1.Protective effects of propofol preconditioning on myocardium against hypothermia ischemia normothermia reperfusion injury on isolated rat hearts

Objective To investigate the protective effects of propofol preconditioning on myocardium against hypothermia ischemia normothermia reperfusion injury on isolated rat hearts. Methods The Langendorff apparatus was used.Sixty SD rat hearts were divided randomly into 5 groups after 20-minute equilibrium(n=12): control(Con) group,hearts were continually perfused with K-H buffer for 175 min;ischemia/reperfusion(I/R) group,hearts were perfused with K-H buffer for 40 min,then subjected to global ischemia at 27 ℃ for 75 min,and followed reperfusion at 37 ℃ for 60 min;propofol preconditioning group 1(P1),group 2(P2),and group 3(P3),hearts were perfused with K-H buffer including 50,100,and 150 ?mol/L propofol for 10 min and followed reperfusion like I/R group,respectively.Heart rate(HR),left ventricular end-diastolic pressure(LVEDP), left ventricular developed pressure(LVDP) and ?dp/dtmax at the end of equilibration,pre-ischemia and at the end of reperfusion were recorded.The contents of creatine kinase(CK) and lactate dehydrogenase(LDH) in coronary effluent were measured at the end of equilibration and 1,10,20,30,and 60 min during reperfusion.The activity of superoxide dismutase(SOD) and the contents of maleic dialdehyde(MDA) were measured at the end of reperfusion.The area of infarct region was determined at the end of reperfusion. 3.Mechanisms of inhibiting the proliferation of triple negative breast cancer cell lines by silencing the CD147 gene

Objective · To explore the inhibitive effect of silencing the CD147 gene on the proliferation of triple negative breast cancer cells and relevant mechanisms. Methods · Normal human mammary epithelial cell line HMEC and three triple negative breast cancer cell lines MDA-MB-231, HCC70 and T4-2 were cultured in vitro. mRNA and protein expressions in cells were measured using realtime-PCR and Western blotting, respectively. A siRNA sequence targeting the coding region of human CD147 gene was designed and used to construct a recombinant lentivirus Lv-shRNA-CD147, which was used to infect the three breast cancer cell lines. The negative control group (Lv-NC infected group) and the noninfected cell control group (cell control group) were simultaneously used. The effects of silencing the CD147 gene were measured with real-time PCR and Western blotting. The proliferation and migration of cells were measured with MTT and Transwell assay, respectively. HCC70 cells were collected 72 h after viral infection and proteins related to proliferation, migration and apoptosis of cells (β-catenin, MMP2, MMP9, and Bax) were measured with Western blotting. Results · mRNA and protein expressions of CD147 were significantly higher in three breast cancer cell lines than in HMEC (P<0.01). The Lv-shRNA-CD147 infected group had lower mRNA and protein expressions of CD147, cell proliferation, and cell migration as compared with the Lv-NC infected group and the cell control group,the differences were statistically significant (P<0.01). The Lv-shRNA-CD147 infected group had lower expressions of β-catenin, MMP2, and MMP9, and higher Bax expression in HCC70 cells 72h after viral infection as compared with the Lv-NC infected group and the cell control group, the differences were statistically significant (P<0.01). Conclusion · Silencing the CD147 gene can inhibit the proliferation and migration of triple negative breast cancer cells.

5.Bioactivity and biocompatibility of hydroxyapatite/DL-poly lactic acid composite: In vivo implantation

BACKGROUND:Hydrolysis in vivo is the key mechanism of degradation in DL-polylactic acid (PDLLA). When it is combined with hydroxyapatite (HA), could the biodegradation and weight loss rate be improved? OBJECTIVE: To observe the changes in the interface and structure of HA/PDLLA composite after in vivo implantation into rabbit femoral defects.DESIGN: Randomized grouping and controlled observation.SETTING: Biomedical Materials and Engineering Research Center, Wuhan University of Technology.MATERIALS: Forty healthy adult Japan White Rabbits of 2.0-2.5 kg, either male or female were provided by the Animal Experimental Center of Hubei Province (No. SCXK. 2003-0005).METHODS: The experiment was conducted in Biomedical Materials and Engineering Research Center, Wuhan University of Technology from June 2005 to March 2006. ①The rabbits were randomly divided into two groups: HA/PDLLA group and PDLLA control group with 20 animals in each group. After anesthetized with ketamine and proazamine, the sample rods of HA/PDLLA and PDLLA were respectively implanted into the drilled bone cavities (φ5 mm × 8 mm) among condyles of femur sites of the rabbits, and the rod could be slightly higher than the surface of bone substance. The samples were covered by periosteum and skin, and then the skin and periosteum were repositioned. ②The complete implants and peripheral bone tissues were taken out respectively after 3, 6, 12 and 24 weeks implantation. The changes in the interface and structure of HA/PDLLA composite after in vivo implantation were observed by using scanning electron microscope (SEM, JSM-5610LV, Japan).MAIN OUTCOME MEASURES: Changes in the interface and structure of HA/PDLLA composite after in vivo implantation.RESULTS: Totally 40 rabbits were involved in the result analysis. After the materials were implanted, HA granules shed from the material surface, some fibroblasts grew into the tissue and a little new osteotylus was formed, indicating HA/PDLLA composite had capabilities of bone-formation and bone-connection. After 24 weeks implantation, the material was divided and wrapped by tissues, neogenetic bone tissue grew into the material, and the fracture healed well,indicating HA/PDLLA composite had good biocompatibility. As for biodegradable PDLLA polymer, hydrolysis in vivo is the most main mechanism of degradation; the degradation speed was decreased owing to being compounded with HA.CONCLUSION: HA/PDLLA composite has capabilities of bone-formation and bone-connection; the biocompatibility of the composite is improved accordingly on account of the decrease of the degradation speed. HA/PDLLA composite is suitable for clinical application as absorbable materials for internal fixation.
6.Effect of Tanshinone ⅡA on nerve conduction of oxaliplatin induced peripheral neuropathy in rats

