conclusion PEDF could decrease the migmtory capacity of the glioma cells even in the presence of VEGF because of the regulation of Laminin-8 in part.

5.Functional Activity and Cognitive Impairment of Patients with Alzheimer Disease in its Early Stage

Objective:to study cognitive impairment and social function of patients with Alzheimer disease (AD). Method:22 patients with AD and 12 with depression in elderly were followed for 3 months, both at the very beginning and the end of 3 months, they were tested with CASI (cognitive abilities screening instrument) and FAQ (functional activities questionnaire).Result:scores of short memory, attention, mathematics, abstract and association of CASI were significantly lower in AD group than that in depression group. AD patients had also poorer outcome in FAQ, such as in that of shopping, learn new things, and recall of engagement.Conclusion:Cognitive impairment and social function of patients with AD were poorer than elderly patients with depression.

Objective To express the pigment epithelium-derived factor (PEDF) in E.coli. and to assess its role in angiogenesis of gliomas.Methods The expression plasmid pET41/PEDF was constructed and the protein was purified by metal affinity tag (His Tag ) and anion-exchange chromatography. Tube formation was used for evaluating its biological activity. The changes of vascular enthothelial growth factor (VEGF) and thrombospondin-1 (TSP-1) were evaluated by the enzyme-linked immunosorbent assay.Results The high purified and inhibitory-actived PEDF proteins were acquired and PEDF had the function of decreasing the VEGF 1.8 times as well as increasing the TSP-1 5.3 times.Conclusions It is possible that PEDF expression is a potent factor for the inhibition angiogenesis in gliomas.
7.Effect of rhein on high glucose-induced mesangial cell Wnt-β-catenin Signaling Pathway

Objective To discuss the effects of high glucose on human mesangial cells (HMC)Wnt-β-catenin signaling pathway,and the intervention of rhein on it.Methods High concentration of glucose (30 mmol/L) combined with different concentrations of rhein were used to intervene cultured human mesangial cells.The activity of mesangial cell proliferation after all the interventions was examined by MTT measurement.Total Wnt、β-catenin RNA was detected By RT-PCR in normal mesangial cells and cells intervened by high glucose and Rhein.Results ①Inhibition effect to human mesangial cells:compared with NG group for 24 h,48 h and 72 h (OD values were 0.169± 0.051,0.228±0.074,0.285±0.075),human mesangial cells proliferation in HG group(OD values were 0.307± 0.074,0.507 ±0.038,0.711±0.075),HG+R1 group(OD value were 0.241± 0.027,0.334±0.015,0.499±0.063),HG+R2 group (OD value were 0.244±0.081,0.386±0.033,0.531±0.011),and HG+R3 group(OD value were 0.277±0.036,0.407± 0.057,0.594±0.042) were iucreased significantly (P<0.05、P<0.01) ; Compared with HG group for 24 h,48 h and 72 h,the HG+R1 group,the HG+R2 group and the HG+ R3 group showed a downward trend at 24 h,but not significantly; but the trend decreased significantly at 48 h and 72 h,and the performance expressed a time,concentration-dependent (P<0.05,P<0.01).② In the normal state,the mesangial cells expressed s certain amount of Wnt and β-catenin,when they were stimulated by high glucose,the expression of Wnt、β-catenin in mRNA increased (P<0.05) ; while the expression of them was significantly reduced in high glucose-induced mesangial cells in HG+R1 group,HG+R2 group and HG+R3 group(P<0.05).Conclusion Rhein may inhibit the proliferation of high glucose-induced mesangial cells through the Wnt and β-catenin gene expression.
8.Imaging features of split cord malformation associated with scoliosis and its correlation with neurologic symptoms

Objective To analyze the imaging features of congenital spinal deformity (CSD) associated with split cord malformation (SCM) and other intraspinal abnormalities, and to investigate the relationship to neurological symptoms. Methods 105 cases CSD with SCM were retrospectively studied. Analysis the imaging features of SCM (including type of SCM, location of SCM, location and apical vertebrae, symmetry of divided cord) and other intraspinal abnormalities. To investigate the relationship of the factors and neurological symptoms using Chi?square test of one factor and multiple factors logistic regression analysis. Re?sults 28 cases (26.7%) were formation failure, 33 cases (31.4%) were segmentation failure, and 44 cases (41.9%) were combina?tion of 2 disorders. 41 cases had neurological symptoms, 64 cases were asymptomatic. The distribution of SCM combined with spi?nal deformities:thoracic (11 cases), thoracolumbar (18 cases) and lumbar (20 cases) in type I SCM, thoracic (31 cases), thoracolum?bar (20 cases) and lumbar (5 cases) in type II, none was in cervical. The location of SCM upper than apical vertebrae 29 cases, on apical vertebrae 25 cases, lower than apical vertebrae 51 cases. Spinal cord was splitted symmetric 27 cases and asymmetric 78 cases. 66 cases combined with other intraspinal abnormalities, lower conus 42 cases, syringomyelia 38 cases, meningocele 10 cas?es and sakrale zyste 5 cases. Associated with intraspinal abnormalities, the rate of neural symptoms was different. According to Chi?square test of one factor and multiple factors logistic regression analysis, lumbar SCM, spinal cord asymmetric and lower conus were related with neurological symptoms. Conclusion The predilection spinal deformity of type I is combination, type II SCM is segmentation failure. When SCM patients associated with other intraspinal abnormalities, the incidence of neurologic symptoms is increased. The lumbar SCM, hemicords asymmetry and lower lying conus have significant relationship with neurologic symptoms.