Objective To investigate the effect of TanshinoneⅡA on nerve conduction of oxaliplatin induced peripheral neuropathy in rats. Methods Fifty Wistar rats were randomly divided into normal control group, model group, treatment group, prevention group, prevention and treatment group. Except for those in model group, Wistar rats were injected i.p. with oxaliplatin (20 mg/kg). The electrophysiological instrument were employed to detect the sciatic nerve conduction velocity, latency, amplitude 6 h, 24 h, 72 h and 7 d after modeling. Results In the model group, velocity of sciatic nerve conduction slowed, and latency prolonged 24 h after modeling (P<0.05) which continued to slow and prolong until 7 d after modeling (P<0.05). After the application of Tanshinone ⅡA, nerve conduction velocity became faster and latency shorter significantly (P <0.05), especially in the prevention and treatment group, in which no significant difference was found when compared with those in the normal control group (P > 0.05). Conclusions During the chemotherapy with oxaliplatin , TanshinoneⅡA can increase the conduction velocity of sciatic nerve , shorten the disease duration and play a protective role for peripheral nerve.

9.Distribution laws of 5 compounds in rhizome and root of Polygonum cuspidate.

To understand the distribution and accumulation rules of polydatin, resveratrol, anthraglycoside B, emodin and physicion in different tissue structure of rhizome and root of Polygonum cospidatum, the content of 5 active compounds were analyzed simultaneously by HPLC, based on plant anatomy and histochemistry. The rhizome and root consist of different tissues, with an increased diameter, the proportions of the secondary xylem and phloem have increased. Resveratrol and polydatin mainly distributed in the pith, the secondary phloem and periderm of rhizome, and the secondary phloem and periderm of the root, while emodin and anthraglycoside B concentrated in the secondary structure and pith of rhizome mostly. In different thickness of the measured samples, the total contents of 5 compounds were correspondingly higher in thinner rhizome and root than those in the coarse ones.

10.Meta-analysis of bifocal lenses for retarding myopia progression in school-aged myopic children

Background Whether the wearing of bifocal lenses can delay the development of myopia in school childhood is in controversy.To assess the effect of bifocal lenses using evidence-based medicine method is of important significance.Objective Present study was to compare the effect of bifocal lenses with single vision lenses in retarding myopia progression in school-aged myopic children.Methods This was a evidence-based medicine study.The systematical literature search was performed from MEDLINE(1966 to October 2010),EMBASE(1974 to October 2010),Cochrane Library,Chinese Biomedical Database(1978 to October 2010),and Chinese Clinical Trial Registry combined with hand searching of related bibliographies of journals and books were applied to collect the randomized-controlled clinical trial about bifocal lenses.Screening,evaluation and data extraction of the retrieved literature were performed by two investigators independently.Mata-analysis was used to assess the progression of refraction and axial length among included randomized clinical trials.Results Three high-quality randomized-controlled clinical trials meeting the inclusion criterion were included in this meta-analysis.The results showed that the weighted mean difference in progression of refraction was 0.22D between bifocal lenses and single vision lenses(95% CI:-0.24-0.67),and the difference was statistical insignificance(P=0.35).The weighted mean difference in progression of refraction during the follow-up durations of 6,12,18,24 and >30 months were 0.15(95% CI:-0.09-0.38),0.17(95% CI:-0.05-0.39),0.42(95% CI:-0.14-0.98),0.23(95% CI:-0.21-0.66) and 0.03(95% CI:-0.40-0.46),respectively without statistical significance.The weighted mean difference in elongation of axial length between two interventions was -0.17mm(95% CI:-0.26-0.08) with a statistically significance(P=0.000).Conclusion Based on currently available studies,bifocal lenses could not significantly slow the progression of myopia in myopic school-aged children in comparison with single vision lenses.Because only few high-quality studies are currently available,this conclusion need to be supported by more large-sample-size clinical trials.
